Unparalleled inventory of the human gut ecosystem – Health Europa

An international team of scientists has collated all known bacterial genomes from the human gut microbiome into a single large database. Their work, published in Nature Biotechnology, will allow researchers to explore the links between bacterial genes and proteins, and their effects on human health.

This project was led by EMBLs European Bioinformatics Institute (EMBL-EBI) and included collaborators from the Wellcome Sanger Institute, the University of Trento, the Gladstone Institutes, and the US Department of Energy Joint Genome Institute.

Bacteria coat the human body, inside and out. They produce proteins that affect our digestion, our health, and our susceptibility to diseases. They are so prevalent that the body is estimated to contain more cells in its microbiome the bacteria, fungi, and other microbes than it has human cells.

To understand the role that bacterial species play in human biology, scientists usually isolate and culture them in the lab before they sequence their DNA. However, many bacteria thrive in conditions that are not yet reproducible in a laboratory setting.

To obtain information on such species, researchers take another approach: they collect a single sample from the environment in this case, the human gut and sequence the DNA from the whole sample. They then use computational methods to reconstruct the individual genomes of thousands of species from that single sample. This method, called metagenomics, offers a powerful alternative to isolating and sequencing the DNA of individual species.

Last year, three independent teams, including ours, reconstructed thousands of gut microbiome genomes. The big questions were whether these teams had comparable results, and whether we could pool them into a comprehensive inventory, says Rob Finn, Team Leader at EMBL-EBI.

The scientists have now compiled 200,000 genomes and 170 million protein sequences from more than 4600 bacterial species in the human gut. Their new databases, the Unified Human Gastrointestinal Genome collection, and the Unified Gastrointestinal Protein catalogue, reveal the tremendous diversity in our guts and pave the way for further microbiome research.

This immense catalogue is a landmark in microbiome research, and will be an invaluable resource for scientists to start studying and hopefully understanding the role of each bacterial species in the human gut ecosystem, explains Nicola Segata, Principal Investigator at the University of Trento.

The project revealed that more than 70% of the detected bacterial species had never been cultured in the lab their activity in the body remains unknown. The largest group of bacteria that falls into that category is the Comantemales, an order of gut bacteria first described in 2019 in a study led by the Bork Group at EMBL Heidelberg.

It was a real surprise to see how widespread the Comantemales are. This highlights how little we know about the bacteria in our gut, explains Alexandre Almeida, EMBL-EBI/Sanger Postdoctoral Fellow in the Finn Team. We hope our catalogue will help bioinformaticians and microbiologists bridge that knowledge gap in the coming years.

A freely accessible data resource

All the data collected in the Unified Human Gastrointestinal Genome collection and the Unified Human Gastrointestinal Protein catalogue are freely available in MGnify, an EMBL-EBI online resource that allows scientists to analyse their microbial genomic data and make comparisons with existing datasets.

The project already has a number of users in the scientific community. As new datasets emerge from research teams around the world, the catalogue might expand to include the microbiomes of other body parts, like the skin or inside the mouth.

This catalogue provides a very rich source of information for microbiologists and clinicians. However, we will likely discover many more novel bacterial species in under-represented geographical areas like South America, Asia, and Africa. We still dont know much about the variation in bacterial diversity across different human populations, explains Almeida.

Lahontan Cutthroat Trout thrive at Paiute’s Summit Lake in far northern Nevada – Nevada Today

Summit Lake in remote northwest Nevada is home to the only self-sustaining, robust, lake population of Lahontan Cutthroat Trout, North Americas largest freshwater native trout species. Research to understand the reasons why this population continues to thrive, where others have not, will be used to protect the fish and its habitat as well as to apply the knowledge to help restore other Nevada lakes that once had bountiful numbers of the iconic fish that historically reached 60 pounds.

A team of researchers from the University of Nevada, Reno and the Summit Lake Paiute Tribe has been studying the watershed ecosystem and recently authored two papers published in scientific journals describing their findings about the relatively small desert terminal lake.

This project is part of a 9-year collaboration to conserve habitats and promote a healthy ecosystem for the lake. University researchers Sudeep Chandra and Zeb Hogan as well as students from their aquatics ecosystems lab and Global Water Center work with the tribes Natural Resources Department, formerly led by fish biologist William Cowan before he retired from the U.S. Fish and Wildlife Service.

An objective to implement holistic management at Summit Lake is to blend science with traditional knowledge to protect and conserve natural ecologic processes, species diversity and tribal cultural practices, Cowan said. The partnership with the Global Water Center, as well as many other researchers, agencies, and organizations has complemented this objective by implementing science-based research and technological advances to investigate the viability of trout in the Summit Lake watershed.

Monitoring data, including climate, hydrology, fish and wildlife population trends and habitat integrity, is used to develop, revise or validate the tribes management plans and regulations. This approach is a stark contrast to when the lake ecosystem and associated resources were at risk of irreversible impacts caused by non-point source pollution, irrigation diversions, livestock grazing, and the unknown affects caused by exporting trout eggs for establishment or supplementation of other populations.

Our team at the University wants to support the efforts initiated by the Summit Lake Tribe," Chandra said. Our goals are to assist them in developing their science-based program to protect Nevada's only strong, self sustaining lake population of Lahontan Cutthroat Trout. We believe that investigations in this robust ecosystem like Summit, where there is little human impact, could improve recovery efforts in other lake systems that are less fortunate and that have lost their trout like the Walker and Tahoe. Surprisingly there are still few comparative investigations of these lake ecosystems and how they could support trout during a time for increasing global changes.

The Lahontan Cutthroat Trout, with its crimson red-orange slash marks on the throat under the jaw and black spots scattered over steel gray to olive green scales, is Nevada's state fish and holds a cultural significance to the Summit Lake Paiute Tribe while providing the tribe with bountiful food and fish resources.

As an important traditional food source, Lahontan Cutthroat Trout composed a large part of Tribal members diets and were the focus of many gatherings held to honor the fish and to learn oral history, traditional practices, and cultural resources from elders of the tribe.

The tribe has exercised their sovereignty to protect, manage and enhance tribal homelands, including the lake ecosystem and associated resources by working with federal agencies and other organizations that enable the tribe to holistically manage and protect the land, water and resources that fish, wildlife and tribal members depend on for survival, Cowan said.

The lake is about one square mile of surface area, has a mean depth of 20 feet with the southern end generally deeper with about 50 feet of depth at the deepest. The lake elevation decreased about 13 feet during the severe drought in the western United States that lasted from 2012 to 2016.

One thing we learned is that the climatically induced drought can change the hydrology, or flow of water and connections of stream to lake, but even with these changes, the trout populations remain relatively stable in the lake, Chandra said. They look for the opportunity to spawn every year and likely wait for better conditions with higher flows for better access to upstream spawning grounds.

So it is critical to support the tribes efforts to protect the watershed and understand how the long term changes in water resources, like the flow of water, will change with pending climate change projections for the Great Basin.

James Simmons, doctoral student with the Ecology, Evolution and Conservation Biology program at the University said the population appears resilient to today's climate disturbances/drought, which is very positive, but should the frequency and severity of drought increase in the future, will the population remain resilient in the face of continued low abundance, survival, spawners and a skewed sex ratio.

I think the key going forward will be for the tribe to try to understand how the long-term flow of water in the watershed will be impacted by the future changing climate in the Great Basin - so that the tribe can formulate a game plan to get ahead of any potential negative repercussions, he said. Like cutthroat populations across the western U.S., this population faces unknown impacts from climate change.

Declining abundance and diverging male and female abundance under changing drought cycles and conditions may have negative long term consequences. The prediction of increased frequency, severity and duration of drought and an increased percentage of rain may decrease abundance, reduce the effective population size and skew the sex ratio at Summit Lake.

The research team found that connections between the upper watershed and the lake are essential for maintaining a healthy population during a drought. During the drought of 2012-2016, Summit Lake had a strong, stable population of naturally reproducing Lahontan Cutthroat Trout. The numbers of trout spawning up Mahogany Creek, one of the lakes only inflow streams, was also relatively stable in number. Some of the trout in the lake migrated all the way to the upper watershed, about eight miles.

Lahontan Cutthroat Trout can live in streams and lakes, Chandra said.The trout that live in lakes need rivers to spawn to keep their populations healthy. The numbers do show with little to no major changes to the watershed by human development, there is still a highly variable amount of spawning from lake to stream.

Adequate stream flow is necessary for spawning and movement to the lake-dwelling component of the population. In rivers where flow is regulated, enough flow must be preserved in the spring to allow lake spawners to come upstream and in the fall to allow juveniles to migrate to the lake.

Healthy habitat and ecological connectivity between habitats, such as no man-made migration barriers and adequate stream flow, should be preserved throughout as much of the watershed as possible (and of course between the stream and the lake) to facilitate movement for both stream- and lake-dwelling fish, and to support a robust overall population, Teresa Campbell, a biologist and staff researcher in the Universitys Global Water Center and lead author of one of the scientific papers, said.

Strong connectivity between healthy stream and lake habitats is crucially important to the long-term survival of the Summit Lake Lahontan Cutthroat Trout because it seems that the exchange of individual fish across habitats contributes to the resilience and vitality of the population as a whole.

The study also found that in drought-prone systems, streams should have adequate pool habitat and cover such as trees and woody debris to provide a refuge area from the drought and cooler temperatures for trout.

During the drought, in the stream, these refuge pools with structure in the form of wood, cobbles, or boulders supported higher densities of stream-dwelling trout, Campbell said. Therefore, this habitat type is an important component of healthy stream habitat for trout.

The lake and surface water on the Reservation are further protected by restricting public access and monitoring resources necessary to sustain endemic species diversity in the area, Cowan said.

The Summit Lake Paiute Tribe Reservation is the most remote Native American reservation in Nevada. Located in the northwest corner of Nevada, the reservation is 50 miles south of the Oregon border and 70 miles east of the California border.

Apple to Updating Developer Ecosystem Language to Make it More Inclusive – The Mac Observer

Apple has announced it is going to update the terminology it uses within its developer ecosystem. It updated its style guide as part of this effort to make it more inclusive.

A news update on the developer website said:

At Apple, were working to remove and replace non-inclusive language across our developer ecosystem, including within Xcode, platform APIs, documentation, and open source projects. These changes began on June 22 with the beta software and developer documentation released at WWDC20 moving to terms such as allow list and deny list, and main as the default SCM branch in Xcode 12. An updated Apple Style Guide reflects these and other changes.

Furthermore, it said that Developer APIs with exclusionary terms will be deprecated as we introduce replacements across internal codebases, public APIs, and open source projects, such as WebKit and Swift. The note added that developers were encouraged to closely monitor deprecation warnings across your codebases and to proactively move to the latest APIs available in the platform SDKs.

The updated style guide contains a number of changes. These include Apple TV+ and the instruction to not use the phrase whitelist. Entries for dock adapter and Hold switch have been removed.

With ‘Another Idea,’ Gray Center steps into virtual programming during the pandemic – Hyde Park Herald

When the COVID-19 pandemic began to seriously affect everyday life earlier this year, museums and galleries that were forced to close responded by moving their collections online, creating viewing rooms for digital visitors. (In Hyde Park, the Museum of Science and Industry launched a website with science resources and activities for children to try at home.)

But several arts spaces have also taken up a different challenge, aiming to show work that responds to life under lockdown, in which our ordinary interactions with one another have become overwhelmingly virtual. Thats the aim of Another Idea, an exhibition at the University of Chicagos Gray Center for Arts and Inquiry thats been running since June 1.

The Gray Center doesnt usually put together large exhibitions its more of a forum for artists and academics to discuss and engage with one anothers work, with public programming like Gray Sound, an experimental music series started in 2019. But Another Idea, which closes at the end of July, was organized in response to the sudden shift that took place with the onset of the pandemic.

It came out of some observations on our part, and some conversations with UChicago Arts, thinking about how to approach moving things online, said Mike Schuh, assistant director of fellowships and operations at the Gray Center, who organized the exhibit with curator Zachary Cahill. Theres a lot of artists working today and going back throughout the 20th century whose work is malleable enough that it's well-suited for a web-based exhibition.

The short introductory paragraph to Another Idea notes that its artworks are inherently ephemeral. That may seem surprising for those who worry about the permanence of information online but as the writer and critic Kate Wagner recently pointed out, the disappearance of old websites, especially popular ones, often leads to a significant loss of data, as if a smallish Library of Alexandria has been burned to the ground.

On a smaller scale, Devin T. Mayss Taxidermy illustrates this point: the page for Mays on the Gray Center website currently displays eight links to expired or deleted Craigslist postings. The hint to their contents comes in the text of the now-defunct URL, which are all some variant on chicago-used-usa-flag-free-delivery. (By the time this has gone to press, of course, it is possible that a ninth, extant link has appeared, in which case a reader of the Herald may soon find themselves the owner of an American flag.)

As the persistence of Mayss hyperlinks show, even something that disappears can leave a trace thats true for other works in Another Idea, too, albeit in less tangible ways. Take the two pieces by British artist Liam Gillick, which instruct the viewer to perform some task, such as: Using a pipe and cable detector locate all the cables and metalwork hidden below the surface of a chosen wall. Loosely mark their location using a yellow pencil.

He doesnt really care how much time you spend with a work in a place. He's much more interested in its lingering effects maybe two weeks later, you stumble across something in your life. The work comes back to haunt you that way, said Schuh. It's really intended to to generate a residue from the experience that isn't so much about a kind of specific time-and-space engagement.

As with Gillicks piece, the mood of engagement is often collaborative, encouraging the visitor to the show to participate somehow in it. Zarouhie Abdalians work, for instance, presents a series of seven etudes, described as prose scores for any willing performers.

The most recent focuses on BlackRock, the international investment company that, according to its own website, manages $6.47 trillion in assets. Abdalian directs willing performers to research the companys investments and contrast it with the social good that might be done if that money were used to hire workers at the wage of $15 an hour. Outside the location of the BlackRock office nearest to you, make a public demonstration of your findings, it concludes.

One of the earlier etudes, State Portrait, has already been performed by another artist, Dena Beard. In Beards video, which is posted on the Gray Center website, she produces documentation from a protest in Oakland, Cal., on June 3. At one point, Beards camera, tilted upwards at the tops of tall buildings, pans around an intersection as a speaker tells a crowd, We cant sit back and watch our babies, our men, our peoples bodies lying lifelessly in the street at the hands of the police.

This connection to current events is on display, too, in Cauleen Smiths COVID Manifesto, which consists of a series of messages written on yellow lined paper, with a new piece at the beginning of each week. Over the course of the exhibition, the texts transition from reflections on life under lockdown (I do love seeing my students faces on the Zoom) to exhortations in favor of prison abolition (Everybody everybody out of jail NOW).

Some of the works were originally conceived for physical shows, and reworked for an online presentation. Food Situation for a Patriotic Banquet, an installation by the Spanish artist Antoni Miralda, consists of a table laid out with eight trays of cooked rice, colored and arranged to resemble the flags of power countries in the 1970s, including Germany, China, the United States, and Switzerland. The rice gradually decomposes over the time its on display.

Originally proposed in 1972, the piece didnt actually show until 2010. For Another Idea, Miralda will submit 61 photographs in total, uploading a new one each day that the virtual exhibition continues. Taken from a 2015 installation of the show, the images include his initial sketches of the work, a photo of a woman ironing a tablecloth at the installation space, and scientific-looking close-ups of the rice grains, fuzzy with mold.

A handful of the virtual pieces, meanwhile, were created for earlier iterations of the internet. Susan Hillers Dream Screens was first launched in 1996 neatly laid out, it allows the viewer to click around on the screen, changing its color along some hidden gradient while a robotic-sounding voice recites a monologue about dreaming.

I'm watching a man who has an amazing psychic power to somehow generate dreams that everyone can see. Works by several famous modernist artists turn into dream sequences in his mind, it begins. On an accompanying page of resources, Hiller, who passed away last year, writes that the text is largely based on loose recollections of films shes seen, each with the word dream in the title. The page also contains a color map, a list of hues used in the piece (Antwerp red to chrysocolla, a shade of turquoise), as well as an extensive bibliography of Hillers sources.

That site provides a lot of sort of supporting information, and gives you a peek into her research. At the same time, the way in which she presents it is still relatively obfuscating, said Schuh. It just leaves you with questions, which is really what you want to have, and have them sort of remain with you. I kept thinking about me sitting here with this piece in my house, and someone else doing the same exact thing. And so that's both this shared thing, and sort of also all my own.

Ihsahn teases new music and hints it’s coming later this week – Louder

Ihsahn appears to be teasing new music, with the Emperor vocalist and guitarist uploading a short audio clip to his Instagram page.

The only piece of information to go along with the clip is the date July 17 this coming Friday. This could indicate a new track or his new five-track EP which would follow Telemark which was released in February this year.

Speaking previously with Prog, Ihsahn said that while Telemark represented his heavier side, the as-yet-untitled companion EP would show his more progressive side.

He said: The plan is to follow up this EP with its aesthetic and musical counterpart, wherein I can distil all those more progressive, experimental and mellow elements of my music.

Ultimately, its all a means for me to get some of those ideas out of my system, giving myself space before I delve into the next album.

Last month, Ihsahn revealed that he and Matt Heafywere continuing to work on theTrivium frontmans long-awaited Mrityu project while in lockdown.

He said: Me and Matt Heafy have been tracking some killer vocals.The current situation prevents us from travelling, but with this technology, it is almost like were in the same studio.

Matt is absolutely killing it and were making great progress every session.

The project first came to light eight years ago when Heafy was looking to explore his love of black metal.

I used the consumer features in Microsoft Teams for one week, and it’s better than you’d think – OnMSFT

In these times, theres a lot of ways to communicate with friends and family. From Facebook Messenger, WhatsApp, iMessage, and so much more, there are no shortages of ways to talk to people.

But riding the momentum of Microsoft Teams in the enterprise and business space, Microsoft recently brought Teams over to the consumer world to enter the fray of messaging platforms. The company envisions Teams as not only something you can use for work, but also for friends and family.

With a beta of these features recently making their way over to the iOS version of the Microsoft Teams app, I thought Id try it out. I really had my doubts, but after a week of using Teams consumer features, I loved it. Heres my story.

Much like signing up for WhatsApp or other services, setting up Teams for consumer was quite easy. For most people who arent already using Microsoft Teams in their work and are trying Teams for the first time this will be an important step. Luckily, Microsoft makes it simple.

Since I already use Teams for work at OnMSFT, I simply went into the hamburger menu, and under accounts and orgs, signed in with my personal Microsoft Account then confirmed my phone number. I also opted to sync my contacts, too, as I wanted to see if anyone else was using Teams. I was then good to go.

Coincidently, an old college friend of mine (who now lives in Kazakhstan) was already using Teams consumer features. After I synced my contacts, I noticed that her name and number showed up when I went to create a list of messages in the chat. My colleague at OnMSFT also showed up too, and we used the app to talk instead of our usual method of communication, WhatsApp, and iMessage. I spoke to both these people on Teams throughout my week with the app thus far, and it is still ongoing.

I even convinced my mother to give Teams a try. As a first-time Teams user, she found the setup process quick and easy. She clicked the Sign up for free button, signed up with her phone number, confirmed it, added a profile picture, synced her contacts, and was good to go!

Just like Teams for business and enterprise, the core experience of Teams consumer features is the chat experience. As someone who is quite used to messaging on WhatsApp and iMessage, I must say this is where Teams consumer experience is just as good.

In the chat, you not only can send out text messages, but you also can record and send voice audio messages. This is a feature that has been present in iMessage and WhatsApp for quite some time, and its something that I am glad made its way to consumer Teams, too. After all, there are situations where written words are not right.

Another thing I liked about the chat experience is how similar it is to WhatsApp. The locations for the user interface are almost exactly the same. A plus icon to the left of the compose box to send files and GIFS, or location. Theres also a camera to the right to quick-send a photo, and then the microphone to record audio.

Composing messages, meanwhile, is the same as WhatsApp. If you opted to sync contacts, youll see everyone with Teams appear under the compose box. You can also add someone to a group chat without them already having Teams. Just tap their phone number, and add them. Theyll be sent an invite link via text to download and join Teams.

The same goes for a chat, too. If you want to chat with someone on Teams who doesnt yet have the app, just tap their number and theyll be sent a unique link to join the later. (This is possible in WhatsApp, but the recipient of the invite wont be able to automatically join the group chat or the chat you had for them.)

As my mother told me when she tried it out, most people will feel right at home when using Teams. Theres not much learning curve to using it. In my one week so far, chatting with my colleague and friend has been as natural as it is on WhatsApp and other apps. Not to mention, the emoji reactions (which arent on Whatsapp) have made for some funny moments and some more prolonged conversations between myself and my friends.

But there are some advanced features in chat that can only be found in Teams. This includes a formatter to bold, italics, and underline things in your messages. Theres also the ability to @mention someone to get their attention (outside of a group chat) or use the Task and the Safe features. Keep in mind, WhatsApp doesnt support these, which is why I will dive deeper into these next.

Asides from chat, another thing that makes Teams consumer unique is the Dashboard. From here, youre able to use Teams for consumer features to your advantage. Theres a lot to get into, including the Safe, location sharing, uploading files, and more. As a summary, though: the dashboard is essentially a hub, showing you all the things that happened within a chat or group chat.

Ill start first by talking about location sharing. This was one of the features Microsoft spoke a lot about when it first unveiled Teams for consumers. Just go to a chat, then tap the Dashboard, then choose Share location. Youll be able to share the location for 30 mins, 1 day, or always on. If your contact is also sharing location, you even can pull up a map to see a comparison of where you are in comparison to each other. You can stop sharing at any time with a Stop Sharingbutton in the UI.

Alas, the point is the person in the chat will be able to see exactly where you are. I used this with my mother to show her where I was when I left the home one day, but its a feature I think will be useful for families, and keeping kids safe. Say, a kid leaving school, etc. Its not exactly exclusive, though, as WhatsApp has the same exact feature, though, as do iPhones with the Find My app.

But what is exclusive to Teams is the Safe. Setting this up is simple, as youll need to enter a PIN (and store an encryption key for it on your MSA.) Youll then need to grant your contact access to your safe, and theyll be able to access it in real-time (or vice versa, too.) This is a great way to share information like passwords, passports, memberships, and more. Once you share your safe, the contact will be able to access it, too, and add content. I used it with my mother to share step by step instructions on how to do things in Teams and to give her the WiFi password. This is quite nifty, and Safe is even locked behind FaceID or TouchID for extra security. I have yet to see something like this on other messaging platforms, and its really Teams big potential.

Then there are the other features. You can create a shared calendar so you and contact can plan an event. You also can create a shared task. This one was useful for me and my mother, as I left her a grocery list, and she was able to check things off as she went to the store. Meanwhile, as I was home and was able to see the things she picked up from the store as she checked it off the list. Again, something that can be useful for families and giving children tasks or chores to do.

Finally, there is the ability to upload files. If you and the person youre sharing the file with both share or have a Microsoft 365 subscription, you can edit and co-edit the documents together by viewing the file in Teams, and then opening it through the Office app. This is great, as it combines the power of Office with Teams, allowing you to get your maximum productivity.

After one week, Im now a believer in Microsoft Teams being used by consumers. Although most features are already on iMessage, WhatsApp, and other platforms, its great to see Teams expand its reach. After all, most people already likely use Teams for work, so why not bring it into their homes, too?

But, not everything is perfect. Currently, Teams consumer features are only available on the mobile apps. There is no way to enjoy it on the desktop. Photos, videos, and other content sent on Teams dont easily transfer to desktop, as they would with enterprise and business.

Also, switching between work and personal accounts could lead to some serious problems. It really annoyed me to get double notifications, from both work and my personal account when using a single app for communication

This experience is still in beta, so more changes could be coming soon. Until then, I am a believer. Heres to hoping that Teams become just as popular as WhatsApp.

Back in 1988, Vin Scully and Joe Garagiola were on the call from Wrigley Field for the first real night game at Wrigley Field as we heard this afternoon on the Marquee Network.

Sometimes, even during a gloomy summer of masks, half-open restaurants, and social distancing, life works just right.

Today, I switched TV providers from YouTube TV to Hulu Live. The first show I found on the Cubs Marquee Network included on Hulu but not YouTube TV was the first night game game at Wrigley Field from August 9, 1988. I attended that game, and the first voice I heard during the replay broadcast today was my own.

Mets catcher Gary Carter was stuck on 299 career home runs, and I built a significant dislike forCarter during his career with the Expos and Mets (when he wrapped his career with the Dodgers, Giants, and Expos again, I pitied his eroding abilities so my dislike waned). I turned the game on this afternoon while the game was in the top of the sixth with Carter at the plate, and heard 299! 299! 299! I had to turn the TV up, but there was my psychotic chanting faintbut audible.

I yelled 299! incessantlyfrom our seats in the 10th row behind the Cubs on deck circle every time Carter came to the plate. A fan sitting in front of us asked why I hated Carter so much. I told him, Because he refers to himself in the third person. He calls himself The Kid all the time, and I dont like self-important people. The guy smiled and nodded. I think it pleased him that I had any reason at all besides the empty beer cups that were stacked in front of me.

Carter failed to hit his 300th home run that night, and I walked home believing I may have exerted some kind of a whammy on him. That delusion was proven flawed two dayslater, as I sat in differentbox seats. As I yelled 300! for the last time, Carter stroked a solo shot into the left field bleachersoff Al Nippers first pitchin the top of the second inning.

It was serendipity I was at the first night game. The game the night before against the Phillies was rained out while Iplayed softball at Welles Park. Somehow, we made to the finals of our league tournament, and I would rather play than watch, so I opted for a game we lost just before the rains came.

Every Cubs fan who was anyone was at Wrigley for the pageantry of that first night game on August 8th, but August 9th was more like a real Cubs game with the fans who had all of the heart but none of the cash of the guys from the night before.

The Cubs won that first night game 6-4. Frank DiPino got the win one of the seven he notched in his two-plus seasons as a mediocre lefty out of the bullpen and the Cubs climbed to within 12 games of the top spot in the NL East.

And now we watch that same game on the Marquee Network as the endless summer of Covid-19 continues and continues and continues. Ten days from today, we are promised a shortened seasonof 60 Cubs games, but there wont be fans anywhere but on the rooftops across Waveland and Sheffield Avenues.

Im thinking of driving up for one of the first games. I can sit in the Cubby Bear, feel a little of the atmosphere, laugh with friends (from a distance), and at least hear the crack of the bat as a little bit of normal creeps back into our lives. Then it occurred to me that I dont even know if the Cubby Bear will be open.

We long for that feeling from 1988, 1998, 2008, and 2018. We want to baskin the splendor of a perfect afternoon or night at Wrigley, not live on the memories created 32 years ago on a beautiful night screaming 299! at a hall of fame catcher because he called himself The Kid.

Black Lives Matter — Manifesto in Ten Points | National Review

Black Lives Matter has delivered a ten-point manifesto of what they want. I have to say, it isnt as bad as I expected. In fact, some of it makes a lot of sense. For example, they ask for the end of broken windows policing, the end of for-profit policing practices such as civil asset forfeiture as well and the end of the police use of military equipment.

What I am, however, surprised about is that the list doesnt at all mention ending the failed Drug War even though many of their demands are to end policies (like the ones mentioned above) that are by-products of the Drug War. It is even more surprising since manyhave noted how the black community suffers disproportionately from the policy.

I find this essay from John McWhorter extremely compelling and moving about the disastrous results the Drug War has had on Black families. Far from finding excuses for the decisions made by those who choose employment in the illegal drug market rather than lower-paying jobs in the legal labor market due to the incentives created by the Drug War, McWhorter explains the consequences and the vicious cycle that follows. He writes:

The War on Drugs destroys black families. It has become a norm for black children to grow up in single-parent homes, their fathers away in prison for long spells and barely knowing them. In poor and working-class black America, a man and a woman raising their children together is, of all things, an unusual sight. The War on Drugs plays a large part in this. It must stop.

I know this is a controversial issue among conservatives, even though acknowledging that the Drug War has failed and needs to be scaled back is different from condoning the use of drugs.National Review even called for legalization of Marijuana back in 1996, long before the New York Times did in 2014.

That being said, no matter what conservatives think about the policy, it remains strange that it doesnt appear on the BLMs list of demands.

Less surprising, but important, is a failure to ask for the end of minimum-wage policies. Over at Cafe Hayek, Don Boudreaux posted a great discussion between George Mason Universitys Walter Williams and the Hoover Institutions Thomas Sowell on, as Boudreaux writes, the minimum wage governments practice of ordering low-skilled workers to remain unemployed if, and for however long as, those workers are unable to persuade or entice employers to hire them at wages at least as high as the wage that government dictates. As Williams says during the interview:

The minimum-wage law has been, and continues to be, one of the most effective tools in the arsenal of racists everywhere around the world.

It is worth also reading the article published a few months ago over at the TNR about the racist origins of the minimum-wage laws. While the intentions behind the law have changed dramatically, it doesnt mean that the policy has become low-income-worker friendly. As we know, the road to hell is paved with good intentions.

Even less surprising is the absence on the BLMs list of the need to reform Social Security. As I have mentioned before Social Security redistributes money from blacks and other minorities to white people. You would think that considering the stakes, BLM would put it on their list.

All this goes to say that while the list wasnt as bad as I expected, it fails to address important policy changes that would really make a difference.

This is about the quality of life for black people, for poor people in this country, said Umi Selah, co-director of Dream Defenders in Miami. Though not officially affiliated, Dream Defenders and similar social justice groups often align themselves with Black Lives Matter.

The conception that all were mad about is police and policing is a strong misconception, Selah said.In fact, Black Lives Matter released a statement last weekcondemning the shooting in Dallas as counter to whatthe movement is trying to accomplish.

Ralikh Hayes of Baltimore BLOC echoed Selah, saying that Black Lives Matter is not inherently anti-police or anti-white, nor does the phrase Black Lives Matter means other lives aren't important.

We are against a system that views people as tools, Hayes said.

Cullors also hears claims that Black Lives Matterlacks direction or strategy. But Cullors said the strategyis clear -- working to ensure that black people live with the full dignity of theirhuman rights.

We are not leaderless, were leader-full, she said. "We're trying to change the world...developing a new vision for what this generation of black leaders can look like."

Filmmaker Emmai Alaquiva of Pittsburgh wanted to talk to his 8-year-old daughter about the racial climate in the US, but was concerned that at her age, she might not be able to fully grasp the issues.

I told her the name George Floyd and we talked about what Black Lives Matter means, he says. I sort of gave her the Cliffs Notes version, to give her some inspiration.

His daughter surprised him, however, and came back to him with a question: She said, hey, Dad, how come theres no people doing sign language or ASL in your photos and videos? It was a great point, we werent. He says he and his daughter take ASL classes together, part of his mission to help her learn to express herself in different mediums.

Over the next few days Alaquiva reached out to professionals in the Deaf and hard-of-hearing community including freelance sign language interpreter Amy Crawford, Danielle Filip of Pittsburgh-area Sign Language Interpreting Professionals, and Greg Pollock, an accessibility officer at a local bank, to be consultants on the script for the video he wanted to do, featuring a message in American Sign Language.

I wanted to make sure the messaging was clear, and that we werent appropriating anything from the community, Alaquiva said.

The result is Unspeakable, a public service announcement featuring members of the Deaf community: Although we may be Deaf, we can hear the world loud and clear, the group signs.

It is my hope that Unspeakable brings attention to the basic human right of communication access as a thread which connects us all, Filip said.

The nuances of communicating in sign language were important to get right in the video, Alaquiva said: for instance, the word black referring to the color can be signed with one finger pointed out in the ASL 1 handshape moving horizontally across the forehead. But when referring to a Black person, the sign uses four fingers pressed together in the ASL b handshape moving horizontally across the forehead, and is generally reserved for use by Black people, Alaquiva said.

Participating in public rallies is often challenging for Deaf and hard-of-hearing people, especially in large crowds with many people wearing face masks that cover their mouths. Activists have worked with the National Alliance of Multicultural Disabled Advocates to push for the #BlackDisabledLivesMatter efforts, to spotlight these challenges, as Teen Vogue columnist Sarah Kim reported. They also seek to call attention to the number of police violence victims who had disabilities.

Alaquiva said he hopes the PSA is inspiration for others involved in protest movements to be more inclusive, not necessarily just for people who are Deaf or hard of hearing, but anyone with a disability. Its not our job to just do something then go about our business, he said, adding he and his production team built a website with additional resources.

Theres a photo I took at a protest in Oakland and the persons sign said Im sorry Im late, I had a lot to learn. That can apply to any movement you want to be a part of. Whether youre on time or late, you gotta show up.

Background: Pseudomonas aeruginosa is an opportunistic pathogen that causes serious nosocomial infections, especially in immunodeficient patients and cystic fibrosis, cancer, and burned individuals. The biofilm that plays an important role in the virulence of P. aeruginosa is under the regulation of quorum sensing and two-component regulatory systems of bacteria. Curcumin, an active phenolic extract of turmeric has shown an inhibitory effect on the biofilm formation of some pathogenic bacteria. Thus, the present study aims to evaluate the effect of Nano-Curcumin on the expression of major regulatory genes involved in biofilm formation of P. aeruginosa.Materials and Methods: The biofilm formation of P. aeruginosa ATCC 10145 was assessed in the presence of 15, 20, and 25 g/mL concentrations of Nano-Curcumin using the microplate titer method. The effect of Nano-Curcumin on the expression level of regulatory genes were determined by relative reverse transcriptase-realtime PCR.Results: In the absence of Nano-Curcumin, P. aeruginosa strain ATCC 10145 strongly produced biofilm (3+) and in the presence of 15 and 20 g/mL, biofilm formation was reduced to moderate (2+) and weak biofilm producer (1+), respectively. Nano-Curcumin at a concentration of 25g/mL inhibited biofilm formation in P. aeruginosa. The expression of regulatory genes was not affected by biofilm inhibitory concentrations of Nano-Curcumin.Conclusion: The antibiofilm mechanism of Curcumin is not related to the downregulation of regulatory systems of P. aeruginosa and probably it prevents the formation of a complete biofilm structure.

The European Commission has launched consultations on its planned strategy for offshore renewable energy which aims to set out best ways to exploit and scale-up the sector's generation.

Offshore wind is expected to play an instrumental role in building the region's largely renewable energy power system between 2030 and 2050 as stated in the European Green Deal communication.

An EU offshore renewable energy strategy is to be published before the end of the year.

The public consultation aims to gather input from all interested stakeholders and is open until 24 September.

A separate four-week consultation has also been launched on an offshore renewable energy roadmap that aims to inform citizens and stakeholders about the Commission's work in order to allow them to provide feedback and to participate effectively in future consultation activities.

Citizens and stakeholders are in particular invited to provide views on the Commission's understanding of the issue and possible solutions and to make available any relevant information that they may have.

This consultation is open until 13 August.
