Biochemist Jennifer Doudna hailed for discovery of ‘holy grail’ of genetic engineering

Some of the biggest discoveries in science often hide away in plain sight for many years before their importance is fully realised. This is certainly true of Crispr (pronounced crisper), which has taken the world of genetics by storm.

Crispr stands for clustered regularly interspaced short palindromic repeats, a devilishly contrived acronym which just about sums up why it was ignored for so long. For nearly two decades after Japanese researchers first discovered Crispr in bacteria in 1987, scientists mostly dismissed it as junk DNA.

In fact, the apparently nonsensical sequences within Crispr, which were repeated in palindromic order (the same backwards as forwards), did have a purpose and were far from junk. About six years ago, scientists discovered that these DNA sequences matched the genetic sequences of various viruses that attack bacteria, which led to the discovery of a sophisticated bacterial immune system.

Far from being junk, Crispr was actually a way of storing the genetic information of an invading virus in the form of a palindromic DNA sequence. The bacteria used this genetic memory to target the viral invader by chopping it up with powerful Crispr-associated (CAS) enzymes capable of cleaving its DNA molecule, just like a pair of molecular scissors.

The mystery of Crispr was finally resolved by Jennifer Doudna of the University of California, Berkeley, a specialist in RNA, the smaller molecular cousin of DNA. About seven years ago, she was asked by a university colleague to look into this genetic peculiarity of bacteria and quickly became fascinated.

The more we looked into it, the more it seemed extremely interesting, Professor Doudna said. Then, in 2011, she met Emmanuelle Charpentier of Umea University in Sweden at a scientific conference. Professor Charpentier told Professor Doudna of another kind of Crispr system that seemed to rely on a single gene, now called CAS9.

Both professors collaborated on the project and in August last year published what is now considered the seminal paper showing that CAS9 was an enzyme capable of cutting both strands of a DNA double helix at precisely the point dictated by a programmable RNA sequence in other words, an RNA molecule that could be made to order. We found that CAS9 has the ability to make a double-stranded break in DNA at sites that are programmed by a small RNA molecule. What was so important was that we could really show how the CAS9 protein worked, Professor Doudna said.

Not only were we able to work out how it worked, we were able to reprogramme it to recognise new DNA sequences. If it could be made to work in eukaryote systems plants and animals then youd have a system where you could effectively decide where to produce a double-stranded break in that cells genome.

It not only worked on plants and animals, it worked beautifully. Professors Doudna and Charpentier had found the holy grail of genetic engineering a method of cutting and stitching DNA accurately and simply anywhere in a complex genome. Until now, this was carried out by modified viruses, which inserted their DNA at random, or by elaborately cumbersome techniques known as zinc fingers or Talens.

You can actually introduce new genetic information at the site of cleavage. So it has become a powerful way of doing genetic engineering. Its a fundamentally different way of recognising DNA target sites, Professor Doudna said.

The Glamour of Futurism: An Interview with Author Virginia Postrel

Futurism and glamour are inextricably linked in American history. The sleek techno-utopian futures of yesteryearthe ones filled with flying cars, jetpacks, and automatic highwayscouldn't exist without the support of this concept that's equal parts intrigue and attraction.

Virginia Postrel's new book, The Power of Glamour: Longing and the Art of Visual Persuasion, looks at glamour as a powerful force in our culture. I talked with Postrel, who writes a regular column for Bloomberg, about the role that glamour has played in shaping our expectations for the futurefrom the Futurama exhibit of the 1939 World's Fair to the Jet Age dreams of the 1960s.

I also asked Postrel about glamour in the futurism of today, and why the two concepts might feel less connected than ever. Can we embrace a glamorous future when the headlines are filled with techno-centric fears? And is there perhaps a dash of glamour mixed in with our modern concerns about tomorrow?

Gizmodo: How do you define glamour?

Postrel: I think of glamour as a form of communication, persuasion, rhetoric. What happens is you have an audience and you have an objectsomething glamorous. It could be a person, could be a place, could be an idea, could be a carand when that audience is exposed to that object a specific emotion arises, which is a sense of projection and longing.

Glamour is like humor. You get the same sort of thing in the interaction between an audience and something funny. Its just the emotion thats different. So when you see something that strikes you as glamorous, or you hear about or see something glamorous, it makes you think, If only. If only life could be like that. If only I could be there. If only I could be that person, or with that person. If only I could drive that car, fly in that spaceship, or whatever.

And there are always three elements that create that sensation: one is a promise of escape and transformation. A different, better life in different, better circumstances. The other is there is a sense of grace, effortlessness, all the flaws and difficulties are hidden. And the third is mystery. Mystery both draws you in and enhances the grace by hiding things.

Another way of thinking about glamour is to think about the origins of the word glamour. Glamour originally meant a literal magic spell that made people see something that wasnt there. It was a Scottish word. A magician would cast a glamour over peoples eyes and they would see something different. As the word became a more metaphorical concept, it always retained that sense of magic and illusion. And where the illusion lies is in the grace; in the disguising of difficulties and flaws.

Gizmodo: Why do you think Americans remember the 1920s and '30s as the Golden Age of Glamour, and what role did futurism play during this era?

Postrel: Glamour was incredibly important in the 1920s and 30sin the interwar period. We remember it as a Golden Age of Glamour partly because of the association with glamour and the movies. But glamour occurred in many different forms during that periodeverything from the first superhero comic books to streamlined design.

Freedom Angel watching over veterans

ROXBURY, Conn. (WTNH) -- A seven foot angel statue now stands near Roxbury Town Hall. It's a "freedom angel," keeping watch over veterans.

"I invented the Freedom Angel as a new national symbol," said Sterett-Gittings Kelsey, Freedom Angel Founder. "And the reason behind that is not because we need another new monument, we don't. But what we need is a way to reach the rest of America."

The creator hopes to reach Americans by one day making angel pins and other memorabilia. The proceeds will benefit veterans organizations.

If you're a hospital and you want to do research on PTSD or whatever, we can fund that," Kelsey said. "If Wounded Warriors say we need more help, we can do that."

The Freedom Angel Foundation is new, unveiling a 3,000 lb. statue Wednesday.

"She's beautiful, just gorgeous," said visitor Linda Chapman. "For people who have family that are in the service. It's just so so important that we have something that we can look up."

The angel will be in the town of Roxbury for six months, the plan after that is to take her touring across the country.

"She will be the most recognized symbol in the country and everybody will know what she stands for," said Kelsey.

The spirit of freedom for the men and women who fight to protect it.

Freedom Alliance Provides College Scholarships to 270 Children of Military Heroes

Freedom Alliance, an educational and charitable organization that supports military personnel and their families, has awarded college scholarships to 270 children of military heroes in 2013. The scholarships total $1.3 million and were awarded to students whose parent is a Soldier, Sailor, Airman or Marine who was killed or permanently disabled in a combat mission or training accident.

"These scholarships are made available," explained Freedom Alliance President Tom Kilgannon, "because these students have a parent who sacrificed life or limb defending our country. Both the parents and the students have made tremendous sacrifices for our country which we hope to recognize and honor with these scholarships."

In total, Freedom Alliance has now awarded more than $7 million in scholarship assistance to students to help them finance their college educations.

"This Scholarship Fund," Kilgannon said, "is a living memorial to military heroes. Each scholarship is a practical act of support for a family that has made incredible contributions to our country. Each scholarship is also recognition for a patriots sacrifice and a reminder to his or her child that the sacrifice of their parent will never be forgotten by a grateful nation."

Among the scholarship recipients are students whose parent served in Operation Enduring Freedom, Operation Iraqi Freedom, the Persian Gulf War, the 1983 terrorist attack on the Marine Barracks in Beirut, the Battle of Mogadishu in 1993, and other military campaigns in defense of our country.

Kalie Walters is one of Freedom Alliances scholarship recipients. She recently graduated from the University of West Florida, and is currently serving an internship with the organization. Walters father, Tech. Sgt. Howard Walters, died in a helicopter crash in November of 2003 in Afghanistan. He was assigned to the 20th Special Operations Squadron in Hurlburt Field, Florida. Walters was 13 years old at the time of her fathers death.

The Scholarship Fund is one of several programs sponsored by Freedom Alliance in support of those who serve our country. To be eligible for the Freedom Alliance Scholarship, students must have a parent that has become 100 percent disabled or deceased due to an operational mission or training accident, and under the age of 26.

Freedom exactly where it expected to be

FREEDOM A year ago when Freedom was marching through the playoffs it surprised many being a No. 8 seed and not having a track record of winning in the postseason.

This year, the Irish is seeded No. 2 and is just where everybody expected it to be Level 3.

No surprises this year but the Irish do want to at least match its accomplishment last season when it reached Level 4. To do that, Freedom, which has outscored its first two playoff opponents by a combined score of 63-8, has to win at No. 1 Roncalli in the GameTime Game of the Week.

"We're playing good right now, everything's clicking, we're peaking at the right time, senior defensive end/tight end Mason Sturm said. Our coach always says just a steady incline all season and we're peaking perfect.

Like last season, Freedom (8-3) is finding success in the playoffs with its running game. Last year, Nathan Peters did it on his own, leading the state with 2,249 rushing yards.

This season, Peters battled an ankle injury and has had help. Peters has 1,185 yards and 20 TDs, while Isaac Gonnering has added 511 yards and 11 TDs.

We have two backs, theyre battling between each other for carries, coach Clint Kriewaldt said. Each one wants more of the carries. Its fun to see these guys duke it out and fight for the carries. Theyve both been running pretty hard for us.

And they will need to Friday, as Freedom will be on the road for the second week in a row after winning last week at Campbellsport 30-8. Notwithstanding, Freedom almost embraces road games.

"If you look at all our losses the last two years, every one is on this field here, Kriewaldt said. I don't know what it is, but for whatever reason it seems we come together more on that bus ride there.

I think we get the kids away from, whether it be their girlfriends, just get them out of their house and get focused on football right away. So going on the road isn't a big deal. It should be a pretty fun atmosphere to play in Friday night."
