The benefit of pelvic lymph node dissection (PLND) at radical prostatectomy (RP) remains unclear given the low prevalence of known nodal disease (pN1) and concerns about its therapeutic utility.

To characterize the impact of PLND and secondary treatment on oncologic outcomes.

Cohort study of men who underwent primary RP with PLND for prostate cancer (PCa) at our institution since 2003. Men stratified by nodal status.

Outcomes include biochemical recurrence-free survival (bRFS), overall survival, and PCa-specific mortality (PCSM). Multivariable Cox regression models used for each outcome.

Of 1,543 men who underwent primary RP, 174 (11%) had pN1 disease. Median follow-up was 34 months (interquartile range, 15-62). Seven-year outcomes were similar whether less than or 14 LNs dissected. Among node-positive patients, 29% had undetectable (UDT) prostate-specific antigen (PSA), 11% had UDT PSA+adjuvant therapy, and 60% had detectable PSA, and 7-year bRFS differed (75% for UDT PSA, 90% for UDT+adjuvant therapy, 38% for detectable PSA, p<.01). Survival outcomes did not differ. In multivariable analysis, detectable PSA (vs. UDT, HR 5.2, 95% CI 2.0-13.3) associated with worse bRFS. After salvage treatment, 7-year outcomes did not differ between groups. Study limited by retrospective review.

$2 Million NIH MIRA Grant Will Support Trailblazing Research in UMass Amherst Lab – UMass News and Media Relations

AMHERST, Mass. Jianhan Chen, a University of Massachusetts Amherst chemistry and biochemistry and molecular biology professor, has received a five-year, $2 million National Institutes of Health (NIH) grant to support research in his computational biophysics lab aimed at better understanding the role of intrinsically disordered proteins (IDPs) in biology and human disease.

The grant falls under the National Institute of General Medical Sciences MIRA program, which stands for Maximizing Investigators Research Award. Its designed to give highly talented researchers more flexibility and stability to achieve important scientific advances in their labs.

The MIRA award enables us to continue working on several central problems regarding the study of disordered proteins and dynamic interactions. The flexibility of this funding mechanism also allows us to follow new research directions as they emerge, Chen says.

Until relatively recently, it was thought that proteins needed to adopt a well-defined structure to perform their biological function. But about two decades ago, Chen explains, IDPs were recognized as a new class of proteins that rely on a lack of stable structures to function. They make up about one-third of proteins that human bodies make, Chen explains, and two-thirds of cancer-associated proteins contain large, disordered segments or domains.

This disorder seems to provide some unique functional advantage, and thats why we have so much disorder in certain kinds of proteins, Chen says. These IDPs play really important roles in biology, and when something breaks down, they lead to very serious diseases, like cancers and neurodegenerative diseases.

In his lab, Chen and colleagues focus on using computer simulations to model the molecular structure and dynamics of proteins. IDPs are a mess; its difficult to determine the details of their properties because they are not amenable to traditional techniques that are designed to resolve stable protein structures, he says.

Because of their chaotic state, IDPs must be described using ensembles of structures, and computer simulations play a crucial role in the quantitative description of these disordered ensembles. Our goal is really trying to combine simulation and experiments in collaboration with other labs to tease out what are the hidden features of these disordered proteins that are crucial to their function, Chen says. Then we can look at how these specific features might be perturbed by disease-related mutations or conditions.

The next step would be to develop effective strategies for targeting disordered protein states. Toward that end, Chens lab will study the molecular basis of how the anti-cancer drug EGCG, an antioxidant found in green tea extract, and their derivatives interact with the p53 gene, a tumor suppressor and the most important protein involved in cancer.

The key, he says, is knowing how to design drug molecules to bind well enough to IDPs to achieve a therapeutic effect. Traditional, structure-based drug design strategies are faced with significant challenges, Chen says, because IDPs do not contain stable, druggable pockets.

We believe that targeting IDPs requires new strategies that explore the dynamic nature of IDP interactions, Chen says. If we can do this, it could really open up a whole class of drugs that were previously thought impossible.

Valedictorian Amanda Salinas and Salutatorian Elizabeth Abila, both students from The Academy of Health Science Professions and STEM, will be graduating with their home school, Juarez Lincoln.Salinas has been chosen as 1 of 8 in the state of Texas to receive early admission into the physician assistant program. She will be graduating with her associate degree in biology from STC and 66 college credit hours.

I had applied for the program this semester and went through an application process and got an interview at UTRGV, Salinas said. Im really glad that I was able to receive this amazing opportunity because this means that Ill have a secured spot on the masters program once I graduate with my bachelors from UTRGV. Having that spot will bring me one step closer to achieving my career goal of becoming a PA.Salinas has always dreamed of working in the medical field, and this past year opened her eyes and pushed her towards that goal even more.My school really focuses on showing their students the different careers in the medical field, Salinas said. And one of them was a PA. When I learned about being a PA, it really inspired me to want to become one. A recent experience that inspired me more was when my dad and sister got sick with COVID. That experience was terrifying for me because I was so worried about them and if they would get better or not. Thankfully because of my dads physician, they did fully recover. I want to do the same for other families and patients in the future.Despite the health scare in her family, Salinas had the support of her school when things got tough.The academy really shaped me into the person I am today because they always encouraged and supported me in my academics, Salinas said. They encouraged me to take rigorous courses and with the support of my high school I was able to apply to the dual enrollment medical science academy at STC. I was attending high school the first half of my day, and then I would attend STC the second half of my day. This really prepared me for college. Once I finish the program, Ill be going to graduate school at UTRGV as well.Salinas love for her family and her community shows in how hard she has worked to achieve valedictorian and pursue her goals.My family means so much to me, especially now, Salinas said. During the pandemic I learned so many lessons about what it means to be a family and I just really love spending time with them as much as I can. We may face challenges that scare us or try to stop us from accomplishing our goals. But in that case, you really have to persevere through those and make the best of the situation. I think weve all learned that from this past year.Salutatorian Elizabeth Abila has plans to attend Michigan State University and double major in Biochemistry and Molecular Biology.

One of the main reasons I chose Michigan State was because I used to have a neighbor that went there, thats how I first heard of it, Abila said. And then I started researching more on the biochemistry program, because thats what I want to major in. They have one of the best in the nation so thats why I decided to apply there, and I got in.Abila credits her future career choice to her kindergarten teacher who went above and beyond for her when she had to miss school for some time.Ive always known that I wanted to be in the medical field since I was in kindergarten, Abila said. I broke my arm when I was in kindergarten and my teacher would always come to my house after school to teach me everything that she taught the class that day so that I wouldnt fail the school year. So that was one of my big influences that kept me going with getting good grades and everything.Abila has a love for children and the medical field and has done exceedingly well first at Juarez Lincoln, and now at the STEM Academy.

I want to be a pediatric heart surgeon, Abila said. My family is really proud of me, especially my mom. Shes telling the whole world.Although school can be tiring and tedious, especially for high achievers, Abila reminds her peers that its not worth it to give up or slack off.You should never give up, Abila said. Even if you feel like you might not get there or if you struggle along the way, at the end of the day, you just need to keep working towards your goals.

Connecticut Guardsman volunteers to support COVID-19 response | Article | The United States Army – United States Army

NEW BRITAIN, Conn. On a cold January day, a team of four Connecticut National Guard Soldiers make their way down a line of vehicles, helping local residents register for COVID-19 tests.

New Britain Stadium, home of a collegiate summer league baseball team and former minor league home to baseball stars like David Ortiz and Joe Mauer, has seen its parking lot converted into a drive-thru COVID-19 testing site in response to the increase in cases associated with the omicron variant.

Among the soldiers supporting the civilian-led site is Pfc. Stefanie Charpentier, a combat medic assigned to the 141st Medical Company.

I joined the Guard because I always wanted to be in the military, and with the Guard, I could serve close to home while going to college, said Charpentier.

In addition to supporting the states response to the COVID-19 pandemic, Charpentier studies biochemistry at Central Connecticut State University also located in New Britain.

I wanted to be a medic in the Guard and be involved in the medical field on the civilian side as well, said Charpentier. I volunteered for this mission so I could help provide access to medical services that are so critical right now.

Charpentier was also activated in 2021 to help administer the COVID-19 vaccine as part of the Guards efforts at vaccination sites across the state.

As Connecticut grapples with the omicron variant, so far approximately 250 Guardsmen have volunteered to assist 24 testing sites and dispense test kits and masks at five regional points of distribution throughout the state. In addition, Connecticut Guardsmen support logistics operations at the states commodities warehouse in New Britain the hub for testing supplies and PPE distributed statewide.

We train to be ready at a moments notice, and throughout this response, weve used our skills to help the people of Connecticut, said Charpentier. It means a lot that Im able to help people in my state, including right here in New Britain, during this time of need.

Charpentier said she hopes the Guards efforts will help the state get tests for everybody who needs one, and that she is appreciative of the support shown by residents at these testing sites.

The Guard is ready to help, and well continue to do everything we can to support our neighbors here in Connecticut, said Charpentier.

Guengerich, Hamm named to inaugural class of ASBMB Fellows – Vanderbilt University News

Fred Guengerich, who holds the Tadashi Inagami, Ph.D. Chair in Biochemistry and is a professor of biochemistry, and Heidi Hamm, who holds the Aileen M. Lange and Annie Mary Lyle Chair in Cardiovascular Research and is a professor of pharmacology, have been named part of the inaugural class of fellows of the American Society for Biochemistry and Molecular Biology.

The newly established honor recognizes the most distinguished members of the professional society.

The ASBMB is the premier scientific society for biochemistry and molecular biology, so recognition as inaugural fellows is a tremendous honor, said Lawrence J. Marnett, Mary Geddes Stahlman Professor of Cancer Research and dean of basic sciences. Both Heidi and Fred are highly accomplished scientists who have done a great deal for the society throughout their careers. Its hard to imagine two more deserving individuals.

Fellows will receive a crystal award and a pin, and will be recognized with their own profiles on the ASBMB website and at the annual ASBMB meeting, held this year between April 27 and April 30.

Guengerich was nominated by Marnett himself. Dr. Guengerichs extraordinary accomplishments in scientific research are matched only by his devotion to the ASBMB. Guengerich has been an active member of the society since 1978.

For Guengerich, the path was always clear. Ever since I was a sophomore in college, he said, the only career I wanted was to be a biochemist. A native of Pekin, Illinois, Guengerich completed a Ph.D. in biochemistry at Vanderbilt University and returned, after a brief postdoctoral fellowship at the University of Michigan Medical School, to join the faculty of the Department of Biochemistry.

Guengerichs research aims to understand the role of cytochromes P450, or CYPs, as central players in both metabolic and biosynthetic pathways and in drug metabolism related to health and disease. CYP enzymes help detoxicateor remove the harmful effects ofchemical carcinogens and toxicants, and have central roles in hormone synthesis and breakdown. Throughout his academic career, Guengerich has published over 1,000 papers, reviews, and book chapters that have collectively been cited over 100,000 times.

Aside from research excellence, Guengerich is committed to the training of newer generations of biochemists. This commitment to mentoring was rewarded in 2019 with the receipt of the Richard Armstrong Mentoring Award. Among his numerous national and international awards are the William C. Rose Award from the ASBMB, an honorary doctorate from the Universit de Paris, an appointment as one of the inaugural American Chemical Society fellows, an appointment as an American Association for the Advancement of Science fellow, and the receipt of the Award for Outstanding Achievement in Chemistry in Cancer Research from the American Association for Cancer Research.

Following years of service on the editorial board of the Journal of Biological Chemistry, published by ASBMB, he became an associate editor in 2006 and a deputy editor in 2013, a role in which he continues to serve. He also served as interim editor-in-chief in 20152016.

Working with the ASBMB and the Journal of Biological Chemistry has been a great experience for me, Guengerich said. I was very thrilled to be named to the inaugural class of fellows. It really means a lot to me to have come to this point, and I thank everyone along the way.

Hamm was nominated by departmental colleague Tina Iverson, who holds the Louise B. McGavock Chair and is a professor of pharmacology. According to Iverson, Heidis long service to ASBMB, her national contributions to science advocacy, and her stature as a world leader in the field make Hamm an ideal member of the first cohort of ASBMB fellows. Hamm has been a member of ASBMB for many years and served as president of the organization from 2006-2008.

Hamm obtained her Ph.D. training at the University of Texas at Austin. Following a postdoctoral fellowship at the University of WisconsinMadison, she held faculty appointments at the University of Illinois College of Medicine at Chicago and Northwestern University before moving to Vanderbilt in 2000. Her research focus has centered on G protein-coupled receptor signaling, as well as on the structure and function of GTP binding proteins. GTP serves as a source of energy used for signal transduction processes.

While serving as the chair of the Department of Pharmacology between 2000 and 2014, Hamm oversaw an increase in its size and a quintupling of its NIH funding, catapulting it as the top NIH-funded pharmacology department in the nation during her tenure.

Hamms research efforts have been recognized through numerous awards, including the Robert R. Ruffolo Career Achievement Award in Pharmacology from the American Society for Pharmacology and Experimental Therapeutics, the Fritz Lipmann Memorial Lectureship from ASBMB, and the Ariens Award from the Dutch Pharmacological Society. Hamm was also elected a AAAS fellow in 2011.
