Greg Peterson, who started at Bethel as a student and later became the schools assistant athletics director and an assistant football and basketball coach, has been named the Royals new Director of Athletics starting June 1, the university announced Wednesday. Peterson, the offensive coordinator for Bethel football, will begin transitioning to his new role on Feb. 15 and officially take over when Bob Bjorklund resigns as the schools AD.

Bethel athletics has been on a steady climb over the last decade and Bob Bjorklund has uniquely positioned our department to continue that trajectory, Peterson said in a statement. Im thrilled to follow someone of Bobs integrity, character, and passion for Bethel University and our student-athletes.

Peterson started at Bethel as a student in 1988, playing football and basketball, and became a part-time assistant coach upon graduation. He currently also serves as the schools assistant athletics director.

Greg has worked full time at Bethel for the last 23 years and brings an in-depth understanding of Bethel athletics, a passion for Christ, and a desire to support and serve student athletes to his new role, Dr. John Addleman, Bethel vice president of student experience, said in a statement.

The trial is being conducted at The University of Texas MD Anderson Cancer Center (MD Anderson) as part of an ongoing clinical collaboration.

Background and Opportunity

According to the World Health Organization, esophageal cancer is currently the sixth most common cause of cancer death in the world and is estimated to have caused over 508,585 deaths in 2018. The American Cancer Society estimates that in 2020 in the United States, there were approximately 18,440 new esophageal cancer cases diagnosed, and approximately 16,170 deaths due to esophageal cancer. Approximately 20% of patients survive esophageal cancer at least five years after diagnosis.

Phase I Study of NBTXR3 Activated by Radiotherapy with Concurrent Chemotherapy for Patients with Esophageal Cancer (MD Anderson Study 2020-0122)

This study is an open-label, single-arm, prospective phase I study consisting of two parts: (i) dose-escalation to determine the RP2D of NBTXR3 activated by radiotherapy with concurrent chemotherapy, and (ii) expansion at RP2D with toxicity monitoring.

The patient population will include adults (age 18 years) with stage II-III adenocarcinoma of the esophagus that are treatment-nave and radiographically non-metastatic at screening. The number of participants enrolled will be determined based on the maximum number required to establish the RP2D of NBTXR3 activated by radiation therapy. Up to 24 subjects will be enrolled, including a maximum of 12 subjects for the dose-escalation part. Twelve additional subjects will be enrolled for the RP2D expansion part. Recruitment is ongoing and the planned enrollment period is 24 months.

[Full text] EDTA-K2 Improves the Detection Sensitivity of SARS-CoV-2 IgM and IgG A | IJN – Dove Medical Press

Introduction

Currently, the widespread threat to human health from SARS-CoV-2 urgently requires the development of fast and accurate analytical methods for early diagnosis. At present, pathogen detection and nucleic acid testing are commonly used to identify COVID-19 in the clinic. In addition, infected patients are mostly positive for SARS-CoV-2-specific IgM antibodies after seven to eight days from disease onset and their IgG titers during the recovery period, are more than four times higher than those in the acute phase,1 indicating that serological detection of specific antibodies is crucial for the rapid identification of SARS-CoV-2. Specifically, this study confirmed that the gold immunochromatographic assay (GICA) could serve as a rapid diagnostic test for RT-PCR-negative highly suspected patients and screening of SARS-CoV-2 carries in limited-resource countries.2,3 Recently, GICA has been widely used to detect antibodies, because the test trips based on colloidal gold particles were fast, convenient, and require minimal equipment. Therefore, we feel that this method is a good candidate for the large scale, rapid screening, of both symptomatic and asymptomatic COVID-19 carriers and the screening of healthy workers following isolation.4

Early in 2014, a study found that different additives in the blood collection tubes, were able to influenced the detection of alcohol concentration.5 Moreover, a study found that DNA samples extracted from blood using two different anticoagulants (heparin tubes and EDTA-K2), gave similar results, especially for PCR based applications.6 These studies revealed that different additives in the vacuum blood collection tubes, showed clear confounding effects clinically. However, it is not clear whether these vacuum blood collection tubes with different additives affect the detection of SARS-CoV-2 IgM and IgG antibodies. Therefore, this study investigated the impact of different vacuum blood collection tubes, when detecting SARS-CoV-2 IgM and IgG antibodies in the plasma and serum using GICA. These results are expected to provide a theoretical basis for the future accurate detection of SARS-CoV-2 IgM and IgG antibodies.

A total of 112 patients diagnosed as having COVID-19 by nucleic acid testing were enrolled from the Respiratory Department of Hankou Hospital of Wuhan, China, from March 6 to March 18, 2020. Nine of the 112 patients had their samples collected into four different types of collection tube. Among the patients, seven of the nine patients diagnosed with COVID-19 were used to quantify the concentration of SARS-CoV-2 IgM and IgG antibodies. Thirty-two of the 112 patients were used to detect SARS-CoV-2 IgM and IgG antibodies using EDTA-K2 as plasma and whole blood anticoagulants. Next, 200 healthy control subjects diagnosed as noninfectious for COVID-19 based on nucleic acid testing, were enrolled from the Department of Clinical Laboratory Medicine of Nanfang Hospital of Guangdong, China, from April 4 to April 5, 2020. Serum and plasma specimens were isolated from 2 mL of blood and placed into four different types of vacuum blood collection tube, which contained different additional reagents, such that the serum was collected into tubes with coagulants present and plasma with different anticoagulants (EDTA-K2, sodium citrate or lithium heparin).

IgG and IgM specific antibodies were detected in blood samples using recombinant coronavirus N protein labeled, and the results were seen using the naked eye. The new coronavirus antibody detection kit (colloidal gold based) and the colloidal gold solution were developed by Innotek (Tangshan) Biotechnology, and the chemiluminescence detection reagents were developed by Mairui Biotechnology Co., Ltd (Guangdong, China). The vacuum blood collection tubes with coagulants and EDTA-K2 were produced by Zhiyuan Biotechnology, Co., Ltd (Guangdong and Hubei, China), and those with sodium citrate and lithium heparin were from Jinxing Biotechnology Co., Ltd (Guangdong and Hubei, China). Blood samples from healthy subjects were collected into EDTA-K2 coated test tubes, and the samples from COVID-19 patients were collected into the test tubes as stated above. They were centrifuged at 1000g at 4C for 15 min and the serum and plasma collected. Finally, the samples were subjected to the GICA, or tested on an ARCHITECT i2000 (Mairui Biotechnology, Shenzhen, China) for the detection of IgM and IgG as specified by the manufacturer.

Immunocapture methodology was used here to detect the SARS-CoV-2 IgM and IgG antibodies. The nitrocellulose membrane was coated with mouse anti-human IgM monoclonal antibodies, mouse anti-human IgG monoclonal antibodies, and goat anti-mouse IgG antibodies. Colloidal gold-labeled recombinant coronavirus antigen and mouse IgG antibodies were then used as a tracer. When the sample was added to the well in the presence of the IgM or IgG detection reagent, it is able to combine with the corresponding antigen to form a complex and then the coated mouse anti-human antibodies will react with this complex, forming a purple-red band, as detected on the card. The colloidal gold-labeled mouse IgG antibodies will form a purple-red band with the goat anti-mouse IgG antibodies.

Here, our aim was to determine the effect of different additives in the collection test tubes on their ability to chelate the colloidal gold. The size of the particle was determined by a Zetasizer Nano ZS90 analysis system (Malvern Instruments, UK, Zetasizer version 7.12). A size polydispersity Index (PDI), for number particle size distribution (PSD), with the x-axis showing the distribution of estimated particle diameter (nm) and the y-axis showing the relative percentage, was created.

Data were analyzed using origin (version 2018), IBM SPSS Statistical software for Windows, version 22.0 (IBM Corporation Armonk, NY, USA) and GraphPad software (GraphPad Software, Inc., La Jolla, CA, USA). Data are presented as the mean standard deviation or median (interquartile range), unless otherwise indicated. The results from the concentration of SARS-CoV-2 antibodies detected by chemiluminescence assay (CA) were assessed by one-way analysis of variance followed by Student-Newman-Keuls test. A two-tailed probability (P) value of < 0.05 was considered statistically significant.

We collected serum and plasma with EDTA-K2 from 112 patients with COVID-19 to detect SARS-CoV-2 IgM and IgG antibodies using GICA. The vacuum collection tubes with coagulants and a separator were used to collect serum. Our results revealed that EDTA-K2 anticoagulated plasma had a higher positive detection rate for detecting SARS-CoV-2 IgM antibodies when compared to the serum group. Specifically, seventeen patients were found to be weakly positive, with an additional fifteen patients testing positive but were negative based on serum testing. Five patients were weakly positive, thirteen patients were positive and three patients were strongly positive, but very weak positive using serum testing. Twenty-one patients were detected as positive and two patient was strongly positive but weakly positive based on serum testing. Similarly, for IgG antibody testing, forty samples were interpreted as strongly positive but were detected as only positive when serum tested (Table 1).

In addition, we analyzed the consistency of the results in detecting SARS-CoV-2 IgM and IgG antibodies when comparing serum and EDTA-K2 anticoagulated plasma. We classified very weakly positive, weakly positive and strongly positive, as positive, and kappa values were used to evaluate the consistency of the positive and negative groups. We found the total coincidence rate of IgG antibodies were all 100% in these two groups (weighted kappa value = 1.0), while the kappa value for the IgM antibody was 0.409 between the two groups (Table 2). Taken together, these findings demonstrated that EDTA-K2 increased the positive detection rate of SARS-CoV-2 IgM antibodies.

Based on our hypothesis above, in order to investigate the impact of different types of vacuum collection tubes on the detection of SARS-CoV-2 antibodies, we collected blood specimens from nine COVID-19 patients with four different vacuum blood collection tubes (serum and anticoagulated plasma with lithium heparin, sodium citrate and EDTA-K2). Then, SARS-CoV-2 IgM and IgG antibodies were detected by GICA. Our results showed that EDTA-K2 anticoagulated plasma had a higher positive detection rate for IgM antibodies, with a darker color being present on the IgG card. However, there were no significant differences among the serum and anticoagulated plasma, when lithium heparin or sodium citrate were used (Figure 1).

In addition, we aimed to further verify that this highly positive effect was attributable to EDTA-K2. Therefore, thirty-two of the 112 patients with COVID-19 had their plasma collected into EDTA-K2 tubes and the whole blood from these patients was tested. Our data showed that there was no difference in the ability to detect SARS-CoV-2 IgM or IgG antibodies when both the anticoagulated blood plasma or whole blood with EDTA-K2 was used (Table 3 and Supplemental Figure 1). These results strongly demonstrated that anticoagulated blood plasma in the presence of EDTA-K2 improved the detection rates of SARS-CoV-2 IgM and IgG antibodies, and this was likely to be due to EDTA-K2, rather than the use of blood serum, plasma, or whole blood. Hence, these findings indicated a marked improvement when EDTA-K2 was used in the ability to enhance the detection of SARS-CoV-2 IgM antibodies.

To explore the ability of EDTA-K2 to improve the detection sensitivity of SARS-CoV-2 IgM and IgG antibodies, four different kinds of blood specimens from seven COVID-19 patients with four different vacuum blood collection tubes (serum and anticoagulated plasma with lithium heparin, sodium citrate and EDTA-K2), were then subjected to the GICA and CA, respectively. The latter assay has a higher sensitivity than the former.7 Our results revealed that when CA was used to detect SARS-CoV-2 IgM and IgG antibodies, the presence of different anticoagulants had no effect on the measurements for serum or plasma (Figure 2A and B). However, when the same samples were detected by GICA, the results became significantly variable (Figure 1). Compared with other anticoagulated plasma and serum specimens, the results using EDTA-K2 anticoagulated plasma detected by GICA were more consistent with those results using CA. For example, in patients from 36 to 39, the concentration of IgM antibodies was found to be 2.710.03, 2.840.09, 1.150.08, and 5.240.11 Au/mL respectively when using CA (the reference interval was 0.01.0 Au/mL). These patients would be expected to test positive or strongly positive using the GICA. However, only the results from plasma treated with EDTA-K2 were consistent with those from the CA. The other anticoagulated plasma samples were judged as negative or weakly positive by GICA (Figure 1 and Table 4).

We found that the anticoagulant EDTA-K2, increased detection sensitivity of the GICA. Next, we determined whether it induced the false positive for the detection of SARS-CoV-2 specific antibodies. We set up a blank group containing simply diluent and EDTA-K2. Surprisingly, our results showed that there was no visible purple-red color on the test line. Additionally, we collected anticoagulated plasma with EDTA-K2 from 200 healthy control patients with no COVID-19 infection, and all 200 healthy individuals were negative for both SARS-CoV-2 IgM and IgG antibodies, providing evidence that EDTA-K2 did not increase the false positive rate.

EDTA-K2 is commonly used in blood testing, as it can chelate calcium and reduce the anticoagulation reaction, However, it is unknown whether EDTA-K2 can also chelate colloidal gold and participate in antigen-antibody reactions. Therefore, we set up four groups: the control group was treated with the colloidal gold solution and coagulants, the test group was treated with EDTA-K2, sodium citrate or lithium heparin and colloidal gold solution. Next, Malvern apparatus was used to evaluate the particle size in the four groups. As expected, our results showed that there were two peaks in the EDTA-K2 group, but only one peak in other three groups.

Specifically, the particle sizes were 47.58 1.67 nm (PDI = 0.247 0.02), 49.62 2.20 nm (PDI = 0.250 0.04) and 48.27 2.65 nm (PDI = 0.209 0.07) in the Au, Au+sodium citrate and Au+lithium heparin groups respectively. However, for the Au+EDTA-K2 group, the particle size of first peak was 164 30 nm, which was larger than in the other three groups by approximately four times, and the second peak was 5399 143 nm (PDI = 0.275 0.03) (Figure 3A). Next, they were observed under a confocal laser microscope. We found that larger particle sizes in the test group with EDTA-K2 (Figure 3E), but no significant differences were seen between the Au group and the test groups containing lithium heparin, or sodium citrate (Figure 3BD). These results provided solid evidence in support of our hypothesis that EDTA-K2 could amplify the positive signal by chelating colloidal gold.

The incubation period of various SARS-CoV-2 infections in humans, is from one to fourteen days, and its transmission channels are diverse. In addition to the main routes of transmission through respiratory droplets and contact, it can also be transmitted through aerosols, the digestive tract, and vertical transmission.8 It has a high infection rate due to its multiple channels of transmission and strong infectivity. Therefore, early recognition of suspected patients, and identification of those with severe illness and the early isolation of those with clinical symptoms, are essential for disease control and prevention. At present, the nucleic acid SARS-CoV-2 RT-PCR tests have become the gold standard assay for the diagnosis of COVID-19 with a high sensitivity and specificity.9 However, this method is a relatively time-consuming and complicated and therefore, is not conducive to the screening of large numbers of samples.

GICA based on colloidal gold particles is widely used to screen for antibodies in the blood against SARS-CoV-2. It has the advantages of convenient, fast detection, low cost and suitable for rapid screening of large numbers of samples, the results are intuitive and there is no requirement for specialized equipment, etc.1012 Early in 2011, a study confirmed that GICA could be combine with the enrichment technique of immunomagnetic nanoparticles, to detect Escherichia coli O157: H7 with speed and sensitivity,13 this study provide powerful evidence for the conclusion that EDTA-K2 could chelate colloidal gold particles and increase the sensitivity of detection on SARS-CoV-2 IgM and IgG antibodies. However, some limitations exist for these assays, such as individual differences in the interpretation of results, which may lead to inconsistent results. In addition, these assays could be affected by several factors, including pH, where it was shown to influence the diagnosis of syphilis using GICA.14 Other studies reported a matrix effect when looking at five kinds of meat, using a colloidal GICA to detect sulfamethazine.15

EDTA-K2 has anticoagulant effects because it can chelate large amounts of calcium ions in the blood.16 Therefore, EDTA-K2 is commonly applied to blood samples as an anticoagulant in the clinical setting, but it can lead to the aggregation of platelets in the blood of individual patients, leading to pseudothrombocytopenia.17 EDTA-K2 has been found to influence the test results of Ca, Mg and Fe compared with the collection tubes excluding anticoagulant.18 Others have reported that EDTA-K2 can impact on the extent of detection of coagulation by PLT.19 However, researchers have found that EDTA-K2 no significant effects on the detection of divalent cation chelators containing anticoagulant detected ITP platelet specific antibodies (GP IIb/IIIa and GPI b) using multi-antigen printing immunoassays.20 Whether EDTA-K2 could affect the detection of antibodies based on GICA remains unknown. In this study, we confirmed that EDTA-K2 could improve the detection sensitivity of anti-SARS-CoV-2 specific antibodies. Previous study found that colloidal selenium melamine test strips could rapidly detect colloidal selenium melamine test strips in contaminated milk products or animal feed, with high sensitivity.21 This finding provided powerful evidence for our results that the method of EDTA-K2 based on colloidal gold could be used in the detection of SARS-CoV-2. Therefore, we concluded that there is a significant effect of EDTA-K2 on colloidal gold, but these mechanisms remain largely unknown, and more studies are needed to clarify this phenomenon.

IgM antibodies are rapidly produced in patients in response to acute infections with pathogens, therefore, it is important to accurately detect IgM antibodies at the time of initial diagnosis. In addition, IgM antibody tilters are important for monitoring recovering and relapsed patients. We found that EDTA-K2 could increase the positive detection rate of SARS-CoV-2 IgM antibodies when using GICAs. In this study, we designed a series of experiments to determine how EDTA-K2 affected the detection of anti-SARS-CoV-2 antibodies. We used confocal laser microscope and a Zetasizer Nano ZS90 analysis system to evaluate the particle sizes of our colloidal gold. The results revealed that EDTA-K2 could amplify the positive signal by chelating the colloidal gold. This suggested that, even if there was only a small amount of antibody in EDTA-K2 anticoagulated plasma, the color of the T line was darker. A deeper understanding of the reaction principle suggested that EDTA-K2 could amplify a weak signal by chelating colloidal gold and this can explain why the SARS-CoV-2 IgM antibodies could not be detected in other anticoagulated blood specimens such as sodium citrate and lithium heparin but could work in EDTA-K2 anticoagulated plasma.

Although its sensitivity is not as high as CA, GICA can easily be used to screen large numbers of samples and is suitable for use in primary hospitals. Currently, with the new coronaviruses spreading worldwide,22,23 it is urgent to improve the sensitivity and speed of detection of SARS-CoV-2 IgM and IgG antibodies. These are major priorities to help curb the spread of the virus, so it is important to ensure that any diagnosis is made promptly and accurately.

Overall, the results of this study provided solid evidence for the use of EDTA-K2 to improve the sensitivity of detection of SARS-CoV-2 IgM and IgG antibodies. However, it should be noted that when results between serum and anticoagulated plasma were inconsistent, we recommend that multiple methods should be used to confirm results, such as nucleic acid detection, CA, ELISA, and imaging examinations, combined with the patients clinical symptoms. These findings provided new insights for improving the detection of SARS-CoV-2 by GICA. However, several problems need to be addressed. First, the samples we used were from patients during the middle and late stages of treatment and rehabilitation. There 112 patients with COVID-19 and 200 healthy people involved in the project, but only seven patients with COVID-19 were enrolled to detect four different tubes. Since there are few patients infection SARS-CoV-2 in China, it is difficult to collect samples from the early stage of infection for verification, which is the limitation of this study. Second, other effects influencing the detection of COVID-19 by GICA remains unclear. Third, only one commercial kit was used for comparison in this study, and whether the pore size of the chromatography membrane had an effect on the test results remains largely unknown.

GICA and equipment-dependent CA were usually used to detect antibodies in clinic, GICA is widely used to detect antibodies for the advantages of convenient, fast, low cost, suitable for screening large sample and require minimal equipment. In this study, we found that EDTA-K2 anticoagulated plasma has higher positive rate than other anticoagulated plasma (sodium citrate and lithium heparin) or serum, especially, the results of EDTA-K2 anticoagulated plasma detected by GICA was high consistent with CA results. Further study shown that EDTA-K2 could amplify positive signal by chelating colloidal gold. Therefore, we suggested that EDTA-K2 anticoagulated plasma maybe more suitable for the detection of SARS-CoV-2 antibodies. However, more positive samples with SARS-CoV-2 were needed to further verify this results.

This study was approved by the Hankou Hospital Ethics Committee (No. HKYY-2020-028), and according to the Ethics Committee review, patients informed consent was not required. It is noteworthy that these specimens taken were part of routine hospital procedure, rather than being taken specifically for this study. There was no additional manipulation or injury performed on the participants. Everything was done to ensure that the patients personal information was maintained under strict privacy. This study complied with the Declaration of Helsinki.

French skincare brand Caudalie has released a new anti-ageing range of products that comes with Harvard Medical School's stamp of approval.

The improved Resveratrol-Lift line was created from a partnership between Caudalie founder Mathilde Thomas and the school's Dr David Sinclair, an acclaimed biologist who specialises in the science of ageing.

They patented a new combination of ingredients, adding vegan collagen booster to the existing formula of vine resveratrol and hyaluronic acid, which is also patented.

A known anti-ageing antioxidant, resveratrol can be found in grape skin - a tribute to Caudalie's roots, which originated in a vineyard in France. The new recipe is said to be twice as effective as retinol and to help the skin produce five times the amount of natural collagen.

Local brands Ying The Label and RISIS have joined hands for a sustainable capsule of accessories.

Called Re:loop, the collection features upcycled fabric necklaces by Ying, paired with RISIS's signature orchid charms. The gold used for the charms was recycled from unused wastage, according to RISIS chief executive Verene Ng.

The necklaces, priced at $228, come in six fabric designs and four gold finishings for the orchid charm. Shoppers can mix and match the fabric and charms.

The collaboration came about after Ying founder Phuay Li Ying discovered during the circuit breaker that her tailor had plenty of uncut fabricleft over. It spurred her to find creative ways to upcycle the fabrics.

French luxury brand Dior has revamped its signature Rouge Dior lipstick with a refillable case. The lipstick's exterior retains its sleek silver trim and "CD" initials, but you can now pop the lipstick bullet in and out of the case.

A Rouge Dior lipstick, available in 35 shades here, costs $56. Refills (price to be confirmed) will be launched on Aug 15. The house also introduced two new finishes - an extra-matte Velvet with long-lasting comfort and a bright Metallic.

Spanish luxury house Loewe has a new member in its Puzzle bag family: the Nano Puzzle, a rescaled version of the iconic silhouette and the smallest iteration of the handbag yet.

Dash Shaws Sophomore Effort Cryptozoo Fascinates on the Surface – Film School Rejects

From 2016 to 2020 in Richmond, Virginia, writer-director-animator Dash Shaw and animation director Jane Samborski hand drew Cryptozoo with a small team of artists. On the surface, so what? The age-old tradition of meticulously hand-drawn animation isnt a modern-day spectacle simply because the practice is almost extinct. But the married couple isnt just bringing hand drawn animation back to cinema. Theyre revolutionizing it. Of course, thats a loaded statement.

The film follows human cryptozookeeper and cryptid (an animal whose existence is disputed or unsubstantiated) rescuer Lauren Gray (Lake Bell) and her new cryptid understudy, Phoebe (the cleverly cast Angeliki Papoulia), a gorgon from Greek mythology (think: Medusa) who must tranquilize her snakes (read: hair) and hide them to fit in. She also has to cover and disguise her eyes to keep from turning others into stone.

Lauren and Phoebe are searching for the ethereal Japanese Baku, a pig-elephant-esque cryptid that emits hundreds of little smoky, pearlescent blue spirals and is known for its omnipotent ability to steal dreams a truly mindboggling creature. They intend to bring the Baku back to the cryptozoo, where it will be safe from deep state lackeys who want to capture it in order to mine its power for military use against the rising 1960s counterculture.

The stacked cast also features a unicorn-meddling hippie voiced by Michael Cera, a wheeling and dealing dive-bar Mr. Tumnus of sorts voiced by Peter Stormare, and more natural and supernatural oddities voiced by Grace Zabiskie, Jason Schwartzman, Thomas Jay Ryan, and Louisa Krause. They bring to mind acid-induced dream logic adventures like Belladonna of Sadness, Fantastic Planet, Yellow Submarine, and that banned Mormon cartoon from the 80s.

Its difficult to land on a lone descriptor that accurately captures the breadth of animated expression in the mythological menagerie that is Cryptozoo. However, any number of words would suffice to describe the experience as a whole: kaleidoscopic, hallucinatory, inventive words that could also apply to John Carroll Kirbys terrific score. The animation is, in Shaws own words, exploiting what drawing can do that live-action cant: depict[ing] what we cant see.

With inspiration from early 20th-century cartoonist Winsor McCay and other pioneers in the field, Shaw and Samborski took a new thin-line approach to Cryptozoo. From the first scenes skinny ribbons of red-blue forestry laid like neon over black slate, the animation of the couples second feature is very different from their 2016 debut, My Entire High School Sinking into the Sea, the thick-black-lined subjects of which looked more like newspaper comic strip characters. Here, outlines are hardly traceable and sometimes invisible to the naked eye.

The near-absence of densely lined barriers between different styles of animation creates a flattening effect that miraculously renders everything on the same plane at once. It allows characters and settings to exist in a fluid transition of color from top to bottom and heightens the gorgeous, trippy aesthetic of the film. Rocks are painted like Pollocks, strewn globs of oil protruding from the surface. Cryptids and humans alike are painted with watercolors (almost all of them done by Samborski herself). Settings are comprised of wild patterns, texture swatches, pencil sketches, impressionistic creations, and so much more. Its insanely impressive, a full-force imagination. But where that nebulous energy thrives in the vast imagination of the animation, it sours in the stale narrative.

Unfortunately, and quite shockingly, another set of less desirable words would also suffice to describe the Cryptozoo experience: tiresome, aimless, strained. I want to be clear: these words do not apply to the animation, which remains wondrous throughout, but to the story at the center. But why? We travel from California to Florida to Kentucky and back with a pursuant sinister military head on our tail and a seemingly infinite cast of newly minted cryptids. On paper, it sounds like a rollercoaster. A seminal tarot card reading sequence stands out as one of the films most compelling. Yet, its those few compelling moments down the road that awaken the realization that, while transfixed with the morphing of colors and unfolding of textures that define Shaws idiosyncratic direction, the story is missing an emotional core.

Racism is paralleled through the tangled relationship between humans and cryptids. The sickening effects of capitalism on non-profit-pursuits-turned-profitable sit center stage thematically as Lauren begins to question the function of the cryptozoo/sanctuary/theme park and its effect on the cryptids on display after Phoebe expresses concerns. Theres no shortage of ambition in what Shaw tries to address in the story, but it feels inert and lacks a sense of navigation when it comes to tackling, or even just touching on, themes like racism, capitalism, and captivity. They fall woefully short of the gripping standard set by the animation.

The spoon-fed themes and narrative predictability can be summed up in an interaction between the naked free spirits that stumble onto the cryptozoo in the opening sequence. There could be magic here, or a utopia, the man says, already scaling the fence to see whats behind it. Utopias never work out, the woman replies in an all too heavy-handed, foreshadowing tone.

Films arent often fascinating and tedious at the same time, but consider the nature of the work, which feels more like wandering through a great museum than watching a movie at times. How often do you stare at your favorite artworks for ninety-five minutes uninterrupted? Or even thirty minutes? Barring outright obsession or a research initiative, it sounds a bit dull, no?

However, it would be ridiculous of me not to recommend it. At the end of the day, its a fever dream unlike any youve seen. It makes Fantastic Planet look tame. And in 2021, dreams are more important than ever. They form the collective imagination that drives us toward change. It is a singular vision such as this even if that singularity only occupies certain aspects of the film that draws out the real-world meaning at Cryptozoos core: Without dreams, there can be no future. And that is a nightmare.

MANILA, Philippines Unsatisfied with the firing of the militarys intelligence chief, lawmakers in the leftist Makabayan bloc in the House of Representatives want more heads to roll over red-tagging.

Rep. Arlene Brosas (Gabriela party-list) said Thursday that it is not enough for Maj. Gen. Alex Luna to be sacked as Defense Secretary Delfin Lorenzana, National Security Adviser Hermogenes Esperon and the militarys southern Luzon commander Lt. Gen. Antonio Parlade Jr. should also be fired.

"More heads must roll over this persistent and shameless red-tagging by the Duterte regime. Luna cannot be just a sacrificial lamb," she said.

Rep. Ferdinand Gaite (Bayan Muna party-list) backed this, adding that if Lorenzana is serious in exacting accountability over red-tagging, then he should dismantle the anti-communist task force created by President Rodrigo Duterte in 2017.

Their spokesperson Parlade has been repeatedly making such blunders. He's been wrongly accusing anyone including activists, church leaders, celebrities and other personalities, and several academic institutions," Gaite said. They should have long been sacked.

Rep. France Castro (ACT Teachers party-list), meanwhile, said red-tagging will persist despite Lunas firing unless the governments anti-communist task force is abolished and the highly-contested anti-terrorism law junked.

Lorenzana announced Thursday that he has fired Luna over a list of University of the Philippines students who were supposedly captured and killed by the military as communist rebels published on the Armed Forces of the Philippines Information Exchange Facebook page.

The list turned out to be false as those identified to have been captured or killed surfaced in a press conference to disprove the claims.

Lorenzana called the publication of the list an unforgivable lapse.

The Makabayan bloc has been repeatedly accused by government officials, including Duterte himself, as being fronts of communist rebels, despite not showing proof.

Stoke City bosss message to Tom Edwards as he heads to New York Red Bulls – Stoke-on-Trent Live

Michael ONeill will be keeping watch of Tom Edwards progress at New York Red Bulls in hope that he can come back ready to fulfil his potential at Stoke City.

Edwards has returned from a loan at Fleetwood Town ready to head over to the United States for the rest of 2021, trying to make sure a career which started so young is going in the right direction.

The right-back, who has just turned 22, made his Premier League debut at 18 and is under contract at Stoke until 2024 and there is hope that this move and the chances and tests it presents will be a big step in his development.

O'Neill said: I spoke to Tom yesterday and I think its a good opportunity for him. He had a loan spell at Fleetwood which went relatively well although there was a bit of upheaval with the manager leaving just before the Christmas period.

This presents a different opportunity for Tom. Hes spent most of his career at Stoke, coming through the club as a local boy. This gives him an opportunity to see a different side of football away from Stoke and Stoke City.

Hes still on a lengthy contract and this is an important opportunity for him and one he should grasp with both hands.

Young Potters defender Nathan Collins is attracting plenty of transfer interest ahead of Monday night's deadline.

If he takes his opportunity and does well, well continue to monitor him in the MLS and hopefully he can come back and try to reignite his career here at Stoke City.

Edwardss exit this month follows his old Academy teammate Thibaud Verlinden and there could be more to come.

But, with four new signings in, there are not likely to be any coming the other way before the 11pm deadline on Monday.

ONeill said: I dont think well be adding to the squad in that period. Our main focus is on players leaving the club and players on the periphery of the team looking at opportunities elsewhere.

Once we got our future editing out of the way, we started in with our questions about what role, if any, this entity had related to the chaotic situation associated with the Houston Texans, how a team that only 12 months ago held a 24 point lead in a divisional round playoff game, primed to host the first NFL conference title game in Houstons history. As we observed to our host in the midst of our research, we kept coming across some interesting names, such as Mephistopheles, Beelzebub and a Mr. Diabloespecially when looking at some very unique correspondence about deals where noted sports figures entered into unique contracts. Can you explain this?

Replied our host, Oh, that one? Well, that DID come from someone in the Texans organization. Yeah, that was back sometime around 2017 I thinkyep, definitely 2017, so we made the deal, and I obliged. Did the Texans not get that franchise quarterback?

(Totally Not Fake News) Well, yes, they did draft Watson and he has played pretty well for the team. However, it feels like nothing has gone right since then. We are pretty sure that the individual in question wanted to actually WIN a Super Bowl!

(Lucifer) Did they? You see, they only asked to get a franchise quarterback. They never asked for an ACTUAL championship. So, the team had a franchise quarterback. Now, as for everything else

(Totally Not Fake News) So, are you saying that you are behind all of the failures since 2017?

(Lucifer) Do I look like the GM, the Head Coach, the owner, for the Texans? No, I am notbut now that you mention it, I kinda recall a few things back some time ago, maybeoh, thats right, 2019. This one guy comes in, one of those preachy types, my personal favorites. Anyway, he tries to challenge me, saying that he can withstand me, casting me out or whatever. Well, I am generally a live and let live type of guy. Unless you ask, and I am always willing for others to ask, I tend to stay out. Well, this guy just kept calling me out. As we processed this information, the Morning Star noted Well, it was really quite easy. I only had to just trigger a little bit of temptation, getting this guy to think Hey, Ive been around football players. I can do this. Heck, I can do the Coach/GM job. I can do this all. Once I set that in motion, it has been working like a charm. Of course, the real work was by this guy. Ya know, it is not quite as fun as I would hope. I suppose I should have let the Texans get further along in their adventures before sparking the preachy guy, but they were so small time in their asks. Besides, this holy guy was really so small potatoesnot really worthy of an epic fall.

I dont say what happens after that fact. I am an entity that fulfills his contract. Nothing more, nothing less.

As we rapidly left our hosts residence, which somehow started to look more like a snakes den as we left, we continue to try to understand what transpired. Perhaps it is true that divine forces are at play behind the struggles of not just the Texans, but of the general situation for Houston sports. Normally, we at Totally Not Fake News subscribe to facts and logic and do not give credence to the spiritual world (especially since our prayer entreaties and live chicken sacrifices did not yield that billion-dollar jackpot last weekend). However, there is so much happening that mere logic is at a loss to explain. Perhaps it is the case. Still, we will continue to monitor the situation and determine just what is at the cause of the issues with the Texans and hopefully, what can be the cure.

Online Poker Launches In Michigan Friday Morning – CardPlayer.com

Online poker launched in Michigan Friday morning after state regulators approved a gaming license for the Little Traverse Bay Band of Odawa Indians.

The tribe, which runs Odawa Casino in Petoskey, partnered with PokerStars to operate online poker in Michigan. With the Michigan Gaming Control Board officially giving them an online license, PokerStars launched its Michigan platform at 11 a.m., according to a report from The Detroit News.

Along with the tribe, the board awarded licenses to Detroits Greektown Casino, which partnered with Penn Gamings Barstool Sportsbook & Casino to offer online gambling. Fox Bet, PokerStars sports betting platform, also launched with the license given to the Odawa Indians.

Michigans online gambling market launched last week, but the operators who received licenses did not have an online poker platform or a partner with one, leaving the initial launch reserved for other online casino games and sports betting.

PokerStars is the only operator in Michigan so far. BetMGM Poker, which has an obvious partnership with the MGM Detroit, could be the second to launch.

PokerStars now has operations in New Jersey, Michigan and Pennsylvania. With the recent ruling on the Wire Act, which allowed for most forms of interstate online gambling, and Gov. Whitmer signing a bill that would allow for online operators to join shared player pools, it opens a very real possibility for Stars to combine its player pools in all three states.

That law has not gone into effect yet, however, and would also need an agreement from the other states involved before it is launched.

Pennsylvania regulators were reportedly awaiting a ruling on the Wire Act before allowing operators from its state to join shared player pools. Some media outlets have reported that Michigan regulators are already in talks with the New Jersey Division of Gaming Enforcement and could see a compact between the states by the end of 2021.

Kevin Mathers, the information coordinator for PokerAtlas, tweeted screenshots of the sites platform shortly after the initial launch.

About an hour after its rollout, there were no-limit holdem cash games running at the $.01-$.02, $.25-$.50 and $.50-$1 stakes. Tables were available all the way up to $25-$50, but nobody was sitting. There were also several tournaments scheduled for the first day of operation, with buy-ins ranging from $3 to $30.

Is There A Future for Nationwide Online Gaming and Poker in the US? – Poker News Daily

With the complete changeover in the federal government in Washington, D. C., many have begun to float the trial balloon that there could be changes in the way online gaming and poker are viewed by the government. Added into this fact is the recent passing of one of online gamings most vocal opponents, the late Las Vegas Sands Corporation founder Sheldon Adelson. There are many moving cogs in this machine, however, and to think that there will be any immediate change in the situation might be just wishful thinking.

Despite Appearances, No Change in Federal Stance

At this point, the U. S. Department of Justice is working off the premise that online gaming is against the law because of the Wire Act of 1961. That law, initially established to prevent the Mafia from using phone lines to transmit sports bets, was used for 50 years as an explanation for preventing online gaming from taking place. But that was changed, reversed, and reversed again in just the last decade.

In 2011, the Office of Legal Counsel for the DoJ ruled that the Wire Act only applied to sports betting, which threw the floodgates open for the creation of intrastate online gaming. Nevada, New Jersey and Delaware quickly jumped on this bandwagon in 2013, but thats where the rush stopped. It would take four long years before another entity joined in with those three states the state of Pennsylvania and the Chinese Water Torture has continued since.

But the last presidential administration tried to throw cold water on the issue of online gambling. In a 2018 ruling from that administrations Office of Legal Counsel, the Wire Act was once again introduced as the standing law, overriding the previous decision. Now, that decision has been overturned.

Earlier in January, the U. S. Court of Appeals for the First Circuit overturned that viewpoint, ruling that the New Hampshire Lottery Commission could continue to sell lottery tickets. With the ascension of Joe Biden to the Presidency, it is unknown as to whether the DoJ will continue to pursue the issue regarding the enforcement of the Wire Act. What is known is that, for the moment, the 2011 decision by the Office of Legal Counsel is the standing opinion of the legal profession.

Continued Fluctuations and Bigger Issues

The continued fluctuations in the legal opinions are one of the reasons that the Commonwealth of Pennsylvania hasnt pursued a compact with any other operations. The law, as it was interpreted by the previous administration, made it illegal to even conduct online gaming. As it is allegedly interpreted now, there are still questions as to the legality of online gaming compacts, or the ability of states to partner together to offer gaming across borders, which would be crucial for the success of online poker.

Even with that said, however, the drive to push for online gaming regulations on the federal level just isnt there. Opposition to the federal efforts might have been removed with the passing of the anti-online gambling Adelson (the company now allegedly is looking into getting into the online gaming market in Nevada themselves), but there are still several strong opponents that would try to prevent online gaming and poker. Supposed family organizations (Focus on the Family prominently) and freedom oriented groups (Let Freedom Ring comes up in the anti-online gaming camp) are still against the industry, while Adelsons Coalition to Stop Internet Gambling is still out there, despite Adelsons death.

Then there is the simple fact that the Biden Administration has several more pressing issues on the table than whether people can put a bet down online. The continued COVID pandemic, financial and medical assistance for those affected by the virus, international relations and a highly wounded nation (especially after the insurrection of January 6) all might take a bit more precedence than my being able to check-raise an opponent while I hold the nuts. And this doesnt count the potential problems lurking in the future.

The federal government just needs to let the issue of online gaming be arbitrated by the states. They should also allow for these states to be able to reach business decisions between each other without threat of federal action (much like the feds have done with marijuana laws on a state-by-state basis). Even though there have been some changes for the positive, dont expect massive changes such as a bill for the federal regulation of online gaming to happen, even under the Biden Administration.

Online poker in the cards for North Dakota? – INFORUM

Rep. Jim Kasper, R-Fargo, introduced on Jan. 14 House Concurrent Resolution 3012, which would put a measure to allow the internet game on the 2022 general election ballot. If the measure is approved by the Legislature and gets a simple majority from voters, the state Constitution would change, and the process of how to bring online poker to North Dakota would begin.

The country is just going more and more into gaming online, Kasper said Thursday, Jan. 21. Lets not leave North Dakota as one of the few states that doesnt allow it.

Kasper said he played poker as a kid, but he became interested again when he saw the World Series of Poker on TV in 2003. It wasnt the typical five card draw he knew. Instead, it was Texas Hold em, which became increasingly popular in the 2000s.

After playing himself, he read in a magazine about the prospect of internet poker. That led to him drafting legislation in 2005 that would have legalized online poker in the state through a constitutional amendment.

The poker industry worldwide was watching my bill, he said.

The amendment passed the House but got a bad flop while in the Senate. The U.S. Justice Department opined that internet gambling of any kind was illegal, thanks to the Interstate Wire Act of 1961 that prohibited betting or wagering over wire communications.

The constitutional amendment was busted when the Senate voted it down 43-3.

The Justice Department has flipped in its opinion on whether online card gambling is allowed, with the latest stance in 2019 stating the Wire Acts ban applies to all forms of internet gambling, and not just sports.

The U.S. First Circuit Court of Appeals ruled last week against the Justice Department, saying the Wire Act does not apply to nonsports betting.

At least five states Nevada, Delaware, New Jersey, Pennsylvania and Michigan have legalized online poker. Kasper said he wanted to introduce the legislation again to see where it goes.

The legislation in North Dakota does not give details on how the game would be taxed or managed. It simply would allow voters to decide if online poker can occur in the state and if North Dakota would license and regulate the game.

Details on taxation and management would come later. The Legislature would set up a committee in 2023 or future sessions to figure out the details if voters approve the measure.

Its possible the state could join a group of states, he said.

There would be a lot of work that goes into it, he said.

Its unclear how much income online poker would draw from North Dakota. The poker industry estimated the 2005 legislation would have stacked up $500 million in annual taxes, with 80% going to reduce property taxes, Kasper said. Annual property taxes back then averaged about $400 million, he added.

Typically, proceeds from gambling in North Dakota go to "educational, charitable, patriotic, fraternal, religious or other public-spirited uses," according to state law.

Kasper said he hasnt been able to gauge support in the Legislature, but the proposed measure has sponsorship from both Republicans and Democrats.

Its just a game I enjoy, I think other people should have the opportunity to play it as well, he said.

The first rule of tapering is dont talk about tapering anytime soon.

Federal Reserve policy makers probably think the prospects for the economy in 2021 look significantly better than they did when they gathered mid-December. For now, though, the economy is facing the surge in Covid-19 cases that began last fall, along with the related stepped up caution and restrictions. Following their two-day meeting Wednesday, they focused on the downside.

They noted in their post-meeting statement that the recovery in the economy and job market has slowed. They again committed to keeping rates near zero until the labor market is tight and inflation has obviously cleared its 2% target rate, and to keep buying $80 billion in Treasurys and $40 billion in mortgage bonds per month until substantial further progress has been made toward those goals.

Yet though the economy has weakened in recent months, economists have been raising their growth forecasts. Following the Feds December meeting, for example, economists at JPMorgan Chase reckoned that in the fourth-quarter of 2021, gross domestic product would be 3.4% above its year-earlier level. Since then, they have raised that forecast to 5.4%.

There are multiple forces driving the improved outlook. First, there is the Covid-19 relief package that was passed late last year. Its extension of jobless benefits, renewed support for small businesses and the direct payments it provided to many Americans assuaged concerns that GDP might actually contract in the first quarter relative to the fourth. Second, with Democrats gaining control of the Senate, another round of fiscal support is looking likely in the months ahead. Throw in expectations that enough people will be vaccinated for life to take on some semblance of normal later this year, and things start looking pretty rosy.
