From California – A new report published in Neurology finds that people who have had pesticide exposure and also suffer a head injury are at significant increased risk of Parkinson #39;s disease. Researchers studied over 1000 individuals from an agricultural area in California, nearly 400 of whom had Parkinson #39;s disease. Participant #39;s exposure to the pesticide paraquat–a weed killer–was determined using geographic data. Participants also reported if they ever had a head injury where they lost consciousness for 5 minutes or more. Results showed that Parkinson #39;s patients were 2x more likely to have suffered a head injury that caused 5 minutes loss of consciousness. They were also 36% more likely to have exposure to paraquat than those without the disease. Edgar Vinyard, a 92 year old legally blind military veteran (and grandfather to our team lead Ryan Riedy) experiences problems with depth perception and other issues that make certain daily activities complicated. One of these problems is difficulty eating, resulting in constant spilling of his food. This is where the concept of the Swivel Spoon was developed. Ryan observed that the difficulty his grandfather was facing was causing him anxiety, embarrassment, and overall discomfort. After many instances of observing this issue, Ryan decided that something could be created to assist Edgar and other individuals that experience similar problems. The Swivel Spoon is envisioned to increase the quality of life for individuals that have disabilities such as vision impairment, illnesses such as Parkinson #39;s Disease, and elderly people in general that have impaired motor skills. After initially conducting 13 face to face interviews, we were able to interview 12 additional individuals for a total sample size of 25. The overall feedback from all 25 people interviewed was positive. All 25 interviewees agreed that our concept was a “good idea” and perceived it to be marketable. The majority of our sample size were found to have no use for the Swivel Spoon for personal use; however, 11 people did know someone that could benefit from the product. An Overview of the Molecular Mechanisms by Which Exercise Impacts Aging

Exercise slows many of the degenerations of aging and - much like calorie restriction - this appears to be the result of changes in a multitude of biological processes and systems. In effect exercise adjusts the operation of your metabolism, moving it into a better configuration.

If you'd like a look under the hood, an overview of what is presently known of the biology that links exercise to improved long term health, you might read this recent open access review paper. It focuses on the heart, but the underlying mechanisms are of general interest:

It is generally accepted that regular exercise is an effective way for reducing cardiovascular morbidity and mortality. Physical inactivity and obesity are also increasingly recognized as modifiable behavioral risk factors for a wide range of chronic diseases, including cardiovascular diseases. Furthermore, epidemiologic investigations indicate that the survival rate of heart attack victims is greater in physically active persons compared to sedentary counterparts. Several large cohort studies have attempted to quantify the protective effect of physical activity on cardiovascular and all cause mortality. Nocon et al. in a meta-analysis of 33 studies with 883,372 participants reported significant risk reductions for physically active participants. All-cause mortality was reduced by 33%, and cardiovascular mortality was associated with a 35% risk reduction. Exercise capacity or cardiorespiratory fitness is inversely related to cardiovascular and all-cause mortality, even after adjustments for other confounding factors.

Physical inactivity is increasingly recognized as modifiable behavioral risk factor for cardiovascular diseases. A partial list of proposed mechanisms for exercise-induced cardioprotection include induction of heat shock proteins, increase in cardiac antioxidant capacity, expression of endoplasmic reticulum stress proteins, anatomical and physiological changes in the coronary arteries, changes in nitric oxide production, adaptational changes in cardiac mitochondria, increased autophagy, and improved function of sarcolemmal and/or mitochondrial ATP-sensitive potassium channels. It is currently unclear which of these protective mechanisms are essential for exercise-induced cardioprotection. ... A better understanding of the molecular basis of exercise-induced cardioprotection will help to develop better therapeutic strategies.

It will lower your odds of living in good health for as long as you might like - and that is enormously important in this age of biotechnology. Mitochondrial Antioxidants Fail in Flies

Mitochondrially targeted antioxidants - such as gene engineering of increased amounts of catalase - are shown to extend life in mice, but here researchers find no such effect (or a negative effect) in flies: "The simultaneous overexpression of multiple copies of Mn superoxide dismutase (SOD) and ectopic catalase (mtCat) transgenes in the mitochondria of the fruit fly, Drosophila melanogaster, was shown previously to diminish the life span. The hypothesis tested in the present study was that this effect was due primarily to the presence of one or the other transgene. An alternative hypothesis was that both transgenes have additive, negative effects. Crosses were performed between five pairs of transgenic lines containing single-copy insertions of either mtCat, Mn SOD, or P element vector control transgenes at unique loci, and the life spans of progeny containing two mtCat, Mn SOD or vector insertions were determined. Increasing amounts of mitochondrial catalase activity tended to be associated with decreases in mean life span. Overexpression of two copies of the genomic Mn SOD transgene had no effect on life span. The results do not support the hypothesis that enhanced mitochondrial SOD or catalase activity promotes longevity in flies." This suggests that it's possible to set up a situation in mammals wherein mitochondrially targeted antioxidants are harmful to life span, but I'm not aware of any examples.

Cranberry Helps Cure Urinary Tract Infection, US Study Says

Higher doses of cranberry can help treat UTI or urinary tract infections by helping remove the bacteria from the surface of the urinary tract tissue.

According to a study performed by researchers from Rutgers University,  72 mg of cranberry (Vaccinum macrocarpon) can help ward off and even treat urinary tract infections or UTIs.

The study directly supports an early health claim from France that states that at least 36 grams of fresh cranberries is effective against UTI because it prevented some of the Escherichia coli (E. Coli) bacteria from sticking to the urinary tract (in males and females).

According to lead researcher Amy Howell, there is still much more to explore in terms of determining the most effective dose to fight off urinary tract infections.  The study was performed in different countries, including Japan, Hungary and even in Spain. Howell reports that they are also trying to determine the link between the adhesion of the bacteria responsible for UTI and the origins of the respondents.

What the study does show presently is that as the cranberry dose increases, the more effective it becomes in fighting off the adhesion of the bacteria.  The study noted 50% less adhesion at eighteen milligrams and 100% less adhesion at seventy-two grams of cranberries.

Polyphenols Found in Green Tea Can Help Save Diabetics

Polyphenols or phytochemicals found in green tea can help fight off bad cholesterol and harmful sugar-derived substances through strong antioxidant activity in the body.

In a recent scientific research published in the journal Food Chemistry in Taiwan, it was found that the compound EGCG or epigallocatechin 3 gallate can help reduce low-density lipoproteins (LDL) or bad cholesterol and also prevents glycatin of blood sugar. Glycation is a process whereby sugar molecules are able to bind with other types of molecules such as protein.  Glycation is characteristic of conditions such as type 2 diabetes or adult onset diabetes.

Aging of the Innate Immune System

The innate immune system declines with age, just like the adaptive immune system. The details are different: "The innate immune system is composed of a network of cells including neutrophils, NK and NKT cells, monocytes/macrophages, and dendritic cells that mediate the earliest interactions with pathogens. Age-associated defects are observed in the activation of all of these cell types, linked to compromised signal transduction pathways including the Toll-like Receptors. However, aging is also characterized by a constitutive pro-inflammatory environment (inflamm-aging) with persistent low-grade innate immune activation that may augment tissue damage caused by infections in elderly individuals. Thus, immunosenescence in the innate immune system appears to reflect dysregulation, rather than exclusively impaired function." Understanding the cause of the problem steers the search for solutions. Dysregulation means that the focus is on fixing errant signaling mechanisms, or on finding ways to directly instruct cells to act or not act. Cell transplants or repairs are not much use if the problem actually lies in the control systems.

As a follow-up to their ground-breaking and successful essential tremor pilot clinical trial, they have launched a new study using focused ultrasound to treat patients with tremor-dominant Parkinson #39;s disease. To be eligible for the study, patients must have symptoms that remain disabling despite optimal doses of medication. So far, the study has treated four patients and is expected to enroll 26 more. The trial is following a randomized double-blinded protocol, and patients do not immediately know if they are receiving an actual focused ultrasound treatment or a sham procedure. The randomization process will ultimately assign 20 patients to the study #39;s treatment arm and 10 to the sham group. Three months after-treatment, assignments will be “un-blinded” and sham arm patients will be eligible to crossover into the treatment group.
