The end of TCU Horned Frogs vs. Gonzaga Bulldogs Sunday in the Round of 32 in the 2023 NCAA Tournament was like a gambling speedball. Regardless of which you side you bet, it was outrageous.

Gonzaga beat TCU 83-81 and the Horned Frogs covered as +4.5 underdogs. But how TCU cashed was bonkers. Just like the rest of this ridiculous tournament.

I mean, do you believe this s***? Gonzaga was anywhere from -4 to -4.5 favorites pregame depending on when you bet it. I gave out Gonzaga as -4 favorites earlier Sunday afternoon and it felt good at the end of the game.

Ill let you be the judge. TCU led 38-33 at the end of the 1st half but the Bulldogs took control in the 2nd half. Gonzagas biggest lead was 9 with 3:28 remaining.

Gonzaga got a key stop and C Drew Timme defensive rebound up 79-73 with :37 remaining before Timme was fouled. The Bulldogs were in the double bonus so Timme was on the line for two.

Timme missed the 1st and made the 2nd to give Gonzaga a 80-73 lead with 0:33 left on the clock. As a Gonzaga -4 bettor, I was pumped. It got even better when the Bulldogs got another stop and defensive rebound in TCUs next possession.

From here the nonsense begins. Gonzaga SF Julian Strawther craps his pants while inbounding the ball. Strawther BOUNCE passes it to TCUs Mike Miles Jr. and then fouls him at the rim to send Miles to the foul line.

Of course, Miles made both to bring the Horned Frogs deficit to 80-75. Then Gonzaga beat TCUs full-court press and SG Hunter Sallis got a breakaway dunk with :09 remaining to give Zaga a 82-75 lead.

Gonzaga -4/-4.5 looks good, right? Ehhh. No. Its never that easy in this racket. TCU charged down the court and banged a 3 to make the game 82-78.

The Bulldogs got fouled after inbounding the ball with less than :01 left to play. For a moment, I got excited. Gonzaga -4 was a freeroll with .07 seconds remaining and a Bulldog shooting two free throws. There is no way TCU covers.

It hurts to type out what happened and you already saw it. For the record, I hate TCU. Those clowns kicked me in the nuts in the College Football Playoff title game and then slapped me in the mouth Sunday.

This was the perfect ending for me in the 1st weekend of the NCAA Tournament. Most of my March Madness futures got nuked and I pretty much lit money on fire this weekend. Im done gambling. Until the Sweet 16.

‘Even more addictive now’: Malverne man’s gambling problem made worse by legalized sports betting – Newsday

Gambling nearly destroyed Joe Ruscillos life.By the time he was 20, he says, he owed bookies at least $100,000.

The Malverne native was a star in high school starting quarterback of the varsity football team at Valley Stream North High School his junior and senior years a decade ago, and a member of the varsity basketball and baseball teams.

His SAT scores were good enough to have SUNY Maritime College recruit him to play football, he said.

But Ruscillos downfall began when he discovered gambling. It started at 16 with small bets placed with bookies on games in just about every sport tennis was a favorite. The wagers,made illegally,quickly escalated into the thousands.

Now 26,Ruscillo said he hit rock bottom last year with the advent of legalized online sports betting in New York State.Over the past decade, he figures he wagered between $2 million and $3 million and lost between $200,000 and $300,000 before he began pulling his life back together with the help of his family and girlfriend.

Your first bet was $20 at 16, then it turns into 50, then it turns into 100. And you lose a couple bets in a row and you get yourself in a hole and things just happen, he said.

All of a sudden youre in thousands of dollars of debtto local bookies that you cant pay back. Youre in debtwith your friends that you cant pay back, he said. Its a dangerous road.

He owed so much, bookiesput him on payment plans.

As the debts mounted, he nearly lost his friends and family. He leaned on them for loans. When he didnt pay them back, tensions grew.

More than once his parents banished him from their home, forcing him to move in with his grandparents. He often did not have enough money to buy food or gas.

Hes a great kid with a bad problem, said his mother, Jennifer Ruscillo.

Besides the loans, he gambled with money from his jobs working in sanitation, his familys car glass businessand waiting tables.

The hard-earned money that you made that week you can lose in literally a minute, he said. Its scary.

His mother said she has taken him to everyone she could think of to get help doctors, psychologists, psychiatrists, Gamblers Anonymous meetings but none were able to cure him.

The beginningof legalized online sports betting in January 2022 in New York sent him over the edge, Joe Ruscillo said.

I was already addicted and then it made it a lot easier to get a bank account, put some money in it, and the deposit is right there in two seconds, he said.

Ruscillo said the ubiquitous ads especially on iPhones made it almost impossible for him to resist.

Me, knowing so much about it, even at the time knowing how bad it is and knowing how detrimental it is to your life, they still got me, he said of the ads.

He figures he lost between $40,000 and $50,000 within the first two months of online sports betting becoming legal in New York.

Nanoparticles used in solar cells are increasing the amount of energy absorbed from sunlight.[19] Solar cells are currently created from layers of silicon that absorb sunlight and convert it to usable electricity.[20] Using noble metals such as gold coated on top of silicon, researchers have found that they are able to transform energy more efficiently into electrical current.[20] Much of the energy that is loss during this transformation is due to heat, however by using nanoparticles there is less heat emitted thus producing more electricity.[20]

Nanotechnology is enabling the use of hydrogen energy at a much higher capacity.[21] Hydrogen fuel cells, while they are not an energy source themselves, allow for storing energy from sunlight and other renewable sources in an environmentally-friendly fashion without any CO2 emissions.[21] Some of the main drawbacks of traditional hydrogen fuel cells are that they are expensive and not durable enough for commercial uses.[22] However, by using nanoparticles, both the durability and price over time improve significantly.[22] Furthermore, conventional fuel cells are too large to be stored in volume, but researchers have discovered that nanoblades can store greater volumes of hydrogen that can then be saved inside carbon nanotubes for long-term storage.[22]

Nanotechnology is giving rise to nanographene batteries that can store energy more efficiently and weigh less.[23] Lithium-ion batteries have been the primary battery technology in electronics for the last decade, but the current limits in the technology make it difficult to densify batteries due to the potential dangers of heat and explosion.[21] Graphene batteries being tested in experimental electric cars have promised capacities 4 times greater than current batteries with the cost being 77% lower.[23] Additionally, graphene batteries provide stable life cycles of up to 250,000 cycles,[24] which would allow electric vehicles and long-term products a reliable energy source for decades.

How is pharmacogenomics affecting medical treatment?

Currently, doctors prescribe drugs based mostly on factors such as a patients age, weight, sex, and liver and kidney function. For a few drugs, researchers have identified gene variants that affect how people respond. In these cases, doctors can select the best medication and dose for each patient.

Additionally, learning how patients respond to medications helps to discern the different forms of their diseases.

For many years, NIH-funded scientists, through the Pharmacogenomics Research Network (PGRN), have studied the effect of genes on medications relevant to a wide range of conditions, including asthma, depression, cancer, and heart disease. The research findings are collected in an online resource called PharmGKB. In addition, the Clinical Pharmacogenetics Implementation Consortium (CPIC) was started as a shared partnership between the PGRN and PharmGKB to help lower the barrier to clinical use of pharmacogenetic tests. CPIC creates, curates, and posts freely available, peer-reviewed, evidence-based, updatable, and detailed gene/drug clinical practice guidelines. Another NIH-funded project, the Clinical Genome Resource, aims to define the clinical relevance of genes and variants for use in precision medicine and research.

Understanding pharmacogenomics would not be possible without sequencing the genomes of many people and comparing them, and then comparing their response to medicines. But we have also learned that a person's genome sequence is not everything when it comes to medication responses. The human body is a very complicated machine, and the instructions written in our DNA are just part of the process.

There are some cases, as with the breast cancer treatment tamoxifen, where a small study showed that there might be a relationship between someone's response to the medicine and a variant in theCYP2D6gene. However, this finding did not appear to be true in a larger study that involved many more people. That's why at this time, the U.S. Food and Drug Administration (FDA) labeling for tamoxifen does not recommendCYP2D6pharmacogenomic testing, butthe issue is still being reviewedas more research is conducted.

Another gene in the sameCYPfamily, calledCYP2C19, has variations which affect how your body can useclopidogrel(more commonly known as Plavix). This medication is a "blood thinner" which helps prevent blood clots, and thus reduces your risk of strokes or some heart attacks. If yourCYP2C19protein is not working properly due to a mutation in the gene, then you will not be able toprocess clopidogrel, and you need either a different dose or a different medication. As it turns out, these variants inCYP2C19are also more common in those with Asian ancestry. Although testing for variants in this gene is also not routinely recommended, you may wish to speak with your healthcare provider about the test if you are given a prescription for clopidogrel, particularly if you have East Asian family members.

PHARMACOGENOMICS: Driving Personalized Medicine

Personalized medicine tailors therapies, disease prevention, and health maintenance to the individual, with pharmacogenomics serving as a key tool to improve outcomes and prevent adverse effects. Advances in genomics have transformed pharmacogenetics, traditionally focused on single gene-drug pairs, into pharmacogenomics, encompassing all 'omics' fields, e.g., proteomics, transcriptomics, metabolomics, and metagenomics. This review summarizes basic genomics principles relevant to translation into therapies, assessing pharmacogenomics' central role in converging diverse elements of personalized medicine. We discuss genetic variations in pharmacogenes (drug-metabolizing enzymes, drug transporters, and receptors), their clinical relevance as biomarkers, and the legacy of decades of research in pharmacogenetics. All types of therapies, including proteins, nucleic acids, viruses, cells, genes, and irradiation, can benefit from genomics, expanding the role of pharmacogenomics across medicine. FDA approvals of personalized therapeutics involving biomarkers increase rapidly, demonstrating the growing impact of pharmacogenomics. A beacon for all therapeutic approaches, molecularly targeted cancer therapies highlight trends in drug discovery and clinical applications. To account for human complexity, multi-component biomarker panels encompassing genetic, personal, and environmental factors can guide diagnosis and therapies, increasingly involving artificial intelligence to cope with extreme data complexities. However, clinical application encounters substantial hurdles, such as unknown validity across ethnic groups, underlying bias in health care, and real-world validation. This review will address the underlying science and technologies germane to pharmacogenomics and personalized medicine, integrated with economic, ethical, and regulatory issues - providing insights into the current status and future direction of health care. Significance Statement Personalized medicine aims to optimize health care for the individual patients with use of predictive biomarkers to improve outcomes and prevent adverse effects. Pharmacogenomics drives biomarker discovery and guides the development of targeted therapeutics. This review addresses basic principles and current trends in pharmacogenomics, with large-scale data repositories accelerating medical advances. The impact of pharmacogenomics is discussed, along with hurdles impeding broad clinical implementation, in the context of clinical care, ethics, economics, and regulatory affairs.

By India Today Science Desk: There is a backup of youth in our cells that we have recently learned about that can be made to work, Dr David Sinclair, Professor in the Department of Genetics at Harvard Medical School said. He added that the technology to reverse aging and trigger bio switches is developing faster than we expected.

"The science of aging and reversal has come a long way. It was considered crazy 20 years ago, but today we understand what drives aging and there is a backup of youth in our cells that we have recently learned that can be made to work. Technology is developing faster," Dr Sinclair added.

David Sinclair was speaking at the India Today Conclave 2023 in Delhi. He discussed the science of age and disease reversal as more and more research pours into understanding the process of aging. The 53-year-old genetics professor, who is also an anti-aging researcher, was recently in the news as he claimed his biological age is 10 years younger.

"What we discovered is that there are genes that control how long we live. Some people are born with the best version of those genes who tend to live for 100 years and what we have discovered is how we live our lives, move and consume, sleep, and how relaxed we are modifies another type of system known as epigenomes," Dr. Sinclair said.

Known for his work on understanding why we age and how to slow its effects, Dr. Sinclair said that the good news is that aging is easier to slow and reverse than we thought. The bad news is that the process gives our bodies a shock.

Speaking about the diet he takes to slow the process, the genetics professor said that there is a set of genes that get turned on when there is not enough energy, like low sugar and protein, and they defend the body against the damage that causes the aging process.

"I skip meals at times, but my goal is to not eat a large meal until dinner and then have a healthy vegan meal and little alcohol. I switched completely to planets and my body has responded well, my skin looks better, my memory is better. The three-meal system plus snacks is aging us faster than we need to," Dr. Sinclair said.

Talking about plant-based meals that he switched from meat and alcohol to slow his aging process, Dr. Sinclair said that "The proteins that are in plants have a ratio of amino acids that stimulate longevity genes, and if you eat meat every meal the body does not fight the way plants help it do."

Whats the secret to looking and feeling younger? For some, its expensive surgeries. For others, its a change in health and wellness habits. But what does science say?

David Sinclair, a 53-year-old genetics professor and anti-aging researcher who claims his biological age is 10 years younger, swears by four key habits:

While these habits can certainly contribute to a healthy lifestyle overall, experts disagree about their link to longevity. For example, research is mixed on whether intermittent fasting can slow the aging process. But Sinclair, who is also the cofounder of Tally Health, a membership-based longevity platform that includes TallyAge testing, an at-home test to determine your biological age, says his habits have dramatically altered his life span.

My calculated biological age has been going down for the past decade or more to a point where Im predicted to live at least a decade longer than I would have if I hadnt done anything, Sinclair said in a recent interview with Insider. So its never too late.

In addition to a plant-based diet and cutting back on alcohol, Sinclair shared that he drinks one to two matcha teas per day and takes supplements that contain resveratrol, an anti-inflammatory compound most commonly found in red wine. But the jury is out on whether people can reap the same benefits in a pill form.

As soon as I see resveratrol in anybodys supplement stack, they lose all credibility, University of Washington longevity researcher Matt Kaeberlein told Insider. Its been disproven over and over and over in the longevity field, at least.

Sinclair, however, maintains that these habits arent just about living longer, but making the most of healthy years.

Nobody wants to be sick for a decade or have cancer that drags on or be frail, he told Insider. What were really talking about is preventing those things, or squeezing them into the last bit of life.

Biotechnology is the use of biology to develop new products, methods and organisms intended to improve human health and society. Biotechnology, often referred to as biotech, has existed since the beginning of civilization with the domestication of plants, animals and the discovery of fermentation.

Modern applications of biotechnology work most often through genetic engineering, which is also known as recombinant DNA technology. Genetic engineering works by modifying or interacting with the genetic cell structures. Every cell in an animal or plant contains genes that produce proteins. By modifying or interacting with genes, scientists can strengthen the characteristics of an organism or create an entirely new organism. Around the same time, humans learned to harness the biological process of fermentation to produce bread, alcohol and cheese. People also began changing the genetic makeup of domesticated plants and animals through selective breeding.

Selective breeding works by breeding parents with desirable characteristics to express or eliminate certain genetic characteristics in their offspring. Over time, species that are selectively bred evolve to be different from their wild ancestors. For instance, during the agricultural revolution, wheat was selectively bred to stay on its stem when harvested instead of falling to the ground like wild wheat. Dogs were selectively bred to be more docile than their wolf ancestors.

However, biotech methods such as selective breeding can take a long time to show changes in species. Biotechnology remained limited to these slow, agricultural methods until the 19th century when biologist Gregor Mendel discovered the basic principles of heredity and genetics.

Also, during that era, scientists Louis Pasteur and Joseph Lister discovered the microbial processes of fermentation. This laid the foundation for biotechnology industries where scientists interact more directly with the molecular and genetic processes of organisms.

Based on the work of these scientists, genetic engineering was developed in 1973. This method is the foundation of modern biotechnology practices and recent advances. It enabled the first direct manipulation of plant and animal genomes, which is the complete set of genes present in a cell.

Over the last 100 hundred years or so, biotechnology emerged with the following discoveries and advancements:

1919. Hungarian scientist Karl Ereky coins the term biotechnology.

1928. Alexander Fleming discovers penicillin, the first true antibiotic.

1943. Oswald Avery proves DNA carries genetic information.
