Finding a job is also an issue. Rodgers went back to college and got certified to work on computers, but he says potential employers dont want to be liable for his medical issues.

I cant get any companies to hire me, mainly because of my health, he said. That needs to be addressed.

Warner noted that many disabled individuals also worry about taking a full-time job because that can mean losing public benefits such as health insurance or Social Security payments.

Im glad for what we do have, as far as being able to be out in the community and gain access to commerce, but I do believe there are further steps that could be taken, Warner said.

Improving work opportunities for the disabled has been a focus for Harkin since he retired from the Senate in 2015. He has urged companies to overhaul training programs and do whatever else is necessary to increase their hiring of disabled Americans.

Congress could help by passing legislation that would address the issue of disabled individuals losing benefits if they find full-time employment, he said.

Sen. Tom Harkin participated in events this week marking the 30th anniversary of the ADA, including a plane ride on Friday. The first armless pilot, Jessica Cox shown with him in the plane was at the controls.

The two met through her advocacy work on the United Nations Convention on the Rights of Persons with Disabilities treaty, which they both continue to push.

Progress still hampered by battles on the fringes – McMinnville News-Register

Wednesday night, Portland Mayor Ted Wheeler, also the citys police commissioner, was simultaneously being gassed by federal agents and berated by protesters cursing his name, labeling his appearance at the 56th night of protest nothing more than a photo opportunity.

The videos of federal agents dispatched by the president gassing moms and clubbing veterans simply wanting to hold a conversation are abhorrent. While the agents have legal standing to protect the federal courthouse, the tactics administered have rightfully sparked a national outcry against a militarized federal government engaging in street clashes against its citizens. It is indeed a radical response from the U.S. Executive Branch, and likely more about politics than law and order as an election nears.

But its also not a surprising occurrence in Portland, where fringe activists have historically been allowed too much leeway to ruin the pure demonstrations for First Amendment rights amid honest attempts at progress.

To quote from a 2017 editorial in this newspaper:

Whether its Republicans on the right or Democrats on the left, allowing the most radical elements of their bases to compromise their principles and mute their voices is repugnant. It helps tighten the grip of raw partisanship, thus serving to thwart reasonable policy initiatives. ...

But when black-masked anarchists infiltrated the otherwise peaceful Portland demonstrations to pelt police, set blazes, smash windows, snarl traffic and engage in random acts of mayhem, organizers were more critical of the police for restoring order than the anarchists for disrupting it. ...

The anarchists were aided and abetted by then-Mayor Charlie Hales, who established a virtually unmatched record of ineptitude during his mercifully brief tenure. They got another boost when the district attorneys office dismissed charges against virtually all the 113 arrested.

Mayoral successor Ted Wheeler, displaying an equal lack of fortitude, allowed the anarchist fringe to bring city council meetings to a grinding halt for weeks with ceaseless demands for the police chiefs ouster. And it took nothing more than an anonymous threat for Portland anarchists to force cancellation of this years 82nd Avenue of Roses Parade.

It should have come as no surprise when anarchists began tossing Molotov cocktails, setting bonfires, defacing storefronts, shattering windows and pelting police again during the union movements annual May Day Parade, staged May 1 in Portland. After all, they openly recruited participants on social media sites.

But the response from the self-proclaimed voice of the American worker was, distressingly, a condemnation of the police rather than the bomb-throwers who spoiled the party and soiled the cause.

Those following the 50-plus days of protests in Portland this summer know they have been mostly peaceful and lawful. The righteous indignation of systematic racism and police brutality is warranted and deserving of cognitive discourse leading to legislation.

But the ongoing inability or unwillingness on both sides to filter extremist views has lead to a fictitious narrative, to the point where all cops are bastards and hundreds of moms wearing yellow are nothing more than Antifa in a new outfit.

With a subsection of protesters and a president eager to fan the flames of the ongoing urban warfare, as Wheeler described what he felt Wednesday night, it is difficult to find hope in positive reconciliation. But find hope, we must.

Throughout history, longevity and eternal life have been the ultimate goal of humanity. From the king of the ancient Chinese Qin dynasty to the European alchemists during the medieval age, countless people have devoted their lives to unveil the secrets of immortality.

Although none of such efforts have been proven to grant eternal life, nowadays, modern scientists succeeded to elongate the lifespan of simple organisms, such as worms and flies, by a considerable amount with meticulous manipulation of our genes. Among many of those studies, there have been three main methods that held their significance even in the human body.

Enzyme regulation

Most notably, the Scripps Research team discovered that the disruption of enzymatic pathways by small molecules can affect the lifespan of an organism. The team used Caenorhabditis elegans, a type of a roundworm, to test their hypothesis.

According to Benjamin Cravatt, Gilula Chair of Chemical Biology at Scripps Research, C.elegans worms were used for their experiment due to their relatively short lifespans (typically lives only a few weeks). Cravatts research involved about 100 compounds that were known to inhibit serine hydrolases in mammals.

In his experiment, Cravatt used each of the 100 molecules to block the enzymatic pathway and observed their effects on the lifespan of the C.elegans. When the team treated the worms that were 1 day into adulthood with the inhibiting compounds, they found that some of the compounds extended the average lifespan of the worm by at least 15%, according to Alice Chen, a graduate student in the Cravatt lab.

Chen elaborated that among the compounds, a carbamate compound called JZL 184, even extended the lifespan by 45% when treated at the optimal dose. Through further analysis, the team concluded that JZL 184 extended the worms lifespan by inhibiting fatty acid amide hydrolase 4(FAAH-4), which is known to break down a molecule called 2-AG, a molecule linked to aging in mammals.

Whats fascinating about this finding is that monoacylglycerol lipase was not present in C.elegans worms. MAGL usually breaks down the 2-AG molecules in mammals, but in the case of Chens experiment, FAAH-4 substituted the role of MAGL.

While the findings only apply to C.elegans worms as of now, the team stated that the FAAH-4 and 2-AG pathways will suggest a new path in extending human life.

Calorie Regulation

In addition, a research team led by David Sinclair, assistant professor of pathology at HMS, found that calorie regulation extends the lifespan of yeast cells. According to Sinclair, the PNC1 protein regulates a vitamin called nicotinamide, which is an inhibitor of the Sir2 molecule. Sinclair further hypothesized that since Sir2 typically extends the lifespan of yeast cells by stabilizing the ribosomal DNA, the regulation of nicotinamide, which is its inhibitor, will consequently prolong the organisms lifespan.

While the team believed that such a regulation process was initiated by the severe calorie restriction in yeast cells, they later discovered that calorie restriction had no impact on the Sir2 level. Thus, Sinclairs team tested the effect of the molecule NAD on PNC1 levels to confirm that NAD was responsible for altering the Sir2 level. It turned out: NAD had no effect on the PNC1 level.

Undeterred, with more in-depth analysis, the team finally reached the conclusion that nicotinamide, which is one of the products of the reaction between Sir2 and NAD, was responsible for the change in Sir2 level. Based on the correlation they found between Sir2 level and nicotinamide facilitated by calorie restriction, Sinclairs team is now investigating human genes that play the same role as PNC1, according to Harvard Medical School.

Mifepristone

Finally, the research team led by John Tower, professor of biological sciences at the University of Southern California, found that the drug mifepristone extends the lives of female flies that have mated.

Before the experiment, Tower had set a premise that the sex peptide in female flies from male flies reduces the lifespan of the female flies as it causes inflammation. In his study, he and his colleagues discovered that the drug mifepristone, also called RU-486, blocked the effects of the sex peptide during reproduction, which retained the female flies health and thus extended their lifespan.

What played the most significant role in the reaction was, according to Tower, a molecule called the Juvenile hormone.

According to Towers research team, the juvenile hormone is responsible for the growth and development of fruit flies throughout their life. The sex peptide, they elaborated, boosts the effect of the juvenile hormone, which causes harmful inflammation in the flies body and enervates the male flies by shifting the metabolic pathways.

Therefore, the inhibition of the sex peptide by mifepristone also regulates the level of juvenile hormone, consequently extending the lifespan of the flies. With their further testing of the drug on C.elegans, which has similar genes as those of humans, Tower suggested that their findings may be applicable to extending the lives of humans.

The life-extending technology is still in its burgeoning stage, as it only applies to simple organisms, such as worms and flies. However, while eternal life still seems improbable, humanity will proceed one step further to unveiling the secrets of longevity with scientists perpetual efforts.

Air Force awards contracts to develop Skyborg combat drones that ‘learn’ how to fight – Stars and Stripes

Four companies have been chosen to develop combat drones that will use artificial intelligence to either fully execute or support manned aircraft on strenuous or dangerous missions, the Air Force said.

Boeing, Northrop Grumman, General Atomics and Kratos Unmanned Aerial Systems were awarded contracts in support of the Air Forces Skyborg Vanguard Program, the Air Force Life Cycle Management Center said in a statement Thursday. The companies will compete for up to $400 million in orders, it said.

The program name evokes the cyborg man-machine hybrids of science fiction, but also perhaps the fictional artificial intelligence network Skynet in the Terminator film series.

Will Roper, the Air Forces assistant secretary for acquisition, technology and logistics, suggested a more endearing fictional creation, Defense News reported. At a conference last year, he likened it to the beep-booping droid R2-D2 of the Star Wars universe that helps Luke Skywalker and his father pilot their fighter spacecraft.

The real-world drones are supposed to include AI upgrades to allow them to learn how to engage enemy defense systems, as well as to share data with manned F-35 and F-15 fighters. This would allow the drones to operate in tandem with the jets in a wingman role, officials said.

Because autonomous systems can support missions that are too strenuous or dangerous for manned crews, Skyborg can increase capability significantly and be a force multiplier, Brig. Gen. Dale White, program executive officer for fighters and advanced aircraft, said in the statement. We have the opportunity to transform our warfighting capabilities and change the way we fight and employ air power.

The new drones, which will eventually replace the MQ-9 Reaper, will be designed to be attritable, meaning they will be inexpensive enough that losing one in combat would be tolerable.

The first Skyborg drones are expected to be operational by 2023. At least two models a ground attack and an air superiority version are expected to be fielded, according to Aviation Week & Space Technology.

The U.S. is not the only country developing stealthy combat drones, although its not clear whether any others are AI-enabled.

In October, China featured a prototype of the GJ-11 Sharp Sword drone at its national day parade. Russia, meanwhile, has released video of a much larger Sukhoi Su-70 drone flying in formation with an Su-35 fighter, indicating that the two are capable of operating in tandem.

And Israel has already used in Syria a stealth loitering drone that can crash into targets if its detected, or return to base if the strike is called off.

The Knicks are reportedly hiring Tom Thibodeau and Knicks Twitter lost it – For The Win

Theres always some sort of drama or controversy with the New York Knicks and their fans. If its not one thing, its the other.

On Saturday, it was their coaching hire. The Knicks are moving forward to make former Bulls and Timberwolves head coach Tom Thibodeau their next head coach, according to a report from ESPNs Adrian Wojnarowski.

The two sides are in the middle of hammering out the final details on a contract that would keep Thibodeau in New York for the next five years. It will be on him to shape the prime of R.J. Barrett and Mitchell Robinsons careers.

You can already hear the sound of creaking knees in the background just reading that tweet.

The Knicks are in a unique space. Theyve got a ton of young talent on that team starting with Barrett and Robinson. They needed a coach who could help bring out their best while also shaping their future.

And Thibodeau definitely has one part of that down. Lets, uh, leave the Timberwolves aside for a sec. He coached Derrick Rose to an MVP season, turned Jimmy Butler and Luol Deng into All-Stars and turned Joakim Noah into a Defensive Player of the Year.

On the other hand? He ran also ran pretty much all of them into the ground Jimmy Butler is an exception because hes a cyborg.

Thibodeau is known to grind his players away with heavy minutes and that concerns a lot of Knicks fans.

The jokes were pretty funny, though.

Its all good, Knicks fans. There are a couple of silver linings, at least! First, at least its not Jason Kidd. Second, we get to use this Vine again!

You would be hard-pressed to find anyone who doesn't harbor incredibly strong feelings about President Trump. Millions of Americans hate him. Millions still support him. "I don't have an opinion about Trump" is not something you'd expect to hear in a casual or political discussion - which is precisely the point.

For decades, Trump has been a lightning rod attracting the electrical discharges of hate and admiration with little neutral voltage in between. After he beat Hillary Clinton in 2016, he began to get zapped by hundreds of thousands of opinionated charges, and then literally millions, emanating from around the globe.

Today we could say these charges have become "matter versus antimatter," creating a dangerous energy field that may not be able to be shut down regardless of the outcome of November's presidential election.

In a speech at the American Enterprise Institute's annual dinner last year, Nikki Haley, a former U.N. ambassador and former South Carolina governor who is still a powerful voice in the GOP, cut to the chase regarding Trump: "President Trump is a disruptor. That makes some people very happy, and it makes some people very mad." She is correct - the president is a disruptor, and the need to disrupt likely is in his DNA. Our nation and the world haven't experienced another president like him.

Forget "The Art of the Deal," the book that introduced many Americans to Trump. For the past five decades, he has used "the art of disruption" to break down seemingly insurmountable walls to achieve his goals. Many times, he succeeded. Sometimes, the bricks and mortar collapsed upon him.

But even when they caved, he crawled out, dusted himself off and went about disrupting the next obstacle in his path. Those who have come to hate Trump must think they are watching a never-ending, real-life version of the "The Terminator," with Trump playing the role of the unstoppable cyborg.

Since Haley made her remarks last November, the world has been turned on its head by a deadly pandemic and our nation has been disrupted by legitimate protests and civil unrest tinged with troubling violence.

But through it all, Trump remains virtually the same. What you see is what you get. He's still that one-man disruptive force of often epic proportions. He goes full speed ahead - damn the (rhetorical) torpedoes!

Most media appear to be left of center in the U.S. and many other countries. They've come to truly loathe Trump and made their low opinions of him abundantly clear. Some world leaders and their staff have scoffed at the president too. Trump the disruptor, marching to his own beat, truly rubs them the wrong way. They want him gone.

On the other hand, the millions of voters in the U.S. who strongly support Trump need him to remain that disruptive force against both major political parties and the entrenched elite establishment. Power centers, they believe, never have had their best interests at heart.

As the election approaches, our nation is in a precarious and volatile position. Millions of Americans seem to be on the edge of despair - whether from the destructive fallout from the pandemic, its politicization, the ugly polarization of our politics or the pockets of anarchy.

The killing of George Floyd in Minneapolis was barbaric, and those who rose up in protest were right to do so; their voices need to be heard. But violence in the streets, destruction of property and calls to "defund the police" are deeply worrisome to many Americans. Such anarchy goes too far even for some Democrats, liberals and minority communities. No one wants violence in their neighborhood, town or city, especially Americans who may be struggling to survive.

Against this backdrop, voters will be asked to choose between our disruptive president or tentative former Vice President Joe Biden. I wonder if Democrats privately have confidence in the polls showing Biden leading Trump - or confidence in Biden himself.

But if the storms continue to rage, I suspect enough voters - frightened by the anarchy and nervous about the reversal of course sought by some on the left - will choose to ride it out with Trump until 2024. Then, they'll take a deep breath, collect their thoughts and elect America's first female president.

The X-Men’s Most Unstable Team Is Killing Each Other – Screen Rant

The X-Men started the Hellions hoping it would be a superhero therapy group... but therapy doesn't usually involve mutant guns and chest blasts.

Spoilers for Hellions #2!

The X-Men have been feeling experimental since the start of the Dawn of X relaunch, and their latest attempt at making the lives of mutants better is not going well. The ruling body of the mutant nation Krakoa, the Quiet Council, put together a team of unpredictable, antisocial, even villainous mutants, then tasked them with performing missions that the Council felt would be cathartic and productive. They called that team the Hellions. InHellions #2, the therapeutic superhero squad takes on their first mission... to zero success and lots of violence between teammates.

Hellionsis written by Zeb Wells, illustrated by Stephen Segovia, and colored by David Curiel. The team roster mixes popular X-Men heroes with classic and obscure villains, all under the banner of the new mutant cause of Krakoa and its belief that even the team's worst villainscan find clemency there. The current roster includes Cyclops' brother Havok, psychic ninja Psylocke, feral fighter Wild Child, indoctrinated soldier Orphan-Maker and his protectivecyborg Nanny, emotional manipulator Empath, and techno-shapeshifter Greycrow, with the supervision of mad geneticist Mister Sinister.

In Hellions #2, the teamsets out to destroy an old lab of Sinisters located beneath a now-abandoned orphanage that Sinister once used as a cover to obtain experimental subjects. Much of the team has ties to Sinister and his baby-snatching operation, and tensions are immediately high. Wild Child attacks a police officer with a K-9 companion in an instinctive effort to become the pack leader.Havok, who grew up there, insists he doesn't belong with the traumatized crew, but Psylocke senses his anxiety is loudest of all since he grew up there under Sinisters' testing schedule.

When Empath decides to blow off steam by nudging Nanny into a frenzy of protective worry, she latches onto Greycrow as if he's a waywardchild. Greycrow doesn't find it funny; he shoots Empath in the head. Since the X-Men have effectively conquered death, this is a matter of paperwork rather than a permanent loss. But stakes get higher the further they delve into the lab, and the team doesn't get any more cohesive.

The real violence begins when the team discovers that diabolical witch Madelyne Pryor has taken up residence in the lab and has been growing half-mindless mutant soldiers from Sinister's leftover, half-fried genetic samples. The enemy mutants may not be highly skilledcombatants, but Madelyne sends each one to fight a Hellion they're perfectly suited to disable. While the others fall, Pryor approaches Havok personally and reminds him of their past romantic history, which leaves him mesmerized enough to leave with her. As her zombifiedclones drag the dead and incapacitatedHellions away, the only two who remain are Psylocke and Wild Child... who sees that the ninja pack leader is wounded and weak, and goes in for the kill.

Hellions is only two issues in and the kinks have clearly yet to be worked out of this roster. Still, since most of the roster is dead, MIA, or trying to kill each other, it might be time to have a long talk between Xavier, Sinister, and--if he ever returns--Havok about the viabilityof this shooting-and-stabbing-based therapy program. Have theQuiet Council given any thought to art therapy? Role-playing? A villains-only dance studio?

Two Immunotherapies Converted Into Single Therapy for Certain Blood Cancers – Technology Networks

Some of the most promising advances in cancer treatment have centered on immunotherapies that rev up a patients immune system to attack cancer. But immunotherapies dont work in all patients, and researchers have been searching for ways to increase their effectiveness.

Now, researchers at Washington University School of Medicine in St. Louis have combined two immunotherapy strategies into a single therapy and found, in studies in human cells and in mice, that the two together are more effective than either alone in treating certain blood cancers, such as leukemia. Evidence also suggests that the new approach could be safer than one of the most recent cellular immunotherapies to be approved by the FDA, called CAR-T cell therapy, in which the immune systems T cells are engineered to target tumor cells. Cell-based immunotherapies are most commonly used against blood cancers but can be harnessed against some solid tumors as well, such as prostate and lung tumors and melanoma.

The study appears online in the journal Blood.

In the new research, the scientists have harnessed the technology used to engineer CAR-T cells and, instead of modifying specialized immune cells called T cells, they have used similar technology to alter different immune cells called natural killer (NK) cells. The resulting immunotherapy combines the benefits of both strategies and may reduce the side effects that are sometimes seen in CAR-T cell therapy. In some patients, for example, CAR-T cell therapy causes a cytokine storm, a life-threatening overreaction of the immune system.

Immunotherapies show great promise for cancer therapy, but we need to make them more effective and more safe for more patients, said co-senior authorTodd A. Fehniger, MD, PhD, a professor of medicine. This combined approach builds on the treatment strategy that we developed for leukemia patients using natural killer cells. We can supercharge natural killer cells to enhance their ability to attack cancer cells. And at the same time, we can use the genetic engineering approaches of CAR cell therapy to direct the natural killer cells to a tumor target that would normally be overlooked by NK cells. It fundamentally changes the types of cancer that NK cells could be used to treat, both additional blood cancers and potentially solid tumors as well.

In past work, Fehniger and his colleagues showed that they could collect a patients own NK cells, expose the cells to a specific recipe of chemical signals that prime the cells to attack tumors, and then return the primed cells to patients for therapy. This chemical exposure is a sort of basic training for the cells, according to the investigators, preparing the NK cells to fight the cancer. When the cells are then returned to the body, they remember their training, so to speak, and are more effective at targeting the tumor cells. Because their training has given the NK cells a memory of what to do when they encounter tumor cells, the researchers dubbed them memory-like NK cells.

In small clinical trials conducted atSiteman Cancer Centerat Barnes-Jewish Hospital and Washington University School of Medicine, such cells were effective in putting some patients with leukemia into a lasting remission, but they didnt work for everyone. Some tumor cells still evaded the memory-like NK cells, despite the cells basic training. To help the cells find the tumor cells, so their basic training can kick in and kill the correct target, the researchers modified the memory-like NK cells with the same CAR (chimeric antigen receptor) molecule that is typically used to target T cells to tumor cells. The CAR molecule is flexible and can be modified to direct the cells to different tumor types, depending on the proteins on the surfaces of the cancer cells.

The resulting hybrid cells were more effective in treating mice with leukemia than memory-like NK cells alone, leading to longer survival for mice treated with CAR memory-like NK cells. The researchers also found the therapy to be effective despite the fact that the mice were given relatively low doses of the cells.

One aspect of this study I find most exciting is how nicely these hybrid NK cells expand in the mice to respond to their tumors, said co-senior authorMelissa Berrien-Elliott, PhD, an instructor in medicine. We can provide a tiny dose and see an incredible amount of tumor control. To me, this highlights the potency of these cells, as well as their potential to expand once in the body, which is critical for translating these findings to the clinic.

Fehniger also pointed out that an advantage of NK cells in general and for biological reasons that the scientists are still working to understand NK cells dont trigger a dangerous immune response or the long-term side effects that T-cell therapy can cause in attacking the patients healthy tissues, a condition called graft-versus-host disease.

In all of the clinical trials exploring any type of NK cells, we dont see the troublesome side effects of cytokine release syndrome or neurotoxicity that we see with CAR-T cells that can profoundly affect patients, Fehniger said. These side effects can be life-threatening and require intensive care. Were still working to understand how NK cells are different. But if you can get the benefits of CAR-T cells with few if any of the side effects, thats a reasonable line of research to pursue. Another benefit of this safer therapy is the potential to give these cells to patients at an earlier stage in their disease, rather than using them as a last resort.

Other groups have developed CAR-NK cells, but a major difference is that other groups NK cells came from donated cord blood or induced stem cells, rather than adult donors or the patients themselves.

The other groups have artificially differentiated stem cells into something that resembles an NK cell, Fehniger said. With that strategy, theres no guarantee that those cells will have all the properties of typical mature NK cells. In contrast, were starting with adult NK cells, so were more confident that they will have all the inherent properties and behavior of adult NK cells, which we have already shown to be effective in certain types of cancer patients, especially those with leukemia. Inducing memory properties adds to their persistence and effectiveness against many cancer types.

Over the next several years, we would like to be able to scale up this process to produce enough cells for a first-in-human clinical trial, and investigate their effectiveness in different types of human blood cancers, he said.

FINDINGS

Researchers at the UCLA Jonsson Comprehensive Cancer Center and UCLA School of Dentistryhave identified a potential new combination therapy to treat advanced head and neck squamous cell carcinoma, the most common type of head and neck cancer.

A study in mice found that using an anti-PD1 immunotherapy drug in combination with PTC209, an inhibitor that targets the protein BMI1, successfully stopped the growth and spread of the cancer, prevented reoccurrences and eliminated cancer stem cells. This is the first preclinical study to provide evidence that targeting BMI1 proteins enhances immunotherapy and eliminates cancer stem cells by activating antitumor immunity.

BACKGROUND

Immunotherapies using PD1 blockade have transformed the way people with difficult cancers are treated. Currently, PD1 blockade combined with chemotherapy is approved for recurrent or metastatic head and neck cancer, giving people whose disease would have otherwise been seen as a death sentence another option. However, response rates are not very high and response duration is relatively short, indicating that this type of cancer might be resistant to PD1 blockade.

To help overcome immunotherapy resistance, UCLA researchers have been studying the role of cancer stem cells and the protein BMI1. Growing evidence suggests cancer stem cells might be responsible for such resistance, as well as for relapse or reoccurrence, and BMI1, which functions in several cancers, including head and neck, has been found to control cancer stem cells self-renewal. Targeting cancer stem cells may be critical for improving the efficacy of immunotherapy and preventing tumor relapse.

METHOD

The team used a mouse model of head and neck squamous cell carcinoma that fully mimicked human cancer development and metastasis, allowing them to perform lineage tracing of BMI1-positive cancer stem cells in an undisturbed tumor immune microenvironment. They then tested whether BMI1 cancer stem cells could be eradicated by PD1 blockadebased combination therapy using both pharmacological and genetic inhibition of BMI1. They found that inhibiting BMI1 not only helped eliminate the BMI1 cancer stem cells but also enhanced PD1 blockade by activating tumor cellintrinsic immunity, which inhibited metastatic tumor growth and prevented tumor relapse.

IMPACT

Many people with advanced head and neck cancers who are treated with PD1 blockade and chemotherapy eventually see their cancer return and become resistant to the therapy. This preclinical study provides an important foundation for developing a new PD1 blockadebased combination therapy with BMI1 inhibitors that have the potential to help overcome resistance to the immunotherapy.

ABSTRACT

Nearly 500000 fatalities due to COVID-19 have been reported globally and the death toll is still rising. Most deaths are due to acute respiratory distress syndrome (ARDS), as a result of an excessive immune response and a cytokine storm elicited by severe SARS-CoV-2 lung infection, rather than by a direct cytopathic effect of the virus. In the most severe forms of the disease therapies should aim primarily at dampening the uncontrolled inflammatory/immune response responsible for most fatalities. Pharmacological agents antiviral and anti-inflammatory molecules have not been able so far to achieve compelling results for the control of severe COVID-19 pneumonia. Cells derived from the placenta and/or fetal membranes, in particular amniotic epithelial cells (AEC) and decidual stromal cells (DSC), have established, well-characterized, potent anti-inflammatory and immune-modulatory properties that make them attractive candidates for a cell-based therapy of COVID19 pneumonia. Placenta-derived cells are easy to procure from a perennial source and pose minimal ethical issues for their utilization. In view of the existing clinical evidence for the innocuousness and efficiency of systemic administration of DSCs or AECs in similar conditions, we advocate for the initiation of clinical trials using this strategy in the treatment of severe COVID-19 disease.

Teslas Next Gigafactory Will Be in Texas – Car and Driver

During its second-quarter 2020 earnings call, Tesla CEO Elon Musk announced that Texas will be the location of the company's next Gigafactory. Musk called the location "GigaTexas" and described the plan for it as an ecological paradise with biking and walking trails.

Construction of the factory already started over the past weekend, according to Musk. The location will be where Tesla will build Model 3 and Model Y vehicles bound for the East Coast. The 2000-acre facility will also be home of the Tesla Semi and the upcoming Cybertruck.

For the Fremont, California, factory, that location will continue to build Model S and Model X for worldwide distribution and will likely be the facility that builds the upcoming Tesla Roadster. It'll also be the home of Model 3 and Y vehicles bound for the West Coast.

Musk said that the location near Austin on the Colorado river would be an ecological paradise with walking and biking paths. The location will also be open to the public. During the call Musk said, "Ive never been more optimistic or excited about the future of Tesla."

TSLA, -6.34% is attracting "greater levels of enthusiasm" from investors thanks to its "demonstrated and perceived" technological dominance, analyst Adam Jonas at Morgan Stanley said in a note Tuesday. Jonas compared Tesla at $300 billion valuation, a milestone reached last week, with Apple Inc. AAPL, -0.24% at $300 billion, which the Cupertino, Calif., company crossed in early 2011, and Amazon.com Inc. AMZN, +0.74% at $300 billion, reached in late 2015. Tesla is a "significantly smaller company" than either companies were at the time, and Tesla's market-cap milestone reflects a "significant valuation premium" compared to when Apple and Amazon reached the same market value, he said. Tesla's revenue is less than half Apple's at the time of the $300 billion milestone, and 70% less than Amazon's at Amazon's market value milestone, Jonas said. Some S&P 500 index's SPX, -0.61% market ratios are higher today, which explains some of the valuation premium for Tesla, he said. Tesla is scheduled to report second-quarter results on Wednesday after the bell, with Wall Street consensus calling for a loss. Some investors, however, remain hopeful the Silicon Valley car maker will surprise markets with a GAAP profit and be on its way to eventually join the S&P 500 index.
