Breakthrough shows how DNA is 'edited' to correct genetic diseases

An international team of scientists has made a major step forward in our understanding of how enzymes 'edit' genes, paving the way for correcting genetic diseases in patients.

Researchers at the Universities of Bristol, Mnster and the Lithuanian Institute of Biotechnology have observed the process by which a class of enzymes called CRISPR pronounced 'crisper' bind and alter the structure of DNA.

The results, published in the Proceedings of the National Academy of Sciences (PNAS) today, provide a vital piece of the puzzle if these genome editing tools are ultimately going to be used to correct genetic diseases in humans.

CRISPR enzymes were first discovered in bacteria in the 1980s as an immune defence used by bacteria against invading viruses. Scientists have more recently shown that one type of CRISPR enzyme Cas9 can be used to edit the human genome - the complete set of genetic information for humans.

These enzymes have been tailored to accurately target a single combination of letters within the three billion base pairs of the DNA molecule. This is the equivalent of correcting a single misspelt word in a 23-volume encyclopaedia.

To find this needle in a haystack, CRISPR enzymes use a molecule of RNA - a nucleic acid similar in structure to DNA. The targeting process requires the CRISPR enzymes to pull apart the DNA strands and insert the RNA to form a sequence-specific structure called an 'R-loop'.

The global team tested the R-loop model using specially modified microscopes in which single DNA molecules are stretched in a magnetic field. By altering the twisting force on the DNA, the researchers could directly monitor R-loop formation events by individual CRISPR enzymes.

Freedom Summer to Empower Youth

Albert Sykes grew up in west Jackson the next block over from the street where Medgar and Myrlie Evers lived, but says he never had a real connection to the Civil Rights Movement until he was in the sixth grade and met Bob Moses.

Moses was the former leader of the Student Nonviolent Coordinating Committee and started the Algebra Project in 1982 to help improve math education for kids of color.

Today, Sykes remains involved with the Young People's Projectan outgrowth of the Algebra Projectand is one of the organizers of the Freedom Summer Youth Congress and Freedom Fest, which coincides with the 50th anniversary of Freedom Summer this year.

During the Freedom Summer of 1964, young people gathered in Ohio for orientation sessions before boarding buses to head to Mississippi to do justice organizing work. Sykes said that this time around, Mississippi will act as the training ground for young organizers before they are dispatched to locales around the nation.

Speaking at Koinonia Coffee House's Friday Forum, Sykes and fellow Freedom Summer Youth Congress and Freedom Fest organizer Jed Oppenheim said the Congress would address the intersectionality of issues facing young people, including the criminalization of normal youth behavior, LGBT bullying, labor rights and challenging voter ID.

A special effort will also focus on raising money to offset the $50 registration fee for local students in Jackson.

Lately, the challenges facing kids in the capital city seem to be getting bigger. In the past few weeks, a rash of homicides, including a 3-year-old who was hit by a stray bullet, has fueled a greater conversation about curbing youth violence in Jackson.

Sykes told Jackson Free Press that people do not commit violence for the sake of committing violence, but that there are often deeper issues that are easier for adults to complain about that to understand.

"People have to dismantle their fear of young people," Sykes said.

MANILA, Philippines The House of Representatives has approved a bill declaring November 23 as Philippine Press Freedom Day.

November 23 marks the commemoration of the grisly massacre in Maguindanao four years ago that killed 58 people, 32 of which were media workers, allegedly at the hands of the Ampatuan clan which wielded a powerful private army in the region.

House Bill 4128 was recommended for approval by the House public information committee, which has yet to consolidate the different versions of the freedom of information bill.

The measure shall promote and uphold freedom and heighten awareness on press freedom so that journalists may freely and securely report the truth no matter who gets hurt by it, said Misamis Occidental Representative Jorge Almonte, who chairs the public information committee.

One of the bills authors, Ako Bicol Rep. Christopher Co, said the country remains to be a dangerous place for journalists even as it has the freest press in Asia.

Remembered as the single deadliest event for journalists in recent history the Maguindanao massacre is and continues to be a symbol of a fight for press freedom and their rights in the practice of their profession, Co said.

The commemoration of Philippine Press Freedom Day shall serve as a fitting reminder that we will never forget the sacrifices of these journalists who gave up their lives in the practice of their profession, he added.

According to the bill, the private sector, government agencies and instrumentalities, including government-owned and controlled corporations and local government units, should encourage and allow their employees to participate in the commemoration of Press Freedom Day within the premises of their respective offices.

The bill also requires the Department of Education (DepEd), the Commission on Higher Education (CHEd) and the Technical Education and Skills Development Authority (Tesda) to lead public and private schools in organizing activities to celebrate the importance of the press, and to call for an end to violence against the media.
