Scientists discover why some proteins are speedier than others

(Phys.org) Scientists from our Department of Biology & Biochemistry have solved a problem that has frustrated biologists for years why different parts of proteins are made at different rates.

Their discovery, published in the online journal PLOS Biology, should help scientists make proteins that would be otherwise tricky to produce in the lab.

Proteins are made in the cell by machines called ribosomes, which read gene transcripts (called mRNA) and 'translate' the genetic code into strands of protein.

The researchers looked at yeast proteins that were made at different rates. They found that the parts of proteins that took the longest to make were composed of mostly positively charged amino acids.

Co-author Laurence Hurst, Professor of Evolutionary Genetics at the University, explained: "If you imagine the ribosome as a doughnut with a hole in it, the mRNA strand is fed through the hole in the centre, translating the genetic code to give the corresponding amino acid chain, which comes out of a tunnel in the side of the doughnut.

"If the protein being created is positively charged, it gums up the negatively charged tunnel and slows down the protein production."

Lead author Katie Charneski, added: "This was a real surprise as most people before had assumed that it was something about the mRNA that caused slowing or speeding of the ribosome."

"Many organisms, including humans, have 'tails' on the ends of their mRNAs that normally the ribosome does not translate. But if mistakes occur in protein production, the tail may be erroneously translated, always producing a string of positively charged amino acids.

"These charges might act as a tagging signal for the cell to destroy the potentially toxic protein."

The speed at which proteins are made has consequences for the cell and has been puzzled over by biologists for years. Changes in this speed can affect how well proteins fold and where they end up in the cell, and hence their eventual function.

ReNeuron Group wins grants for stem cell therapy development

LONDON (SHARECAST) - ReNeuron Group has won two grants worth 1.2m pounds to develop its stem cell therapy.

The grants were awarded by the UK Biomedical Catalyst, a programme of public funding jointly managed by the Technology Strategy Board and the Medical Research Council.

The first award of 0.4m will be used for ReNeurons ReN009 stem cell therapy candidate for critical limb ischaemia, a severe blockage in the arteries which markedly reduces blood-flow to the extremities including hands, feet and legs.

The second award of 0.8m relates to the company's ReN003 stem cell candidate for the treatment of retinitis pigmentosa, an eye disease in which there is damage to the retina.

Michael Hunt, the Chief Executive Officer of ReNeuron, said: "We are delighted to have been successful in winning the two grants we applied for in the current round of the Biomedical Catalyst funding competition.

"This highly competitive scheme involves a rigorous peer review of applicants' technologies and commercial development plans.

Stem Cell Therapy Now In Davao

With its growing popularity in the country, the so-called stem cell therapy, which was recently approved by the Department of Health (DOH), is now available in Davao City.

Acosta said that based on what was stipulated in the approved directions for stem cell therapy, the stem cell could just come from the person himself and not from the other sources like black sheep or aborted fetus, adding that sources of stem cells are fats, blood, bone marrow and umbilical cord.

"It could take five to six hours for one to undergo stem cell therapy. Harvesting is done in the first two hours, then another one to two hours for stem cell processing and activation, and the last one to two hours for treatment of stem cells back to the same patient," she said.

Popular personalities who have already publicly admitted that they had undergone stem cell therapy are former President Joseph Estrada, Senate President Juan Ponce Enrile and former Senator Ernesto Maceda, who said that they spent millions to pay for the treatment.

But Acosta said that it could be a lot cheaper here than abroad, as the cost of treatment will be in accordance to the guidelines of the Phil. Society for Stem Cell Medicine.

She said there have been very good clinical outcomes from stem cell treatments on autism, auto-immune diseases, cerebral palsy, diabetes, heart disease, liver cirrhosis, macular degeneration, multiple sclerosis, nerve damage, osteoarthritis, spinal cord injury and stroke.

"But we have to clarify here that we are not claiming stem cell therapy as a cure for cancer, though it can alleviate pain and improve patients' wellbeing while undergoing cancer therapy," she said.

Chartwells School Dining Services Provides Nutrition Education and Awareness Through Kids Eat Right Campaign Grants

RYE BROOK, N.Y.--(BUSINESS WIRE)--

With March being National Nutrition Month, Chartwells School Dining Services is pleased to report the successful results of 25 mini-grants that were awarded to Chartwells schools in September 2012 as part of the Kids Eat Right campaign. Founded by the Academy of Nutrition and Dietetics and the Academy of Nutrition and Dietetics Foundation, the Kids Eat Right campaign is dedicated to supporting the efforts of the White House to end the childhood obesity epidemic by providing resources that will help educate families, students and communities about the importance of quality nutrition.

The $200.00 mini-grants gave Chartwells grant recipients access to five toolkit presentations on the topics of healthy breakfast, healthy snacking, family meals, cooking activities and nutrition myth-busters. Each toolkit included information such as ready-to-use PowerPoint presentations, nutrition handouts, cooking demo recommendations, activity sheets, recipes and talking points and scripts.

Grant recipients were each required to deliver two presentations in their school for students, parents and members of the community. A total of 61 presentations were completed and approximately 3,300 people nationwide attended. After the presentations were delivered, 92 percent of recipients said they would be very likely to use these materials again in the future to help educate their student population about good nutrition habits. A second round of twenty-five additional grants will be implemented in late Spring 2013.

The Kids Eat Right toolkits really enabled us to change attitudes and behaviors around healthy eating, said Margie Saidel, RD, VP of Nutrition and Sustainability for Chartwells School Dining Services. After attending the presentations, students said they would make time to eat breakfast in the morning now that they knew how important it is. Older students were taught what is in the food they are eating and how to incorporate protein and other beneficial nutrients into their diets. Biological wires carry electricity thanks to special amino acids

Slender bacterial nanowires require certain key amino acids in order to conduct electricity, according to a study to be published in mBio, the online open-access journal of the American Society for Microbiology, on Tuesday, March 12.

In nature, the bacterium Geobacter sulfurreducens uses these nanowires, called pili, to transport electrons to remote iron particles or other microbes, but the benefits of these wires can also be harnessed by humans for use in fuel cells or bioelectronics. The study in mBio reveals that a core of aromatic amino acids are required to turn these hair-like appendages into functioning electron-carrying biological wires.

"It's the aromatic amino acids that make it a wire," says lead author Derek Lovley of the University of Massachusetts, Amherst. Lovley and his colleagues removed the pivotal amino acids from the pili and replaced them with smaller, non-aromatic amino acids. Without these key components, Lovley says, the pili are nothing more than protein strings. "We showed it's not good enough to just make the string - you've got to make a wire," says Lovley.

G. sulfurreducens "breathes" by removing electrons from organic materials and funneling them to iron oxides or to other microorganisms, much the way humans pull electrons out of organic molecules in food and dump them on oxygen. The bacteria use their pili to reach out to iron oxides or other microbes, transferring the "waste" electrons along the structure to the destination. Geobacter's pili are only 3-5 nanometers wide, but they can be 20 micrometers long, many times longer than the cell itself.

Trafficking in electrons is how all living things breathe, but it is normally carried out by discrete proteins or other molecules that act like containers for shuttling electrons from one place to another. Lovley says earlier results showed the pili in G. sulfurreducens possess metallic-like conductivity, the ability to carry electrons along a continuous structure, a controversial finding in biology.

To investigate how pili accomplish this singular feat, Lovley says they looked to non-biological organic materials that can conduct electricity. "In those synthetic materials, it's aromatic compounds that are responsible for the conductivity. We hypothesized that maybe it's similar in the Geobacter pili. In this case, it would be aromatic amino acids." Aromatic compounds have a highly stable ring-shaped structure made of carbon atoms.

Turning to the pili, Lovley says his group looked for aromatic amino acids in the parts of the pili proteins that would most likely contribute to the conductivity. Using genetic techniques, they developed a strain of Geobacter that makes pili that lack aromatic amino acids in these key regions, then they tested whether these pili could still conduct electricity. They could not. Removing the aromatic amino acids was a bit like taking the copper out of a plastic-covered electrical wire: no copper means no current, and all you're left with is a string.

Removing aromatic amino acids from the pili prevents the bacteria from reducing iron, too, says Lovley, an important point because it adds further proof that Geobacter uses its pili as nanowires for carrying electrons to support respiration.
