Intarcia Enrolls First Patients in " FREEDOM " Global Phase 3 Program for ITCA 650 in Type 2 Diabetes

BOSTON, March 21, 2013 /PRNewswire/ -- Intarcia Therapeutics, Inc. announced today the enrollment of the first patients in FREEDOM-1, the first of four planned Phase 3 clinical trials studying the safety and efficacy of ITCA 650 (continuous subcutaneous delivery of exenatide) for the treatment of type 2 diabetes (T2D). Intarcia is conducting the FREEDOM clinical program with strategic partner Quintiles, Inc., the world's leading clinical research organization, who will be responsible for the global implementation of all four trials. This collaboration, along with strategic input from a world-class group of renowned diabetes experts, is designed to achieve the highest standards of quality, safety and operational excellence.

The FREEDOM clinical trial program aims to demonstrate that optimizing GLP-1 therapy with once- or twice-yearly dosing of ITCA 650 results in effective, well tolerated glycemic control that virtually ensures longer-term compliance a transformational approach to controlling T2D that aims to free patients from the frequent self-injections that all too often result in poor therapeutic compliance and control. The FREEDOM-1 study is a placebo-controlled, double-blind study involving T2D patients whose HbA1c is not controlled on zero to three oral anti-diabetes drugs and whose HbA1c measures between 7.5% and 10.0%. The 450 patients will be randomized into three groups in a 1:1:1 ratio, evaluating ITCA 650 40 mcg/d and ITCA 650 60 mcg/d versus placebo. Subjects in the active arms will be treated with 3-month devices for the first 13 weeks that deliver an initial dose of 20 mcg/d, and then treated with 6-month ITCA 650 at doses of 40 or 60 mcg/d. The primary endpoint will be change in HbA1c at 9 months; secondary endpoints include changes in weight, FPG, PPG, tolerability, and other measures of safety and efficacy.

Michelle Baron, MD, FACE, Vice President and Chief Medical Officer of Intarcia, commented on the initiation of the FREEDOM Phase 3 program: "We named our program 'FREEDOM' in recognition of the burden that patients with type 2 diabetes face every day of their lives, and our sincere hope that ITCA 650 will alleviate some of that burden by achieving glucose control in a manner that optimizes patient adherence to therapy. It is well documented that sub-optimal adherence to medication results in poor outcomes and disproportionately higher medical costs. Our goal is to demonstrate that ITCA 650 can successfully deliver up to a full year of treatment from a single placement of the mini-pump a potential breakthrough in addressing a huge unmet need in this chronic disease. We expect these studies to support regulatory submissions in both the U.S. and the EU, and anticipate the first clinical results in the second half of 2014."

"Our vision is to unleash a game-changing type 2 diabetes treatment that is so innovative and disruptive that it could truly reshape the standard of care and enable far better outcomes for the many patients suffering from this devastating disease," said Kurt Graves, Chairman, President and CEO of Intarcia. "Years of innovation and tenacity have paved the way for the first potential once-yearly type 2 diabetes medicine and the start of the FREEDOM program. We firmly believe that a once-yearly GLP-1 therapy that virtually ensures long-term compliance can do extremely well in any real-world outcomes comparative trials and in the payor-driven and outcomes-focused healthcare environment of the future. As a Company committed to delivering superior outcomes for patients, physicians, and payors, the enrollment of the first patient in our FREEDOM clinical series is very exciting and gratifying milestone for us."

Concurrent with FREEDOM-1, Intarcia will also be conducting an open-label study, called FREEDOM-1 HBL (high baseline), in patients who meet all eligibility criteria for FREEDOM-1, but whose baseline HbA1c is greater than 10%. All patients in this study will be treated with ITCA 650 20 mcg/d for the first 3 months and with ITCA 650 60 mcg/d for the next 6 months.

About the FREEDOM Program

The FREEDOM program is designed to evaluate the safety and efficacy of ITCA 650 (continuous subcutaneous delivery of exenatide) for the treatment of type 2 diabetes. Intarcia expects to enroll greater than 4,000 patients at more than 500 clinical trial sites in more than 30 countries. The clinical studies will cover a broad range of patients whose diabetes is not controlled by oral anti-diabetes medications including metformin and metformin-based combinations. FREEDOM-1 is a U.S.-only, placebo-controlled, double-blind study comparing ITCA 650 doses of 40 mcg/d and 60 mcg/d to placebo; the FREEDOM-2 study will be a 500-patient, global, active-comparator controlled, double-blind, double-dummy study in patients on metformin only, comparing ITCA 650 20 mcg/d for 13 weeks plus ITCA 650 60 mcg/d for 39 weeks to patients taking sitagliptin (Januvia, Merck); FREEDOM-CVO is a global, placebo-controlled cardiovascular outcomes study that will examine the safety of ITCA 650 at 60 mcg/d vs. placebo in approximately 3,000 patients on a variety of approved anti-diabetes therapies.

About ITCA 650

ITCA 650 (continuous subcutaneous delivery of exenatide) is being developed for the treatment of type 2 diabetes. The investigational therapy employs Intarcia's proprietary technology platform involving a matchstick-size, miniature osmotic pump that is inserted subcutaneously to provide continuous and consistent drug therapy, and the company's proprietary formulation technology, which maintains stability of therapeutic proteins and peptides at human body temperatures for extended periods of time.

Cyborg Santos Blasts Twitter Haters, Says She’s Open to Fighting Fallon Fox

As long as the fighter is licensed by a commission, Cristiane "Cyborg" Santos is willing to step into the cage against anyone, including male-to-female transgender fighter Fallon Fox.

The former Strikeforce women's featherweight champ recently spoke with Josh Gross of ESPN.com about the controversy surrounding Fox's quest to become a licensed female fighter.

She wants to be a girl. I don't agree. I think you're born a girl, you're a girl. You're born a guy, you're a guy. But I don't choose opponents. The commission needs to check and make sure she doesn't have testosterone.

In a story broken by Loretta Hunt, Fox revealed to Sports Illustrated she had undergone sex reassignment surgery back in 2006, which also included supplemental hormonal therapy.

The fallout from the story has included a heated debate involving the ethical standards set by the sport. During an episode of The Joe Rogan Experience podcast, UFC commentator Joe Rogan argues that while Fox has undergone the necessary surgical augmentations, she still has the bone density and physical makeup of a man.

Former Strikeforce women's bantamweight champ Miesha Tate told ESPN.com she wouldn't fight Fox based on fears and questions involving her own personal safety.

Cyborg, on the other hand, has no qualms about fighting Fox, as long as she passes the necessary requirements for licensure.

Even though she was born a woman, Cyborg can relate to some of the cruelties faced by Fox in the media and amongst MMA fans.

People tell me on Twitter: 'I think you have a d---.' A lot of bad things, they say. I think people have a small mind. They don't think a girl can punch hard like a man. I think people are ignorant. People are stupid. I don't want to be the same as people who do that.

...I'm not going to judge other people. If the commission says she can fight, why not?

Rockaway Residents To City: Rebuild Our Beaches

A home along the shore in the Rockaways ravaged by superstorm Sandy Nov. 2, 2012 (credit: Spencer Platt/Getty Images)

NEW YORK (CBSNewYork) Residents of Rockaway Beach descended on City Hall in Lower Manhattan Saturday, demanding that their beaches be rebuilt right now.

As WCBS 880s Jim Smith reported, residents of the Rockaways said they feel ignored in the wake of Superstorm Sandy, which devastated the peninsula in Queens.

Theyve worked on Coney Island. Theyve worked on Long Beach. Were just as entitled. We pay taxes just like everybody else, said Milly Orenga. We need our beaches.

Orenga is one of about 50 residents taking the message directly to the steps of City Hall after they said a previous rally on the beaches fell on deaf ears.

The beaches shouldve been built by now. The sand should be there, the dunes, the water walls, Orenga said. We have nothing absolutely nothing.

Residents said what the city now has is plenty of federal money. And Eddy Pastore, founder of Friends of Rockaway Beach, demanded action before another hurricane season.

Were in imminent danger of flooding again, Pastore said.

Beaches from N.C. to Miss. critical loggerhead turtle habitat

CHARLESTON, S.C. (AP) A federal agency is designating 740 miles of beaches from North Carolina to Mississippi as critical habitat for loggerhead sea turtles, including 175 miles in the Carolinas.

While loggerheads have been listed under the Endangered Species Act since 1978, the U.S. Fish and Wildlife Service in 2011 changed the loggerhead listing from a single worldwide designation to nine distinct groups to focus on need for conservation in specific areas.

So the nesting areas are now being designated as critical habitat for the recovery of the species threatened in the northwest Atlantic. The areas selected have the highest densities of loggerhead nests.

Only occupied habitat is being proposed as we determined this is adequate for the conservation of the species, Sandy MacPherson, the national sea turtle coordinator for the U.S. Fish and Wildlife Service, told reporters Friday.

She said that about 19 percent of the shoreline in the six states is owned by the federal government; 21 percent is owned by state governments; and the rest is private.Continue Reading

We do not envision additional landowner use restrictions that will result from this critical habitat designation, she said, adding most coastal landowners are aware of state and federal rules designed to protect sea turtles.

Its an informative process that identifies the habitats necessary for the recovery of the species, said Jennifer Koches, a spokeswoman for the Fish and Wildlife Service in Charleston. Anytime there is a federal project such as beach renourishment or dredging activities things that are funded or permitted by the federal government applicants have to consult for impacts on federal species.

At the time the agency changed the loggerhead designation to nine groups, it said it did not have enough data to identify the critical habitat. In January, three nonprofit groups, including Oceana Inc., sued in federal court in San Francisco seeking to have those designations made.

Its something were required to do ... and it was something we were working on when we received the complaint, MacPherson said.

Amanda Keledjian, a marine scientist with Oceana, said the National Marine Fisheries Service should now designate (as critical) off-shore areas as well as waters directly adjacent to nesting beaches if they want these vulnerable populations to recover. Sea turtles often get tangled in trash such as fishing lines or are hit by boat propellers and injured.

Florida beaches proposed as turtle “critical habitat”

Nearly 150 miles of Florida beaches, including all of Palm Beach County and stretches in northern Broward and the Florida Keys, could be designated by federal wildlife managers as critical habit for loggerhead turtles.

The U.S. Fish and Wildlife Service, which released the proposal on Friday, stressed that the designation wouldnt affect landowner rights and was largely intended to draw attention to the importance of 90 beaches in six southeastern states which are prime nesting areas for the threatened species.

In Florida, much of the proposed area runs from Brevard County south to Palm Beach County. An eight-mile section of northern Broward would be included as well as Long Key and Bahia Honda in the Keys.

Identifying this habitat will help us work with coastal communities to protect loggerhead nests and ensure that more hatchlings reach the water and begin their lives at sea, said Cindy Dohner, the Services Southeast Regional Director. Major threats to turtles including artificial lighting, beach erosion and climate change.

All of Folly Beach and Kiawah Island among 41 miles of beaches in Charleston County would be made critical habitat for the threatened loggerhead sea turtle under a proposal made Friday by the U.S. Fish and Wildlife Service.

To read the U.S. Fish and Wildlife Service critical habitat proposal or for more information, go to fws.gov/ northflorida.

What difference that would make is anybodys guess.

New homes, businesses or other structures to be built at the beaches would be scrutinized closer by federal regulators before permits could be issued, said a news release by Oceana, an environmental advocate.

Fish and Wildlife staff members said the designation was required by law and shouldnt greatly affect how permits are handled now.

It might be that both are true. The designation is being proposed for beaches that hold the most turtle nests year to year. Those beaches already are protected somewhat under the Endangered Species Act and other federal rules.

But if nothing else, the designation would put more pressure on regulators to get it right when it comes to modifying projects or requiring mitigation or it would give environmental opponents more ammunition in lawsuits to stop a project.

More than 700 miles of beaches from North Carolina to Mississippi would be made critical habitats if the proposal is approved.

More than 40 beaches crucial to loggerhead sea turtles targeted for critical habitat list

More than 40 Florida beaches considered vital for loggerhead sea turtles including Canaveral National Seashore could be named as critical habitat under a federal proposal announced Friday.

Several Volusia and Brevard beaches could be designated by the U.S. Fish and Wildlife Service as vital areas to be protected for the turtles, which lay their eggs in the beach dunes.

The proposal isn't expected to change anything for local beachgoers, but it could affect federal permits or projects on such beaches, such as beach renourishment projects on eroded areas, said Sandy MacPherson, the service's national sea-turtle coordinator.

For example, this would require that a dredging or sand project at a critical beach cannot change the beach so that it would no longer serve as a nesting area for turtles.

Florida beaches are among the world's most important nesting grounds for loggerhead sea turtles. Last year, more than 58,000 nests were counted at the state beaches tracked for long-term trends, according to the Florida Fish and Wildlife Conservation Commission.

Beaches included in the federal proposal include Canaveral, Merritt Island National Wildlife Refuge, Kennedy Space Center and Brevard beaches south of Patrick Air Force Base. In Volusia, beaches north of State Road 40 in Ormond Beach and Ormond-by-the-Sea are included.

However, the federal proposal doesn't include beaches in the Daytona Beach region, including the areas where beach driving is permitted. Officials said that doesn't mean other beaches aren't good habitat for turtles, but that the federal designation focuses on the areas with the highest densities of nests which are considered crucial for the species' survival.

Environmental groups had sued the federal government for the lack of such habitat designations and praised the federal proposal.

"It will benefit the turtles because it will be easier to ensure that the quality and quantity of beaches will not be degraded over time," said Amanda Keledjian, a marine scientist at Oceana, a nonprofit conservation group.

Aerospace plan in progress

KUALA LUMPUR: Malaysia is on track to become a global aerospace player by 2015 in line with the primary target of the country's National Aerospace Blueprint.

Launched in 1997, the blueprint was prepared by the Malaysian Industry-Government Group for High Technology (Might).

Its senior vice-president, Might Industry Intelligence, Datuk Kamarulzaman Zainal said since the formation of the steering body, the Malaysian Aerospace Council (MAC) in 2001, the industry had rapidly expanded to post annual growths of between 6% and 7%.

In respect of implementing the proposals contained in the National Aerospace Blueprint, we have completed 80%, and are on track to attain the goal of becoming an important global player. This is due to the emergence of local companies which are gaining prominence in the field. It is also supported by four key segments, namely, services maintenance, maintenance, repair and overhaul (MRO), manufacture of aeronautical aircraft components and systems integration, as well as human capital training and development, he told Bernama.

He said overall, the industry last year recorded an income of RM30.2bil, with 65,000 jobs having been successfully created to date.

Kamarulzaman said for the MRO sector, an income of RM5bil was recorded by industry players, close to the RM8bil target in 2015, while for the manufacture of aircraft components, about 8,000 workers with a variety of skills and 1,500 engineers are expected to be produced.

For aeronautics and systems integration, we have set the key performance index such that local companies can effectively expand and compete at the top tier from their present second spot. For human capital training and development in the aerospace industry, we have set no targets in respect of income, but have projected for it to become a regional centre, he added.

Commenting further, he said the sectors are also the key focus of Might in creating high-income job opportunities, alongside technopreneurs.

To further strengthen the industry, Might, as the secretariat to the MAC, would sponsor an industry forum at the Langkawi International Maritime and Aerospace Exhibition (LIMA 2013) from March 26-30.

It would focus on the development of logistics based on performance, with a representative of Boeing as the main speaker.

Maternal And Family Health Services WIC Program Celebrates National Nutrition Month

During National Nutrition Month, MFHS is raising awareness about the public health value of the WIC Program, the compelling evidence of its success in ensuring healthy outcomes, and the strong public support for the program. Maternal and Family Health Services administers the WIC Nutrition Program in Northeast Pennsylvania, serving more than 57,000 people each month at WIC Nutrition Centers in 16 counties.

Wilkes-Barre, Pennsylvania (PRWEB) March 21, 2013

During National Nutrition Month, MFHS is raising awareness about the public health value of the WIC Program, the compelling evidence of its success in ensuring healthy outcomes, and the strong public support for the program.

WIC is a preventative public health nutrition program designed to influence lifetime nutrition and health behaviors in a targeted, high-risk population. WIC provides nutrition and breastfeeding education, nutritious foods, and improved health care access to low-income pregnant women and children under age 5 with, or at risk of developing, nutrition-related health problems.

Established to promote good nutrition for children and expectant mothers, WIC has earned the reputation of being one of the most successful, cost-effective, federally-funded nutrition programs in the United States. More than half (53%) of all children born in the United States are served by the WIC Program.

The WIC food package, which was updated in 2009 to include a greater variety of healthy choices, has a positive impact on how children eat. A recent study by Public Health Solutions in New York showed that following the improvements with the food package, children participating in WIC are more likely to have increased consumption of healthy foods, including low / non-fat milk, and infants enrolled in WIC are more likely to be breastfed. In a study completed for the Mid-Atlantic WIC Assessment Project showed that the majority of participants (nearly three-quarters) interviewed report that they have made changes in eating practices and physical activity since being a part of WIC. Improvements include: increased physical activity, increase consumption of fruits and vegetables, whole grains and low-fat milk, and decreased consumption of soda and fast food.

Participation in WIC improves nutrition, resulting in overall healthier pregnancies, healthier birth outcomes, and better growth and development of young children. WIC helps ensure normal physical growth for infants and children, and has been shown to improve cognitive development, reduce levels of anemia, improve access to regular health care and social services, improve diets and household health behaviors, and improve breastfeeding rates. WIC children arrive at school with a healthy foundation and ready to learn.

The scientifically based WIC food package reinforces the nutrition education provided by WIC nutritionists, and provides supplemental foods with essential nutrients. WIC approved foods include whole grains, low-fat dairy, fresh fruits and vegetables, soy and tofu, eggs, baby food, tuna, salmon, sardines, and mackerel, canned and dry beans, peanut butter, juice, and iron-fortified infant formula.

WIC promotes breastfeeding as the most perfect infant food and provides support for new mothers who choose to breastfeed. The MFHS WIC Breastfeeding Peer Counselor Program works to increase breastfeeding initiation and duration rates for WIC participants. Peer counselors provide new mothers with support and encouragement during the early post-partum period and help moms make the transition back to work or school.

With all the public health benefits of the WIC Nutrition Program, it should not come as a surprise that WIC is highly regarded among American voters. A recent national bipartisan poll conducted by the Mellman Group and American Viewpoint regarding voter attitudes toward the WIC Program showed that showed that Americans - across party lines - strongly support WIC, and not only do voters oppose cuts to WIC, they want the government to invest more money in programs like WIC. When ranked by favorability, WIC is among the most popular government programs including programs such as Social Security, Medicare, and National Defense. Opposition to cutting WIC is strong across party lines: Democrats (80%), Independents (55%), and Republicans (45%). Instead of cuts, a majority of voters want the federal government to invest more money in programs like WIC that provide nutrition and healthcare to pregnant women and children. Opposition to cutting WIC remains strong even after voters hear detailed arguments for and against such spending reductions.

Pete Thamel: Longevity defines Blue Devils assistants run with Krzyzewski-led Duke

Chris Collins (right) and Steve Wojciechowski (left) have spent nearly three decades combined as assistants under coach Mike Krzyzewski.

Rich Barnes/Icon SMI

PHILADELPHIA -- Perhaps the best way to quantify Duke's generation of dominance in college basketball is through NCAA seeding: In the past 17 years, the Blue Devils have made the tournament 17 times, receiving a No. 1 or No. 2 seed 14 times.

Coach Mike Krzyzewski's presence represents the obvious constant, but an underrated factor in Duke's supremacy has been the continuity of the staff.

Duke assistants Chris Collins and Steve Wojciechowski have a combined 27 years of experience under Krzyzewski -- Wojo 14 years and Collins 13 -- an unusually long stretch for assistants at an annually successful high-major program. Consider assistant coach turnover at programs like Louisville, Florida and Pittsburgh and the longevity of Duke's assistants is appreciated.

"Continuity is kind of king in this profession," said Duke Athletic Director Kevin White.

With the Chicago Tribune reporting that Collins will interview for the open Northwestern job, the question again looms as to why Duke's assistant coaches stick around so long. Johnny Dawkins spent 11 years at Duke before taking the Stanford job five seasons ago. Collins and Wojciechowski, who both hold the title of associate head coach, have flirted with opportunities but have never left.

"All of us have aspirations to be head coaches," Collins said. "It has to be something that we all believe in because we do have such a good job."

Jeff Capel, the staff newbie at just two years, recalls hearing people criticize Collins and Wojciechowski because they haven't taken head coaching jobs. Collins turns 39 in April. Wojo is 36.

"Those guys have gotten knocked because they haven't left," Capel said. "I know when I was a head coach, I heard that. It's easy to say on the outside when you don't know. This is a great job. You don't leave this job just for any job."
