CRISPR Used to Silence Crucial Hepatitis B Gene – Medscape

The CRISPR gene-editing tool can be used to silence an important hepatitisB virus gene, a proof-of-concept invitro study suggests.

"It's the first time we've seen CRISPR editing done in a hepatitisB model," said Douglas Dieterich, MD, director of the Institute of Liver Medicine and professor of medicine at the Icahn School of Medicine at Mount Sinai in New York City.

HepatitisB can lead to liver disease and is the primary cause of hepatocellular carcinoma. In 2015, more than 250million people around the world were infected with the virus, according to the World Health Organization.

For their study, investigator Hao Zhou, from The First Hospital of Jilin University in China and the Department of Medicine at the University of Minnesota in Minneapolis, and colleagues targeted the Sgene. Zhou presented the findings at the Liver Meeting 2019 in Boston.

The Sgene gives rise to the hepatitisB surface antigen, the presence of which indicates that a person is infected with the virus. "The question is whether it's the right target," Dieterich told Medscape Medical News.

Reducing the amount of the hepatitisB surface antigen is a "good idea" because that's what is believed to inhibit the immune system from clearing the virus. Doing so might help the immune system recover and clear the virus, "with a little help from some antivirals," explained Dieterich, who was not involved in the study.

However, "the surface is not the only DNA that's integrated into the host genome," he pointed out. "I think maybe a broader application might be necessary to actually get the hepatitisB genome out of the hepatocytes."

Zhou's team used a newer CRISPR approach, called CRISPR-STOP, for their gene-editing procedure.

"The idea is that CRISPR-STOP can be as efficient as standard CRISPR editing, but it's safer," said Kiran Musunuru, MD, PhD, associate professor of cardiovascular medicine and genetics at Penn Medicine in Philadelphia, who was not involved in the study. Musunuru is cofounder of and senior scientific advisor at Verve Therapeutics, a company using gene editing to prevent cardiovascular disease.

The standard CRISPR-Cas9 approach requires a double-strand break in the genome, and the problem with that is it introduces the possibility for "mischief," he explained. "If you have more than one double-strand break occurring in the human genome at the same time, you have the potential for different parts of different chromosomes coming together in the wrong ways and then causing problems."

Instead of creating a double-strand break, CRISPR-STOP uses a base editor to chemically modify the DNA base from one base to another and introduce a stop codon into the target gene sequence, effectively hamstringing the ability of the target gene to produce a functional protein.

This is a very nice, clean way to turn off a gene effectively.

For their CRISPR-STOP procedure, Zhou's team first transduced liver cells infected with the hepatitisB virus using a base editor called AncBE4max. Next, to activate the base editor so that gene editing could begin, they transduced the cells with one of two lentivectors: one encoded for single-guide RNA that targets the Sgene; and an empty one, which served as the control.

With the gene-editing approach, 71% of the liver cells that expressed the base editor gained the desired stop codon in the target gene.

"That's a very robust number," said Musunuru.

In addition, hepatitisB surface antigen secretion was reduced by 92% with the gene-editing approach.

The investigators report a high degree of conservativity for hepatitisB genotypesB, C, F, and H. Specifically, 94% of the Sgene sequence was conserved for genotypeB, 92% for genotypeC, 91% for genotypeF, and 71% for genotypeH.

Your hatred of heart-healthy veggies could be genetic – WTVR CBS 6 News

If certain vegetables have always made you gag, you may be more than a picky eater. Instead, you might be what scientists call a super-taster: a person with a genetic predisposition to taste food differently.

Unfortunately, being a super-taster doesnt make everything taste better. In fact, it can do the opposite.

Super-tasters are extremely sensitive to bitterness, a common characteristic of many dark green, leafy veggies such as broccoli, cauliflower, cabbage and Brussels sprouts, to name a few.

The person who has that genetic propensity gets more of the sulfur flavor of, say, Brussels sprouts, especially if theyve been overcooked, said University of Connecticut professor Valerie Duffy, an expert in the study of food taste, preference and consumption.

So that [bitter] vegetable is disliked, and because people generalize, soon all vegetables are disliked, Duffy said. If you ask people, Do you like vegetables? They dont usually say, Oh yeah, I dont like this, but I like these others. People tend to either like vegetables or not.

In fact, people with the bitter gene are 2.6 times more likely to eat fewer vegetables than people who do not have that gene, according to a new study presented Monday at the annual meeting of the American Heart Association.

We wanted to know if genetics affected the ability of people who need to eat heart-healthy foods from eating them, said study author Jennifer Smith, a registered nurse who is a postdoc in cardiovascular science at the University of Kentucky School of Medicine.

While we didnt see results in gene type for sodium, sugar or saturated fat, we did see a difference in vegetables, Smith said, adding that people with the gene tasted a ruin-your-day level of bitterness.

Our sense of taste relies on much more than a gene or two. Receptors on our taste buds are primed to respond to five basic flavors: salty, sweet, sour, bitter and umami, which is a savory flavor created by an amino acid called glutamate (think of mushrooms, soy sauce, broth and aged cheeses).

But its also smelling through the mouth and the touch, texture and temperature of the food, Duffy said. Its very difficult to separate out taste from the rest. So when any of us say the food tastes good, its a composite sensation that were reacting to.

Even our saliva can enter the mix, creating unique ways to experience food.

When we come to the table, we dont perceive the food flavor or the taste of food equally, Duffy said. Some people live in a pastel food world versus others who might live in a more vibrant, neon food world. It could explain some of the differences in our food preference.

While there are more than 25 different taste receptors in our mouth, one in particular has been highly researched: the TAS2R38, which has two variants called AVI and PAV.

About 50% of us inherent one of each, and while we can taste bitter and sweet, we are not especially sensitive to bitter foods.

Another 25% of us are called non-tasters because we received two copies of AVI. Non-tasters arent at all sensitive to bitterness; in fact food might actually be perceived as a bit sweeter.

The last 25% of us have two copies of PAV, which creates the extreme sensitivity to the bitterness some plants develop to keep animals from eating them.

When it comes to bitterness in the veggie family, the worst offenders tend to be cruciferous vegetables, such as broccoli, kale, bok choy, arugula, watercress, collards and cauliflower.

Thats too bad, because they are also full of fiber, low in calories and are nutrient powerhouses. Theyre packed with vitamins A and C and whats called phytonutrients, which are compounds that may help to lower inflammation.

Rejecting cruciferous or any type of vegetable is a problem for the growing waistline and health of America.

As we age as a population, vegetables are very important for helping us maintain our weight, providing all those wonderful nutrients to help us maintain our immune system and lower inflammation to prevent cancer, heart disease and more, Duffy said.

Food scientists are trying to develop ways to reduce the bitterness in veggies, in the hopes we can keep another generation of super-tasters from rejecting vegetables.

Theres been some success. In fact, the Brussels sprouts we eat today are much sweeter than those our parents or grandparents ate. Dutch growers in the 90s searched their seed archives for older, less bitter varieties, then cross-pollinated them with todays higher-yielding varieties.

People who already reject vegetables might try to use various cooking methods that can mask the bitter taste.

Just because somebody carries the two copies of the bitter gene doesnt mean that they cant enjoy vegetables, Duffy said. Cooking techniques such as adding a little fat, a little bit of sweetness, strong flavors like garlic or roasting them in the oven, which brings out natural sweetness, can all enhance the overall flavor or taste of the vegetable and block the bitterness.

Updated Alta Trial Results Support SB-525 Gene Therapy for Hemophilia A – Hemophilia News Today

Updated results from the Alta trial show that a single infusion with the highest dose of SB-525, an investigational gene therapy, yields dose-dependent and durable increases in clotting factor VIII (FVIII). The trial, in adults with severe hemophilia A , found no bleeding episodes up to 24 weeks following the infusion.

That highest dose of SB-525 31013 vector genomes, vg/kilogram, kg led patients to reach normal FVIII activity. Participants no longer needed replacement therapy following a short preventive course post-SB-525-administration.

With these promising results, Pfizer has initiated a lead-in study (NCT03587116) to support SB-525 advancement to a Phase 3 registrational clinical trial. The six-month study will evaluate the current efficacy and safety of preventive replacement therapy in the usual care setting. It is currently recruiting participants worldwide.

The Alta trials most recent findings will be shared at the upcoming 61st Annual Meeting of the American Society of Hematology (ASH), to be held Dec. 7-10 in Orlando, Fla.

Data will be featured in a poster titled Updated Follow-up of the Alta Study, a Phase 1/2, Open Label, Adaptive, Dose-Ranging Study to Assess the Safety and Tolerability of SB-525 Gene Therapy in Adult Patients with Severe Hemophilia A.

SB-525 is a gene therapy candidate to treat hemophilia A thats being developed by Sangamo Therapeutics in collaboration with Pfizer. It consists of a DNA sequence coding for the production of a working FVIII the clotting factor missing in hemophilia A. That FVIII is carried and delivered to liver cells, where clotting factors are produced, using a harmless adeno-associated viral (AAV) vector.

The goal of the therapy is for patients to regain the ability to continuously produce the coagulation factor, and reduce or eliminate the need for FVIII replacement therapy.

The therapys safety and effectiveness for the treatment of adults with severe hemophilia A are currently being evaluated in the open-label Phase 1/2 Alta trial (NCT03061201).

The study is testing a single infusion into the vein (intravenous) of one of four ascending doses of SB-525: 91011 vg/kg; 21012 vg/kg; 11013 vg/kg; and 31013 vg/kg. Two people have been dosed per group, except for the highest dose group, which was expanded to five patients.

Updated trial data now released show the results for the two patients dosed in the third group those given 11013 vg/kg and the five individuals receiving the highest dose of 31013 vg/kg.

In the third group, a single infusion of SB-525 resulted in stable and clinically relevant increases in FVIII activity.

Stronger results were seen with SB-525s highest dose. Of the five patients treated, data were available for four. For these participants, a single infusion with the highest dose of SB-525 led to normal FVIII levels with no bleeding events reported up to 24 weeks post-administration. These individuals no longer needed replacement therapy after the initial prophylactic period of up to about three weeks after SB-525 dosing.

In addition, preliminary tests from the high-dose group indicate similar activity of SB-525-derived FVIII and the clotting factor provided by Xyntha, Pfizers recombinant therapy for hemophilia A.

As to safety, one patient had treatment-related serious adverse events, namely low blood pressure and fever, occurring about six hours after infusion. These effects resolved with treatment within 24 hours, with no loss of FVIII expression.

Some patients also showed elevated blood levels of liver enzymes(ALT, alanine aminotransferase). However, these were reported to be mild and temporary increases, which were treated in a timely manner with corticosteroids.

Dosing in the fourth group is ongoing. At the upcoming meeting, Sangamo will disclose additional analyses of the trial data, including a follow-up of approximately 4 to 11 months after treatment.

The rapid kinetics of Factor VIII expression, durability of response, and the relatively low intra-cohort variability in the context of a complete cessation of bleeding events and elimination of exogenous Factor VIII usage continues to suggest SB-525 is a differentiated hemophilia A gene therapy, Bettina Cockroft, MD, MBA, chief medical officer of Sangamo said in a press release.

UPDATED: Decrying ‘arbitrary and capricious’ action, RegenxBio sues FDA over clinical holds on gene therapy – Endpoints News

When RegenxBio disclosed that the FDA had placed a partial clinical hold on one of its lead gene therapies, execs outlined several customary next steps: continuing assessment and monitoring, delaying a related IND filing, and working with the FDA to address the matter.

As it turned out, they were planning something much less mundane. Two days after announcing the hold in its Q3 update, RegenxBio filed a lawsuit seeking to set it aside, the FDA Law Blog noted.

The lawsuit shed light on the interactions between the biotech and regulators, revealing that there was actually a full clinical hold on the diabetic retinopathy trial in addition to the partial hold on wet age-related macular degeneration and RegenxBio withdrew the IND.

According to the complaint, the company had discussed results from its Phase I/IIa trial with the FDA and was on track to begin the next phase before the end of 2019.

But on October 18, 2019, without notice or explanation, FDA placed RGX-314 on a clinical hold, effectively halting REGENXBIOs development of this potentially life-altering treatment for retinal diseases that are leading causes of adult blindness. Since issuing the clinical hold order, FDA has rebuffed REGENXBIOs repeated attempts to obtain an explanation of the basis for the clinical hold.

Failing to provide advance warning or explanation for the hold, RegenxBio claims, is in violation of the FDAs own regulations. And that led to an arbitrary and capricious final decision, they wrote.

But thats not it.

Since they didnt get a chance to review or rebut the reasons for the hold, which harmed their reputation and property interest in RGX-314, RegenxBio believes the FDA violated the Fifth Amendments Due Process Clause. Furthermore, they charged a particular section of the Food, Drug, and Cosmetic Act represented an unconstitutional vesting of legislative power in the Secretary of Health and Human Services.

Along with the agency, the federal government, HHS Secretary Alex Azar, acting FDA commissioner Brett Giroir (who has taken over from Ned Sharpless while Stephen Hahn goes through the confirmation process), and FDA regulatory project manager Edward Thompson were also named as defendants. Thompson allegedly first notified RegenxBio of the holds citing issues associated with [RGX-314s] delivery systems.

Following multiple exchanges, the FDA apparently told RegenxBio that it would provide a written explanation of the basis for the hold by this Friday, November 15.

Why risk the ire of regulators when an update is due so soon? As a procedural matter to preserve their rights, the company said.

This action was taken on the recommendation from counsel as we continue to work with the FDA to address this matter, and we hope this step will help ensure the FDA will provide RegenxBio with their specific concerns about the unspecified device, it wrote in an email to Endpoints News.

Attorney Deborah Livornese offered this take on the FDA Law Blog:

While it seems unlikely that the merits of the clinical hold will be resolved through the judiciary process in a helpful timeframe, the complaint has likely increased the chances that FDA will respond with a thorough explanation of its reason for the hold when it does provide the written basis.

The biotech added that the plan is still to start the wet AMD trial and file an IND for diabetic retinopathy in Q2 2020, as we believe that there are readily available and suitable alternatives for all of the devices used in our studies.

Comparative genomics, bioinformatics, and biochemical assays enable the pairing of endogenous peptides with orphan GPCRs.

Although there is increasing evidence of the clinical importance of GPCRs that bind to peptide hormones and neuropeptides, there remain more than 100 GPCRs for which the endogenous ligands are unknown. These are referred to as orphan GPCRs (oGPCRs). Foster et al. combined comparative genomics, bioinformatic analysis, and functional assays to pair 17 endogenous peptide ligands with five oGPCRs, as well as identify additional peptides for nine GPCRs that had previously characterized ligands. The authors used comparative genomics to identify peptide precursors and peptides, showing that almost all precursors contained an N-terminal signal peptide, which is required for secretion. Investigation of the characteristic features of class A GPCRs showed distinct structural differences between those that bind to peptide ligands and those that bind to nonpeptide ligands. The authors then mined the entire human proteome to identify peptides with characteristics that made them plausible GPCR ligands. They made a library of synthetic peptide ligands and generated cell lines expressing different oGPCRs that were predicted to be potentially activated by peptides. Last, the authors performed pharmacological and biochemical assays to assess receptor internalization, -arrestin recruitment, and select second messenger generation to identify peptide-receptor pairs that resulted in signaling responses. A number of these peptides and GPCRs have previously been implicated in disorders of the reproductive and nervous systems, among other diseases. Thus, this combinatorial approach has deorphanized five GPCRs, expanded the known peptide-receptor network, and facilitated the future investigation of the roles that these peptides and GPCRs play in physiology and disease.

How Donald Trump Jr. Landed Smack in the Middle of a Right-Wing Civil War – The Daily Beast

Donald Trump Jr. had just started talking about his new book at UCLA on Sunday when someone in the audience began imitating actor Joaquin Phoenixs unsettlingly loud laughter from Joker.

As the cackles filled the auditorium, Trump Jr. looked into the crowd, unsure whether what he was saying was really that funny.

Things only got worse from there.

Audience members began demanding a chance to question Trump Jr. and the events host, young conservative star and Turning Point USA chief Charlie Kirk. Over chants of Q&A!, Trump Jr.s girlfriend, former Fox News host Kimberly Guilfoyle, took the mic to call her boyfriends detractors losers.

I bet you engage in online dating, because youre impressing no one here to get a date in person, Guilfoyle said.

Unsurprisingly, that didnt win over Trump Jr.s critics. The trio of embattled conservative stars eventually left the stage amid shouts of Q&A and America First.

And their fringe-right hecklers, who call themselves Groypers in a nod to an obese alt-right cartoon toad thats like Pepe the Frog but more racist,notched another win in their new battle against mainstream conservatives.

What a HUGE victory today, 22-year-old white nationalist Nick Fuentes wrote on encrypted messaging app Telegram after the event. Cannot be understated what an incredible win we saw at UCLA.

Whether he knew it or not, Trump Jr. had walked straight into a right-wing civil war that pits Fuentes and his allies against more mainstream conservative stars, including Kirk, podcaster Ben Shapiro, and Rep. Dan Crenshaw (R-TX).

Fuentes, who marched in the 2017 white-supremacist rally in Charlottesville and has made remarks denying the Holocaust, has encouraged his far-right fans to confront conservative figures at Q&A sessions across the country since October. Fuentes and other anti-Semitic personalities coach their supporters on what questions to ask and even how to behave at the events, encouraging them to use Donald Trumps America First rhetoric but aim it at questioning legal immigration, the role of LGBT people in the Trump movement, or United States support for Israel.

Ahead of the UCLA event, Fuentes told his fans to avoid questions about Israel, instead focusing on embarrassing Kirk and hurting his relationship with the Trump family.

The name of the game tomorrow is to expose Kirk in front of Don Jr., Fuentes wrote.

The campus Q&A has long been an asset for figures like frequent White House guest Kirk and right-wing comedian Steven Crowder, who stars in a meme featuring him sitting next to a sign urging his political foes change my mind. Compilations of Shapiro shutting down campus liberals are big draws on YouTube, often captioned with the phrase Ben Shapiro Thug Life to underscore his rhetorical skills.

But this fall, Fuentes and his white-nationalist associates have turned the viral potential of the Q&A sessions against mainstream conservatives. Instead of deftly batting away campus leftists, Kirk and his allies are left to struggle to respond to questioners couching racist, homophobic, or anti-Semitic messaging in Trumpism.

Turning Point has dealt with racists in the past, but usually theyre coming from inside the group. In 2017, The New Yorker reported on racist messages sent by a top Turning Point official. In May, the group banned one of its campus leaders after he appeared in a video yelling white power and using racial slurs.

A Turning Point spokesman said in a statement that the Q&A portion of Trump Jr.s speech had been canceled days before the event.

This was a Turning Point USA event and it was our organizations decision to cancel the Q&A portion days before after we were made aware of a pre-planned effort to disrupt the event, the statement reads. The event was not cut short or ended early, as in lieu of the Q&A we simply extended the amount of time given to our VIPs for their remarks.

Trump Jr. wasnt their first target. On Friday, a Turning Point event featuring Crenshaw at Arizona State University was besieged by Groypers, egged on by Charlottesville marcher and alt-right internet video personality Timothy Baked Alaska Gionet.

One Crenshaw antagonist kicked off the Q&A by asking Crenshaw, a Navy veteran, about the 1967 Israeli attack on the USS Liberty that left 34 crew members dead. Inquiries in the United States and Israel found that the attack was an accident, but it has become a popular talking point for Fuentes and his supporters.

What do you call yourselves? Crenshaw responded. The Groppers? Its a very strange name. These guys are the Alt Right 2.0.

As Crenshaw faced increasingly hostile questioning, friendly audience members tried to create a separate question line to get around Crenshaws critics. As the event ended, Gionet and his allies attempted to reach Crenshaw at the stage but were blocked by Turning Point members.

Fuentes feud with Turning Point appears to date back to at least March, when he tried to give a speech at Iowa State University on a purported invite from a Turning Point chapter there. Then, in September, Turning Point fired one of its ambassadors, MAGA personality Ashley St. Clair, after she appeared in a picture smiling with Fuentes and other far-right internet personalities. In late October, Fuentes tried to take a picture with Kirkapparently in an attempt to embarrass himbefore being stopped by security guards.

Fuentes attacks have put some pressure on Turning Point. Since the Groyper war began, two the leaders of two Turning Point chapters have dissolved their groups, citing sympathy for Fuentes or claiming that Turning Point is stifling their free speech.

Last week, Shapiro devoted much of a speech sponsored by another campus group, Young Americas Foundation, to denouncing Fuentes and his cohort. Shapiro highlighted a video in which Fuentes denied the Holocaust using an elaborate metaphor about the Sesame Street character Cookie Monster, as well as a post immediately after the Charlottesville march where Fuentes attacked a rootless transnational elite.

Shapiro also brought up Fuentes attack on Matt Walsh, a columnist at Shapiros Daily Wire, whom Fuentes called a shabbos goy race traitor for attacking white supremacists in the wake of the El Paso shooting.

But maybe he was just being ironic, bro, Shapiro said, mocking Fuentes defenders.

The fight has drawn in a host of conservative personalities on either side. Conspiracy-theory hub Infowars has been generally supportive of Fuentes, describing his targets as grifters from Conservatism, Inc. Columnist Ann Coulter has retweeted messages supporting the attacks on Kirk, while writer Michelle Malkin praised the questioners in a radio appearance and downplayed the racist and anti-Semitic nature of their group.

Its breaking out on college campuses now, Malkin said. There are many young, and very intelligent and sharp nationalists that are challenging Charlie Kirk and some other members of the open-borders lobby in the Republican Party.

Former White House adviser Sebastian Gorka has blasted Fuentes and his fans, calling Fuentes a Holocaust Denying scumbag on Twitter.

First initiated by opposition to an extradition bill passed in March, these protests have begun to draw international support such as the American Congress passing of bills in support of the defense of human rights and liberties in Hong Kong. On June 15, after 13 weeks of protests, Chief Executive of Hong Kong Carrie Lam announced that the government would suspend the bill.

The protests did not end, however. Many believed that the government had not done enough and a broader democractic regime change movement ensued. As of October 29, these protests show no sign of stopping anytime soon. But how has this movement been able to continue with increasing support?

Very early in the movements lifespan, cartoonists quickly began satirizing the extradition bill that Lams government had passed. Popular cartoonists such as Badicuao and Zuni released images depicting all sorts of rhetoric in favor of the protestors.

President Donald Trumps eldest son found himself caught in the middle of an alt-right takeover of a libertarian group with close ties to mainstream conservatism, and video of the encounter provided an embarrassing start to his book tour.

Right-wing activists led by white nationalist Nick Fuentes have been turning up at campus events sponsored by Turning Point USA and other conservative groups to boost their racist, anti-LGBT and anti-Semitic messages, reported The Daily Beast.

More mainstream right-wingers like podcaster Ben Shapiro and Turning Points Charlie Kirk have used campus Q&A events to spread their own messages and more importantly, to set the stage for dramatic confrontations with lefty protesters in hopes of creating a viral moment that further amplifies their conservative views.

Fuentes, a 22-year-old Holocaust denier who marched at Charlottesville, has encouraged his followers who call themselves Groypers, after a cartoon toad similar to the alt-rights Pepe the Frog mascot to reframe Trumps America First rhetoric at these same events in opposition to mainstream conservatism.

Thats where Donald Trump Jr. enters the story.

The names of the women in Liza Birkenmeiers Dr. Rides American Beach House will automatically make you cast your mind back 30 years. Not just the Dr. Ride in the title the physicist Sally Ride who was the first American woman in space in 1983 but the quartet of fictional Missouri ladies who do-si-do around each other and a summer rooftop in St. Louis. Primarily theres Harriet (Kristen Sieh) and Matilda (Erin Markey), best friends who at first seem interchangeable in their mauve waitress uniforms. Their old-fashioned names will remind you of the plucky heroines from books you read when you had scraped knees and bugbites girls who were spies, rebels, avengers, and role models. We dont talk about men on the roof, says Harriet at one point, extremely inaccurately (she has recently finished a shaggy-dog story about a shaggy dude). But everything about the setting high above an unseen street below and the womens joshing, unguarded voices makes it seem like were peeping into a No Boys Allowed treehouse.

Harriet and Matilda are, though, adults, having come as a matched set through Catholic-school upbringing and twin (unused) graduate degrees in poetry. Theyre often entwined with each other, talking close, and its unclear which one is dominant: Is it charismatic loudmouth Matilda or Harriet of the hidden depths? Both refer to male partners, a Luke and an Arthur, who barely figure; Matilda even has a daughter who has diarrhea like a leaking Hot Pocket, she announces while squirming out a window and onto the tar-paper roof. But sick kid or no, Matilda wouldnt miss an installment of the Two Serious Ladies Book Club, her social hour with Harriet, when they can sing A Groovy Kind of Love and drink beer and eat Breyers out of the tub. Since its the night before Rides blastoff, we often hear the male news announcers through a crackly radio, talking about the countdown. Occasionally Harriets landlady, Norma (Susan Blommaert), pops onto the roof with concerns about a dripping air conditioner, and eventually Meg (Marga Gomez) arrives, a new book-club recruit. But theyre satellites to Harriet and Matilda, Matilda and Harriet.

Your queerness Geiger counter should be shorting out by now. Two Serious Ladies is a sapphic classic by Jane Bowles about shucking convention and living as you wish; Sally Ride herself lived privately, coming out of the closet only in her obituary. Shes a lesbian, announces Meg confidently, recognizing a sister, though the others still believe what theyve read in Time magazine. Meg is the one who actually asks about the Harriet and Matildas relationship, who nudges them about sex, who can see that the women are thrashing emotionally together like fish in a net. God knows Markeys raspy drawl could generate heat with a standing lamp, and Sieh has a way of seeming very rigid while also swaying, barely, towards the person speaking.

Birkenmeier and the director, Katie Brook, have built the show as a slow burn the sun dies in the blue-painted sky (Kimie Nishikawa designed the set), and a trick of Ben Williamss sound and Oona Curleys light make the place seem hot and sticky and exactly midwestern. The women do start to melt into each other, but its not some straightforward romance: Birkenmeier and Brook are interested in all the nuances of liking and knowing and needing. Theres the ebb and flow of realistic talk, a bit of light binocular voyeurism, and Harriet relays some stories from her recent trip to Florida. The stories kind of wander into each other Harriet often fades off at the end of sentence but the parable is clear: Shes learned at last to ask for what she wants. Then, after 80 luscious minutes of conversation, the company somehow has material for a whole nother play, a ten-minute scorcher between Sally Ride (Sieh) and an old friend Molly (Markey). NASAs phallic rocket waiting on its launchpad is forgotten; the women stare into each others eyes. The play itself just keeps soaring up and up and up.

Im still working out whats happening in Dr. Ride, because despite its casual humor and dizzying erotic atmosphere, theres something clean-lined and literary under it. Everythings been so carefully considered that it plays out like a formal court dance: each gesture mirrored, every movement countered by a partner. Even the way the women talk about poetry calls your attention to how landlady Norma talks in syncopated rhythms, pausing as if her thoughts contained the long dashes and caesuras out of Dickinson. In fact, Markey, Blommaert, and Gomez all speak with a kind of metered, emphatic artificiality theyre real and funny, but theyre deliberately letting us hear their line breaks as line breaks. Sieh, on the other hand, seems to be having all these thoughts for the first time. Ive been watching Kristen Sieh for more than a dozen years, and I keep thinking she cant surprise me again. I already know walking in that shes great. But then shell go and be the best Viola in Twelfth Night that Ive ever seen, or shell slay me playing Teddy Roosevelt in a big old mustache. Here shes at her most pure and radiant, a narrow, determined candle flame. I think I must have looked too long at her face in the blue light of the final scene, because its been stamped on my retina like it was burned there.

If your theater appetite hasnt been slaked by 90 minutes of beers and Breyers with Dr. Ride, then Id recommend the Radiohole show Now Serving, back at Collapsible Hole in the West Village. I saw this short production in January, when it was swamped by the several experimental theater festivals that fight it out in the first weeks of the year, but now theres nothing of comparative weirdness to steal its thunder. Radiohole, to be honest, doesnt have a lot of competition in its genre: elegantly repulsive gorgeosity, something only avant-garde art vampires could come up with.

In Now Serving, a perverted banquet with live violin accompaniment, the Radiohole gang invites the audience (or part of it) to dine with them. Theres an elaborate intake situation, in which three women in severe 1940s hats greet the theatergoers at teller windows. Eventually, were ushered to our seats or to the table, a Twin Peaks orgy of black and red, where participants decant wine from IV bags and nod politely as the ladies (Amanda Bender, Maggie Hoffman, and Erin Douglass) talk Dadaistically about the frustrations of the patriarchy. Theres a man in a Pepe the Frog costume (Eric Dyer) who is kind of a servant? Or a sex toy? Or a sacrifice? Or a meatloaf? I remember some upsetting stuff around arugula, and Kristin Worrall go-to chef for downtown experimenters presents desserts that are meant to taste like mascarpone and clitoris. The dialogue frequently lurches into body horror: Nobody else in the world knows how to kill the long pig is the sort of line that keeps you up at night. (Long pig is supposedly what Pacific Islanders called human flesh.) And why do they keep describing the food as consensual?

I admit I didnt have the stones to sit at the table in January, but Ive worked up my courage and Id do it now. Its a chance to be absorbed into a Peter Greenaway film, into a Terry Gilliam landscape, into the weird fantasies of dreamers from David Lynchs Black Lodge. If youre a Radiohole virgin, this would be the right show to take your first steps with, since its swift and service-oriented and willing to feed you. The danger is, the tastes addictive. Im hoping to go back, and I dont usually see things twice. So how do you kill the long pig? Shouldnt I face the arugula and find out?

Dr. Rides American Beach House is at Ars Nova at Greenwich House through November 23.Now Serving is at the Collapsible Hole through November 16.

When Bob Lazar, the original Area 51 whistleblower, emailed me late last month to tell me his thoughts on the Storm Area 51 meme, he wrote that he was glad my information has reached so many people.

The meme he was talking about grew out of a mock Facebook event called Storm Area 51, They Cant Stop All of Us, which jokingly proposed that people ambush a military base in Nevada to find the aliens the government was supposedly hiding there.

The event spawned countless alien rescue memes, whose levity, Lazar thought, sent the wrong message. The meme absolutely cheapens a serious matter, he wrote. I liken it to the Weather Service giving local residents a warning about a dangerous hurricane approaching, and the residents responding with a Woo-hoohurricane party! There are secrets in the Nevada desert, and its a crime to keep the truth from the public.

Sharing the full story, not just the headlines

Lazar, 60, is known for saying that he had reverse-engineered alien spacecraft at a classified military base in the Nevada desert. He first delivered this information during a 1989 interview with George Knapp, a reporter for KlasLas Vegas, under the pseudonym Dennis.

In late June, Lazar was on Joe Rogans podcast, and the interview inspired Matty Roberts, a 21-year-old petroleum engineering student, to create the mock Facebook event.

In addition to the memes, it spurred non-digital activity: three separate, ET-centric events in Nevada between 19 and 22 September: an EDM concert in downtown Las Vegas thrown by Roberts; a Burning Man-like event in the approximately 35-person town of Rachel hosted by the towns only business, the Little AleInn; and a gathering of UFO enthusiasts at the Alien Research Centre, a glorified gift shop in Hiko.

Since then, other Storm copycat events have emerged, from an unserious proposal to Storm Loch Ness to find the notorious cryptid, to the possibly real protest Storm BlizzCon 2019, They Cant Censor Us All, targeting video game company Activision Blizzards censorship of a gamer who spoke in support of Hong Kong protesters.

The event that started it all (Facebook)

There was even a Halloween stoop devoted to the whole shebang in the Fort Greene neighbourhood of Brooklyn. Its 2019, and no one can stop the spread of an internet joke gone too far.

After all, were living in the era of Pizzagate, a time when outrageous Twitter claims about a child enslavement ring involving Hillary Clinton inspired a man to fire a gun into an unsuspecting pizza parlour in Washington, DC.

Politicians and journalists repeat and retweet hoax news stories on the regular, like the one about Syrian refugees being relocated to Navajo reservations, repeated by Sean Hannity and Donald Trump.

Yet you can hardly blame people for taking digital jokes seriously, a practice codified in 2005, when someone with the username Nathan Poe posted to a Christian forum: Without a winking smiley or other blatant display of humour, it is utterly impossible to parody a Creationist in such a way that someone wont mistake for the genuine article.

Washington: Researchers used nanoparticles to identify the presence of deadly microbes present on medical devices, like catheters, and make them infection-free. This study was conducted as an interdisciplinary collaboration between microbiologists, immunologists, and engineers led by Dr Simon Corrie from Monash University`s Department of Chemical Engineering and Professor Ana Traven from the Monash Biomedicine Discovery Institute (BDI).

It was recently published in the American Chemical Society journal - ACS Applied Interfaces and Material. Candida albicans, a commonly found microbe, can turn deadly when it colonises on devices such as catheters implanted in the human body. While commonly found in healthy people, this microbe can become a serious problem for those who are seriously ill or immune-suppressed.

The microbe forms a biofilm when it colonises using, for example, a catheter as a source of infection. It then spreads into the bloodstream to infect internal organs."The mortality rate in some patient populations can be as high as 30 to 40 per cent even if you treat people. When it colonises, it`s highly resistant to anti-fungal treatments," Professor Traven said.

"The idea is that if you can diagnose this infection early, then you can have a much bigger chance of treating it successfully with current anti-fungal drugs and stopping a full-blown systemic infection, but our current diagnostic methods are lacking. A biosensor to detect early stages of colonisation would be highly beneficial," added Professor Traven.

The researchers investigated the effects of organosilica nanoparticles of different sizes, concentrations and surface coatings to see whether and how they interacted with both C. Albicans and with immune cells in the blood. They found that the nanoparticles bound to fungal cells, but were non-toxic to them."They don`t kill the microbe, but we can make an anti-fungal particle by binding them to a known anti-fungal drug," Professor Traven said.

The researchers also demonstrated that the particles associated with neutrophils - human white blood cells - in a similar way as they did with C. Albicans, remaining noncytotoxic towards them."We`ve identified that these nanoparticles, and by inference a number of different types of nanoparticles, can be made to be interactive with cells of interest," Dr Corrie said. "We can actually change the surface properties by attaching different things; thereby we can really change the interactions they have with these cells - that`s quite significant," added Dr Corrie.

Dr Corrie said while nanoparticles were being investigated in the treatment of cancer, the use of nanoparticle-based technologies in infectious diseases lags behind the cancer nanomedicine field, despite the great potential for new treatments and diagnostics.

Bricks of ‘very pure’ cocaine keep washing up on French beaches – New York Post

It takes a lot to shock on a French beach, where topless sunbathing is the norm.

But giant bricks of cocaine littering the beaches have managed to raise a few eyebrows.

Packages of very pure cocaine and other drugs have been washing up on Frances southwestern shores over the past several weeks, the Guardian reports. The drug parcels wrapped in clear or black plastic and labeled diamante or brillante have been plaguing a 310-mile or so strip of the countrys Atlantic coast, stretching from the regions Loire-Atlantique (which includes Nantes) to Landes (near the Spanish border).

New bundles are appearing daily. So far, about 1,675 pounds of the dangerous substances valued in excess of $66 million have surfaced on French shores. On Sunday alone, 30 pounds of drug packages were found in Pornic, a beach town about an hours drive from Nantes.

French authorities believe the drugs very probably originated in South America, and are investigating the possibility that a drug traffickers boat sank in the ocean.

In the meantime, French police are urging beachgoers not to touch any of the packages, and to report them immediately to the authorities.

Its a very pure product that must not be consumed in this form because there is a very high risk of overdose, Philippe Astruc, a public prosecutor from the French town of Rennes, told France 2. There is absolutely an immediate health risk.

Sea of devotees throng beaches – The Hindu

Over one lakh devotees arrived at the three beaches along Machilipatnams coast in Krishna district on Tuesday morning to take a holy dip on the occasion of Kartika Pournami.

The devotees started arriving at the Manginapudi, Pedapatnam and Hamsaladeevi beaches from across the State on Monday night itself.

A large number of swimmers and a battalion of the National Disaster Response Force (NDRF) were deployed along the beaches to prevent untoward incidents.

Nearly 500 policemen and 60 women police personnel were also posted along the beaches to keep an eye on devotees and dissuade them from wading into deep waters while taking the holy dip.

District authorities deployed 120 swimmers at the Manginapudi beach alone while some swimmers were deployed on the 17 boats that patrolled the area.

At least 50 special bus services are being run between Machilipatnam and the Manginapudi and Pedapatnam beaches while special autorickshaws were provided for physically-challenged devotees and senior citizens.

Krishna Assistant Superintendent of Police (ASP) M. Sattibabu said that there were no reports of untoward incidents from any of the three beaches that were thrown open to the devotees. Milk has been arranged for children while changing rooms have been provided for the convenience of devotees, Mr. Sattibabu said.

By evening, at least 1.50 lakh devotees are expected to take the holy dip on the Machilipatnam coast.

Trump Administration Makes It Easier to Dredge Protected Areas to Restore Beaches – The New York Times

WASHINGTON The Trump administration changed a 25-year-old policy to make it easier for coastal communities to take sand from protected ecosystems to improve their beaches.

The shift makes it cheaper for some of the wealthiest communities in the country to replenish their beachfronts, which are increasingly under threat from more frequent and intense storms, rising seas and other effects of climate change. Critics say that comes at the expense of vulnerable coastal ecosystems.

Undeveloped coastal islands and beaches will now be opened up to sand mining that will imperil birds and other wildlife, destroy important habitat and reduce the protections these places provide against impacts of storms and erosion, said Karen Hyun, vice president for coastal conservation at the National Audubon Society, in a statement.

In 1982, Congress established the Coastal Barrier Resources System, which protects 1.4 million acres of land around the country from development. The Interior Department has long said that the law prohibited using federal money to remove sand from those zones to replenish beaches elsewhere.

In a letter this week to Jeff Van Drew, a New Jersey congressman, Interior Secretary David Bernhardt said he had reversed that position. Last month, Representative Van Drew met with the secretary to request the change.

This flawed interpretation of the law has prevented a number of coastal storm damage reduction projects, Secretary Bernhardt wrote this week to Representative Van Drew, who represents communities in Southern New Jersey that stand to benefit from the new policy.

The Trump Administration is committed to protecting our coastlines and utilizing our available resources to restore, enhance or stabilize our beaches consistent with the law Congress wrote, Secretary Bernhardt said in a statement released by Representative Van Drews office this week.

Late last month, Representative Van Drew was one of two Democrats to vote against opening an impeachment inquiry into President Donald Trump.

A spokeswoman for the Interior Department, Melissa Brown, said in a statement that There is absolutely no connection whatsoever between the vote and the letter from Mr. Bernhardt.

The communications director for Representative Van Drew, Mackenzie Lucas, wrote in an email, These things have nothing to do with one another. Noting that Representative Van Drew has long opposed impeachment, the spokesperson wrote, it is absurd to think that the Congressman would trade his vote for some sand.

In his news release this week, Representative Van Drew said Mr. Bernhardts decision proves that we can still come together in a bipartisan fashion and seek common-sense solutions to improve the lives of Americans.

Beyond the threat posed to protected coastal zones, critics said the policy shift would also encourage more property development in beach towns at risk of flooding, or even becoming uninhabitable in the long term, because of climate change.

They say the cost of that development will fall on taxpayers, who fund not just the beach renourishment projects but also the programs required to support those communities, such as federal flood insurance and disaster recovery programs. In many cases, beach nourishment is subsidizing the most vulnerable and exposed property in the United States, said Robert S. Young, director of the Program for the Study of Developed Shorelines at Western Carolina University.

For the 2nd day in a row, some Hawaii beaches are closed because of shark sightings – Q13 News Seattle

Beaches on the Hawaiian island of Maui were closed for the second straight day Tuesday after several tiger sharks were spotted in the water.

Maui County officials closed about two miles of shoreline, which includes the Kamaole Beach Parks.

They saidin a Facebook postTuesday one of the tiger sharks was 10-15 feet in length and was 300 yards off the beach, while another one, about 10 feet in length, was about 100 yards off the beach.

Authorities posted signs directing people to stay out of the water because of the sightings, the county said, and the Maui Fire Department was assessing the safety of the waters throughout the day Tuesday.

The beaches were closed Monday after three to five tiger sharks were spotted. A helicopter crew communicated with people still in the water to tell them to return to shore.

Tiger sharks are among the top three species of shark frequently involved in shark attacks, and they should be treated with extreme caution when encountered, according to theFlorida Museum of Natural History.

There are no reports of shark-related injuries on this stretch of coastline from the past two days, Maui County said.

In April, a65-year-oldwoman was attacked by a shark off Hawaiis Big Island.

A month later, a 65-year-old man, optometrist Dr. Thomas Smiley, was killed while swimming off Mauis Kaanapali Beach, about 30 miles up the coast from Kamaole, where this weeks sharks were seen.

A 16-year-old boy, Max Keliikipi,survived an encounteroff Oahu in August after a shark took a bite out of his surfboard.

At the time, Keliikipi told CNN he didnt have any plans to quit venturing into the waves, and he said knowing surfers whove been attacked by sharks was not common, but its not uncommon, either.
