Reawakened immune cells attack cells tied to diseases of aging – STAT – STAT

Scientists have started to test whether natural killer, or NK, cells can be trained to go after hard-to-cure blood cancers in human patients. But making these sentinels of the innate immune system a potential boon to human health spans might be simpler: Rather than needing to be genetically engineered or primed with synthetic antibodies, they just need to be turned on.

In mice, researchers reported on Monday in the journal Med, activating NKT cells can eliminate the senescent cells partly responsible for many diseases of aging. If the results hold up, they could offer a promising alternative to senolytics experimental drugs that destroy these zombified cells that pile up and pollute your tissues as you get older. Although dozens of such drugs have postponed or even reversed diseases of aging in mouse experiments, clinical trials have thus far underwhelmed.

Its an interesting approach that works in experimental animals with two different conditions, said geriatrician James Kirkland of the Mayo Clinic, whose discovery that giving old mice senescent cell-crushing compounds makes the animals live longer, healthier lives, helped take senolytics from backwater to boomtown. Were going to need multiple ways of getting at senescent cells, he said. Any step forward is important, and this is quite a nice step forward. But he cautioned that a single senolytic strategy is unlikely to work for all age-related conditions.

For decades, scientists had largely ignored senescent cells old and arrested in a permanent state of suspended animation dismissing them as a quirk of evolution, a clever way for the body to keep damaged cells from proliferating into cancer. But more recently, Kirkland and other researchers established that senescence is actually a driver of the decrepitude that comes with old age. As cells stop dividing, they dont exactly go dormant. In their zombie-like state, they start spewing a cocktail of toxic molecules that cause inflammation, damage surrounding tissues, and contribute to diseases like osteoarthritis, atherosclerosis, diabetes, and Alzheimers.

That realization spurred the creation of at least two dozen companies developing ways to systematically purge the body of senescent cells. Senolytics attracted this wave of investment because it promises a scintillating and fundamental shift in medicine away from the one-drug-one-target-one-disease paradigm of the last century, toward correcting a root cause behind many of them with a single treatment.

One of those researchers is the new studys senior author, Anil Bhushan of the University of California, San Francisco. In 2019, his lab traced the progression of type 1 diabetes in mice and human pancreatic cells. They discovered that signatures of senescence preceded the onset of disease. When his team removed the senescent pancreatic cells in mice, their metabolism stabilized and their diabetes symptoms went away.

What Bhushan took away from that study was that senescence didnt just happen when the bodys biological clocks wound down too far. It occurred in acute diseases too. Thered been talk in the field that an immune surveillance system maintained tissue homeostasis, said Bhushan. We postulated that that system was failing in the disease state and gradually failing in aging, so the only time wed see senescent cells is when this system fails.

To test that hypothesis, his team first went looking for clues as to the identity of their indolent immune cells. By comparing the transcriptional profile which genes were turned on and off in senescent pancreatic cells to healthy ones, they uncovered that the senescent ones boosted production of their antigen-presenting machinery. These are the proteins that, if a cell were infected with a pathogen, would shuttle little bits of the bacteria or virus to its surface, displaying them for immune cells to find. Then they cross-referenced those results with an analysis of senescent stem cells that accumulate in the fat tissues of obese mice who are fed a chronic, high-fat diet. Those cells also upregulated antigen-presenting molecules, and one in particular: CD1d. That was the lock, said Bhushan. And once wed found it, the key was then obvious.

Only one kind of immune cell binds to CD1d invariant natural killer T cells, or iNKTs. Comprising less than 1% of all peripheral blood immune cells, iNKTs are rare but critical components of the bodys surveillance system, scanning for infected and defective cells in need of removal. When they find them, iNKTs expel torrents of cytokines, which signal to other immune cells to do the dirty work. Bhushan figured that something was interfering with that process. And though he wasnt sure what it was, he knew there was a way to fix it.

In the early 1990s, Japanese scientists from Kirin Brewerys pharmaceutical research lab, looking for anticancer treatments in the porous bodies of marine sponges collected in the Okinawan sea, purified a lipid compound called -galactosylceramide. And they discovered that when CD1d grabs onto this -GalCer lipid, it turns on iNKTs like crazy in mice.

So Bhushans team shot up some of their diet-induced obese mice with -GalCer. Within days, the levels of senescent cells in the mouse fat tissues had dropped. So did their fasting glucose. Their insulin sensitivity also improved. Their metabolism started to look normal.

To see how generalizable the effect was, they repeated the experiment with mice whose lung tissues had been damaged by a chemotherapy drug a common model for idiopathic pulmonary fibrosis, a serious and incurable human lung disease, and one of the nastier complications of Covid-19. In those mice, -GalCer successfully activated iNKTs, again resulting in the removal of senescent cells. The treated mice had fewer damaged cells, and they also lived longer than the control group.

Finally, Bhushan and his colleagues looked at how well activated iNKT cells could tell senescent human cells from healthy ones when cultured together. After 18 hours, 100% of the senescent cells had been destroyed; the vast majority of healthy cells went unscathed. That could give the iNKT approach a potential advantage over the senolytics drugs already in development.

Most of them are repackaged cancer drugs that work by flipping on senescent cells self-destruct buttons. But because zombie cells share a lot of molecular features with their fully animated counterparts, those drugs run the risk of creating lots of collateral damage. Clinical trials of one such drug, 17-DMAG, were abandoned due to toxic side effects in the kidney and brain. Other groups are trying to solve this by engineering a different kind of immune cell, the CAR-T cell, to become a better anti-aging treatment. But CAR-Ts come with their own dangerous side effects and are expensive to make.

Bhushan is optimistic that by returning the cells best trained to suss out senescent cells to the ranks of active immune duty, both these safety and cost concerns can be ameliorated. We have this built in specificity of the immune cells part of their job is telling senescent cells apart from healthy ones were just helping them do their job, he said.

Its still an open question. But clinical trials to answer it could be underway by the end of next year. Bhushans initial discoveries are now being developed by a biotech startup called Deciduous Therapeutics, which he co-founded in 2018. Deciduous is backed by 8VC, CRV, and Laura Demings Longevity Fund, and has until now, been operating in stealth. CEO and co-founder Robin Mansukhani told STAT that the company has been focusing on developing compounds that can best stimulate human iNKTs, whose receptors are structurally a bit different from those of a mouse. He expects Deciduous to file its first investigational new drug application to start human testing within the next 18 months, likely for a metabolic disease or fibrotic lung disorder.

Investors pumped the brakes on senolytics after one of the biggest and brightest stars of the nascent sector and another Longevity Fund portfolio company, Unity Biotechnology, announced last August that its lead drug candidate had failed to reduce knee pain in patients with osteoarthritis. The experimental drug was immediately and unceremoniously dumped, along with nearly one-third of Unitys staff.

But despite the recent slowdown, Mansukhani remains optimistic. The issue in the field has always been what is the actual immune system process behind senescence clearing?he said. And I feel like weve uncovered that.

Mayos Kirkland cautions that deciphering one chapter of the immune system users manual isnt likely to be the whole story. Senescence can be caused by lots of things aging, yes, but also obesity, chemotherapy drugs, and radiation. There are about 40 to 50 different things that can push a cell into a death spiral, said Kirkland. That makes it really hard to define what a senescent cell is, because its molecular makeup depends on how its senescence was induced.

But the good news, according to Kirkland, is that all the fundamental aging processes mitochondria powering down, oxygen radicals disfiguring DNA, rampant inflammation, the spiral toward senescence appear to be tightly interlinked. Its looking increasingly like if you hit one part of this network of things going on, you affect all the rest, and usually in a positive way, he said.

Kirkland, together with his team at Mayo, have had some success with a cocktail of dasatinib and quercetin. In 2019, they reported positive results from a Phase 1 pilot study of nine diabetic kidney disease patients senescent cells were reduced. A Phase 2 study is now underway. And his team has several more trials for serious conditions, including osteoporosis and Alzheimers disease, in the pipeline. Kirkland serves as a scientific adviser to a new senolytics company called NRTK Biosciences that has yet to receive funding and anti-aging supplement company Elysium Health.

But the real question for the future, said Kirkland, is which interventions can you combine to get an additive, synergistic effect? Something that actually alleviates not just one disease of aging, but many, or even all of them? Figuring out how immune cells interplay with senescence is going to be the start of looking for those combinations.

Thats whats next for Bhushans lab a painstaking process of deleting different antigen-displaying peptides across dozens of types of immune cells, and mapping out the almost infinitely complex network of interactions brewing in the toxic stew surrounding senescent cells.

We know we only have part of the story, Bhushan said. We know we can fix whatever is going wrong with iNKTs, but we still dont know exactly what that is.

CT team developing COVID drug to reverse aging … really – CT Insider

The older you are, the more likely your COVID-19 infection will be more severe, and even fatal. Of the nearly 5,000 COVID-related deaths in Connecticut to date, 81 percent are people over 70 years old.

But what if you could turn back the clock? A team of researchers at UConn are working on ways to literally reverse the effects of aging, as a way to mitigate the severity of a COVID infection.

The goal here is not to erase aging, but rather were testing interventions that are designed to slow the rate of biological aging, and therefore help prevent severe COVID, according to George Kuchel, director of UConns Center on Aging and chief of geriatric medicine at UConn Health.

One drug in particular, called compound RTB101, can theoretically not only slow aging but boost the immune response, helping the body and any potential vaccines do their jobs better.

Thats important because, as Kuchel said, these vaccines dont work as well on older adults.

Even if the vaccine turns out to be as effective, as the initial studies suggest, theres a very strong likelihood that its going to be less effective in older adults, he said. These kinds of approaches are going to be important as additional therapies to the vaccine, for the vaccine alone is not going to be sufficient.

Another weakness of any potential vaccine is their specificity. The COVID-19 pandemic may be contained, but the virus could mutate, or another virus might emerge.

At some point, were going to be done with COVID-19, hopefully, Kuchel said. But I think we all agree that there could very well be another COVID, whether its COVID-21, 22, whatever, or another pathogen out there, that we dont even know about yet. And if older adults are more vulnerable to get this and to die from it, then we believe this approach will be helpful irrespective of what the infection is.

The compound known as RTB101 doesnt actually make you younger. Theres a difference, Kuchel explained, between chronological age how many years you have been alive and biological age.

Whereas biological aging is an effort to capture the rate of aging, how fast somebody ages, Kuchel said. As you can imagine, you could have two people who are both 70 years old, and one could have a biological age which would be younger than most 70-year-olds, and another one who has a biological age that would be older than most 70-year-olds.

RTB101 targets the biological pathways that control the aging process, the so-called mTOR pathway, for example.

Early versions of the compound have been studied for decades at Connecticuts Jackson Labs and elsewhere and have been shown to slow aging in mice.

While inhibiting the natural processes that lead to biological aging, the compound also boosts the bodys ability to fight disease.

There are a lot of genes that are involved in our ability to fend off viral infections, Kuchel said. There are these inflammatory molecules called interferon gamma. That whole family of molecules are very important to our ability to fend off infection, particularly viral infections. What this compound does is it boosts those responses. So it down-regulates mTOR and it up regulates the interferon gamma responses.

In a sense, it kind of resets the body to behave the way it would have at a younger age, and to boost immune responses, Kuchel said.

The work on RTB101 is being funded by the National Institutes for Health and the first step which the team at UConn is currently recruiting subjects for is a small-scale feasibility study involving people over the age of 65 who have either been recently diagnosed with a COVID-19 infection or who lives with someone diagnosed with COVID in the last few days.

Were in a very tight timeline, Kuchel said. We need to do this in the next two months.

But dont expect your doctor to prescribe an anti-aging drug any time soon.

After the feasibility study will come a large-scale, multi-site clinical trial. Only then will Kuchel and his team seek FDA approval.

I know that sometimes the progress of science seems maddeningly slow. But unfortunately, its the only way we can show that it works, Kuchel said. In terms of this being available at physicians offices, its not going to be within the next year or two. This is going to take some time.

But, again, this sort of research could be important, Kuchel said, because it is not a therapy to treat or prevent any specific disease.

We believe that will be transformative not only for the current COVID pandemic, but for future pandemics, future infections, which we know are going to happen, he said. We know that future infections will happen, we know that older adults will be more vulnerable. Thats absolutely clear. The only thing we dont know is what the pathogen will be. With this approach, the argument can be made that we dont need to know what that pathogen is because were not targeting the pathogen, were targeting the person.

Question: I am here with the greatest tight end in NFL history who now calls himself Mr. Recovery. After a litany of injuries, you are coming out of retirement this year and making your comeback with the Tampa Bay Buccaneers. How did this all happen?

Gronk: Thank you! I am super excited, super pumped up. I feel so good, you know. I let my body heal and recover. Ive been playing football for fifteen plus years, plus along with other sports, and with all this action Ive seen, my body just needed a little rest. I had to investigate and find connections to heal. Dr. Rick turned everything around for me. Its a full-time job as an athlete. Athletes dont just show up. We have to take care of our bodies and find solutions that work. The recovery is cake with Dr. Rick.

Question: Your journey back to health is unprecedented. Few NFL players return to action after retiring like you did. Allografts are the new buzz words for lasting recovery. Is this the type of therapy and injections you received from Dr. Rick at REGENCare?

Question: How does your focused mindset play a role in your healing? Is it an importantpart of your treatment process?

Gronk: Yeah, definitely! Some people can be telling you to do this, and you just got to stick with your gut. You got to stick with your heart, knowing what is best for your body. To know that, you have to go through a lot of experiences. Ive tried many, many things, and some do work for me. I focus on all the natural treatments that you could do for yourself. I find the best ones that suit me and enjoy em while doing em.

What would you tell your friends and teammates who are suffering from chronic pain and injuries about these regenerative treatments?

Gronk: Just get started on it now! The longer you wait, the harder it is going to be to heal that injury and nagging pain. The longer you wait, it gets more settled into your brain that you have that nagging pain. So hop on it ASAP, get moving, get cruising on it, find some good workouts, strengthen your core tight, and start small. Thats where it begins. You dont got to go big at all. Just start small and make little changes.

Question: So whats next for you? You have an exciting season with Tampa Bay, what are some of the things that youre doing to prepare?

Gronk: You know, Im just hanging out! Throwing footballs, doing football workouts, doing band workouts, doing some strength workouts, just doing it all at high speed. Its a lot of expectations for sure. Just have to go in and be a consistent player and youre just ready to go. Im just excited to get back out into the field and play some football and help out the team.

Question: If you were to leave a legacy behind, what would it be?

Gronk: Just working hard, being the best teammate that I can be and doing the best that I can to help out the team. Now that Dr. Rick has helped me to recover, I can achieve this and more.

Device Reads Paralyzed People’s Thought Through Their Veins – Anti Aging News

Synchron recently announced the publication of a first-in-human study demonstrating the successful use of their device called Stentrode which is a brain-computer interface neuroprosthesis. The brain implant that is inserted in the jugular allows ALS patients to type with their minds.

Synchrons CEO, Thomas Oxley, MD, Ph.D., describes the implant as Bluetooth for the brain. The big difference between what we have done with what previous approaches have done is that we do not put any needles into the brain or on top of the brain. The device stays inside the blood vessel.

The study findings published in the Journal of NeuroInterventions Surgery specifically show the devices ability to enable patients with severe paralysis to resume daily tasks such as texting, emailing, shopping and banking online all through direct though without the need for open brain surgery.

This study is the first to demonstrate that a BCI that has been transplanted via patient blood vessels is able to restore the transmission of brain impulses out of the body wirelessly; patients successfully used their impulses to control digital devices without using a touchscreen, mouse, keyboard or voice-activated technology.

This is a breakthrough moment for the field of brain-computer interfaces. We are excited to report that we have delivered a fully implantable, take home, wireless technology that does not require open brain surgery, which functions to restore freedoms for people with severe disability, said Thomas Oxley MD Ph.D., founding CEO, Synchron. Seeing these first heroic patients resume important daily tasks that had become impossible, such as using personal devices to connect with loved ones, confirms our belief that the Stentrode will one day be able to help millions of people with paralysis.

Using the Stentrode neuroprosthesis device patients controlled the Microsoft Windows 10 operating system, in combination with an eye tracker for cursor navigation without a mouse or keyboard. Patients undertook machine learning assisted training to control multiple mouse click actions, and the first 2 patients were able to achieve an average click accuracy of 92 and 93% with typing speeds of 14 and 20 characters per minute with predictive text disabled. The device was used to resume daily tasks independently following implantation and training to commence unsupervised home use of the system from day 86 and 71.

Using the Stentrode has been life-altering, said Graham Felstead, the first patient enrolled in the first Stentrode clinical study and the first person to have any BCI implanted via the blood vessels. The device has allowed me to be productive again, including shopping, banking, and delegating tasks among the Rotary Club members with whom I volunteer. Its incredible to gain this level of independence back. By using the Stentrode, Graham was able to achieve his goals of remotely contacting his spouse, increasing his autonomy and reducing her burden of care. He was able to maintain contact with other family members, medical professionals and people in his community. Graham, a 75-year-old man living at home with his wife, has experienced severe paralysis due to amyotrophic lateral sclerosis (ALS), and received the Stentrode implant in August 2019. He continues to use it today.

It is truly amazing, and very rewarding, to see the participants use the Stentrode to control a computer with their minds, independently and at home, said Nicholas Opie, associate professor and co-head of the vascular bionics laboratory at the University of Melbourne and founding CTO of Synchron. The trial participants have been fantastic, and my colleagues and I are truly honored to make a difference in their lives. I hope others are inspired by their success.

The second participant, Philip OKeefe, a 60-year-old man with ALS, working part-time and living at home with his wife and two children, was able to control computer devices to conduct work-related tasks and other independent activities after receiving the Stentrode in April 2020. Functional impairment to his fingers, elbows, and shoulders had previously inhibited his ability to engage in these efforts.

While the Stentrode is a completely novel concept and design, the procedure to implant it draws upon techniques that I use on a daily basis and which have become routine in our industry. The Stentrode technology exemplifies the progress and potential of neurointervention departments to deliver new therapies to patients, said Professor Peter Mitchell, Director of Neurointervention, The Royal Melbourne Hospital, a pioneering neurointerventionalist who performed the first clinical implantations of the Stentrode device. The intricacies of the brain have always fascinated me and enabling a patient with paralysis to continue to use their mind in a productive way is immensely rewarding.

This device is small and flexible enough to safely pass through curving blood vessels, the implantation procedures are similar to that of a pacemaker and does not require open brain surgery. This may reduce the risk of brain tissue inflammation and rejection of the device which has been an issue for devices in the past using techniques that require direct brain penetration.

The device is delivered into the jugular vein, using a catheter to thread the device upward until it is adjacent to the motor cortex. Once implanted the device is able to pick up nearby brain activity using tiny electrodes that dot its surface. A second device embedded into the patients chest transmits data wirelessly to a computer or other smart device that is running the machine learning software which in turn converts that information into computer commands and functions.

Other approaches to brain-computer interfaces have been focused on high data rates, which create technical barriers to clinical translation. Synchron has focused on building a product that measurably improves patients lives. Our first-to-market product will focus on smart device control using brain impulses, without the need for the use of the hands. This alone has potential to address an unmet market need in 30 million patients in advanced economies, added Oxley.

The US FDA granted Stentrode Breakthrough Device designation, and the safety and efficacy data from this feasibility study will be used to finalize the protocols for a future FDA approval trial that will eventually guide the evaluation of the device for American marketing approval.

The neurovascular bioelectronic medicine company presented preliminary data on the first patient at the Society of NeuroInterventional Surgery Annual Meeting In August 2020, and the long term safety as well as ability to pick up specific electrical frequencies emitted by the brain was established by preclinical studies that were published in Nature Biotechnology, Nature Biomedical Engineering and the Journal of Neurosurgery.

Eventually, the company plans to use this technology to help paralyzed patients control robotic limbs in order to provide them even more freedom and have an even bigger positive impact on the everyday lives of people who use it.

Two Pitt Faculty Members Inducted into National Academy of Medicine for Work on Aging and Neuro-Rehab – Newswise

Newswise PITTSBURGH, Oct. 19, 2020 Two physician-scientists at the University of Pittsburgh School of Medicine have been elected to the prestigious National Academy of Medicine.

Among the class of 100 new inductees honored at todays annual meeting are Toren Finkel, M.D., Ph.D., professor of cardiology and director of the Pitt Aging Institute, and Amy Wagner, M.D., professor of physical medicine and rehabilitation and neuroscience at Pitt.

This is a wonderful honor from my peers, made possible through my good fortune of working with so my talented people along the way, as well as the unwavering support of my family, Finkel said.

Finkels work centers on how oxidative stress and the function of mitochondria alter the rate of aging. The long-term goal is to uncover the molecular basis of aging and age-related diseases through the study of a variety of different cellular pathways. Recently, Finkels focus has been on autophagythe bodys natural way of clearing out old and damaged parts of cellsas a target for anti-aging therapeutics.

Wagner uses biomarkers and statistical modeling to predict how well patients will recover from brain injury and to guide clinical decisions along the waya strategy she calls personalized rehabilitation medicine. This clinical research informs her studies using in vivo pre-clinical models aimed at identifying intervention strategies that promote neurorecovery. She also treats patients with neurological conditions who are undergoing inpatient rehabilitation, and she is a consultant for neurologically devastated patients during their acute hospitalization at UPMC.

One of the things that I look forward to the most with my Academy membership is being able to use my voice to represent and serve all rehabilitation populations, but particularly our brain injury population, which is underserved, Wagner said. Disability is such a huge issuethere are over 50 million people in this country who have some type of disability. Being able to advocate for them and to bring that to a national stage showing what we have to offer is really important.

Election to the Academy is considered one of the highest honors in the fields of health and medicine and recognizes individuals who have demonstrated outstanding professional achievement and commitment to service. New members are elected by current members through a process that recognizes individuals who have made major contributions to the advancement of medical sciences, health care and public health.

Preclinical Study Shows Improvement In Recovery From Heart Attack With Exosomes – Anti Aging News

Recovery from experimental heart attacks can be improved with an injection of a mixture of heart muscle cells, endothelial cells and smooth muscle cells, but results are limited by poor engraftment and retention, plus there are concerns about potential tumorigenesis and heart arrhythmia.

Recent animal research in pigs has shown that using the exosomes naturally produced from a mixture of heart muscle cells, endothelial cells, and smooth muscle cells derived from human induced pluripotent stem cells yielded regenerative benefits that were the equivalent to the injected hiPSC-CCs.

Exosomes are membrane-bound extracellular vesicles that contain biologically active proteins, RNAs and microRNAs that are well known to participate in cell to cell communication, and are actively studied as potential clinical therapies for a wide range of conditions.

The hiPSC-CC exosomes are acellular and, consequently, may enable physicians to exploit the cardioprotective and reparative properties of hiPSC-derived cells while avoiding the complexities associated with tumorigenic risks, cell storage, transportation and immune rejection, said Ling Gao, Ph.D., and Jianyi Jay Zhang, M.D., Ph.D., University of Alabama at Birmingham corresponding authors of the study, published in Science Translational Medicine. Thus, exosomes secreted by hiPSC-derived cardiac cells improved myocardial recovery without increasing the frequency of arrhythmogenic complications and may provide an acellular therapeutic option for myocardial injury.

Studies involving large animals are required to identify, characterize and quantify all responses to potential treatments, prior to this study the feasibility of hiPSC-CC exosomes for cariad therapy had only been shown to be effective in mouse models and in vitro work.

The UAB studies involving juvenile pigs with experimental heart attacks had 1 of 3 treatments injected into the damaged myocardium: a mixture of cardiomyocytes, endothelial cells, and smooth muscle cells derived from human induced pluripotent stems cells, exosomes extracted from three cell types, and homogenized fragments from the cell types.

There were 2 primary findings from this study. Measurements of left ventricle function, infarct size, wall stress, cardiac hypertrophy apoptosis and angiogenesis in the animals treated with hiPSC-CCS, hiPSC-cc fragments or hiPSC-cc exosomes were found to be similar and significantly improved compared to those that recovered without any of the 3 treatments. Additionally, exosome therapy was found not to increase the frequency of arrhythmia.

During experiments with cells or aortic rings that were grown in culture, exosomes produced by hiPSC-CCs were found to promote blood vessel growth in cultured endothelial cells and isolated aortic rings. The exosomes also protected the cultured hiPSC-cardiomyocytes from the cytotoxic effect of serum-free lox oxygen media by reducing the programmed apoptosis cell death and by maintaining intracellular calcium homeostasis which had a direct beneficial effect on heart conductivity. Additionally, the exosomes also increased cellular ATP content which is beneficial as deficiencies in cellular ATP metabolism are believed to contribute to the progressive decline in heart function in those with left ventricle hypertrophy and heart failure.

Some of the in vitro beneficial effects were found to also be mediated by synthetic mimics of the 15 most abundant microRNAs that were found in the hiPSC-cc exosomes. It was noted that knowledge of the potential role of microRNAs in clinical application requires more research as it is far from complete.

Panax ginseng is one of the most widely used and studied species of ginseng, and it has been a staple herb in Traditional Chinese Medicine to help boost immunity, increase strength and blood volume, lower blood sugar levels, promote life, appetite, as well as helping to treat anemia and impotence.

Acetylcholinesterase, monoamine oxidase, and the N-methyl-D-aspartate receptor macromolecules were selected by the researchers to be the target proteins for AUF-MS screening which revealed 19 potential neuroactive compounds present in Panax ginseng.

AChE enzymes are responsible for breaking down acetylcholine neurotransmitters and are mainly found in the nerves and muscles. MAO-B enzymes break down brain chemicals including dopamine. The NMDA receptor protein is the primary excitatory neurotransmitter in the brain that glutamate binds to, and the dysfunction of this receptor is linked to the onset of Alzheimers disease.

All three macromolecules were found to have significant neuroactivity after evaluation of the bioactivities of the Panax ginseng components by AChE-inhibitory testing and DPPH assay, and the AUF results were confirmed with the molecular docking results; based on their findings the team concluded that Panax ginseng exerts a therapeutic effect against Alzheimers disease through the interaction between active components and biological targets.

Ginsenoside triterpene saponins are the main active components of this herb and the majority of research attributes the medicinal properties of the herb to these compounds. These compounds are believed to be potent neuroprotective agents that have antioxidant, anti-inflammatory and anti-tumour activities as well as the ability to inhibit cell apoptosis.

Rice water is the latest beauty trend taking over the internet right now but what actually is it?

As part of a Korean skin care routine, rice water has been used to maintain youthfulness and make hair appear more lustrous.

As a clean beauty ingredient, rice water is now used globally to brighten sensitive skin, and as a hair rinse, its been touted for its ability to strengthen and enrich hair with vitamins and carbohydrates.

While its true that many cultures have historically utilized rice water for hair growth, the scientific jury is still largely out on exactly how effective rice water may be.

There was also a study of a natural, herbal shampoo made in northeastern India that uses rice water as a key ingredient. The self-reported effects of this shampoo were anti-aging, as well as increased shininess, and softness. Rice bran has also been shown to help stimulate hair growth, as well.

But the truth is, everyones hair is different and responds to ingredients in its own way. If youre curious about using rice water for hair, its definitely a clean, nontoxic, inexpensive ingredient to experiment with.

We would love to see more controlled studies on the benefits of rice water for skin, but there have been some promising early studies regarding how beneficial this time-honored ingredient can be in a clean beauty routine.

Its shown in clinical trials to be nonirritating and unlikely to cause allergic reactions however, if you know you have a rice allergy, of course, steer clear. Its also been demonstrated to be hydrating, and has been shown to have some anti-aging and anti-inflammatory properties, as well.

Rinse and strain the rice to remove any impurities. If youre using nonorganic rice, then you may want to rinse it 2 to 3 more times to remove pesticides.

Mix the strained rice in a bowl with the water until the water becomes cloudy. Strain the rice, reserving the water in the bowl, and cover. Let the water sit at room temperature for 12 hours to allow it to ferment, then refrigerate.
