A clone of the endangered Przewalski’s horse is born of DNA saved for 40 years – CNN

The colt is a clone of a male Przewalski's horse and the first successful cloning of the species, San Diego Zoo officials said in a news release on September 4. It was born August 6 to a domestic surrogate mother. Przewalski's horse are known as the last wild horse, according to the National Zoo. They were originally native to Europe and Asia, but the expansion of humans and environmental changes depleted their numbers.

Scientists said that the horse was formally extinct in the wild, and has been surviving for the last 40 years primarily in zoos. Some herds have been found in Mongolia.

"This birth expands the opportunity for genetic rescue of endangered wild species," said Ryan Phelan, executive director of Revive & Restore, a wildlife conservation organization promoting biotechnologies that's partnering with San Diego Zoo Global and ViaGen Equine on the cloning project.

"Advanced reproductive technologies, including cloning, can save species by allowing us to restore genetic diversity that would have otherwise been lost to time."

The DNA used for the colt was cryopreserved in 1980 at the San Diego Zoo Global (SDZG) Frozen Zoo. The cloned stallion was born in 1975 in the United Kingdom and was transferred to the US in 1978. The zoo said he lived until 1998.

"As the new clone matures and successfully breeds, he can provide a valuable infusion of genetic diversity for the Przewalski's horse population," the news release said.

The baby horse was named "Kurt," in honor of Kurt Benirschke, who was instrumental in founding the Frozen Zoo and the conservation research program at San Diego Zoo Global.

Kurt will be moved to the San Diego Zoo Safari Park when he is older to be integrated into a breeding species.

"This colt is expected to be one of the most genetically important individuals of his species," said Bob Wiese, chief life sciences officer at San Diego Zoo Global.

"We are hopeful that he will bring back genetic variation important for the future of the Przewalski's horse population."

SAN DIEGO, Calif. - About 70 companies worldwide are working on therapies for COVID-19 using cloned antibodies, according to an estimate by the International AIDS Vaccine Initiative. A few of these treatments, known officially as monoclonal antibody therapies, have advanced to clinical trials.

Most require an injection, but San Diego-based Diomics and its partner Active Motif, based in Carlsbad, are developing a once-a-day spray that could be easily self-administered.

Early research suggests the coronavirus primarily enters the body through the nose. The spray, called Dioguard, is designed to coat the lining of the nasal cavity with cloned antibodies that are held in place for 24 hours or more using a proprietary polymer material developed by Diomics.

Diomics CEO Anthony Zolezzi said he believes the spray holds the key to allowing many aspects of life to resume until the day comes when theres an effective vaccine in widespread use.

Diomics is also developing two tests for COVID-19 antibodies using its polymer beads, including a device that looks like a nicotine patch that is designed to monitor for infections for a week or more.

Active Motif is providing the cloned antibodies for the nasal spray. With a lab in Shanghai and other relationships in China, the Carlsbad company was able to clone antibodies from 11 Chinese survivors in February, before the World Health Organization declared a pandemic.

When the company started the cloning project, they thought the virus would probably disappear in three to six months, said Active Motif CEO Ted DeFrank. Then people started realizing, no this is going to be with us for a while.

The plasma from the 11 Chinese patients contained thousands of antibodies, and scientists with Active Motif set about selecting the one that was most effective, dubbed 414-1. The company says it can neutralize SARS-CoV-2 virus particles with 98 percent effectiveness.

Monoclonal antibodies have some similarities to convalescent plasma, which is a complex cocktail of antibodies and other immune molecules drawn from the blood of recovered patients. One of the key differences is that cloned antibodies can be mass produced in a lab.

Cloned antibodies have been used in treatments for more than 30 years, primarily for cancer. One such treatment famously helped former U.S. President Jimmy Carter beat melanoma.

But of the more than 100 monoclonal antibody therapies licensed for use, only seven are for communicable diseases, according to IAVI.

Historically, the treatments have been expensive and difficult to produce, but Diomics said its targeting a price of about $2 a spray for Dioguard, roughly $45 per bottle.

We do not want to have huge profits from a pandemic, thats just wrong, Zolezzi said. Were going to price this as effectively as we can for the masses. We want to get this out to the masses.

Animal testing is about to begin and the companies said they hope to progress to human trials soon. Their goal is to release the spray in early 2021, when a vaccine may be on the market but not yet widely available.

If the spray works, it could be adapted to other viruses to help fight future pandemics, Zolezzi said.

"Thats our real goal," Zolezzi said. "That we never get caught flat-footed like this time."

The last confirmed sighting of a horse of Przewalski in the wild it was in 1969.

Fortunately, some of the horses still remained in zoos. But not many. A total of 12 horses formed the ancestors of a captive breeding program: 11 horses of Przewalski captured in freedom between 1899 and 1902, and another captured in 1947. Przewalskis horses are critically endangered animals that are found in Mongolia, per Smithsonians National Zoo. Theyre considered the last species of truly wild horses and are distant cousins of modern day domestic horses, having likely split from a common ancestor around 500,000 years ago, per the Smithsonian.

Przewalskis horses were once extinct in the wild, and while intensive breeding programs helped revive the species and reintroduce them into the grasslands of China and Mongolia, nearly all can be traced back to 12 Przewalskis horses that were born in the wild, the San Diego Zoo said in its press release. The successful cloning of DNA collected 40 years is meant to introduce key generic diversity into the species that could benefit its survival. The zoo said the cloned Przewalskis horse will eventually be transferred to the San Diego Zoo Safari Park and integrated into a herd of other Przewalskis horses for breeding.

The work to save endangered species requires collaborative and dedicated partners with aligned goals, Paul A. Baribault, president of San Diego Zoo Global, said in a statement. We share in this remarkable achievement because we applied our multidisciplinary approach, working with the best scientific minds and utilizing precious genetic material collected and stored in our wildlife DNA bio bank.

The cloned Przewalskis horse was named Kurt after Dr. Kurt Benirschke, the creator of the San Diego Zoo Global Frozen Zoo, which has been collecting and preserving genetic material of endangered animals since 1975.

San Diego Zoo Global partnered with Revive & Restorea wildlife conservation group that aims to incorporate biotechnology into conservation effortsand ViaGen Equinea company that clones horses and petsto successfully clone the Przewalskis horse.

This birth expands the opportunity for genetic rescue of endangered wild species, said Ryan Phelan, executive director of Revive & Restore, in a statement. Advanced reproductive technologies, including cloning, can save species by allowing us to restore genetic diversity that would have otherwise been lost to time.

Przewalskis horses are not the only species Revive & Restore is trying to revive via biotechnology. The group is attempting to revive at least six endangered or extinct species, including the Wooly Mammoth, which went extinct around 4,000 years ago. Other organizations and research facilities are also attempting to revive extinct or endangered species with biotechnology, including the International Islamic University Malaysia, which plans to revive the recently extinct Sumatran rhino.

The US Open is known for its often raucous, vocal fans who make themselves heard everywhere.

But when it was announced that this years US Open would not have spectators on site, many tennis watchers wondered how that might impact not only the players, but the overall experience of the event.

Tournament organizers turned to IBM to help them provide a solutionAI (artificial intelligence) generated crowd noise. Using technology called AI Sounds, engineers digested video from last years US Open and then ranked the excitement level of various video clips. These clips were then turned into a reel and then classified to create a crowd reaction score.

From a player hitting an ace, to saving a break point, or winning a match, all these various sounds were then made available to in-stadium and television production crews covering this years event to use the sounds depending on whats happening in a match during real-time.

COVID-19 brought disruption to sports as a whole, and the ability of fans to experience live sporting events has been heavily impacted in 2020. At the same time, the pandemic accelerated the need for engaging technologies using AI and underpinned by a scalable hybrid cloud," said Noah Syken, Vice President of Sports & Entertainment Partnerships, IBM. "As the technology partner to the USTA, we transformed our offerings to meet tennis fans where they are this yearexperiencing the sport through the US Open digital properties everywhere.

These sounds are definitely noticeable in matches being played at the site two biggest courtsArthur Ashe Stadium and Louis Armstrong Stadium. Without them, it sometimes feels like the players are competing inside an empty parking lot, rather than inside one of the sports most prestigious stages.

Using digital sounds in sporting events is nothing new; even before the current pandemic prompted many sports leagues to not allow fans at their respective matches or games.

According to the New York Times, 20 years ago CBS used taped nature sounds during a PGA golf event that drew criticism from bird experts over the use of such sounds. Nowadays, the practice is more common.

European football leagues when they returned to action this summer are employing virtual stadium noises created for FIFA video games. The National Football League (NFL) is considering using artificial crowd sounds during football games this fall.

Getting back to tennis, fans watching at home are divided over the non-natural sounds being used in New York. Some fans like that it adds some atmosphere for them as they cheer on their favorite pros. Others dont like the artificial sounds and would prefer experiencing the court as it is.

For the players themselves, although they would prefer to have fans on-site, not having fans this year is just one more thing they have to adjust to during this very different U.S. Open.

It's digital fans. It's 2020, Serena Williams said when asked about not having fans on site. Im all about the future. I'm glad we're able to have the Open.

"It's really cool that we can have it. It's so important that this is justwe don't know what the new normal is going to be. So it's just good that we can have virtual fans and be able to have sport.

Why chase after an ideal or a lost love? Each love is unique and cannot be cloned, no matter what!

You can never love any two or more people in the same manner and with the same intensity. Love is an emotion of a myriad hues, never experienced in exactly the same way by different people. And never again even by the same two people in an unchanged form. This is because love is dependent on so many variable factors that it cannot possibly be replicated.

The love that two people share stems from their individual needs, mutual chemistry, and also from their own capacity to give and receive the emotion. It is dependent on individual personal characteristics, and each persons experiences and unique circumstances also have a deep bearing on the outcome. This accounts for the rather changeable nature of love.

Not all loves, nor indeed all relationships, can feed the same need in you. Physical need is just one aspect; we also have emotional needs, spiritual and intellectual needs, and some needs that we ourselves may not understand. Hence you get attracted to other people and at any given time, could theoretically have loving relationships with multiple people. Needs vary, and so a relationship based on need, would vary too.

The only people who can love everyone in exactly the same manner, repeating the same experiences, are almost certainly psychopaths, or at least sociopaths, using their bookish understanding of the emotion to exploit others. In short, these are people just acting out the emotion.

We waste a lifetime in the impossible pursuit of an ideal love or the recreation of a love we once had and lost but cloning is not part of the Love DNA! You cannot ever experience the love of a Romeo and Juliet however hard you try, because that love was the product of their circumstances and the interplay of their individual characteristics and chemistry at a certain time in history, placed in a unique geographical setting. How can you possibly love like them, when you are not them? Heck, even if Romeo and Juliet were themselves to step out of Shakespeares pages, with changed variables, they cannot possibly love the same way again! This time round they might end up happily married, growing old together or splitting paths along the way!

You can certainly love several times in your lifetime, sometimes simultaneously too, but never ever in the exact same manner, clocking the same experiences and emotion. That is impossible because we change and evolve as we go along in life, and our needs and dependencies change too. Time and experience teach us better control of our emotions, patience, forgiveness and self-sufficiency. Conversely, bad experiences at love and relationships may lead to bitterness and cynicism in some.

So, if people change so much and so do their needs, how then can our experience of love remain the same? Love changes too. And that is what makes it so precious, so unique to each. Why look for a clone when you can create your own unique love?

‘They Cloned Tyrone’ Adds Jamie Foxx and Teyonah Parris to the Netflix Sci-Fi Mystery – /FILM

John Boyega may have walked away disappointed with his experience working on the new Star Wars trilogy, but that hasnt turned him off sci-fi in general. The actor is slated to star in the Netflix original sci-fi mystery movie They Cloned Tyrone, and hes finally getting some company in front of the camera. Jamie Foxx and Teyonah Parris (Chi-raq, Candyman) have joined the cast of the movie.

The Hollywood Reporter has news on the They Cloned Tyrone cast expanding with Jamie Foxx and Teyonah Parris, but it hasnt been specified who their characters are. John Boyega is leading the film, and it seems that these two new additions to the cast will fill out the trio at the center of the movie who investigates a series of eerie events, alerting them to a nefarious conspiracy lurking directly beneath their hood. We dont know the entire conspiracy, but were betting it has something to do with cloning someone named Tyrone.

They Cloned Tyrone will mark the directorial debut of Creed II writer Juel Taylor, who co-wrote the script for this new movie with Tony Rettenmaier. Theyll both also be producing with Charles King and his Macro production banner, along with Stephen Dr. Love and Kim Roth. Poppy Hanks and Mark R. Wright are also executive producing for Macro.

Back in 2019, They Cloned Tyrone was one of the films named on The Black List, a collection of the best unproduced screedplays in Hollywood. Thankfully, Netflix snatched up the movie that has been described as a pulpy, sci-fi mystery caper that is like a combination of Friday and Get Out.

John Boyega has already shown his strengths in comedy and sci-fi action thanks to Attack the Block and the Star Wars franchise. Foxx is just coming off another Netflix sci-fi movie, Project Power, and we know he can handle comedy very well. And finally Teyonah Parris has proven to be quite the diverse actress too, with supporting roles in Mad Men, They Came Together, If Beale Street Could Talk, and Empire. Plus, shes also playing a grown Monica Rambeau in Marvels upcoming WandaVision series on Disney+. So having all three of them together for a pulpy movie like this should be something worth watching whenever it arrives on Netflix.

Tahlequah has plenty of medical marijuana dispensaries for those with state medical cards, but patients could save money in the long run and enjoy the fruits, or buds, of their labor by growing their own.

A patient with an Oklahoma Medical Marijuana Authority license can legally possess six mature marijuana plants, and six seedling plants. The start up for those new to "gardening" depends on how much cannabis they would like to grow, and which method they would like to use. A grower could plant seeds or clone indoors using the traditional soil method, or could take the hydroponic route by growing it in a reservoir of water - or take the clone or seeds and throw them in the dirt in the backyard.

Jackie Guinn, owner of Jus Grow Me in Tahlequah, said she would start beginners with a few pots, lights to use inside their homes, and soil for under $500. However, if people want to take it further, they could get a setup that costs upward of $1,000.

"The price is going to depend on how many plants you're going to do," said Guinn. "It depends on what lights you want to get. I've got lights that start at $130 and on up. I would probably start them off with a little clone in a 1-gallon pot."

Whether it's a potted plant or using the hydroponic method, Guinn said a grower should give the plant 18 hours of light and six hours of darkness while it is in its vegetative state. She would water it about every other day and give it a variety of nutrients. Once the plant reaches about knee high, it can be transplanted into a 5-gallon bucket.

"You would keep that in its vegetative state until it reaches about 4 feet high," said Guinn. "When it starts to flower, it's going to double in size, and if you're growing indoors, that's kind of hard for most people."

Once a plant reaches about 4 feet, the grower should transition it to its flowering stage. That requires a different set of nutrients, which would help the plant yield more buds and avoid starvation.

"When you go into flowering, you need your blooming nutrients to help produce the buds, the THC, the CBDs, and your trichomes," said Guinn. "Those are always important, because the higher CBD and THC are going to eliminate your pain."

Miracle-Gro should not be used on cannabis. It contains to much nitrogen, which would give the plant issues once it reaches flowering stage. Guinn said another factor is making sure the plant's water has the appropriate PH balance.

"Tap water is the worst for your plants," she said. "It can burn them up. The PH needs to be between 6.5 and 7. If you use distilled water, you've got to make sure you add Cal-Mag, because distilled water does not have it and your plants need that. Rain water is the best for them."

Those who plant their marijuana outside should make sure the plants don't get too much water, or else they will be susceptible to root rot. Guinn recommends raisers a tarp to cover the plants if it rains too much.

Growers need to look out for mites and other bugs. There are a variety of organic pesticides or other natural ways of getting rid of pests, such as using lady bugs. Cultivators should also be on the lookout for mold and mildew.

"If that happens, you need to just throw them all away. It's not healthy to make any kind of medicine out of that at all," said Guinn. "You've got to keep your humidity in your tent or your room just right. That's a very important step, because if the humidity gets too high, it can cause mold or mildew on your plants."

After the plant reaches flowering stage, the grower can start to harvest anywhere from 60 to 90 days. It depends on what he would like to get out of the plant. Guinn said examining the plant's trichomes, which are small resin glands found on the flowers, will help one decide when it's time to harvest.

"Your trichomes will start out [in the flowering stage] as clear," said Guinn. "Then, it goes to cloudy, and then it can go to ember. When it goes to cloudy, you can harvest. It's higher in THC when it's cloudy. You get higher CBD when it turns to the ember color. The trichomes are the little crystals all over your leaves."

Jus Grow Me offers free classes for people looking to start their own small grow operations. Students can learn about cloning, hydroponics, starting a plant in soil and more. While it may seem complicated at first, Guinn said it takes little time to get the hang of it.

"Every day check on your plant," she said. "Once you start growing your plant, it gets pretty easy. You get to know your plant and you can tell if you have a deficiency. All they've got to do is call me and we'll get them through this. We're all about people growing their own natural medicine. It's the way to go."

Purpose: Human butyrylcholinesterase (BChE) serves as a bio scavenger to counteract organophosphate poisoning. It is also a potential drug candidate in several therapeutic fields. Therefore, in the present study, we constructed a new dual-promoter plasmid consisting of Cytomegalovirus (CMV) and human elongation factor 1 (EF-1) promoters and transfected that into HEK-293 cells using Lipofectamine to enhance the BChE secretion.Methods: The new dual-promoter construction (pBudCE dual BChE) including two copies of the BChE gene was designed and transfected into cells by liposomal structures. The cloned plasmids were evaluated by enzyme digestion and gel electrophoresis analysis. Experimental groups were categorized into the cells transfected by pBudCE dual BChE (treatment), pCMV (positive control) vectors, and nontransfected cells (negative control). BChE gene expression was evaluated by qRT-PCR and the enzyme activity was assessed using modified Ellmans method. The freeze-thaw process was carried out for analyzing the stability of the pBudCE dual BChE vector.Results: Validation examination of the cloned plasmids confirmed the successful cloning process. The gene expression level and Ellmans method value in pBudCE dual BChE was higher than the other groups. CMV promoter has also increased the enzyme activity, although the difference was not significant compared with the control group. Interestingly, freeze-thaw cycles followed by several passages did not affect the enzyme activity.Conclusion: The designed construction with CMV and EF-1 promoters could increase BChE gene expression and the activity of the BChE enzyme in HEK-293 cell line. Large-scale production of BChE enzyme can be achieved by using dual-promoter plasmid construction compared to a single-promoter vector to be used in clinical trials.
