Genetic variation may mediate the increased risk of cardiovascular disease (CVD) in chemotherapy-treated testicular cancer (TC) patients compared to the general population. Involved single nucleotide polymorphisms (SNPs) might differ from known CVD-associated SNPs in the general population. We performed an explorative genome-wide association study (GWAS) in TC patients. TC patients treated with platinum-based chemotherapy between 1977 and 2011, age 55 years at diagnosis, and 3 years relapse-free follow-up were genotyped. Association between SNPs and CVD occurrence during treatment or follow-up was analyzed. Data-driven Expression Prioritized Integration for Complex Trait (DEPICT) provided insight into enriched gene sets, i.e., biological themes. During a median follow-up of 11 years (range 3-37), CVD occurred in 53 (14%) of 375 genotyped patients. Based on 179 SNPs associated at p0.001, 141 independent genomic loci associated with CVD occurrence. Subsequent, DEPICT found ten biological themes, with the RAC2/RAC3 network (linked to endothelial activation) as the most prominent theme. Biology of this network was illustrated in a TC cohort (n=60) by increased circulating endothelial cells during chemotherapy. In conclusion, the ten observed biological themes highlight possible pathways involved in CVD in chemotherapy-treated TC patients. Insight in the genetic susceptibility to CVD in TC patients can aid future intervention strategies.

Both complaints address the infringement of Alleles patented mNeonGreen technology, an important reagent used in the development of therapeutics for COVID-19.

Alleles mNeonGreen protein is considered the worlds brightest monomeric fluorescent protein, and the technology behind that protein was patented in 2019. Prominent scientific journals have touted the use of mNeonGreen as the gold standard for use in assays testing neutralizing antibody and vaccine candidates. Regeneron, Pfizer, and BioNTech used mNeonGreen commercially without authorization from Allele.

I am pleased that mNeonGreen has played a pivotal role in the fight against COVID-19. In no way does Allele want to prohibit, or slow down development of vaccines or therapeutics discovered using this technology, says Dr. Jiwu Wang, Founder and CEO of Allele. SAN FRANCISCO, Oct. 09, 2020 (GLOBE NEWSWIRE) -- Vir Biotechnology, Inc. (Nasdaq: VIR) today announced the publication of preclinical research in an influenza animal model highlighting a new mechanism for enhancing the efficacy of monoclonal antibodies to treat viral infection and induce a protective response. Data demonstrate that selective engagement of an activating Fc receptor on dendritic cells by antiviral monoclonal antibodies induced protective CD8+ T cell adaptive responses. The paper, entitled Fc-optimized antibodies elicit CD8 immunity to viral respiratory infection, was published in the October 8, 2020 online edition of Nature.

In the past several years, we've gained a better understanding of how integral Fc mediated effector functions of monoclonal antibodies are for their therapeutic efficacy in pre-clinical models of neoplastic, infectious and inflammatory diseases, said Jeffrey V. Ravetch, M.D., Ph.D., study senior author and Theresa and Eugene M. Lang Professor and Head of the Leonard Wagner Laboratory of Molecular Genetics and Immunology at The Rockefeller University. These approaches have been successfully applied to anti-tumor therapeutics and have resulted in improved clinical outcomes in a variety of oncologic diseases. Our present studies have uncovered a significant new mechanism by which antibodies, through their Fc region, can not only engage innate immune responses but activate adaptive T cell responses, thereby stimulating protective anti-viral immunity in these models.

The research published in Nature focuses on the role of the Fc domain of monoclonal antibodies, regions with the capacity to bind to other immune cells through a family of receptors (the Fc receptors). By engineering antibodies with modified Fc domains to enhance binding to specific Fc receptors on innate immune cells, investigators observed an enhanced protective immune response. Certain modifications (GAALIE variants) were associated with activation of dendritic cells, as well as antiviral effector T-cells, indicating induction of the adaptive arm of the immune system, which is responsible for long-term immunity. Based on this research, monoclonal antibodies programmed with improved effector function represent a potential new approach in the design of therapeutic antibodies for both the prevention and treatment of infectious diseases.

By observing and learning from our bodys powerful natural defenses, we have discovered how to maximize the capacity of antibodies through the amplification of key characteristics that may enable more effective treatments for viral diseases, said Herbert Skip Virgin, M.D., Ph.D., study co-author and executive vice president, research, and chief scientific officer of Vir. Department of Biotechnology lab to be part of global Covid-19 vaccine assessment project – ETHealthworld.com

The Coalition of Epidemic Preparedness for Innovation (CEPI), a global initiative to develop vaccines against emerging epidemics, has named Translational Health Science and Technology Institute (THSTI), an autonomous institute of the Department of Biotechnology (DBT), as part of the Global Network of Laboratories for centralized assessment of Covid-19 vaccine candidates that are under development.

The Faridabad-based laboratory, under DBT, is one of the six laboratories that have been recognised by the CEPI. The CEPI network will initially involve six labs - one each in Canada, Britain, Italy, the Netherlands, Bangladesh and India, for the assessment of Covid-19 vaccine development.

Under the CEPI Global network, all the laboratories will use the same reagents and follow a common set of protocols to measure the immune response of multiple vaccine candidates under development and trial.

The Department of Biotechnology, Government of India, is supporting efforts for Covid-19 vaccine development and testing. THSTI runs comparability studies with other laboratories internationally and will form an important part of the global network, said DBT Secretary Renu Swarup.

We are very pleased that the bioassay laboratory at Translational Health Science and Technology Institute, an autonomous institute of Department of Biotechnology, has been identified by CEPI for centralised immunoassay analysis for COVID 19 vaccines under development globally. It is well-positioned to run comparability studies with other laboratories internationally and will form an important part of the global network. Homeostatis is the maintenance of the bodys overall inner resistance to change. This resistance stabilizes the body by regulating the internal environment, even as the external environment changes. A stable internal environment is needed for normal physiological function and survival of a living system.

Maintaining a stable internal environment requires constant monitoring, mostly by the brain and nervous system. The brain, more specifically the hypothalamus, receives information from the body and responds appropriately through the release of chemical messengers such as neurotransmitters, catecholamines, and hormones. These chemical messengers signal individual organs to change their functions in order to maintain homeostasis for the whole body.

For instance, if blood oxygen levels are too low, the brain signals the muscles controlling the lungs to breathe faster to increase oxygen intake. The brain also signals the heart to beat faster so other organs and tissues receive the oxygen they need. When oxygen levels return to normal, the brain signals the lungs and heart to return to their normal rates of function, a process called feedback.

Traditionally, the academic discipline of physiology views the body as a collection of interacting systems, each with its own combination of functions and purposes. Each system contributes to the homeostasis of other systems and of the entire organism. No system works inisolation, and the well-being of the person depends upon the well-being of the interactions between body systems. The traditional divisions by system are somewhat arbitrary. Many organs participate in more than one system (such as the heart and kidney), and systems might be organized by function, by embryological origin, or by other categorizations.

For instance, the neuroendocrine system is the complex interactions of the neurological and endocrinological systems. Together, the neuroendocrine system regulates many physiological processes, including those that maintain homeostasis. Furthermore, many aspects of physiology are not easily categorized by traditional definitions of organ systems because they are composed of interactions between organs in multiple organ systems.

The study of how physiology is altered in disease is pathophysiology. Pathophysiology focuses on how physiological processes fail to maintain normal function, resulting in the manifestation of disease symptoms.

Human anatomy deals with how all parts of the human body interact to form a functional whole. The study of anatomy is separate from the study of physiology, although the two are often taught together. While anatomy studies the structureof the parts of an organism, physiology is concerned with the way those parts function together. For example, an anatomist may study the types of tissues found in different parts of the heart, while a physiologist may study how the heart regulates blood flow to supply oxygen to other organs in the body. While anatomy and physiology study different aspects of human biology, together they provide a more complete picture of what the human body is and how it works.

The Haunting of Bly Manor and the Anatomy of an Easter Egg – Den of Geek

The nameless child youll see behind the staircase, when people migrate up and down the stairs, which is a lot. The ghost doctor we see in the study and then a couple of other places. And Viola, we see her in the attic, Kniest says.

By keeping the hidden ghosts consistent and identifiable throughout, Flanagan and the show create a feeling of lived-in history for Bly Manor. These are real inhabitants damned to spend their days standing around a cavernous manor, unnoticed by anyone other than the ultra observant Miles and Flora Wingrave. But of course, the reality of bringing in actors for Easter egg appearances is a touch more mundane than that, according to Kniest.

Its funny because the actors who play the parts come in, but then when we use (the characters) as ghosts, a lot of times theyre sort of background actors or stand-ins, or stunt doubles, the cinematographer explains. So they wait around a lot for us to get set up. But then I think theyre super excited when we get to actually put them on camera.

The scattered ghosts arent the only Easter eggs for The Haunting of Bly Manor. For one, Danis room at the hostel is 217, which is the number of a fateful room in The Shining (the book, not the film, though as director of Doctor Sleep, Flanagan is clearly well-read on both). Nor are the ghosts the only bit of visual flair for these nine episodes. Kniest, who previously shot horror film Hush alongside Flanagan, describes Bly Manors black and white eighth episode as one of the biggest draws of this particular job.
