AI-based brain monitoring and robotic muscle projects win 11.5m EPSRC boost | Imperial News – Imperial College London

Two Imperial College London projects have won a total of 11.5 million to transform healthcare technologies by 2050.

The UKRI EPSRC Transformative Healthcare Technologies for 2050 funding aims to foster the development of revolutionary new technological approaches to transform care and treatments in the NHS by 2050.

A new Imperial-led project will monitor the brain in a way never achieved before.

The project team, led by Professor Dario Farina at the Department of Bioengineering, will develop a new technology, known as Non-Invasive Single Neuron Electrical Monitoring (NISNEM), with the aim of helping scientists better understand neurological diseases like stroke and Parkinsons.

The researchers hope the technology will enable new neurotechnology-based treatments that are non-invasive and more effective than current ones.

Current approaches to monitoring the brain, like using surgical implants, are invasive. However, NISNEM technology uses electrical signals recorded on the skin surface from extremely high-density electrode probes to create images of the brain, which AI algorithms then scan to find traces of individual neuron activities. These decoded neural activities could reveal signs of neurological impairments like stroke and Parkinsons, and help drive brain-computer interfaces for assistance and therapy.

Stroke affects more than more than 100,000 people per year in the UK, and occurs when the supply of blood to the brain is reduced or blocked. It most commonly arises in the cerebral artery, which directly affects the motor cortex, and can cause substantial impairments in the motor abilities of patients affecting walking and speech.

Parkinsons disease affects 1 in 500 people, causing parts of the brain to become progressively damaged over many years. It develops when dopamine-producing cells in the brain stop working properly and are lost over time. This causes a lack of dopamine in the brain, which is important for movement and speech, and causes tremor, slowness of movement, and muscle stiffness.

Stroke and Parkinsons are only two examples of the impairments targeted by the new NISNEM technology. The researchers hope the new technology will detect brain abnormalities to help early diagnosis of these progressive diseases as well as developing non-invasive treatments.

Professor Farina said: The current inability to access single neuron activity in the brain not only limits our ability to understand and diagnose neurological conditions, but also prevents the broad adoption of neurotechnologies in healthcare. We hope our technology, funded by the EPSRC grant, will provide a breakthrough in understanding, diagnosing, and treating increasingly prevalent neurological conditions, such as stroke, Parkinsons disease, and Alzheimers.

A University of Bristol-led project involving Imperials Professor Molly Stevens aims to develop artificial robotic muscular assistance to help restore strength in people who have lost muscle capability. This could include patients who have suffered a stroke or are living with degenerative diseases such as sarcopenia and muscular dystrophy.

Using these highly targeted robotics could help overcome the limitations of current wearable assistive technologies in regenerative medicine. Often, these technologies can be bulky and uncomfortable to wear, and can require two people to put on and take off. Users can also find the movements too slow.

Through using robots to boost the performance of such technologies, the project, called emPOWER, could provide life-changing benefits for sufferers by restoring their confidence, independence and quality of life - all while reducing the cost to the NHS.

Co-investigator Professor Stevens of the Department of Materials said: Loss of strength and muscle wastage is currently an unenviable part of getting older and has a significant impact on health and quality of life. emPOWER will explore how artificial muscles could radically transform treatment options in the future and effectively turn back the body clock.

emPOWER is led by Professor Jonathan Rossiter at Bristol, in partnership with Imperial, UCL and the NIHR Devices for Dignity MedTech Co-operative, hosted by Sheffield Teaching Hospitals Foundation Trust.

Science minister Amanda Solloway announced the funding as part of her keynote speech at London Tech Week 2020.

She said: The pioneering projects we are backing today will help modernise healthcare, improving all of our lives now and into the future. This announcement is part of our ambitious R&D Roadmap and underlines our commitment to back our incredible scientists and researchers and invest in ground-breaking research to keep the UK ahead in cutting-edge discoveries.

Petit Institute to Host Race and Racism Event on Covid-19 – Georgia Tech News Center

The Petit Institute for Bioengineering and Bioscience will host a conversation next week with two campus leaders on the topic of Covid-19 Is a Health Disparity. The event will feature Kaye Husbands Fealing, dean and Ivan Allen Jr. Chair in the Ivan Allen College of Liberal Arts, with an opening presentation by Manu Platt, associate professor in the Wallace H. Coulter Department of Biomedical Engineering.

The conversation will take place Monday, Oct. 19, from 1 to 2 p.m. via Microsoft Teams, with a maximum capacity of 250 attendees, limited to those logged in with a Georgia Tech email address. Attendees will have the chance to ask questions, and a recording will be made available on thePetit websitefollowing the event.

In addition to the many research angles of Covid-19 including therapeutics, diagnoses, co-morbidities, public health, public policy, and healthcare systems this session will highlight the health disparities associated with Covid-19, providing an additional perspective into this public health crisis.

The discussion has been planned and organized by a committee for diversity, equity, and inclusion within the Petit Institute. The group was formed in September following a June town hall and a collective desire within the community to take action following national race-related tragedies.

The goal of our committeeis to create a safer, more inclusive, and more highly prosperous environment for our historically underrepresented minority faculty, trainees, and staff, said Ed Botchwey, committee chair and associate professor in the Coulter Department. We hope to maintain a focus on anti-racism action within our Institute.

In support of this goal, the group began hosting town halls within the Petit community on ways to think about systemic racism across disciplines.

We must confront the routines, biases, and contradictions that preserve the status quo, Botchwey said. This town hall is a fresh call to antiracist action in the bioengineering and bioscience community. Im looking forward to our conversation as we look for ways in which all of our members can answer the call.

How disassociation occurs in the brain – Medical News Today

Researchers may now have gained a better understanding of how the brain causes the disorienting, disruptive sensation of being outside of ones body.

Most of the time, the mind and body operate seamlessly as a single entity. At times, however, a person may experience a disconcerting sense of disassociation, during which they feel as if they are somewhere outside of their body looking in.

According to Dr. Karl Deisseroth who is a professor of bioengineering, psychiatry, and behavioral sciences at Stanford University in California almost 3 out of 4 people who experience trauma report this phenomenon occurring either during or some time afterward.

Also, around 210% of the general population experience it at some point in their lives, he adds.

Recently, Dr. Deisseroth and colleagues conducted a study that allowed them to identify, for the first time, the brain mechanism that initiates disassociation.

They have now reported their findings in the journal Nature.

The feeling of disassociation begins with nerve cells in the brains posteromedial cortex firing synchronously at a specific rate.

Disassociation can be both troubling and disruptive, and it may become chronic. In order to develop treatments, and to understand the biology, we needed to know more, says Dr. Deisseroth.

Now, he adds, This study has identified brain circuitry that plays a role in a well-defined subjective experience.

Beyond its potential medical implications, it gets at the question, What is the self? Thats a big one in law and literature, and important even for our own introspections.

The study also describes the molecular foundation underlying the mechanism that causes disassociation.

Dr. Deisseroth describes disassociation metaphorically as the perception of being on the outside looking in at the cockpit of the plane thats your body or mind and what youre seeing you just dont consider to be yourself.

Indeed, the study is, in part, based on observations of a person who described the experience as being outside the pilots chair, looking at, but not controlling, the gauges.

The study authors were working with this individual as part of the Stanford Comprehensive Epilepsy Program. He experienced disassociation during preseizure auras, which refers to the time period directly preceding seizures.

The researchers recorded electrical signals from the individuals cerebral cortex in an attempt to identify the brain activity causing his seizures. They noticed a particular pattern of electrical activity that coincided with the aura. It was occurring in the posteromedial cortex.

The researchers observed a particular rhythm being generated by nerve signals firing synchronously. The signal pulsed at 3 Hertz (Hz), or three times per second.

They subsequently found that if they deliberately stimulated this area of the brain electrically, the individual experienced a disassociation aura but no subsequent seizure.

Previous research has suggested that mice experience disassociation as indicated by changes in their behavior.

For example, if a mouse located on an uncomfortably warm surface simply lifts its foot without licking it to soothe it as would be expected it is likely experiencing disassociation. Scientists can induce this state using the drug ketamine.

In the recent study, the researchers attempted to cause disassociation in mice by using light to optogenetically stimulate neurons in the rodent equivalent of the human posteromedial cortex.

When they applied stimulation at the same 3-Hz rate they saw in the human brain, they saw disassociative behavior without the presence of ketamine.

Additional research led to the discovery that a specific protein type called an ion channel is necessary for the generation of the 3-Hz pulse.

Recognizing the role of these proteins could lead to the development of new therapies that more effectively address disassociation and the conditions with which it is often associated, such as epilepsy, borderline personality disorder, and PTSD.

Better Vaccines Are in Our Blood: Using Red Blood Cells to Generate Targeted Immune Responses – SciTechDaily

New platform technology uses red blood cells to generate targeted immune responses in mice.

Red blood cells do more than shuttle oxygen from our lungs to our organs: they also help the body fight off infections by capturing pathogens on their surfaces, neutralizing them, and presenting them to immune cells in the spleen and liver. Now, a team of researchers from Harvards Wyss Institute for Biologically Inspired Engineering and John A. Paulson School of Engineering and Applied Sciences (SEAS) has harnessed this innate ability to build a platform technology that uses red blood cells to deliver antigens to antigen-presenting cells (APCs) in the spleen, generating an immune response. This approach successfully slowed the growth of cancerous tumors in mice, and could also be used as a biocompatible adjuvant for a variety of vaccines. The technology, called Erythrocyte-Driven Immune Targeting (EDIT), is reported in Proceedings of the National Academy of Sciences (PNAS).

The spleen is one of the best organs in the body to target when generating an immune response, because it is one of the few organs where red and white blood cells naturally interact, said senior author Samir Mitragotri, Ph.D., a Wyss Core Faculty member who is also the Hiller Professor of Bioengineering and Hansjrg Wyss Professor of Biologically Inspired Engineering at SEAS. Red blood cells innate ability to transfer attached pathogens to immune cells has only recently been discovered, and this study unlocks the door to an exciting array of future developments in the field of using human cells for disease treatment and prevention.

Using red blood cells as delivery vehicles for drugs is not a new idea, but the vast majority of existing technologies target the lungs, because their dense network of capillaries causes cargoes to shear off of red blood cells as they squeeze through the tiny vessels. Mitragotris research team first needed to figure out how to get antigens to stick to red blood cells strongly enough to resist shearing off and reach the spleen.

They coated polystyrene nanoparticles with ovalbumin, an antigenic protein known to cause a mild immune response, then incubated them with mouse red blood cells. The ratio of 300 nanoparticles per blood cell resulted in the greatest number of nanoparticles bound to the cells, retention of about 80% of the nanoparticles when the cells were exposed to the shear stress found in lung capillaries, and moderate expression of a lipid molecule called phosphatidyl serine (PS) on the cells membranes.

A high level of PS on red blood cells is essentially an eat me signal that causes them to be digested by the spleen when they are stressed or damaged, which we wanted to avoid. We hoped that a lower amount of PS would instead temporarily signal check me out to the spleens APCs, which would then take up the red blood cells antigen-coated nanoparticles without the cells themselves getting destroyed, said Anvay Ukidve, a graduate student in the Mitragotri lab and co-first author of the paper.

To test that hypothesis, the team injected red blood cells coated with their nanoparticles into mice, then tracked where they accumulated in their bodies. 20 minutes after injection, more than 99% of the nanoparticles had been cleared from the animals blood, and more nanoparticles were present in their spleens than their lungs. The higher nanoparticle accumulation in the spleen persisted for up to 24 hours and the number of EDIT red blood cells in the circulation remained unchanged, showing that the red blood cells had successfully delivered their cargoes to the spleen without being destroyed.

Having confirmed that their nanoparticles were successfully delivered to the spleen in vivo, the researchers next evaluated whether the antigens on the nanoparticles surfaces induced an immune response. Mice were injected with EDIT once a week for three weeks, and then their spleen cells were analyzed. Treated mice displayed 8-fold and 2.2-fold more T cells displaying the delivered ovalbumin antigen than mice that were given free nanoparticles or were untreated, respectively. Mice treated with EDIT also produced more antibodies against ovalbumin in their blood than either of the other groups of mice.

Red blood cells could be used as a safe alternative to foreign adjuvants to increase vaccine efficacy and speed vaccine creation. Zongmin Zhao

To see if these EDIT-induced immune responses could potentially prevent or treat disease, the team repeated their three-week prophylactic injection of EDIT into mice, then inoculated them with lymphoma cells that expressed ovalbumin on their surfaces. The mice that received EDIT had about three-fold slower tumor growth compared with the control group and the group that received free nanoparticles, and had lower numbers of viable cancerous cells. This outcome significantly increased the window of time during which the tumor could be treated before the mice succumbed to the disease.

EDIT essentially is an adjuvant-free vaccine platform. Part of the reason why vaccine development today takes so long is that foreign adjuvants delivered along with an antigen have to go through a full clinical safety trial for each new vaccine, said Zongmin Zhao, Ph.D., a Postdoctoral Fellow in the Mitragotri lab and co-first author of the paper. Red blood cells have been safely transfused into patients for centuries, and their ability to enhance immune responses could make them a safe alternative to foreign adjuvants, increasing the efficacy of vaccines and speed of vaccine creation.

The team is continuing to work on understanding exactly how an immune response that is specific to the antigen presented by EDIT is generated by the spleens APCs, and plans to test it with other antigens beyond ovalbumin. They hope to use this additional insight to drive their pursuit of the optimal clinical setting(s) for the technology.

The human body is a treasure trove of elegant solutions to healthcare problems, and while medicine has come a long way in understanding those mechanisms, we are still in the early stages of being able to harness them to improve the length and quality of human life. All of the proposed digital health tools will be developed using multiple data sources, and each participating organization will share data and other assets through an NIH-supported central data hub, in ways that protect individuals privacy. In addition, researchers developing further projects will have access to data stored in the hub, the NIH said.
