Notre Dame researcher's paper examines the biology of tumor-derived microvesicles

A new paper by Crislyn D'Souza-Schorey, professor of biological sciences at the University of Notre Dame, discusses the biology of tumor-derived microvesicles and their clinical application as circulating biomarkers. Microvesicles are membrane-bound sacs released by tumor cells and can be detected in the body fluids of cancer patients.

The World Health Organization (WHO) estimates that the incidence of cancer will reach approximately 9 million deaths in 2015. The rising prevalence of the disease is a major factor that drives the growth of the oncology biomarkers market. Biomarkers can be defined as any biological, chemical or physical parameter that can be utilized as an indicator of physiological or disease status. Thus, biomarkers are useful in cancer screening and detection, drug design and also to boost the effectiveness of cancer care by allowing physicians to tailor therapies for individual patientsan approach known as personalized medicine.

The new paper discusses the potential of microvesicles to present a combination of disease and tissue-specific markers that would constitute a unique, specific and identifiable biosignature for individual cancers.

"As such, it would make their sampling over time a preferred method to monitor changes to the tumor in response to treatment, especially for tissues such as the ovary or pancreas, where repeated biopsies of these organs is impractical," D'Souza-Schorey said.

Profiling of microvesicles could form the basis of personalized, targeted cancer therapies, especially as more reliable and rapid profiling technologies become available.

"For example, certain markers like HER2/neu, in addition to being elevated in breast cancer, is also increased in a relatively smaller subset of other cancers such as ovarian cancer," D'Souza-Schorey said. "This latter group of patients would benefit from existing treatment strategies that target the HER2 receptor."

The approach could be advantageous over currently used approaches of profiling whole tissue or un-fractionated body fluid particularly if circulating microvesicles indeed concentrate molecular changes that occur in the tumor, as it would increase the sensitivity of detecting critical markers of cancer progression.

"One complicating factor, though, is the presence of shed vesicles from other non-tumor cell types also in direct contact with these body fluids," D'Souza-Schorey said. "Thus, equally significant is the development of strategies to selectively capture tumor-specific markers that separate from other shed vesicle populations."

Genome Canada Launches Competition to Strengthen Canada's Leadership in Bioinformatics and Computational Biology

The competition involves $5 million from Genome Canada and $1.25 million from CIHR. Of this, $4 million will support large-scale projects by multi-disciplinary teams to develop robust, user-friendly tools needed by the genomics research community. As well, $2.25 million will support small-scale projects by individuals or groups to propose innovative ideas with the potential for significant impact. CIHR's funding preference will be in support of the smaller scale projects. The projects, in collaboration with Canada's six regional Genome Centres, are expected to secure an additional $4 million in co-funding for the large-scale applied projects.

A new model to understand the supertasting phenomenon

Supertasting describes the ability to strongly detect food flavors such as bitter and sweet, and it can affect a person's food preferences. For example, supertasters are often averse to green vegetables because their bitter taste is amplified. Supertasters may also prefer foods lower in sugar and fat. Approximately one out of four people is a supertaster, and a supertaster's avoidance of sweet and fatty foods may have protective cardiovascular effects.

Christopher Nosrat and colleagues at the University of Tennessee Health Science Center and the Monell Chemical Senses Center in Pennsylvania have developed a new mouse model that may be useful to study supertasting. The team's work was reported in the Journal of Biological Chemistry.

Nosrat's group developed mice whose taste buds overexpress brain-derived neurotrophic factor, a growth factor for neurons and a protein that is important for the distribution of nerves to sensory organs, such as taste buds. These mice had larger taste buds, an increased number of taste cells per taste bud, and a greater supply of nerves in the taste buds compared with the control mice. These features suggest that the mice could be a model for supertasters, whose tongues have an increased number of fungiform taste buds (a specific kind of taste bud on the front and sides of the tongue that detects the five basic tastes).

"By generating the supertaster rodent model," Nosrat reports, "we are able to study the supertasting phenomenon in detail." Furthermore, brain-derived neurotrophic factor is important for proper development of the nervous system, Nosrat explains, and this mouse model can facilitate the development of therapies for nerve injuries in which taste signaling to the brain has been damaged.

Behavioral challenges are a common problem for children with an autism spectrum disorder (ASD). Many children with ASD have challenges managing certain behaviors such as aggression or not following directions in addition to experiencing limitations in communication or social skills.

The experts of the Autism Speaks Autism Treatment Network (ATN) and the AIR-P have addressed how parents and families can help improve their child's behavior with The Introduction to Behavioral Health Treatment which provides an overview of in-home strategies as well as tips to teach and increase desirable behaviors and decrease behavior problems. This tool kit also provides an overview of professional behavioral treatments families can seek out from qualified professionals.

The accompanying Applied Behavioral Analysis (ABA) tool kit, also released today, is an informational guide designed to provide parents with a better understanding of ABA, how their child can benefit, and where and how they can find ABA services. ‘Master molecule’ may improve stem cell treatment of heart attacks

Johns Hopkins researchers have discovered that a single protein molecule may hold the key to turning cardiac stem cells into blood vessels or muscle tissue, a finding that may lead to better ways to treat heart attack patients.

Human heart tissue does not heal well after a heart attack, instead forming debilitating scars. For reasons not completely understood, however, stem cells can assist in this repair process by turning into the cells that make up healthy heart tissue, including heart muscle and blood vessels. Recently, doctors elsewhere have reported promising early results in the use of cardiac stem cells to curb the formation of unhealthy scar tissue after a heart attack. But the discovery of a "master molecule" that guides the destiny of these stem cells could result in even more effective treatments for heart patients, the Johns Hopkins researchers say.

In a study published in the June 5 online edition of the journal Science Signaling, the team reported that tinkering with a protein molecule called p190RhoGAP shaped the development of cardiac stem cells, prodding them to become the building blocks for either blood vessels or heart muscle. The team members said that by altering levels of this protein, they were able to affect the future of these stem cells.

"In biology, finding a central regulator like this is like finding a pot of gold," said Andre Levchenko, a biomedical engineering professor and member of the Johns Hopkins Institute for Cell Engineering, who supervised the research effort.

The lead author of the journal article, Kshitiz, a postdoctoral fellow who uses only his first name, said, "Our findings greatly enhance our understanding of stem cell biology and suggest innovative new ways to control the behavior of cardiac stem cells before and after they are transplanted into a patient. This discovery could significantly change the way stem cell therapy is administered in heart patients."

Earlier this year, a medical team at Cedars-Sinai Medical Center in Los Angeles reported initial success in reducing scar tissue in heart attack patients after harvesting some of the patient's own cardiac stem cells, growing more of these cells in a lab and transfusing them back into the patient.

Using the stem cells from the patient's own heart prevented the rejection problems that often occur when tissue is transplanted from another person.

Levchenko's team wanted to figure out what, at the molecular level, causes the stem cells to change into helpful heart tissue. If they could solve this mystery, the researchers hoped the cardiac stem cell technique used by the Los Angeles doctors could be altered to yield even better results.

DNA points to link in drug beating trial

The DNA of a man allegedly involved in a violent drug-related beating was found on a zap strap and a piece of electrical tape used to bind the victim, an expert testified at trial Monday in North Vancouver.

The victim's blood was also found on a picture frame inside another accused man's home, the expert said.

The DNA is among the physical evidence linking Robin Landrew Pryce, 42, of Surrey and Paul Joseph Defaveri, 50, of Squamish to a violent beating of Ronald Perry, 68, in a North Vancouver home almost three years ago.

Bryce and Defaveri are on trial for the attempted murder and aggravated assault of Perry before provincial court Judge Steven Merrick.

Both men have pleaded not guilty to the charges.

Earlier in the trial, Perry - who was involved in the criminal underworld -described how he was attacked with hammer and had his eyes gouged out after he tried to blackmail people who ran a marijuana North Vancouver grow op. Perry described in horrific detail how he was violently beaten inside the bungalow in the 1600-block of Phillip Avenue by two men on Sept. 2, 2009.

A passerby found him early the next morning, with a bag over his head, bound with zap straps, wire and tape, inside his car, parked on Vancouver's Oxford Street.

In court, Perry identified Defaveri as one of the people he'd gone to speak to in the house on Sept. 2, 2009. Pryce, Defaveri, and Defaveri's wife Marney King were living in the rented house at the time.

One of Defaveri's fingerprints was found in blood on an interior door of the house, a police investigator testified.

On Monday, DNA expert May Clark from the RCMP's forensic lab in Edmonton identified that blood - and samples taken from several other places in the Phillip Avenue house - as matching Perry's.

UAH Named Nation’s Most ‘Crime-Rattled’ College, According to Website

The February 2010 biology department shootings helped put the University of Alabama in Huntsville on top of a national websites ranking of Americas 25 Most Crime-Rattled Colleges. The unofficial ranking, based on an analysis of crime reports, wasreleased Tuesday by the website The Daily Beast.

UAH spokesman Ray Garner called the ranking skewed in an interview with WHNT News 19 news partner, The Huntsville Times:

It is no secret that we had a tragic shooting incident on our campus in 2010, and that event heavily skewed this ranking, Garner said Tuesday. If you look at our overall crime statistics, you will find outside that isolated event that our university offers a safe environment for the thousands of students, faculty and staff that travel around our campus.

While taking a UAH campus tour Wednesday, parent of a prospective 2013 freshman Julie DiLiberty told WHNT News 19 she find the report ludicrous. DiLiberti says she has no qualms about safety in considering sending her son to study computer science at the university next year.

The reporting period was 2008-2010, which included the period of a triple homicide committed during a biology faculty meeting on the UAH campus. In humans, amyloid fibers form biological nanostructures that house pigments and other molecules, and may also play an important role in long-term memory. These fibers are one of the most stable protein-based structures in nature, so when they are harmful in diseases, such as Parkinson's, they are extremely difficult for cells to break down.

As a result, Martin Duennwald and AnaLisa Echeverria, at the Boston Biomedical Research Institute, and James Shorter, assistant professor of Biochemistry and Biophysics at the University of Pennsylvania, set out to find ways to promote beneficial amyloid fiber assembly or to reverse its pathogenic assembly, at will. The study is published in PLoS Biology.

Yeast have a protein called Hsp104 that can quickly disassemble amyloid fibers, and this activity is significantly enhances by a group of small heat shock proteins. However, humans and other animals do not have the Hsp104 protein, thus raising the question of whether human cells are also capable of disassembling amyloid fibers?

In this study, the researchers found that when Hsp104 is absent, the yeast small heat shock proteins work together with other proteins to disassemble amyloid fibers. The proteins slowly remove each subunit one by one from the tips of the fibers. The team were surprised by this activity as these proteins are best known for their role in preventing protein clumping.

According to the researchers, the proteins of the amyloid-disaggregating machinery in yeast are also present in humans. Therefore, human small heat shock proteins are able to work together with other proteins to disassemble amyloid fibers, even without Hsp104.

They state that these findings could lead to the development of new therapies for different neurodegenerative disorders.

Their aim is to activate the machinery in humans to pull apart disease-causing amyloid fibers where and when needed by increasing the expression of heat shock proteins.

Double the pain: RUB biologists find the cause of pain in the treatment of fair skin cancer

Apply the ointment, light on, light off that's how easy it is to cure various forms of non-melanoma skin cancer. However, the majority of patients suffer severe pain during the so-termed photodynamic therapy. Why the treatment with ointment and red light can be so painful has now been uncovered by researchers from the RUB. They identified the ion channels involved and signalling molecules secreted by the cancer cells. "The results may provide a starting point for suppressing the pain", says Dr. Ben Novak of the Department of Animal Physiology.

How the photodynamic therapy works

In contrast to normal cells, cancer cells are equipped with different enzymes and have a much higher metabolism. If you apply a molecule called aminolevulinic acid to the skin in the form of a gel, cancer cells take up considerably more of this substance than healthy cells. If aminolevulinic acid accumulates in the cells, the mitochondria the power plants of the cells form the molecule protoporphyrin IX. When irradiated with red light, protoporphyrin IX reacts with oxygen. This produces highly reactive oxygen species, free radicals, which destroy the cancer cells. Approximately ten minutes of red light irradiation is sufficient to successfully treat superficial forms of non-melanoma skin cancers such as actinic keratosis. Doctors also remove warts in this way.

Painful therapy

"The catch is: it's terribly painful", says Ben Novak. Forty percent of those treated experience pain during the light irradiation, which they assess on a scale of 0 to 10 (whereby 10 corresponds to an excruciating pain like a heart attack) as 7 to 8. Using injections, like at the dentist, it is possible to numb the nerves involved. "But that also always involves a risk", says the Bochum biologist. The scientists led by the RUB professor Dr. Hermann Lbbert have now solved the mystery as to why the treatment hurts at all.

Pain-sensitive cells in the skin are stimulated

The pain is generated by two mechanisms. In a cell culture experiment, the team showed that not only cancer cells but also pain-sensitive nerve cells in the skin take up aminolevulinic acid and its derivative methyl aminolevulinic acid from the ointment. Using calcium imaging, the animal physiologists followed the activity of nerve cells which they had treated with aminolevulinic acid and cells that were not exposed to the substance. Treated nerve cells fired when the researchers exposed them to light. In a living organism, this would mean that the cells would send a pain stimulus to the brain. Without the aminolevulinic acid, the pain-sensitive cells remained inactive under red light. In further experiments, the scientists showed that the activity of the nerve cells is caused by sodium channels and voltage-gated calcium channels in the cell membrane. "A drug that targets these channels would, conceivably, be able to suppress the pain but that's still in the future", says Ben Novak.

Center for Productive Longevity Urges People to Shed Negative Stereotypes About Older Age

BOULDER, CO--(Marketwire -06/20/12)- As people continue to live longer and retire earlier, the term "Third-Stage Adulthood" has more relevance than ever before. Referring to people 62-85 years old, this stage of life is chock-full of senior workers who are qualified and ready to continue working after the traditional retirement age of 65 or sooner. This means that there is a large and growing talent pool with experience, expertise, seasoned judgment, and proven performance (referred to by CPL as EESP), to meet the workforce needs of the 21st century.

The concept of "Third-Stage Adulthood" was first conceived in a paper, "The Evolution of Adulthood: A New Stage" co-authored in 2000 by Dr. Elliott Jaques, who conceived the mid-life crisis in 1965, and CPL Founder and CEO William Zinke. Paper, Blog Heats Up GMO Discussion

There's an interesting exchange on genetic engineering at the Food Politics blog, http://tinyurl.com/, featuring a review by industry critic Marion estle of an anti-GE paper, GMO Myths and Truths. I've skimmed the paper, which you can find at http://tinyurl.com/, and I confess to lacking the expertise to evaluate the claims. It would take more time than I have at the moment to dig into the claims, although I hope to do so in the future.

Nestle says the authors of the paper, who find nothing to like in genetic engineering, "have put a great deal of time and effort into reviewing the evidence for the claims. This is the best-researched and most comprehensive review I've seen of the criticisms of GM foods." She asks whether the pro camp can "produce something equally well researched, comprehensive, and compelling?" and concludes, "I doubt it but I'd like to see them try." She says there's enough evidence in the paper to justify labeling, at the very least.

It is, of course, the position you'd expect her to take, and several of the comments following her post challenge both her and the paper. One claims there are indeed well-researched, comprehensive and compelling pro papers. Others say the paper she cites cherry-picks evidence and relies on papers that have been debunked. An example cited in one of these critical comments asserts that it relies on a study of Bt found in human blood that used a test that couldn't detect blood at the levels the study's authors said they found.

My suspicion is most won't read these papers and will continue to think what they already think about the issue. An even worse fear is that reading the papers on both sides wouldn't convert anyone on either side. Still, I may give it a try at some point.

DNA Evidence Ties Griffin to Murder Victim, Her Son

With a rare possibility of exceptions, the DNA of Stanley Griffin was found under the fingernails of Jennifer Hailey, according to a DPS Crime Lab analyst.

Griffin is on trial for the September 2010 murder of Hailey in her College Station apartment. Griffin faces a capital murder charge, and the State of Texas is seeking the death penalty if they secure a conviction.

On Wednesday, the third day of testimony in the trial, analyst Allison Heard also told the jury a garden trowel found in an outdoor trash can near the Hailey home the morning of the murder was covered in the blood of Hailey's son. Heard could match any DNA on the handle.

Authorities allege Griffin attacked the then-nine-year-old shortly after he saw his mother being strangled in her bedroom. Monday, the child testified that he saw Griffin on his mother's back choking her out, then ordered him to go back to his room. When he reemerged 15 minutes later, the boy said Griffin choked him and struck him in the head and neck with the trowel.

Again with a rare possibility of exceptions, the blood of the child was found on the shoes Griffin was wearing when he was arrested hours after the murder, this according to Emma Becker, a second crime lab analyst who testified Wednesday morning.

Prosecutors contend Hailey used her right hand to try and rip Griffin's left arm from around her neck. When authorities found Griffin later the day of the murder, he had fresh scratch marks on his left arm. He said they were caused two days earlier from riding his bike through brush.

A baseball cap found in the Pedernales Drive home of Hailey and her son was also tested for DNA. Hailey's DNA almost certainly was found on it, Heard testified. With other DNA found on the cap, Griffin could not be excluded as a possibility, but it was not as conclusive.

Shortly before the murder, Griffin was seen in surveillance video at a Navasota convenience store with a friend, who testified to dropping Griffin off near Hailey's home. He was wearing a similar hat.

Prosecutors elevated a murder charge against Griffin to capital because they allege he kidnapped Hailey's son. By the letter of the law, they claim the kidnapping was by virtue of Griffin's order to the boy to go back to his room as Griffin attack the boy's mother.
