Influence of the microbiome, diet and genetics on inter-individual variation in the human plasma metabolome – Nature.com

Untargeted plasma metabolites in Dutch cohorts

In this study, we examined plasma metabolomes in 1,679 fasting plasma samples from 1,368 individuals from two LLD5 sub-cohorts (LLD1 and LLD2) and the GoNL6 cohort (Extended Data Fig. 1 and Supplementary Table 1). The LLD1 cohort was the discovery cohort, with information about genetics, diet and the gut microbiome available for 1,054 participants. Moreover, 311 LLD1 subjects were followed up 4years later (LLD1 follow-up). We also included two independent replication cohorts: 237 LLD2 participants for whom we had genetic and dietary data and 77 GoNL participants for whom only genetic data were available (Extended Data Fig. 1 and Supplementary Table 1). Untargeted metabolomics profiling was done using flow-injection time-of-flight mass spectrometry (FI-MS)10,11, which yielded plasma levels of 1,183 metabolites (Supplementary Table 2). These metabolites covered a wide range of lipids, organic acids, phenylpropanoids, benzenoids and other metabolites (Extended Data Fig. 2a). As we observed weak (absolute rSpearman<0.2) correlations among the 1,183 metabolites (Extended Data Fig. 2b), data reduction was not required and, consequently, all metabolites were subjected to subsequent analyses. We validated the identification and quantification of some metabolites (for example, bile acids, creatinine, lactate, phenylalanine and isoleucine) by comparing their abundance levels from FI-MS with those previously determined by liquid chromatography with tandem mass spectrometry (LC-MS/MS)12 or NMR13 (rSpearman>0.62; Extended Data Fig. 2c,d).

To compare the relative importance of diet, genetics and the gut microbiome in explaining inter-individual plasma metabolome variability, we calculated the proportion of variance explained by these three factors for the whole plasma metabolome profile and for the individual metabolites separately. We have detailed information on 78 dietary habits (Supplementary Table 3), 5.3million human genetic variants and the abundances of 156 species and 343 MetaCyc pathways for each individual of the LLD1 cohort. Diet, genetics and the gut microbiome could explain 9.3, 3.3 and 12.8%, respectively, of inter-individual variations in the whole plasma metabolome, without adjusting for covariates (see the Methods section Distance matrix-based variance estimation; false discovery rate (FDR)<0.05; Fig. 1a and Supplementary Table 4), whereas intrinsic factors (age, sex and body mass index (BMI)) and smoking collectively explained 4.9% of the variance. Together, these factors explain 25.1% of the variance in the plasma metabolome (Fig. 1a).

Next, we tested for pairwise associations between each metabolite and the dietary variables, genetic variants and microbial taxa. We observed 2,854 associations with dietary habits (Supplementary Table 5), 48 associations with 40 unique genetic variants (metabolite quantitative trait loci (mQTLs); Supplementary Table 6), 1,373 associations with gut bacterial species (Supplementary Table 7) and 2,839 associations with bacterial MetaCyc pathways (Supplementary Table 8) (see the Methods sections Associations with dietary habits, QTL mapping and Microbiome-wide associations). In total, 769 metabolites were significantly associated with at least one factor (Fig. 1b and Supplementary Tables 58). We then performed interaction analysis to assess the role of dietmicrobiome, geneticsmicrobiome and dietgenetics interactions in regulating the human metabolome using an interaction term in the linear model (see the Methods section Interaction analysis). Among these, 185 metabolites were associated with multiple factors and seven were affected by either geneticsmicrobiome, geneticsdiet or dietmicrobiome interactions (Supplementary Table 9).

As interactions were limited, we further assessed the proportion of variance of each metabolite that was explained by these factors using an additive model with the least absolute shrinkage and selection operator (lasso) method (see the Methods section Estimating the variance of individual metabolites). In general, the inter-individual variations in 733 metabolites could be explained by at least one of the three factors (FDRF-test<0.05; Supplementary Table 10). In detail, dietary habits contributed 0.435% of the variance in 684 metabolites; microbial abundances contributed 0.725% of the variance in 193 metabolites; and genetic variants contributed 328% of the variance in 44 metabolites (adjusted r2; FDRF-test<0.05; Supplementary Table 10). We also estimated the explained variance of metabolites using Elastic Net14, which is designed for highly correlated features, and found that the estimated explained variances were comparable between linear regression and the Elastic Net regression (Supplementary Fig. 1).

We further compared the variance explained by each type of factor (diet, genetics or the microbiome) and assigned the dominant factor for each metabolite if one factor explained more variance than the other two. Inter-individual variations in 610 metabolites were mostly explained by diet, 85 were explained by the gut microbiome and 38 were explained by genetics (Supplementary Table 10). Hereafter, we refer to these as diet-dominant, microbiome-dominant and genetics-dominant metabolites, respectively. The dominant factors of metabolites highlight their origin. For instance, ten out of the 21 diet-dominant metabolites for which diet explained >20% of the variance (FDRF-test<0.05; Supplementary Table 10) were food components based on their annotation in the Human Metabolome Database (HMDB)15. Similarly, of the 85 microbiome-dominant metabolites, 23 were annotated in the HMDB as microbiome-related metabolites (including 15 uremic toxins). Furthermore, out of the 38 genetics-dominant metabolites, ten were lipid species and eight were amino acids. Taken together, our analysis highlights that one factoreither dietary, genetic or microbialcan have a dominant effect over the other two in explaining the variances of plasma metabolites, with diet or the microbiome being particularly dominant. However, we also found that the variances in 185 metabolites were significantly attributable to more than one factor (Supplementary Table 10), including six metabolites associated with both genetics and the microbiome and 153 metabolites associated with both diet and the microbiome. For example, genetics and the microbiome explained 4 and 5%, respectively, of the variance in plasma 5-carboxy--chromanol (Fig. 1c)a dehydrogenated carboxylate product of 5-hydroxy--tocopherol16 that may reduce cancer and cardiovascular risk17. Another example is hippuric acida uremic toxin that can be produced by bacterial conversion of dietary proteins18, with 13% of its variance explained by diet and 13% explained by the microbiome (Fig. 1c).

Temporal changes in plasma metabolites can reflect changes in an individuals diet, gut microbiome and health status. When assessing the plasma metabolome in the 311 LLD1 follow-up samples, we indeed observed a significant shift in the plasma metabolome, with a significant difference in the second principal component (PPC1 paired Wilcoxon=0.1 and PPC2 paired Wilcoxon=1.3105; Fig. 2a). Baseline genetics, diet and microbiome, together with age, sex and BMI, could explain 59.4% of the variance in the follow-up plasma metabolome (PPERMANOVA=0.004) (Supplementary Fig. 2). We also observed that temporal stability can vary substantially between different metabolites (see the Methods section Temporal consistency of individual metabolites; Supplementary Table 11). Previously, we had assessed the changes in the gut microbiome in the LLD1 follow-up cohort and linked these to changes in the plasma metabolome7. Here, we further checked the temporal variability of the plasma metabolome and assessed the stability of diet-, microbiome- and genetics-dominant metabolites over time. Interestingly, the temporal correlation of the microbiome-dominant metabolites was similar to that of the genetics-dominant metabolites (PWilcoxon=0.51; Fig. 2b), whereas the temporal correlation between diet-dominant metabolites was significantly lower than between microbiome- and genetics-dominant metabolites (PWilcoxon<3.4105; Fig. 2b). However, the dominant dietary, microbial and genetic factors identified at baseline also explained similar variance in metabolic levels in the follow-up samples (Extended Data Fig. 3 and Supplementary Table 10). Our data also revealed a positive correlation between stability and the amount of variance that could be explained: the more variance explained, the more stable a metabolite is over time (Fig. 2c). For a few metabolites, we could not replicate the variance explained at baseline at the second time point, and these metabolites also showed weak or no correlation in their abundances between the two time points. For example, N-acetylgalactosamine showed very weak correlation between the two time points (r=0.13; P=0.02), and its genetic association was not replicated at the second time point.

Having established the variances in metabolites explained by diet, genetics and the gut microbiome and the dominant factors that explained most of this variance, we focused on detailing specific associations and on the potential implications of our findings for assessing diet quality and improving our understanding of the genetic risk of complex diseases and the interaction and causality relationships among diet, the microbiome, genetics and metabolism.

We observed 2,854 significant associations (FDRSpearman<0.05) between 74 dietary factors and 726 metabolites (Fig. 3a and Supplementary Table 5; see the Methods section Lifelines diet quality score prediction). Associations with food-specific metabolites can, in theory, be used to verify food questionnaire data. For instance, the strongest association we observed was between quinic acid levels and coffee intake (rSpearman=0.54; P=1.61080; Fig. 3b). Quinic acid is found in a wide variety of different plants but has a particularly high concentration in coffee. Another example is 2,6-dimethoxy-4-propylphenol, which was strongly associated with fish intake (rSpearman=0.53; P=1.51076; Fig. 3c). This association is expected as this compound is particularly present in smoked fish according to HMDB annotation15. In addition, we also detected associations between dietary factors and metabolic biomarkers of some diseases. For example, 1-methylhistidine is a biomarker for cardiometabolic diseases including heart failure19 that is enriched in meat, and we observed significant associations between 1-methylhistidine and meat (rSpearman=0.12; P=7.2105) and fish intake (rSpearman=0.11; P=3.1104) as well as a lower level of 1-methylhistidine in vegetarians (rSpearman=0.15; P=9.7107; Fig. 3d).

Given the relationship between diet, metabolism and human health, we wondered whether the plasma metabolome could predict diet quality. For each of the Lifelines participants, we constructed a Lifelines Diet Score based on food frequency questionnaire (FFQ) data that reflected the relative diet quality based on dietdisease relationships8. To build a metabolic model to predict an individuals diet quality, we used LLD1 as the training set and LLD2 as the validation set. The resulting metabolic model included 76 metabolites, 51 of which were dominantly associated with diet. The diet score predicted by metabolites showed a significant association with the real diet score assessed by the FFQ in the validation set (r2adjusted=0.27; PF-test=3.5105; Fig. 3e). We also tested four other dietary scores (the Alternate Mediterranean Diet Score20, Healthy Eating Index (HEI)21, Protein Score22 and Modified Mediterranean Diet Score23) and found that the HEI predicted by plasma metabolites was also significantly associated with the FFQ-based HEI (r2adjusted=0.23; PF-test=6.5105; Supplementary Table 12).

Genetic associations of plasma metabolites may provide functional insights into the etiologies of complex diseases. After correcting for the first two genetic principal components, age, sex, BMI, smoking, 78 dietary habits, 40 diseases and 44 medications, QTL mapping in LLD1 identified 48 study-wide, independent genetic associations between 44 metabolites and 40 single-nucleotide polymorphisms (SNPs) (PSpearman<4.21011; clumping r2=0.05; clumping window=500kilobases (kb); Fig. 4a and Supplementary Table 6). All 48 genetic associations were replicated in either LLD1 follow-up or the two independent replication datasets (LLD2 and GoNL; Supplementary Fig. 3 and Supplementary Table 6). We also assessed the impact of physical activity, as assessed by questionnaires24, on the genetics association of metabolism, but found its influence to be negligible (Supplementary Fig. 4). Functional mapping and annotation (FUMA) of genome-wide association studies (GWAS)25 analysis revealed that the identified mQTLs were enriched in genes expressed in the liver and kidney (Extended Data Fig. 4) and related to metabolic phenotypes (Supplementary Table 6).

The strongest association we found was between the caffeine metabolite 5-acetylamino-6-formylamino-3-methyluracil (AFMU) and SNP rs1495741 near the N-acetyltransferase 2 (NAT2) gene (rSpearman=0.52; P=1.71066; Fig. 4b), which showed strong linkage disequilibrium (r2=0.98) with a SNP, rs35246381, that was recently reported to be associated with urinary AFMU26. AFMU is a direct product of NAT2 activity and has been associated with bladder cancer risk27. Interestingly, the plasma level of AFMU was associated not only with coffee intake (rSpearman=0.29; P=9.21022; Supplementary Table 5) and the genotype of rs1495741, but also with their interactions (Supplementary Table 9). Individuals with a homologous AA genotype had a similar level of coffee intake, but their correlation between coffee intake and plasma AFMU level was significantly lower compared with individuals with GG and GA genotypes (Fig. 4e,f).

Pleotropic mQTL effects were also observed at several loci, including SLCO1B1, FADS2, KLKB1 and PYROXD2 (Supplementary Table 6). For example, three associations (related to three metabolites, two of them lipids) were observed for two SNPs (rs67981690 and rs4149067; linkage disequilibrium r2=0.72 in Northern Europeans from Utah) in SLCO1B1, which encodes the solute carrier organic anion transporter family member 1B1. Expression of the SLCO1B1 protein is specific to the liver, where this transporter is involved in the transport of various endogenous compounds and drugs, including statins28, from blood into the liver. The SLCO1B1 locus has also been linked to plasma levels of fatty acids and to statin-induced myopathy29. Furthermore, we detected a geneticsmicrobiome interaction between rs67981690 and microbial fatty acid oxidation pathways in regulating plasma levels of 5-carboxy--chromanol (P=1.5103), where the association of the bacterial fatty acid oxidation pathway with plasma levels of 5-carboxy--chromanol was dependent on the genotype of rs67981690 (Fig. 4g,h).

To identify novel mQTLs, we performed a systematic search of all published mQTL studies from 2008 onwards (Supplementary Table 13). This approach identified three novel mQTLs in our datasets (Supplementary Table 13) that were either not located close to previously reported mQTLs (distance>1,000kb) or not in linkage disequilibrium (r2<0.05). The first two novel SNPsrs13100173 at HYAL3 and rs11741352 at ARSBwere associated with N-acetylgalactosamine-4-sulfate (Fig. 4c,d), which is associated with mucopolysaccharidosis30. Interestingly, N-acetylgalactosamine-4-sulfate can bind to HYAL proteins (HYAL1, HYAL2, HYAL3 and HYAL4), suggesting that mQTLs can also pinpoint potential metaboliteprotein interactions. The third novel mQTL was rs17789626 at SCLT1, which was associated with mizoribinea compound used to treat nephrotic syndrome31.

We established 4,212 associations between 208 metabolites and 314 microbial factors (114 species and 200 MetaCyc pathways) (FDRLLD1<0.05; PLLD1 follow-up<0.05; Supplementary Tables 7 and 8). Interestingly, many of the metabolites that were associated with microbial species and MetaCyc pathways are also known to be gut microbiome related based on their HMDB annotations15. For instance, we observed 919 associations with 25 uremic toxins, 142 associations with thiamine (vitamin B1) and 117 associations with five phytoestrogens (FDR<0.05; Supplementary Tables 7 and 8). Uremic toxins and thiamine have been shown to be related to various diseases, including chronic kidney disease and cardiovascular diseases32,33. Phytoestrogens are a class of plant-derived polyphenolic compounds that can be transformed by gut microbiota into metabolites that promote the hosts metabolism and immune system33,34.

To assess whether gut microbiome composition causally contributes to plasma metabolite levels, we carried out bi-directional MR analyses (see the Methods section Bi-directional MR analysis). Here, we focused on the 37 microbial features that were associated with at least three independent genetic variants at P<1105 and with 45 metabolites (Supplementary Table 14). At FDR<0.05 (corresponding to P=2103 obtained from the inverse variance weighted (IVW) test)35, we observed four potential causal relationships at baseline that could also be found in the follow-up in the microbiomes to metabolites direction (Fig. 5ad and Supplementary Tables 15 and 16) but not in the opposite direction (Supplementary Table 17), and these outcomes were maintained following weighted median testing (P<0.03; Supplementary Fig. 5). To ensure that the data followed MR assumptions, we performed several sensitivity analyses, including checking for horizontal pleiotropy (MR-Egger36 intercept P>0.05; Supplementary Table 15) and heterogeneity (Cochrans Q test P>0.05; Supplementary Table 15) and leave-one-out analysis (Extended Data Fig. 5). We did not use causal estimates derived using the MR-Egger method to filter the results, as its power to detect causality is known to be low36. These sensitivity checks further confirmed the reliability of these four MR causal estimates.

We further found that increased abundance of microbial adenosylcobalamin biosynthesis (coenzyme B12) was associated with reduced plasma levels of 5-hydroxytryptophol (Fig. 5a)a uremic toxin related to Parkinsons disease37. We also found that plasma hydrogen sulfite levels were related to Eubacterium rectale (Fig. 5c)a core gut commensal species38 that is highly prevalent (presence rate=97%) and abundant (mean abundance=8.5%) in both our cohorts and in other populations39,40,41. As a strict anaerobe, E. rectale promotes the hosts intestinal health by producing butyrate and other short-chain fatty acids from non-digestible fibers42, and a reduced abundance of this species has been observed in subjects with inflammatory bowel disease39,43 and colorectal cancer44 compared with healthy controls. As a toxin, hydrogen sulfite interferes with the nervous system, cardiovascular functions, inflammatory processes and the gastrointestinal and renal system45. Our results thus reveal a potential new beneficial effect of E. rectale.

To further investigate the metabolic potential of individual bacterial species, we applied newly developed pipelines to identify microbial primary metabolic gene clusters (gutSMASH pathways)46 and microbial genomic structural variants (SVs)47. These two tools profile microbial genomic entities that are implicated in metabolic functions. By associating 1,183 metabolites with 3,075 gutSMASH pathways and 6,044 SVs (1,782 variable SVs (vSVs) and 4,262 deletion SVs (dSVs); see Methods), we observed 23,662 associations with gutSMASH pathways and 790 associations with bacterial SVs (FDRLLD1<0.05; PLLD1 follow-up<0.05; Supplementary Tables 1820). These associations connect the genetically encoded functions of microbes with metabolites, thereby providing putative mechanistic information underlying the functional output of the gut microbiome. In one example, we observed that the microbial uremic toxin biosynthesis pathways, including the glycine cleavage pathway (in Olsenella and Clostridium species) and the hydroxybenzoate-to-phenol pathway (in Clostridium species) responsible for hippuric acid and phenol sulfate biosynthesis, were associated with the hippuric acid (Olsenella species: rSpearman=0.15; P=9.3107; Clostridium species: rSpearman=0.18; P=5.9109) and phenol sulfate (rSpearman=0.17; P=4.2108; Extended Data Fig. 6a) levels measured in plasma, respectively (FDRLLD1<0.05 and PLLD1 follow-up<0.05; Extended Data Fig. 6b).

Next, we carried out a mediation analysis to investigate the links between diet, the microbiome and metabolites. For 675 microbial features that were associated with both dietary habits and metabolites (FDR<0.05), we applied bi-directional mediation analysis to evaluate the effects of microbiome and metabolites for diet (see the Methods section Bi-directional mediation analysis). This approach established 146 mediation linkages: 133 for the dietary impact on the microbiome through metabolites and 13 for the dietary impact on metabolites through the microbiome (FDRmediation<0.05 and Pinverse-mediation>0.05; Fig. 6a,b and Supplementary Table 21). Most of these linkages were related to the impact of coffee and alcohol on microbial metabolic functionalities (Fig. 6a).

Coffee contains various phenolic compounds that can be converted to hippuric acid by colonic microflora48. Hippuric acid is an acyl glycine that is associated with phenylketonuria, propionic acidemia and tyrosinemia49. We observed that hippuric acid can mediate the impact of drinking coffee on Methanobrevibacter smithii abundance (Pmediation=2.21016; Fig. 6c). We also observed that hulupinic acid, which is commonly detected in alcoholic drinks, can mediate the impact of beer consumption on the Clostridium methylpentosum ferredoxin:NAD+ oxidoreductase (Rnf) complex (Pmediation=2.21016; Fig. 6d)an important membrane protein in driving the ATP synthesis essential for all bacterial metabolic activities50.

Of the dietary impacts on metabolites through the microbiome (Fig. 6b and Supplementary Table 21), one interesting example is a Ruminococcus species vSV (300305kb) that encodes an ATPase responsible for transmembrane transport of various substrates51. This Ruminococcus species vSV mediated the effect of fruit consumption on plasma levels of urolithin B (Pmediation=2.21016; Fig. 6e). Urolithin B is a gut microbiota metabolite that protects against myocardial ischemia/reperfusion injury via the p62/Keap1/Nrf2 signaling pathway52. Taken together, our data provide potential mechanistic underpinnings for dietmetabolite and dietmicrobiome relationships.

Expediting IND applications with drug master files – BioPharma Dive

Completing an investigational new drug (IND) application can be a long and tedious process, one known to bog down timelines and frustrate sponsors looking to take their cell or gene therapy (CGT) to the clinic. In a tight race to trials, program sponsors look for efficiencies from bench to bedside.

Using off-the-shelf materialssuch as plasmid DNA products and CRISPR-associated nucleases is one way to potentially speed things up particularly if they come with a drug master file, or DMF.

Drug master files are reference packages that report to the FDA information about the processes involved in new drug products. Theyre not FDA-required nor approved nor denied but they can help supplement INDs and other applications as a way to cross-reference new processes with pre-filed submissions.

And as a concept, DMFs arent new. Download the DMF list and youll see tens of thousands of submissions, some dating to 1939. Theyve always been used to support various materials, even ibuprofen and acetaminophen. But as normalized as theyve become, DMFs are still novel among CGTs, which only recently began using off-the-shelf materials that would benefit from a DMF.

With the growing standardization of cell and gene therapies, interest has emerged in DMFs as a tool to expedite the path to clinical trials. If you use an off-the-shelf reagent that has a DMF, the application links to that preexisting file without the filer having to provide redundant information.

As more off-the-shelf materials such as plasmids become available for cell and gene therapies, suppliers are submitting DMFs for those products as a benefit to customers. Doing so gives researchers a dual offering: They can not only access standardized materials at GMP-grade with the cost and time savings that come with that but they can also cross-reference their INDs to the already prepared DMF.

Two examples of these DMF-ready products are Aldevrons SpyFi Cas9 nuclease and pALD-X80 helper plasmid, each available from research grade to GMP. When new cell and gene therapy products use these materials, users automatically get the benefit of a pre-filed DMF that references Aldevrons manufacturing processes involved in making them.

The reason IND submissions have long embraced DMFs is the same reason DMFs are likely to rapidly expand among CGT applications in the months and years ahead: Theyre not just efficient in saving time, but theyre also potentially more comprehensive with less of a risk of missed information.

If you use the SpyFi Cas9 nuclease, for example, or others offered from Aldevron, you can skip the drug substance part for the Cas9 product when filing the IND because Aldevron has already prepared the DMF.

And when linking to the DMF, the FDA gets complete access to manufacturing information without the filer having to do so. This avoids overlooking information the FDA might need, which can delay application review.

Moreover, translational medicine organizations may find particular value in DMF-ready products because they overcome resource barriers inherent to hospitals and academic institutions. Unlike big pharma or biotech companies, many such organizations may not have easy access to field experts. Turning to DMFs gives academic outfits the economies of scale they might not get otherwise.

By 2025 thats just two years away the FDA expects to approve up to 20 cell and gene therapy products per year. With all that IND activity in a crowded market, sponsors will need any differentiating efficiency they can get along the pathway to clinical trials and beyond.

By taking advantage of pre-filed DMFs, researchers benefit from the value, time savings and reduced complexities of having someone else do the heavy lifting for key components of the final drug product. After all, this concept isnt new in drug development. However, the science in this so-called new frontier of medicine is. As it becomes more standardized, everyone wins including future patients.

What doctors wish patients knew about breast-cancer prevention – American Medical Association

Its natural for women to worry about breast cancer, especially since many people know someone who was touched by the disease. While there is no foolproof way to prevent breast cancer, there are things you can do to lower your risk. Some factors you cant change, but knowing what can help is key to lowering your risk of breast cancer.

Breast cancer is the second most common cancer among women in the U.S.some types of skin cancer are the most common. Between 1989 and 2020, breast cancer death rates decreased by 43%, but racial and ethnic inequities still exist. While breast cancer incidence is lower among Black women than white women, the death rate is 40% higher among Black women than white women, according to the American Cancer Society. That is because about one in five Black women with breast cancer have triple-negative breast cancermore than any other racial or ethnic group. Meanwhile, Asian American and Pacific Islanders have the lowest death rate from breast cancer while Native Americans and Alaska Natives have the lowest rates of developing breast cancer. And while rare, men can get breast cancer too.

The AMAsWhat Doctors Wish Patients Knew series provides physicians with a platform to share what they want patients to understand about todays health care headlines.

In this installment, Jill Jin, MD, an internist at Northwestern Medicine and clinical assistant professor of medicine at Northwestern University Feinberg School of Medicine, took time to discuss what patients need to know about what to do to reduce their risk of breast cancer. She is also a senior physician adviser for the AMA and an associate editor forJAMA.

Know the risk factors

While risk factors for breast cancer are broad, we think primarily about agearound the age of menopause and after menopause is when breast cancer risk goes up in women, said Dr. Jin. Family history, of course, is another big one. That includes genetic mutations that we know of such as BRCA1 and BRCA2.

Theres also this whole concept of estrogen exposure, which can be both endogenouswithin the body or how much your body producesversus exogenous, from medications she said. Then other things like alcohol and smoking are thought to be associated somewhat with breast cancer as well.

Start screening between 40 and 50

Overall, the recommended age to start screening for breast cancer in average-risk women would be anywhere from 40 to 50 years old, said Dr. Jin. It is important to convey to patients that most professional societies do recommend later than 40, either 45 or 50, as the age to start screening. But most physicians are still starting on the earlier end of this spectrum because it can be a tough sell for patients to say, wait until 50 years old when, to be honest, most people around them are probably getting screened earlier.

Almost everyone knows somebody these days who has had breast cancer, whether its a friend or a family member, and when you have that personal connection its scary, she added. Thats why I usually tell women who are at average riskwho dont have family history of breast cancerthat I am comfortable waiting until 45 years old to start screening.

If they do have a family history of breast cancer or other risk factors, we certainly can and should start screening earlier, Dr. Jin said. Its very individualized at the end of the day.

Different screening tests are available

There are several different screening modalities, said Dr. Jin, noting that a mammogram is the most common one. Other methods of screening include ultrasounds, as well as a breast magnetic resonance imaging.

But for most people, we start with mammograms, she said.

Earlier screening isnt always better

It always begins with getting to know the patient, asking about their history and their lifestyleits definitely an individualized risk assessment first, said Dr. Jin. And if there is nothing that suggests they are at higher risk than average, then you can have a discussion with patients about potentially waiting to start screening.

It comes down to the benefits versus the harms of screening, she added, noting the younger you start screening, the more lives you will save because you will catch more cancers at earlier stages, especially the more aggressive ones.

But on the flip side, the younger people are, the more you pick up things that are not cancer, which is called a false positive finding. Younger women have denser breast tissue, and when breast tissue is dense, it is very hard to differentiate normal tissue from something that may look like cancer, Dr. Jin explained. And then you go down this whole path of follow-up testing which includes additional mammograms and sometimes biopsy, which very often ends up being an unnecessary biopsy because everything will turn out normal.

This causes a lot of anxiety. It upends patients lives for a couple months while this whole process is going on, and that amount of anxiety affects many other parts of patients livesit is not trivial, she added. And then you do it all over again the next year. The younger you start, the more the potential harms of these false positives start to outweigh the potential benefit of earlier diagnosis.

Maintain a healthy lifestyle

For all womenreally for everyoneit is important to maintain a healthy lifestyle, said Dr. Jin. That means eating a balanced diet, not drinking too much alcohol, not smoking, maintaining regular physical activity and a normal body mass index.

All of those things are likely helpful for prevention of not just breast cancer, but other cancers as well, along with cardiovascular diseasea lot of things, she added.

There are medications to reduce risk

Chemoprevention, or the use of medications, is another option to reduce breast-cancer risk, said Dr. Jin. For chemoprevention, there are two classes of medications that are used. One is called selective estrogen receptor modulators, or SERMs.

Tamoxifen is probably the most common one that is used. SERMs medications block the effects of estrogen in the breast, she added. Another class is called aromatase inhibitors. Those are usually used in older women after menopause and stop other hormones in the body from becoming estrogen.

However, both have other side effects. While tamoxifen blocks the effects of estrogen in breast tissue, it can actually enhance estrogen effects in other parts of the body, so we do worry about blood clots as well as uterine cancer, said Dr. Jin. And then aromatase inhibitors can cause other side effects related to low estrogen such as hot flashes, bone pain, decreased bone density, and increased risk of osteoporosis and fractures.

Thats why we dont use these medications in everyone to decrease breast cancer risk, and reserve them for high-risk women only. Again, as with every decision in medicine, we want to make sure the balance of potential benefits versus harms is in favor of benefits, she said.

Surgical prevention is also an option

The other kind of prevention would be surgical prevention, said Dr. Jin. This is also done for women who are high risk, most commonly because of the BRCA gene mutation.

People who have a known BRCA gene mutation, which puts them at an increased risk for both breast and ovarian cancer, are candidates for surgery to remove the breasts. Thats called prophylactic mastectomy, she said. They also may be candidates for surgery to remove the ovaries to decrease the risk of ovarian cancer as well.

Test for the BRCA gene mutation

There are calculators that can be used to calculate whether someone, based on their family history and ethnicity, should get genetic testing for the BRCA gene mutation, which is a blood test said Dr. Jin. If you have a first-degree family membersuch as your mom or siblingwho has breast cancer and is known to have BRCA, then you should get tested for it.

If you just have a family history of breast cancer with unknown BRCA status, thats when the calculators come into play, she added, noting they look at how many first-degree and second-degree relatives, whether you are of Ashkenazi Jewish descent, and certain other risk factors to decide whether you should get the genetic testing.

Breastfeeding may reduce risk

While there are no clinical trials on this topic, there is observational data that does suggest that breastfeeding is protective against breast cancer, said Dr. Jin. The same goes for having children versus not having children; pregnancy does seem to be protective as well.

Were not saying go get pregnant and breastfeed to reduce your risk of having breast cancerits not practical, she added. But it does seem to be an association.

Birth control is OK to take

This is also somewhat controversial, but overall, the link between birth control and breast cancer is very small to none, Dr. Jin said. When I talk with my patients about this, I share that using birth control pills most likely does not increase the risk of breast cancer in a clinically significant way.

Furthermore, this very small potential increase in risk is limited to the time that youre actually taking birth-control pills, she said. So, its not a permanent effect. Its temporary.

Be cautious with self-breast exams

There has not been any good evidence to show that self-screening has any overall benefit in mortality, said Dr. Jin. Breasts are just lumpy to begin with and a lot of people end up feeling lumps that just end up being normal breast tissue.

And you may end up, again, going down that path of all the imaging and the biopsies and in the end, it is nothing, she added. So, self exams are not recommended by clinical guidelines.

However, some people are going to be wanting to do that anyways and that is fine. If someone really wants to stay on top of their body, I will explain that breasts can feel lumpy or bumpy, and what they are looking for is a change from baseline. At the end of the day, you still know your own breasts and your own body the best, said Dr. Jin. So, if you feel something that is different, that you have not felt before, then you should let me know and we can decide at that point what to do,

If they are in the office with me, I am happy to do a quick exam of the breast and tell them this is what your normal breast tissue feels like, dont be alarmed if you feel this or if you feel this. It is just normal.

Dont hesitate to talk to your doctor

While screening is recommended to start between 40 and 50 years old, if at any point you do notice something like a lump or you see something weird on the skin or if you have pain or any symptoms that are different than normal, that takes you out of the typical screening category, she emphasized. As with all cancer screening, when a symptom is detected that is different, and you should never hesitate to bring that up to your doctor.

UNC School of Medicine Awarded $3 Million to Lead Study to Reduce PTSD Frequency, Severity | Newsroom – UNC Health and UNC School of Medicine

Led by Sam McLean, MD, MPH, University of North Carolina at Chapel Hill researchers and collaborators were awarded $3 million from the U.S. Department of Defense for to evaluate the efficacy of a therapeutic to reduce the frequency and severity of acute stress disorder and posttraumatic stress disorder.

The UNC Institute for Trauma Recovery in the UNC Department of Psychiatry has been awarded a $3-million grant from the U.S. Department of Defense (DoD) to investigate the potential of a therapeutic agent to reduce the frequency and severity of acute stress disorder and post-traumatic stress disorder (PTSD). Acute stress disorder refers to the bodys immediate response to trauma, whereas PTSD is the long-term effects of trauma.

Historically, we have been able to provide emergency care to address immediate and long-term problems after visible wounds using tools such as sutures and antibiotics. However, we still have nothing to offer trauma survivors, whether in the emergency department or on the battlefield immediately after trauma, to prevent the development of invisible wounds, said principal investigator Samuel McLean, MD, MPH, professor of psychiatry and emergency medicine and director of the Institute for Trauma Recovery at the UNC School of Medicine. We need to investigate potential treatments like ACER-801 in an effort to better address these challenges.

The proposed OASIS trial will examine the safety and efficacy of ACER-801 (osanetant) to reduce acute stress response symptoms, posttraumatic stress disorder symptoms and behavioral changes among patients presenting to the emergency department after a motor vehicle collision.

Participating sites will include Washington University in St. Louis, University of Massachusetts Chan Medical School, Rhode Island Hospital, University of Florida College of Medicine Jacksonville, and Indiana University School of Medicine. The study, proposed to begin in the first half of 2023, will evaluate the efficacy of ACER-801, which Acer Therapeutics licensed from Sanofi in 2019.

The OASIS trial builds upon a foundation of knowledge and infrastructure developed through the UNC-led, $40 million AURORA initiative. The AURORA study is a major national research initiative to improve the understanding, prevention, and recovery of individuals who have experienced a traumatic event. AURORA is supported by funding from NIH, One Mind, private foundations, and partnerships with leading tech companies such as Mindstrong Health and Verily Life Sciences, the healthcare arm of Googles parent company Alphabet.

We are proud to be partnering with a leading academic institution in the field of trauma recovery as we begin exploring ACER-801 as a treatment option to reduce the frequency and severity of acute PTSD, said Adrian Quartel, MD, FFPM, Chief Medical Officer of Acer. The data from thousands of motor vehicle collisions collected through the AURORA initiative should allow us to better predict the correlation of the emergence of acute stress disorder or PTSD symptoms following a motor vehicle collision.

Acute and chronic stress disorders can affect both civilian and military populations. According to the National Center for PTSD, in the US about 60% of men and 50% of women experience at least one trauma in their lives. In the United States alone, one-third of emergency department visits (40-50 million patients per year) are for evaluation after trauma exposures, and in a 2014 study involving 3,157 US veterans, 87% reported exposure to at least one potentially traumatic event during their service.3 Moreover, as many as 500,000 US troops who served in wars between 2001 and 2015 were diagnosed with PTSD.

Scientific rationale for OASIS:

TheTacr3gene encodes tachykinin receptor 3 (NK3R), which belongs to the tachykinin receptor family. This family of proteins includes typical G protein-coupled receptors and belongs to the rhodopsin subfamily. NK3R functions by binding to its high-affinity ligand, Neurokinin B (NKB), which is encoded by the Tac3 (human) gene.

The role ofNKB-NK3R in growth and reproduction has been extensively studied, butNKB-NK3R is also widely expressed in the nervous system from the spinal cord to the brain and is involved in both physiological and pathological processes in the nervous system. In animal models, Tac2 mRNA levels are rapidly up-regulated during fear consolidation 30 minutes after fear conditioning, and subsequent NKB-NK3R activation can lead to over stress sensitization and the consolidation of fear, and treatment with osanetant has been shown to block a critical fear/stress sensitization step in the brain. An effective therapeutic to reduce acute and persistent/long-term psychological and somatic symptoms would fulfill a large unmet need.

The Evolution of Godless Practices: Eugenics, Infanticide, and Transhumanism – The Epoch Times

There is a straight line that runs from eugenics through infanticide to transhumanism. All three are the devils work in the material world.

Let us explore the issue.

From biblical times to the present, there has been an ongoing battle between good and evil that has been waged through the centuries. On one side, there are those who believe in a higher power and that mankind was made in the image of God, which is an immutable constant not to be trifled with or corrupted by human beings. Another way of saying that is that people in this camp believe that human nature itself has been immutable and constant through the millennia and that only by looking to God can mankind improve his condition.

On the other side are those who deny the existence of God and believe that mankind is the supreme intelligence in control of human destiny, is malleable and can be shaped through science, and can evolve to a superior form of man through the planning and experimentation by natural leaders over time. A corollary for the people in this camp is that there are no moral or religious constraints on their practices intended to evolve mankind toward some conceivedand ever-changingfuture vision.

Many in the first camp would characterize the ongoing battle as good versus evil. Mankind has been arguing about, defining, and redefining what it means to be good for millennia. The ancients defined good in terms of normal versus different, knowledge versus ignorance, and later right versus wrong in the context of defining laws and justice. Societies and governments were organized around these concepts.

The concept of evil has also been defined by many cultures through the ages. The dictionary now defines evil as morally wrong or bad; immoral; wicked. Most Americans (and most people in general) have an innate understanding of what is evil on a personal level. Some refer to that understanding as a conscience. Even the people in the second camp who believe that mankind is the supreme being have vestiges of consciences informed by religion and experience.

Governments have been organized to reflect, monitor, and control the cultural norms of the people governed. Logic dictates that those governments would also implement and enforce those cultural norms from a different philosophical framework from each other, partly as shaped by their respective religious philosophies and the various ideologies that have been developed and tested over the centuries, such as monarchical rule, Marxism, fascism, socialism, communism, corporate capitalism, patriarchy, oligarchy, philosopher kings, etc.

The totalitarian ideologies that were developed and tested during the 20th century (particularly fascism and communism) denied the existence of God so that the governments formed that exercised those ideologies would be unconstrained by religious and moral boundaries in their pursuit of developing and grooming their version of modern man in their societies.

Examples of those perversions of mankind include the following:

The Nazis were obsessed with racial theories that resulted in a pseudoscientific racial classification system in which Aryans (people of German and Nordic descent) were considered to be the master race at the apex of the human pyramid and destined to rule the world (on Nazi terms, of course). Jews were by Nazis to be on the lowest level of the hierarchy.

Nazi society was organized around these concepts to develop and promote those with the purest Aryan blood at the expense of those with lesser classifications in the hierarchy. For example, German boys were educated, inculcated, and groomed in Nazi principles through the Hitler Youth program. Girls were brainwashed through two parallel programs: Young Girls was an organization for girls aged 10 to 14, while the League of German Girls was for girls aged 14 to 18, with the latter being focused on comradeship, domestic duties, and motherhood.

Soviet Man was to be the ultimate proof that communism worked and that mankind could evolve for the better (as defined by the communists) without Gods guidance. The Russian communists attempted to shape the individual consciousness, character, and social practices in order to get the people to conform to the Marxist view of the perfect citizen.

The Soviets experimented with telepathic research, cybernetic simulations, and mass hypnotism over television to control the minds of their citizenry. This was a forerunner of the mass formation psychosis being exploited by the left and globalists these days.

The Chinese Communist Party (CCP) has controlled education in China for decades to politically indoctrinate students at all levels according to CCP ideology, principles, history, racial theories, global objectives, etc., and, most importantly, conditioning everyone to acquiesce to the CCPs control of all aspects of Chinese society.

CCP leader Xi Jinping said in 2019, We need to strengthen political guidance for young people, guide them to voluntarily insist on the Partys leadership, to listen to the Party and follow the Party.

And the CCP is leading the world in implementing social controls to monitor compliance with CCP directives by all Chinese citizens. Educate, monitor, control, and discipline: the perfect world with perfectly compliant citizens envisioned for all by the Chinese communists.

Theories initiated in the 19th century bore spoiled fruit in the 20th and 21st centuries. Karl Marxs theories (Marxism) begat the Communist Manifesto, whose godless adherents continue to plague the world today. Charles Darwins theory of evolution begat eugenics, which the National Institute of Healths Genome Research Institute defines as an immoral and pseudoscientific theory that claims it is possible to perfect people and groups through genetics and the scientific laws of inheritance. John Deweys theory of progressive education similarly haunts U.S. public education to this very day.

Eugenics was/is particularly evil. Its adherents included the Nazis and Americans associated with the Population Society, the Committee on Eugenics, which studied selective and restrictive human breeding, and the American Eugenics Society.

Former President Franklin D. Roosevelt even promoted Nazi sympathizer and eugenicist Franklin Osborn to positions in government, including chairman of the Civilian Advisory Committee on Selective Service, chairman of the Army Committee on Welfare and Recreation, and chief of the Morale Branch of the War Department.

The Nazis used eugenics to justify the sterilization and ultimately the elimination of undesirables, including Jews, homosexuals, gypsies, Slavs, and others. Eugenics theories led directly to the Nazi genocide that killed millions in Europe in the 1930s and 1940s.

Eugenics was also the basis for implementing sterilization laws in the United States in over 30 states, with some of those laws persisting on the books until the 1980s, according to the NIH Genome Research Institute. Over 60,000 people deemed to be idiots, imbeciles, promiscuous (females), or feebleminded were sterilized in the United States in the 20th century.

Eugenics also heavily influenced Margaret Sanger. A eugenicist and racist, Sanger founded the Birth Control League (1921) and its successor, Planned Parenthood (1942). She supported several eugenics initiatives: sterilization of people with mental and physical disabilities, segregation in concentration camps of undesirable criminals (for example, prostitutes, paupers, drug addicts, and the unemployed), and mandatory birth control training for mothers with serious disease (choice was not part of the equation).

Another of Sangers initiatives was the Negro Project, in which predominantly black neighborhoods were targeted for birth control programs. Its purpose was evident, as she later disclosed in a private letter in December 1939: We dont want word to go out that we want to exterminate the Negro population.

Planned Parenthood was, of course, one of the main organizations behind the passage of Roe v. Wade in 1973. The U.S. Supreme Court decision claimed that a womans right to an abortion was implicit in the right to privacy protected by the 14th Amendment of the U.S. Constitution. As a selling point, its advocates first claimed that abortions should only be performed in rare instances, such as rape, incest, or to save the mothers life.

Once the federal government funded Planned Parenthoods abortion clinics, abortion advocates incentivized by federal money pushed the boundaries of acceptable abortions from within the first trimester to eventually the evil practice of partial-birth abortions, in which a child being birthed is halted halfway and killed with scissors to the neck. Thus, simple therapeutic abortions have evolved to infanticidethe barbaric killing of children.

It should be noted that Adolf Hitlers Nazis were rightfully accused of committing genocide by killing 6 million Jews during the Holocaust. Similarly, communist China has been accused of committing genocide against 2 million Uyghurs in Xinjiang. However, these numbers pale in comparison to 63 million, which is the number of children aborted in the United States since the passage of Roe v. Wade in 1973. That number includes over 19 million black babies aborted. That is real genocide, whose underpinning philosophy is evil, as eugenics has become infanticide.

The newest effort by the godless camp to meddle with the natural course of humanity is the transhumanism movement, which seeks to accelerate human evolution through advanced technologies. It is a segue from eugenics because it aims to enhance the human species through the addition of advanced biological and physical (mechanical, bio-mechanical) technologies or, as Britannica puts it, to augment or increase human sensory reception, emotive ability, or cognitive capacity as well as radically improve human health and extend human life spans.

In short, the goal is to create super-humans who will live forever. And just like eugenics, there is no natural selection involved, but rather a selective implementation by and for those willing to pay the cost (and to be experimented upon). And there is no limit to the experimentation and no ethical constraints on applying the technologies.

God versus man. Good versus evil. The eternal struggle.

First came eugenics, then came abortion on demand and infanticide, and now there is the new horizon posed by transhumanism. That is the evolution of an evil kind! None of these have moral, ethical, or religious constraintsall are arbitrary as determined by those holding political power.

The Biden administration has cleared the decks for transhumanism through the signing on Sept. 12 of the Executive Order on Advancing Biotechnology and Biomanufacturing Innovation for a Sustainable, Safe, and Secure American Bioeconomy. Into the Brave New World we gowith much trepidation!

A new Policy Review by researchers from the International Agency for Research on Cancer (IARC), the International Atomic Energy Agency (IAEA), the World Health Organization (WHO), and partner institutions presents the evolution of the IAEA, IARC, and WHO joint advisory service to help countries assess national capacities and the readiness of the health system to plan and implement cancer control strategies. These assessments are known as integrated mission of Programme of Action for Cancer Therapy (imPACT) Reviews. The Policy Review was published in The Lancet Oncology.

The researchers describe the methodology of imPACT Reviews and present several country case studies. imPACT Reviews consist of a standardized assessment of the different aspects of cancer control prevention, early detection, diagnosis and treatment, palliation of symptoms, and survivorship as well as cancer surveillance and governance. Each agency is responsible for specific topics; the IARC assessment covers cancer surveillance and early detection.

The joint imPACT Reviews programme supports national health authorities in planning an integrated and specific approach to cancer control and measuring progress in implementation. Since the programme began in 2005, 111 imPACT Reviews have been implemented in 96 countries and more than 800 experts have been deployed.

Bitcoin’s role in the evolution of money with University of Exeter’s Dr. Jack Rogers – CoinGeek

Is Bitcoin just another kind of money? An undergraduate module dedicated to Bitcoin at the University of Exeter sheds light on the question by delving into the history of money. The module, called Bitcoin, Money and Trust, was launched in 2018 after a high demand from students to learn about Bitcoin.

Dr. Jack Rogers, a senior lecturer in economics at the University of Exeter launched the Bitcoin module as a precursor to an MSc Fintech course which he leads. He says participation increased tenfold in recent yearsfrom less than 50 students when it launched to 700 this year.

Despite the impressive turnout, Jack believes that probably the big number is partly driven for the wrong reasonsall the various hype and sense that you could get rich from this.

On this weeks episode of CoinGeek Conversations, Charles Miller talks to Jack about the University of Exeters Bitcoin teaching, the evolution of money and the role Bitcoin plays.

Jack points to the emergence of central banking as a step change in history. He quoted Felix Martin, the author of Money: The Unauthorized Biography, whom he recalls speaking of a compromise power structure between the central bank and the government. For the first time in centuries, a decoupling between money and the state is happening before our eyes, Charles suggests. Jack agrees, saying a new technology that allows people to potentially pay each other without using existing fiat-based systems has indeed raised fundamental questions. He pertains to the authors view on cryptocurrency and how its lead to a disruption in the payment system which central banks have been in control of for a long time.

Jack believes the disruption in central banking was inevitable eventually: I think this stuff, central bank, digital currencies and things that you see now, maybe it was coming anyway I think the emergence of Bitcoin and all the hype and everything has kind of brought that forward.

Based on Jacks comments, its safe to say that the future of money will depend on the outcome of the competition between blockchain-based payment systems. For now, he admits no one is certain as to where Bitcoin is heading.

One of my students did a great dissertation on this, speculating that in 20 years time, will there be loads of different types of money? What does it look like? I mean, no-one can really say, Jack said.

Dr. Jack Rogers is co-authoring a textbook alongside Brendan Lee and Neil Smith. The book will be out by the end of 2023.

Pace began as the in-house advertising department of Heaths Motors, a Ford dealership, and was established in 1964.

Since those foundational years, the Geelong-based agency has grown, developed, and evolved into the modern and diverse company it is today. It is one of the longest-serving agencies in the country that is also family owned and operated.

Account directorEd Jonesspoke toMediaweekabout Paces long-standing partnerships with clients, being part of the family business and the agencys future in sustainability.

While his family ran the agency, Jones shared that his journey to his role wasnt direct. He shared that he initially studied medicine before pursuing aviation and then doing private consultations and marketing strategies in the field.

Then I ended up finding myself enjoying everything here at the agency. Its a very diverse place, and my grandfather started it up.

It was never my plan to go into marketing, advertising, events, or anything like that. But funnily enough, I just stumbled across happiness, which was very unpredicted, he said.

Unlike most, Jones dipped his toe into the media industry growing up.I used to get dragged in here during school holidays because of the family tie-ins. I was brought into the art department or whatever I could be useful in between school holidays, so call it child labour, he joked.Jones shared that he officially began working with Pace 13 years ago temporarily while studying medicine. He said: Before long, I had found myself intertwined with a number of our key accounts, and it was just fun. So, I stuck around.

Pace is led by Jones uncleNicholas Heathwho has been at the agencys helm for over two decades. He noted that during the 1990s, Heath focused on sustainability and long-term planning for the agency.

He (Heath) was trying to get the agency to a point where you didnt have to think where next weeks wages were coming. With retail, automotive and other clients at that time, work was seasonal, Jones explained.

You had four extremely painful peak workloads a year and often some lulls in between, So cash flow was not part of the operation, he added.

Jones said that his uncle examined retainer base models and long-term contracts. He also noted that Heath landed key accounts in sectors such as healthcare, government, and tourism, which gave the agency plenty of continued momentum.

Jones continued: The agency will be 60 years old in 2024 and has just been going through the most excruciatingly painful but most rapid period of growth in the agencys history. Its pretty gun-ho at the moment.

Jones shared that he is often his uncles right-hand man to their team of 24, along with two recent full-time additions and eight regular subcontractors.

Pace has four facets as an agency marketing, advertising and media, events and digital that work collaboratively.

Its probably the most diverse and comprehensive range of services all under one roof. Its done and dusted in there, Jones said.

Jones noted many of their clients go to them with complex problems that require a consolidated team to fix the issue and deliver while working as an integrated team with their organisation.

That broad skill set is super useful for our strengths. It is pretty rare, he said.

Jones noted that over the years, they adopted strategic recruitment to hire people and build a solid team of hardworking and passionate people who bring intelligence, integrity and capability to the team.

Were able to solve very complex things and quite nimbly have small or large organisations. Were finding that many small and large organisations with complex problems need people like us. Its busy, which is a good thing.

Jones shared the agency still has some of its foundation clients to this day, in addition to maintaining long-standing client partnerships, noting their average client tenure is over ten years.

Of course, we have projects and things that come and go at a much shorter timeline than that, but our client relationships last long, he said.

Jones noted the agencys previous work with the likes of Shell, Alcoa, Viva Energy, and Haymes paint, as well as healthcare clients and ASX-listed companies.

Some clients may have reshaped or changed along the way because were talking decades. But we have many clients that have been around a long time, to the point where they have more changeover than we do, and we end up being the basket of knowledge, he said.

Jones continued: There have been some accounts where we have held the baton as theyve gone through change and disruption. So, weve been the one consistent part along the way, which is rare.

When youve got clients you work with that long, you get to know them very personally and professionally. It makes for a good, strong working relationship. You do better work because you care, he added.

Jones remained tight-lipped on Paces recent wins but noted that over the decades, the agency has worked with more than a thousand projects, many of which are a part of the agencys history.

Jones shared that they often re-winning old clients and projects theyve previously worked with. He explained that sometimes such relationships are forced to change.

Jones said: You find that you recross paths, and so thats that to us is a win when you end up working with some of the more sexy, reputable brands. They give us a bit of bragging rights.

Jones added that this is often an opportunity for Pace to reprove ourselves or reimagine things for the brand. He noted: Because its not a brand-new client from scratch, often its rediscovering an existing relationship thats got some sort of history to the place.

Pace did well during the pandemic, according to Jones. But he noted that it was overwhelmingly well, that it almost got hard to keep up.

Were about getting the job done, right to a high standard, collecting the odd award along the way, but its not why we do it.

You adapt. You are whatever is required on the day. When clients have a downturn, or theres some economic decline, he said, referring to COVID and the 2008 GFC.

At the end of the day, we are a value-for-money based agency, and were always a fair price tag and hand on heart, were going to do what we said were going to do.

Jones added: So being reliable and predictable to a degree, but also providing high-quality output and the agencys diversity. We can go wherever the flavour is and adapt as we need as opposed to just being a digital or niche agency.

In an economic downturn, we thrive because we know how to cut money and create opportunity, Jones added.

Sustainability is an area the agency has its eyes set on expanding in as it looks ahead. Jones said: Its not just flavour of the month, but the whole sustainability sector, particularly as everyone hits for net zero. Theres a lot of stuff were doing in that kind of space.

We also do a lot of economic development work and in tourism. Weve taken upon ourselves to provide a bit of leadership in that space, Jones added, highlighting a recent state government event led by Pace on zero-emission vehicles.

Jones noted that the event showcased plenty of technological innovation their existing clients are interested in as they adapt and reshape their footprints.

Jones also noted that the agency has been keeping busy with the demand for content as businesses catch up post-pandemic. He said: We cannot produce enough. At the moment, we think were on top of it and getting asked to do more.

The flavour is shifting from clicks and basic metrics against engagement to the quality of attention were generating and what that means, he said.

Pace re-joined as a member of the IMAA earlier this year at the suggestion of the agencys head of growth,Simon Larcey.

As a member, Jones noted that the agency has enjoyed a range of benefits with the industry body, including industry awareness, staff training and industry representation.

Jones added: Having a network and shoulders to tap on for various resources or requirements or knowledge is always good.

Once upon a time, screens acted as cold separations from the warmth of other people.However, they have now become an integral part of our daily lives that offer us the chance toconnect with people that we otherwise would not have. The arrival of Covid-19 also saw anincrease in the usage of video platforms as educational tools. Children attended school fromtheir living rooms and students completed degrees from their beds.

Language learning especially has benefited from the transition to digital platforms in a waythat other subjects have not. Online video software has provided a platform for people in theUK to access native speakers as far away as Japan. Distance is no longer an object inaccessing native speakers of a learners target language.

Ukrainian startup Preply has wagered a considerable amount of time on the development oftheir own digital platform to teach languages and its paying off, with the companyexperiencing considerable growth.

We spoke with Preplys Campaign Manager, Daniele Saccardi, about how the languagelearning sector has evolved in the past decade and how the pandemic has influenced thetransition to digital learning:

Learning a language used to be all about memorising vocabulary and understandinggrammatical theory. However the world has adapted to a new communicative approach,emphasising the importance of being able to express oneself and share information in thetarget language. Not only is this approach more practical, as it allows the learner to beginspoken practice immediately, it is also beneficial for improving confidence, motivation andmemorisation.

Language learning technology has evolved to adapt to this communicative approach and fitinto an ever shrinking world. Far from the days of Software DVDs that we used in the earlypart of the century, we now have access to a variety of digital tools, such as mobileapplications and video conferencing platforms. The requirement to remain at home duringthe Covid-19 pandemic led to an increase in the usage of these conferencing platforms andopened up opportunities for students despite their geographical locations. Although I believethat we would have begun to rely more on technology in education, the pandemic definitelyacted as a catalyst to trigger this transition sooner.

Apps, such as Memrise and Anki, focus on providing students with key vocabulary by turningmemorisation into a game. These applications make great support tools to complementmore formal learning but can also be used as stand alone platforms to learn the basics.Some of the downfalls of these applications are dehumanisation and a lack of deeperunderstanding due to their surface level approach. Technology can only take us so far beforeassistance from native speakers is needed. Without communicating with native speakers,learners also miss out on learning colloquialisms and culture. What Preply does is bringtogether technological advances with humans, in order to bridge the disconnect between thetwo and to provide a more comprehensive and personal learning experience. Our tutors arewell versed in the merging of these two faces of language learning, allowing them to focuson building a connection with their students and tailoring the teaching to their particularneeds.

The future of language learning technology looks bright and it is worth watching this space

for advances in AI technologies. At the moment, AI is limited to spaced repetition, meaningthat the same word is repeated at regular intervals to improve memorisation. However, thereare endless possibilities to how AI can be integrated into language learning technologies inorder to improve the user experience. One such suggestion that I am aware of is the use ofchatbots that respond to your messages with personalised responses and can even providefeedback on your performance. Although this is convenient and can provide learners withmore confidence to make mistakes without being judged, I still strongly believe thatconversations with native speakers cannot be replaced due to the cultural underpinning oflanguage learning.

This is according to private and public stakeholders from the continent and beyond who gathered at industry eventTransport Evolution Africa Forum & Expoin Durban last week to explore opportunities to connect Africas transport system through sustainable infrastructure.

Celebrating its 10thyear, the trade show has established itself as Africas largest transport event, hosting the regions port, rail and road authorities all under one roof.

TheTransport Evolution Africa Forum & Expo2022was opened by Ms. Fikile Sithole, Deputy Director General: Transportation Services, KZN Provincial Government, and attracted big industry game changers, like Transnet, Export Credit Insurance Corporation of South Africa (ECIC), Briggeman International, Afreximbank and Dube Tradeport. The event welcomed over 2,350 people over the two-day period and included over 100 exhibitors who showcased innovative solutions across the transport industry.

Co-located with The Big 5 Construct KZN, as well as several other events including the Women in Transport Awards and The Transport CEO forum, it successfully gave visitors access to the entire African Transport Infrastructure value chain. In addition, the strategic transport forum tackled African Continental Free Trade Area implementation, infrastructure, investments and cross border policies head on, while the B2B networking platform and exhibition gave solution providers the opportunity to showcase their global innovations and products.

Highlighting the transport sectors contribution of almost one quarter to total global greenhouse gas emissions,Yaa Agyare-Dwomoh, Consultant at Frost & Sullivan Africasaid that in South Africa, over 60% of South Africas 1.2 million vehicles on the road are diesel-powered, with the remaining powered by petrol. To achieve the energy transformation required for decarbonisation, she suggested the sector would need to rely increasingly on renewable energy sources, such as biofuels, natural gas, ethanol, propane, hydrogen and electricity.

Continuing the conversation of coordinating the transformation of the energy and transport sectors was Dr Marcelo Blumenfeld, an Assistant Professor in Future Transport Systems from the University of Birmingham, United Kingdom, and an industrial fellow for Introducing Innovation at the Birmingham Centre for Railroad Research and Education (BCRRE).

While Blumenfeld admitted that many low-income countries may struggle to economically justify the implementation of electric powered railways, he said that alternative options such as battery and hydrogen were readily available, that provide viable and cost-effective ways to decarbonise railways, by upscaling existing assets without the need for fossil fuels. Using a case study carried out by BCRRE in Tanzania as an example, he illustrated how hydrogen-powered trains could reduce carbon emissions by as much as 10,000 tonnes every year.

Jrgen Klopp praised Darwin Nez and Liverpools new-look attack as his team comfortably eclipsed Rangers to strengthen their Champions League qualification hopes at Anfield.

Liverpools victory arrived courtesy of a Trent Alexander-Arnold freekick plus a Mohamed Salah penalty but it was the understanding between a reshaped forward line that enabled Klopps team to dominate Rangers, who remain without a point and a goal on their return to the Champions League stage.

Nez, making his first European start for Liverpool and only his third in total, was denied several times by the veteran Rangers goalkeeper Allan McGregor but impressed throughout. Klopp insisted it is only a matter of time before the Uruguay international is rewarded with his first goal at Anfield.

The Liverpool manager said: The way the boys up front moved together was extremely, extremely good, especially for only one session working on it. And that was a lowintensity session because of their recent workload. We never did it before. You saw how good a striker Darwin is to be constantly in those situations. Everybody saw tonight that this will happen. All good.

I think our setup was the bigger surprise tonight. We surprised ourselves and these situations can then happen. It was difficult to defend. Diogo Jota moved particularly well between the lines. It was a difficult game and occasion for Rangers but we respect them a lot. We dont think we are halfway through. Next week will be different. It was 2-0, a top game, good result, lets go from there.

Alexander-Arnolds goal was a timely reminder of his attacking quality following recent criticism of his defending and the doubt over his England place at the World Cup.

He played a good game, especially defensively, and scored a wonderful goal, Klopp said. Trent didnt have a defensive problem, we had a defensive problem. We were not in the perfect moment so we had to adjust. It was a wonderful goal. Its a long time since he scored a free-kick so I am happy he had that still in his locker.

Rangers manager, Giovanni van Bronckhorst, admitted Liverpool were on a different level and his team have to improve when the teams meet again at Ibrox next week. I think we improved in the second half but the level we face is very high, he said. Dont forget the level of the opposition. Before the game everybody said Liverpool was out of form but you can see the quality. The gap is obvious.

We are competing against one of the best sides in Europe. I think we competed really well. We will learn from this game and we should take those experiences into the next game and the league.

Can Litecoins latest developments make LTC an investor favorite in Q4 – AMBCrypto News

Litecoin [LTC] had a difficult seven days, as the cryptocurrency couldnt perform as per investor expectations. The alt registered a mere 1% seven-day gains. According to CoinMarketCap, LTC at press time, was trading at $53.39 with a market capitalization of more than $3.8 billion.

In an interesting revelation, CRYPTOWZRD, a popular crypto analyst, pointed out that Litecoin was following Bitcoins [BTC]overall sentiment and movements.

The findings of this analysis were evident by looking at both the charts of both the coins as they registered similar growth in the last few days.

For instance, at press time BTCs and LTCs one-hour growth was 0.03% and 0.16% respectively. Nonetheless, the Litecoin ecosystem witnessed a few upgrades and developments. These may have the potential to help the coin move up in the days to come.

Recently, Litecoin developers pushed a new update called MimbleWimble Extension Blocks. According to them, this is the largest upgrade in the LTC blockchain as it will increase LTCs scalability over ten fold. This development looked pretty promising as it increased the blockchains capability considerably.

However, despite the effort, things in the short-term didnt seem to be in LTCs favor as several metrics suggest a possible downfall in the coming days.

For instance, though the Market Value to Realized Value (MVRV) ratio registered a slight uptick, it was still considerably down. Litecoin also didnt manage to gain much traction from the crypto community, which was evident by looking at LTCs social volume as it decreased over the last week.

Earlier this month, Santiments data also revealed that LTC gained the third spot on the list of cryptocurrencies with the most daily active addresses. However, things might have changed a bit over the last few weeks, as LTCs daily active addresses declined as compared to late September. Nonetheless, LTCs development activity had spiked lately, which was a green flag.

Furthermore, data from on-chain intelligence platform Glassnode pointed out that LTCs reserve risk decreased in the last week. This meant that traders could invest in LTC.

LTCs daily chart portrayed an ambiguous picture as a few market indicators supported a price hike, while the rest suggested otherwise. For instance, the Moving Average Convergence Divergence (MACD) displayed a bullish crossover, which might help the coin climb the ladder in the coming days.

The Bollinger Bands (BB) data revealed that LTCs price might soon enter a high volatility zone after registering a weeks-long sideways monument. This may increase the chances of a northbound breakout.

The Relative Strength Index (RSI) and Chaikin Money Flow (CMF) were both near neutral positions, suggesting that the market could head in any direction. However, the Exponential Moving Average (EMA) Ribbon indicated sellers advantage in the market as the 20-day EMA was resting below the 55-day EMA. This may minimize the chances of an uptick in the short-term.

What Is Litecoin Halving – BanklessTimes

Litecoin has now been one of the most popular cryptocurrencies on the market for a very long time. It has a great brand image, along with sustained popularity and a valid use case. Often dubbed as the silver to Bitcoins gold it shares many of the same characteristics as the current no.1 crypto but with some of its own unique features too.

One of the most significant features that these two crypto-assets share is they undergo a process called Halving. Halving can have a dramatic effect on price predictions, miner activity, and the inevitable hype that surrounds a cryptocurrency.

Halving is definitely a concept that everyone wanting to learn about crypto should understand, so we have created this easy-to-follow guide to explain what Litecoin is, how Halving works, and the effects this can have on markets and miners.

Founded in 2011 by former Google engineer and software developer Charlie Lee, Litecoin enjoyed success as one of the most popular Altcoins on the market. After his time at Google Charlie Lee also worked as a director of engineering at Coinbase and was one of the first employees that the company had. His experience in the crypto space then led to the creation of Litecoin with a vision that was very similar to that of Bitcoins.

In fact, Charlie Lee launched Litecoin with the aim to be different from many of the pumps and dumps found on the market. It was unique in the way that no developers received any LTC before its ICO and similarly, there was no pre-mine.

The vision of Litecoin was essentially to be a light version of Bitcoin. If people viewed Bitcoin as virtual gold then Litecoin would be a virtual silver, facilitating cheap and faster transactions than the slower jugunaught that Bitcoin is. Litecoin also differed from BTC in its mining algorithm; instead of the SHA-256 proof-of-work system that would require miners to have specialized hardware, Litecoin would be able to be mined on any regular computer.

This is certainly a valid use-case as even today we see protocols competing to lower transaction costs and increase throughput whilst still keeping the principle of decentralization intact. In tandem with this Litecoin had a capped supply that would be halved every 4 years, just as Bitcoin. This seemed a sure-fire way to protect the value of LTC over time and mirror the deflationary characteristics of Bitcoin.

To understand what halving is we must first touch on mining. Mining is the process of contributing to the decentralized network that supports a protocols ecosystem and receiving financial rewards for doing so. People often dont understand how a network can be decentralized, and offering rewards to miners for providing that physical infrastructure that a network need is exactly how it works.

Litecoin uses a proof-of-work system that essentially means miners must solve complex mathematical equations to create a block. This block is then verified if solved, meaning it has become a verified part of the blockchain it supports and now further blocks can be built on top of it. Creating blocks is costly due to the computer power required, so miners receive rewards for doing so in the form of the native currency - in this case in LTC.

Halving is the deflationary element of the Litecoin ecosystem. It is an inbuilt function within the network that means every four years the rewards miners receive for mining blocks will be cut in half. Of course, this is not ideal for miners but in terms of tokenomics, it helps to reduce the supply of new coins. This essentially increases the value of those LTC that can be mined as now there are half as many available; this can also have a direct impact on the market price of the coin as speculators can see that there is no dilution of value. Essentially, it's a control on the supply.

Halving can have a dramatic impact on price and this is certainly the area most looked at. If the supply of LTC is reduced then usually you will see a correlative effect of an increase in market value. Vise versa, an increase in supply is usually coupled with a decrease in market value. There are also clear patterns to be seen in any halving period with the market price. Usually, miners will increase their workload before the halving period thereby reducing the market value of the asset in question. Following this and the subsequent halving prices will begin to stabilize, and can even dramatically increase under some circumstances.

The first Litecoin halving occurred in August 2015. Originally rewards for miners per block were 50 LTC, this was therefore reduced to 25 LTC. The valuation of LTC slightly rose in the period before the halving, before dropping slightly and then leveling out after the halving had taken place. LTC had previously reached highs of $32 before this first halving but seemed to then level out around about $4 after it.

Within the time between the first and second halving, LTC had significantly changed. What was around a $4 coin at the time of the first halving had now reached far over $250, so the economic environment had changed significantly. August 2019 saw the second halving take place which reduced the miners reward down to 12.5 LTC. Since this event, LTC has somewhat stabilized at its $100 support level having fallen steadily for the past four months.

Litecoins next halving is supposed to take place in August 2023 and will see the rewards for miners drop again by another 50% to 6.25 LTC. The general consensus of the crypto community is that the price of an asset will rise after a halving - but this is somewhat up for debate.

While in theory, a halving should increase the value of an asset this does not seem to always be the case in practice. We have seen with Litecoin that its halving is not necessarily the biggest catalyst in its price change. In fact, after being designed as the silver to Bitcoins gold, it seems that the BTC price is the biggest catalyst for LTC price change.

The price of an asset is determined by two factors: supply and demand.

While halving does clearly have a part to play in this there are too many other contextual factors at play that also could influence this.

Halving is an integral part of the Litecoin ecosystem; it is one of its core tools to regulate the supply of the coin and ensures that it isn't unlimited as is with some fiat. As well as this critical aspect of tokenomics it also acts as a reward system for the people that help provide the infrastructure for the Litecoin network: miners. Halving directly affects miners due to the 50% reduction of their reward for creating blocks but in tandem helps balance the financial ecosystem of the network.

While halving is theoretically supposed to lead to an increase in the value of an asset we have seen this is not historically the case with Litecoin Halving. In fact, it seems that its relationship to BTC is a much more prevalent factor and should be viewed as more meaningful than the 4-yearly halving cycle of Litecoin. We can therefore conclude that the upcoming Litecoin Halving in 2023 is unlikely to dramatically influence the value of LTC.

Can I mine Dogecoin and Litecoin at the same time?

Yes. Dogecoin is mergemined with LTC which means it is possible to mine two coins simultaneously, without requiring any extra computing power. This partnership was made to avoid Doge holders manipulating its market value.

What happens when there are no rewards for miners?

This is one of the most interesting questions in the crypto community. Technically speaking, miners will no longer receive rewards for their services in the form of new supply coins, but instead with transaction fees.

Will the value of Litecoin go up after its halving?

Historically, it looks as though Litecoin Halving does not affect the value of Litecoin to a large degree. Instead, it is its close relationship to BTC that seems to be the most significant factor.

The festival says it will host its biggest event yet when it returns on July 15, 2023.

Headliners are still to be announced for the event, but acts confirmed so far are 90s and 00s favourites Five, N-Trance, Ultrabeat, Flip & Fill, A1 and 911.

Organiser Alex Hutchinson said: As a local team, we are delighted to bring the biggest pop party to Sunderland year after year and 2023 is going to be the biggest year to date. Well be bringing some huge names to Sunderland for the very first time and we know youll be as excited as we are.

New for 2023 is a partnership with the University of Sunderland which will bring an added dimension to the festival.

Earlier this year, an estimated 15,000 people attended the festivals pop and dance day on Saturday, July 9, with Ultrabeat, Vengaboys, Aqua and headliners Steps bringing the summer vibes to Wearside.

The following weekend, the festival hosted a rock and indie weekend featuring UB40, Stiff Little Fingers, Ash and more.

Its set to be a summer of events in Sunderland in 2023.

The US superstar will be getting the party started when she returns to Sunderland on Saturday, June 10 and Sunday, June 11, 2023.
