Cecilia Marzabadi, Ph.D., professor in the Department of Chemistry and Biochemistry, has been named the 2020 chair of the North Jersey Section of the American Chemical Society (ACS). A member of the North Jersey Section for more than 20 years, she becomes only the second Seton Hall University faculty member to be named its chair since 1930.

Founded in 1876 and chartered by the U.S. Congress, the American Chemical Society is the world's largest scientific society. Its mission is to advance the broader chemistry enterprise and its practitioners for the benefit of Earth and its people, ultimately improving lives across the globe.

The North Jersey Section is one of the largest local Sections of the ACS in the country, with a range of programming and year-round activities to engage its 4,300 members. It features a diverse array of topical/discussion groups that hold meetings in areas such as organic chemistry, NMR spectroscopy, mass spectrometry, drug metabolism and chromatography. There are also discussion groups specifically for minority chemists, female chemists, younger chemists, senior chemists and teacher affiliates.

In a recent issue of the Section's monthly e-newsletter, "The Indicator," Marzabadi addressed the members of the NJACS and discussed her objectives for the upcoming year, which included:

"I look forward to this opportunity to work with colleagues at other universities and at local companies as an advocate for chemistry," says Marzabadi. "It is a both a great honor and a challenge to serve for such a large Section. I hope to have an impact on the recruitment of young scientists and on increasing participation in chemistry of underserved groups."

Marzabadi earned her bachelor's and master's degrees from Saint Louis University and her Ph.D. from the University of Missouri, Saint Louis. Her research interests include synthetic organic and carbohydrate chemistry, having done work specifically on the synthesis of small, carbohydrate-based molecules to be used as potential therapeutics. She and her team have targeted molecules for the treatment of epilepsy and other central nervous system disorders, as well as cancer. In addition, she studies issues related to the recruitment and retention of women in the sciences and mathematics. She is a founding member of the Center for Green Chemistry and Catalysis at Seton Hall.

Background: Platelets play important roles in cancer progression and metastasis, as well as in cancer-associated thrombosis (CAT). Tyrosine kinases are implicated in several intracellular signaling pathways involved in tumor biology, thus tyrosine kinase inhibitors (TKIs) represent an important class of anticancer drugs, based on the concept of targeted therapy.Purpose: The objective of this study is the design and synthesis of analogues of the TKIs imatinib and nilotinib in order to develop tyrosine kinase inhibitors, by investigating their molecular requirements, which would express antiplatelet properties.Methods: Based on a recently described by us improved approach in the preparation of imatinib and/or nilotinib analogues, we designed and synthesized in five-step reaction sequences, 8 analogues of imatinib (IIV), nilotinib (V, VI) and imatinib/nilotinib (VII, VIII). Their inhibitory effects on platelet aggregation and P-selectin membrane expression induced by arachidonic acid (AA), adenosine diphosphate (ADP) and thrombin receptor activating peptide-6 (TRAP-6), in vitro, were studied. Molecular docking studies and calculations were also performed.Results: The novel analogues VVIII were well established with the aid of spectroscopic methods. Imatinib and nilotinib inhibited AA-induced platelet aggregation, exhibiting IC50 values of 13.30 &Mgr; and 3.91 &Mgr;, respectively. Analogues I and II exhibited an improved inhibitory activity compared with imatinib. Among the nilotinib analogues, V exhibited a 9-fold higher activity than nilotinib. All compounds were less efficient in inhibiting platelet aggregation towards ADP and TRAP-6. Similar results were obtained for the membrane expression of P-selectin. Molecular docking studies showed that the improved antiplatelet activity of nilotinib analogue V is primarily attributed to the number and the strength of hydrogen bonds.Conclusion: Our results show that there is considerable potential to develop synthetic analogues of imatinib and nilotinib, as TKIs with antiplatelet properties and therefore being suitable to target cancer progression and metastasis, as well as CAT by inhibiting platelet activation.

It seems as if every year a new study rears its head in the newspapers, declaring that coffee is good, or bad, for us. Americans eat up or, drink up, rather coverage on the topic because so many of us are addicted: 64 percent of American adults drink coffee regularly, according to a 2018 study.

Beyond questions of physical health, coffee clearly could affect our mental health, too. After all, it is a mood-altering stimulant. Could coffee be making us anxious? The question is particularly timely as millennials have normalized open discussions about mental health, particularly anxiety or depression.

Now, more of them have begun debating whether coffee could be a major source of anxiety.

Nicole Cole Hernandez, an entrepreneur and certified hypnotist, told Salon she stopped drinking coffee in August because of extreme anxiety that precipitated stomach pains and hair loss.

Within a month [of quitting coffee], my stomach pain subsided without having to take regular medication," Hernandez said. "When I began feeling better, I did opt for a regular cup of coffee, and it was then that I noticed how much it impacted my anxiety, she added. With my heart and mind racing, I realized that coffee contributed to a sense of overwhelm and dread.... I felt rushed to complete tasks as if I was on a deadline.

Katie Ziskind, a licensed marriage and family therapist, also quit drinking coffee due to bouts of anxiety.

I choose to stop drinking coffee because it gave me jitters it actually caused me to have more anxiety, Ziskind said. Now, because I dont drink coffee, my anxiety level has decreased drastically. I feel much calmer and have much more long-term energy.

Given that we live in what has been deemed theAge of Anxiety,perhaps it makes sense that the anxiety-ridden dont indulge in coffee which, of course, is known to cause jitters and make your heart race. But the anecdotal stories aside, does drinking a cup of coffee actually cause anxiety?

Here's what the biochemistry says. When one consumes caffeine, it binds to adenosine receptors in the brain, which allows dopamine to move more smoothly. (This is partly why some studies have found that drinking coffee may reduce the risk of depression.)

However, a study by the Institute of Neuroscience of the Universitat Autnoma de Barcelona and the Karolinska Institutet (KI) in Sweden found evidence that long-term caffeine consumption can worsen neuropsychiatric symptoms, such as those found in anxiety. Caffeine also promotes the release of cortisol, the stress hormone, which can lead to a stress response in the body. For some people, a stress response is synonymous with anxiety and panic. This can contribute to mood swings and ruminations.

The Diagnostic and Statistical Manual of Mental Disorders (DSM5) which is the guide published by the American Psychiatric Association and used by healthcare professionals for the diagnosis of mental disorders lists four caffeine-related disorders, including an anxiety-induced anxiety disorder. A 2005 study found that excessive caffeine consumption can lead to symptoms that resemble to psychiatric conditions like anxiety and psychosis.

In 2018, researchers from Italy and the US reviewed dozens of studies to try to come to a definitive conclusion about caffeine, coffee and health. They concluded that coffee can be part of a healthful diet.

Still, because some individuals react differently than others, it can be hard to generalize caffeine's effects.

It varies from individual to individual, but consuming more than your normal amount of caffeine could make you feel nervous, anxious, irritable or jittery, and may cause excessive urine production or irregular heartbeat, caffeine researcher Maggie Sweeney told the Washington Post . And for those who have anxiety or insomnia, it could worsen their symptoms.

Glasgow biochemists create UKs first hangover cure in a can but you have to drink it before bed – The Scottish Sun

GLASGOW biochemists have created the UKs first "hangover cure" after-alcohol revival drink.

Expert scientists Dhruv Trivedi and Vandana Pillai came up with the formula for Bounce Back.

Users are supposed to drink the can before bed after a night of boozing, so that they can wake up refreshed in the morning.

The drinks website states it was created with an aim to create innovative, scientifically validated formulations that have a real impact in peoples lives and solve specific problems.

It contains 16 unique ingredients all aiming to ensure party-goers feel replenished after a good sleep.

Biochemists Dhruv and Vandana first moved to Scotland to pursue the innovative idea after being scouted by the UK government's Sirius Programme.

They first launched a prototype drink back in 2015.

A spokesperson for Bounce Back said: "We'd came up with a powdered prototype, but marketing research told us people wanted a soft drink in a can, for convenience.

"Work on the can has taken 22 months, with 54 different versions. It's been a long journey."

Explaining how the product works, they said: "After you've had alcohol, it supports your body with the nutrients you need."

"It's something people can drink instead of a glass of water that offers them an extra boost."

And the company are looking to continue moving forward - as they plan a big marketing push early next year.

They said: "We'd say the product has performed as expect so far. We're planning on doing a big marketing push in March."

CONTEXT:

Clinical studies suggest that AL-TENS, acupuncture-like transcutaneous electrical nerve stimulation (TENS), is effective for treating many types of pain and physical dysfunction. To date, only a few studies have compared the TENS and AL-TENS forms of stimulation, and no studies have compared the efficacy of conventional TENS and AL-TENS in patients who have undergone total knee arthroplasty (TKA).

The study intended to determine (1) the efficacy of conventional TENS and AL-TENS for TKA patients and (2) which outcomes-pain at rest, movement-evoked pain, and physical function-were most likely to be affected by conventional TENS compared with AL-TENS for people with pain, to inform the design of future studies.

The research team designed a single-blind, randomized clinical trial with randomized treatment allocation.

The study took place at the Orthopedic and Rehabilitation Hospital of Jeonju (Jeonju, South Korea).

Participants were 30 patients at the hospital who had undergone TKA.

Participants were randomly assigned to 1 of 2 groups, TENS or AL-TENS, and received the relevant intervention at selected points for knee pain. Each group received treatment for 30 min per visit, 5 times per wk, for 2 wk during the study.

Outcome measures were pain intensity, measured with a visual analogue scale; knee functional mobility, measured with the Western Ontario and McMaster UniversitiesOsteoarthritisIndex and with the Timed Up & Go Test; and inflammation, measured by the C-reactive protein level. Data were collected at baseline and postintervention.

Changes in pain, knee function, knee mobility, and inflammation between baseline and postintervention were statistically significant for both groups (P < .05). Changes in pain, stiffness, and inflammation between baseline and postintervention were significantly greater for the AL-TENS group compared with TENS group (P < .05).

AL-TENS was more effective than TENS with respect to pain, stiffness, and inflammation relief for patients following TKA.

What causes neurons to die in Parkinsons disease?

Parkinsons disease is a long-term (chronic) neurological condition that affects about 12,000 people in Ireland and between seven and 10 million people worldwide.

The disease affects the way the brain co-ordinates body movements like walking and talking, but cognitive abilities are also affected.

There is currently no cure for the disease, but researchers at Trinity have recently published findings of a study which may lead to better treatments for this debilitating illness. The paper has been published in the international Cell Press journal Structure.

Neurons in the part of the brain called substantia nigra (dark matter) produce and release a hormone called dopamine. This hormone acts as a messenger between these cells in the substantia nigra and other parts of the brain which control body movements.

If these specialised neurons become damaged or die, the amount of dopamine in the brain is reduced. This means that the parts of the brain that control movement cease to function normally, said Amir Khan, associate professor, School of Biochemistry and Immunology at Trinity.

The only treatment for Parkinsons disease in the last 20 years has been dopamine replacement therapy. This involves providing a substitute to try to increase the levels of the hormone in the brain. However, the treatment is not completely effective and can wear off over time, and it also has side effects.

The main reason why we lack new treatments is that we dont understand the fundamental mechanism of how neurons become sick and die. No one knows why these particular neurons in the substantia nigra are affected.

In the last few years, the field has completely changed. We have new insight into a gene called LRRK2, which is the most common cause of inherited Parkinsons disease. Although only 10% of Parkinsons cases are inherited, the enzyme that is produced by the LRRK2 gene seems to be overactive in both inherited and sporadic cases.

In other words, afflicted individuals may not have an LRRK2 mutation, but the enzyme runs amok in their neurons anyway. Inhibitors of this enzyme are now in late clinical trials for treatment of Parkinsons disease.

The team at Trinity has studied the effects that LRRK2 has on other proteins in neuronal cells. To understand how LRRK2 affects the brain and leads to Parkinsons disease, the team has simulated the activity of the enzyme in the laboratory.

The research allowed us to visualize the 3D structure of a protein complex that is formed when LRRK2 is overactive. From these structural studies of proteins, we can understand how LRRK2 is able to impose its profound effects on neurons. We are the first group to report the effects of LRRK2 in 3-D detail using a method called X-ray crystallography, said Prof Khan.

An overactive LRRK2 runs loose in neurons and wreaks havoc on motor and cognitive abilities. In a way, we are chasing the footprints that LRRK2 leaves in the brain to understand what it does, and find ways to stop it.

We are hopeful that these studies may eventually lead to new treatments for Parkinsons disease, for which there is currently no cure.
