Preclinical Study Shows Improvement In Recovery From Heart Attack With Exosomes – Anti Aging News

Recovery from experimental heart attacks can be improved with an injection of a mixture of heart muscle cells, endothelial cells and smooth muscle cells, but results are limited by poor engraftment and retention, plus there are concerns about potential tumorigenesis and heart arrhythmia.

Recent animal research in pigs has shown that using the exosomes naturally produced from a mixture of heart muscle cells, endothelial cells, and smooth muscle cells derived from human induced pluripotent stem cells yielded regenerative benefits that were the equivalent to the injected hiPSC-CCs.

Exosomes are membrane-bound extracellular vesicles that contain biologically active proteins, RNAs and microRNAs that are well known to participate in cell to cell communication, and are actively studied as potential clinical therapies for a wide range of conditions.

The hiPSC-CC exosomes are acellular and, consequently, may enable physicians to exploit the cardioprotective and reparative properties of hiPSC-derived cells while avoiding the complexities associated with tumorigenic risks, cell storage, transportation and immune rejection, said Ling Gao, Ph.D., and Jianyi Jay Zhang, M.D., Ph.D., University of Alabama at Birmingham corresponding authors of the study, published in Science Translational Medicine. Thus, exosomes secreted by hiPSC-derived cardiac cells improved myocardial recovery without increasing the frequency of arrhythmogenic complications and may provide an acellular therapeutic option for myocardial injury.

Studies involving large animals are required to identify, characterize and quantify all responses to potential treatments, prior to this study the feasibility of hiPSC-CC exosomes for cariad therapy had only been shown to be effective in mouse models and in vitro work.

The UAB studies involving juvenile pigs with experimental heart attacks had 1 of 3 treatments injected into the damaged myocardium: a mixture of cardiomyocytes, endothelial cells, and smooth muscle cells derived from human induced pluripotent stems cells, exosomes extracted from three cell types, and homogenized fragments from the cell types.

There were 2 primary findings from this study. Measurements of left ventricle function, infarct size, wall stress, cardiac hypertrophy apoptosis and angiogenesis in the animals treated with hiPSC-CCS, hiPSC-cc fragments or hiPSC-cc exosomes were found to be similar and significantly improved compared to those that recovered without any of the 3 treatments. Additionally, exosome therapy was found not to increase the frequency of arrhythmia.

During experiments with cells or aortic rings that were grown in culture, exosomes produced by hiPSC-CCs were found to promote blood vessel growth in cultured endothelial cells and isolated aortic rings. The exosomes also protected the cultured hiPSC-cardiomyocytes from the cytotoxic effect of serum-free lox oxygen media by reducing the programmed apoptosis cell death and by maintaining intracellular calcium homeostasis which had a direct beneficial effect on heart conductivity. Additionally, the exosomes also increased cellular ATP content which is beneficial as deficiencies in cellular ATP metabolism are believed to contribute to the progressive decline in heart function in those with left ventricle hypertrophy and heart failure.

Some of the in vitro beneficial effects were found to also be mediated by synthetic mimics of the 15 most abundant microRNAs that were found in the hiPSC-cc exosomes. It was noted that knowledge of the potential role of microRNAs in clinical application requires more research as it is far from complete.

People With HIV Have High Burden of HIV-Associated Neurocognitive Disorder – Neurology Advisor

People with HIV, particularly patients in sub-Saharan Africa and Latin America, have a high burden of HIV-associated neurocognitive disorder (HAND). This indicates the need for earlier neurologic care and initiation of antiretroviral therapy in this population, according to study results published in Neurology.

HIV, being a neurotropic virus, can invade the central nervous system during early infection, which is often associated with neurological complications like HAND. Study researchers sought to evaluate the burden and prevalence of HAND, as well as related factors in the global community living with the HIV.

This study was a meta-analysis of data from 123 cross-sectional and cohort studies that reported the prevalence of HAND in 35513 adults with HIV from 32 different countries. The pooled prevalence of HAND was estimated, and the overall worldwide burden of the disorder was evaluated.

Across studies, the diagnostic criteria for HAND consisted of the 2007 Frascati criteria in 64 studies, Global Deficit Score in 25 studies, and mental status examinations in 34 studies. Overall, the estimated prevalence of HAND in adults living with HIV was 42.6% (95% CI, 39.7-45.5). The prevalence of asymptomatic neurocognitive impairment (ANI) was 23.5% (95% CI, 20.3-26.8). Additionally, the prevalence of mild neurocognitive disorder (MND) was 13.3% (95% CI, 10.6-16.3) and of HIV-associated dementia (HAD) was 5.0% (95% CI, 3.5-6.8).

The prevalence of HAND was lower in individuals with a high level of nadir CD4 count (mean/median CD4 nadir <200, 45.2%; 95% CI, 40.5-49.9) compared with individuals with a low level of nadir CD4 count (mean/median CD4 nadir 200, 37.1%; 95% CI, 32.7-41.7).

The estimated worldwide number of patients with HIV and HAND was approximately 16,145,400 (95% CI, 15,046,300-17,244,500). The majority of cases were in sub-Saharan Africa, which was estimated to be in 72% of this population (n=11,571,200; 95% CI, 9,600,000-13,568,000).

Limitations of this meta-analysis were the inclusion of studies with varying definitions and diagnostic criteria for HAND and the use of screening tools that were limited when applied to certain target populations. Additionally, the use of Frascati criteria and the inclusion of patients with comorbidities that potentially contribute to cognitive impairment may have led to an overestimation of HAND prevalence in adults with HIV.

Based on the findings, the study researchers concluded that HAND should be prioritised among policy makers and HIV health-care providers for improved detection and efficient management of HAND integrated into routine clinical care in people with HIV.

The Duke Neurology Departments Nada El Husseini, MD, and Andrew Spector, MD, both appeared in the American Academy of Neurologys (AAN) AAN Alumni Leadership Newsletter for October 2020, where they discussed transforming telestroke during a global health crisis and how neurologists can reduce racial health disparities.

El Husseini, a 2020 graduate of the AANs Transforming Leadership Program (TLP), spoke with Daniel Jose Correa, MD, MSC, about her year-long project to optimize telestroke care. El Husseini discussed her efforts to add standardized, patient-centered measures of outcomes in Dukes stroke network, improve wellness and satisfaction of telestroke providers, and include telestroke education in our Vascular Neurology Fellowship. Correa and El Husseini also discussed North Carolina and Dukes response to the COVID-19 pandemic, and how that pandemic affected (and continues to affect) telemedicine and the needs of patients with neurological conditions.

Two recent studies have explored the neurological effects of COVID-19 on patients. While a research published in the journal Neurology has pointed out the various neurological manifestations, another published in the Annals of Clinical and Translational Neurology says that neurologic manifestations were present in nearly a third of the COVID-19 patients studied.

What does the new research tell us?

The first study, published in Neurology, was conducted across 11 hospitals and included 64 patients, out of which 43 were men and 21 were women. The median age of the patients was 66. Out of these, the MRIs of 36 patients (54 per cent) were considered abnormal and ischemic strokes, (27 per cent), leptomeningeal enhancement (17 per cent) and encephalitis (13 per cent) were the most common neuroimaging findings.

Confusion was the most common neurologic manifestation experienced by 53 per cent of the patients, followed by impaired consciousness (39 per cent), presence of clinical signs of corticospinal tract involvement (31 per cent), agitation (31 per cent) and headache (16 per cent).

Significantly, half of the patients considered for this study had acute respiratory distress syndrome (ARDS), while 11 per cent died.

The second study examined neurological manifestations in over 509 patients in a hospital network in Chicago, Illinois. This study found neurological manifestations in 215 patients (42.2 per cent) at the onset of the disease, in 319 patients (62.7 per cent) at hospitalisation, and at any time during the course of the disease in 419 patients (82.3 per cent).

As per the study, the most frequent neurologic manifestations were myalgias (44.8 per cent), headaches (37.7 per cent), encephalopathy (31.8 per cent), dizziness (29.7 per cent), dysgeusia (15.9 per cent), and anosmia (11.4 per cent). Further, the researchers found that strokes and movement disorders were uncommon among this cohort of patients, while 26.3 per cent of the patients required mechanical ventilation.

Significantly, researchers say that independent risk factors for developing neurological manifestations include severe COVID-19 and younger age.

So what does this mean?

The findings from the first study imply that patients with COVID-19 can develop a wide range of neurologic manifestations, which may be associated with severe and fatal complications such as stroke and encephalitis.

On the other hand, the findings from the second study imply that neurologic manifestations occur in most hospitalised COVID-19 patients. Researchers also conclude that encephalopathy (a broad term used for diseases that alter brain structure and function) was found to be associated with increased mortality and morbidity, independent of disease severity.

As per a report in The New York Times, only 32 per cent of the patients with altered mental function in the second study were able to handle routine daily activities like cooking and paying bills.

Kinesiology researcher creates smart way to assess and track neurological function – UCalgary News

Testing someone for nerve damage is done manually by a clinician who determines what a patient can detect on their skin, most often using a specialized set of monofilaments or a tuning fork. It can be a long and uncomfortable process for the patient and the clinician. But using wearable technologies, Dr. Ryan Peters, PhD, in the Faculty of Kinesiology and member of the Hotchkiss Brain Institute at the Cumming School of Medicine, is developing new ways for performing neurological testing anytime, anywhere, and with greater accuracy.

Peters is a co-founder and chief science officer of Vibratus Inc., which is bringing these new neurological wearable technologies to market. You can think of Vibratus as your wearable neurologist, says Peters. He is pictured above, right, with members of his team.

The COVID-19 pandemic provides an ideal proving ground for such technologies, when in-person access to the neurologist can be more restrictive.

Patients with nerve damage have increased risk of injuries and falls, and yet months can go by before they can visit a neurologist to have their condition re-assessed. With this app, both the doctor and patient will quickly know if personalized treatment strategies are working or if they need to be adjusted, says Peters.

Peters says the technology is useful for nerve disorders such as peripheral neuropathy (nerve damage caused by conditions such as Type 2 diabetes or chemotherapy), peripheral nerve or spinal cord injuries, cerebral palsy, Parkinsons disease and stroke. The clinical tests that Vibratus is looking to improve upon are part of the standard neurological exam, making them useful for a broad range of neurological disorders.

Though still in the testing phase now, Peters plans to launch the app within the next year and has filed U.S. provisional patents for these technologies. It would be great to see our devices in the hands of everyone who does neurological testing. We have the potential to improve outcomes for people with neurological disorders while saving time and money for the health-care industry.

Vibratus will be the first startup business to come out of the Faculty of Kinesiologys new specialization in wearable technology, referred to as We-TRAC. Dr. Reed Ferber, PhD, who leads the We-TRAC program,says Peters has been a fantastic addition to the collaborative group.

He has shown success in obtaining grants, working with companies, and now as the first startup company to come out of We-TRAC, says Ferber. Peters says Ferber has been a tremendous mentor to him, and that it was Ferber who suggested he apply for the grants that are now supporting this endeavour.

As well as receiving a $101,500 grant from the National Sciences and Engineering Research Council of Canada, or NSERC, Peters received a $200,000 Life Science Fellowship from Innovate Calgary. These grants are for research and development funding, and the creation, entrepreneurship and talent development for emerging life science-based companies.

Scotland allocates 500,000 for treatment of neurological conditions – Health Europa

The investment, which totals 492,535, will be directed specifically towards initiatives which focus on delivering treatment in partnership with the NHS and the third sector; and forms part of the governments 4.5m five-year action plan to ensure patients living anywhere in Scotland can access personalised neurological care and support.

The Neurological care and support: framework for action 2020-2025 programme sets out the governments vision for improving the quality of life of adult patients with neurological conditions by raising the standard and availability of care and treatment which they are able to access; as well as by sourcing and distributing information and support around living with neurological issues.

Joe FitzPatrick, Scotlands Minister for Public Health, said: Around one million people in Scotland live with a neurological condition ranging from common conditions such as migraines to life-limiting illnesses such as motor neurone disease (MND). We recognise the scale, variety and context of neurological conditions and how distressing they can be for the person with the condition and their family and the framework for action aims to support those responsible for providing care and support to people affected. This latest round of funding will ensure this vital work not only continues but is strengthened despite the challenges faced during the pandemic.

One recipient of the new funding is the PSP Association (PSPA), a charity which offers information and support to people affected by progressive supranuclear palsy (PSP) and corticobasal degeneration (CBD). PSPA CEO Andrew Symons said: Neurological Care and Support Framework Funding will allow us to map provision for PSP and CBD in Scotland, simultaneously highlighting the information and support that we can offer from PSPA. This will contribute to the aim of earlier diagnosis of these rare conditions and reduce the time spent by people in the wrong part of the system with the result they can benefit from the provisions of the Framework as soon as their disease is recognised.

Scientific American reported: labs are turning to gene-based vaccines. Scientists use information from the genome of the virus to create a blueprint of select antigens. The blueprint is made of DNA or RNAmolecules that hold genetic instructions. The researchers then inject the DNA or RNA into human cells. The cells machinery uses the instructions to make virus antigens that the immune system reacts to. Cells respond to the instructions as a normal part of their daily existence. This is the same trait infectious viruses exploit; they cannot reproduce on their own, so they use a cells machinery to make copies of themselves. They burst out of the cell and infect more cells, widening the infection.

Some researchers insist DNA vaccines will genetically modify humans, many reputable scientists dispute this claim.

Reuters reported in a fact check: A future COVID-19 vaccine will not genetically modify humans. DNA vaccines do not integrate the virus DNA into the cell nucleus of its recipient but instead inject part of the virus DNA/RNA into tissues to stimulate an immune response in the body.

There is a spreading fire about the fact thatBill Gateshas teamed up with the Department of Defense and DARPA, which is the special project research and development arm of the Pentagon. The team-up was a call to develop a brand new type of vaccine which is being rushed into production, an alarm for med-industry benefits.

Especially considering this new kind of DNA / RNA vaccine would normally take 15-20 years to roll out, while this one is being rolled out in a matter of months and is already undergoing human clinical trials. The urgency and rolled back regulations to streamline this new vaccine is, of course, justified by the crisis.

Will these nanoparticles be inserted into millions upon millions of people around the globe? I do not know, however, the technology is there, it exists and this is not some far fetched conspiracy theory.

The Boys season finale: Victoria Neuman head exploder twist reveals the limits of the show’s nihilism. – Slate

This article contains spoilers for the Season 2 finale of The Boys.

Watchers of The Boys got a real jolt of a reveal at the end of the shows second-season finale. Victoria Neuman, the truth-speaking young congresswoman played by Claudia Doumit, who had worked with the vigilante Boys in their efforts to bring down the evil Vought Enterprises, turns out to be a secret superhero. She also seems to be pretty evilshes the one responsible for all the disgusting random head explosions that have made the show so hard to watch this season. In fact, the terrible scene we were subjected to in Episode 7a Congressional hearing Neuman calls in order to bring Vought Industries to justice ends in a huge mess as the star witness and multiple attendees get their gourds squished by an invisible forcewas all Neumans doing.

Why did this surprise retain the power to jar and upset me, since Im generally a huge fan of what Slates Matthew Dessem has described as the shows deep and abiding misanthropy? Because Neuman is clearly an Alexandria Ocasio-Cortez analog. The comic-book series the shows drawn from had a character named Victor Neuman, but he was a dim-witted Vought exec turned politician, apparently inspired by George W. Bush. This Victoria Neuman is totally different. Shes a young woman of color (Doumit is Lebanese, Italian, and Australian). Shes plain-spoken, sharp, and brimming with charismaand just like AOC, Neuman knows her way around a statement lipstick.

The show even gestures to the similarity explicitly in Episode 5, when Neuman convenes a rally outside Vought after the once-hidden fact that the company has been manufacturing a superhero serum goes public. Homelander (Antony Starr), the evilest superhero, drops in on the rally, paying Neuman a series of smarmy compliments. Didnt you love that little Walk Like an Egyptian dance she did online? he asks the crowd, a seeming reference to that time a video of AOC dancing adorably to Phoenixs Lisztomania on a rooftop in college went viral. So fun! I loved it! Homelander, an old-school sociopath deeply uncomfortable with the workings of online celebrity, did not in fact love itbut the crowd, full of people hoisting signs that say We love you, Victoria!, clearly did.

Not everyone is sold on Neuman, even before the big reveal. The Boys meet with Neuman at a mutual allys house to prepare for those hearings that end in blood. Their leader, Billy Butcher (Karl Urban), doesnt want to collaborate; he is, to put it politely, not a believer in electoral politics. Weve never had Congress on our side before, says the ally (Grace Mallory, ex-CIA, played by Laila Robbins), asking Butcher to play along. To which Butcher spits, in his classic brutal form: Congress? Please! What a bunch of corrupt fucking cunts they are! Neuman replies, casually: Youre not the first to call me a cunt, Mr. Butcher; Im starting to think its like a badge of honor. Rep. Ted Yoho called Ocasio-Cortez a fucking bitch on the Capitol steps in July of this year, so this cant have been an explicit connection since this season of the show was already in the can, but Ocasio-Cortez certainly gets enough abuse online to make the connection resonate.

Looking back at the times Neuman appeared this season, Im not sure why I didnt call this turn ahead of time. This particular show is not about to put its faith in a politician, no matter how apparently virtuous. The reveal that Victoria Neuman is a head-squishing supe works within the universe of The Boys, because nobody but the Boys (and maybe the good superhero, Starlight, and Grace Mallory) are allowed to have pure hearts. But this twist of the story makes me look twice at the shows nihilism, and wonder whether I want to be along for this particular ride.

When we find out shes a supe secretly working against the Boys, Victoria Neumans force of personalitythe leadership qualities that draw people to attend the rallies she convenes, toting signs that say Victoria Neuman, Political Badassbecomes instantly suspicious. Its just too reminiscent of online right-wingers making up fables about AOCs secret nature (Snopes.com has debunked conspiracy theories from AOC is an actress playing a congresswoman to AOC wants to ban motorcycles.) Clearly jealous of her youth and beauty, these real-life conspiracists insist shes too good to be true. I wish this show hadnt done the same.

Neumans nature isnt known to the Boys yet (just to the shows viewers), and the finale ends with Hughie (Jack Quaid), the most idealistic of all the vigilantes, joining up with Neumans campaign. Season 3 is supposed to start filming in early 2021, COVID willing. I suppose theres some room for a turnaroundmaybe well find out the congresswoman had reasons for busting the heads she busted. But Im not holding out much hope.

The Second Season of The Boys Was Fueled by the Nihilism of Reality – The Ringer

The penultimate episode of The Boys second season begins in unnerving fashion, even by the shows standards. A lonely young white man who lives at home with his mother is slowly enveloped, and radicalized, by the dangerous rhetoric spewed by the Vought Corporation superhero and newest member of the Seven, Stormfront. Over time, and with Stormfronts warnings of super-terrorists invading America from beyond our borders constantly booming from his phone and computer screens, the loner begins to suspect that his local bodega owner is one of these threats in hiding. (The owner is, of course, a person of color.) The tragic sequence culminates with the man shooting the innocent owner in the face.

While The Boys is a pessimistic thought exercise exploring what would happen if super-powered beings actually existed among usspoiler: yes, superheroes would absolutely abuse their powersthis particular moment, unfortunately, feels very much rooted in our current reality. As other on-screen superhero stories have confronted historical examples of fascismHYDRA, originally an experimental scientific branch of the Nazi Party, is a recurring antagonist in the Marvel Cinematic UniverseThe Boys portrays more contemporary racist ideologies. Modern fascism comes by way of social media, memes, and conspiracy theories.

Stormfront, a being who the second season reveals was originally created by Heinrich Himmler (a side effect of her powers is that she doesnt really age), is the evolution of the white supremacist rebrand in practice. Once a literal Nazi from Nazi Germany, she shaves part of her head, is undeniably crafty with her faux-feminist/anti-corporate messaging, and succinctly underlines her philosophy for recruiting people like the aforementioned loner. You cant win the whole country anymore, so stop trying, Stormfront tells Homelander, the shows sociopathic Superman/Captain America stand-in. You dont need 50 million people to love you, you need 5 million people fucking pissed. Anger sells. You have fans; I have soldiers.

That The Boys season finale, What I Know, premiered the day after the FBI uncovered a terrorist plot to kidnap Michigan Governor Gretchen Whitmer is further proof that, existence of superpowered humans wearing dorky costumes aside, the series has a firm grasp on how actual cults of personality are formed. Reality these days can be just as strange, and depraved, as fiction. (If the second season wasnt put together in 2019, Stormfront may well have given a shoutout to the same white supremacist group that the president told to stand by just two weeks ago; The Boys is rarely subtle.)

Viewers expecting a crass, irreverent take on superhero culturethe tone of Deadpool in the form of a TV seriescertainly wont be disappointed by The Boys, but Season 2 sure goes down with an un-Deadpool-like bitterness. Amazon Studios originally sent critics the entire second season in advance over the summer, so my enjoyment of the show was distilled in a few afternoons. But The Boys actual release modelafter the first three episodes premiered on September 4, Amazon went with a weekly rollouthas its pros and cons.

On the one hand, the series reaped the rewards of sustaining interest for weeks on end; its popularity is such that not only was The Boys renewed for a third season, but a spin-off about superpowered kids in college is also on the way. (Suggestion: Cast Nicholas Braun for all the Sky High heads out there.) On the other hand, stretching out this particular season into weekly morsels is asking fans to become gluttons for punishment. Even though her name was an obvious reference, Stormfront doesnt reveal her true nature until the end of the third episodeat which point, the show takes its time laying out the full extent of her awfulness (an ageless Nazi who, in a previous superhero iteration under the name Liberty, viciously murdered a young Black man). Its compelling, up to a point: Eventually, you just want to see Stormfront, like Ramsay Bolton on Game of Thrones, get her comeuppance.

To the credit of The Boys, What I Know does deliver a satisfying rebuke to, as many characters end up dubbing her, the Nazi bitch. After A-Train discovers the real reason why Stormfront doesnt want him back in the Sevenbecause hes Blackhe goes about stealing buried, classified documents of her Nazi past. With Hughie and Starlights help, Stormfronts real identity is leaked to the press, giving Vought another PR nightmare to deal with. (One of the many effects of the revelations of Stormfronts Nazi ties is that A-Train is let back into the Seven so the company can try and save face.) As for Stormfront, who confronts the Boys in the finale, she pivots very quickly from decrying the information as a deepfake to acknowledging that people like what she has to saythey just dont like the word Nazi.

And then, thankfully, she gets walloped. Queen Maeve, whos spent much of the series wallowing in self-pity, leads a Stormfront beatdown, joined by Starlight and Kimiko. The sequence is immensely cathartic and scored to the on-the-nose tune of Peaches Boys Want to Be Her. The Boys loves to take jabs at Marvelwhether intentional or not, three female heroes beating the crap out of a Nazi feels like the shows answer to Avengers: Endgames cringey and entirely unearned girl power moment. Stormfront eventually does her best Anakin Skywalker getting roasted on Mustafar impressioncourtesy of Homelanders superpowered son Ryan nailing her with his laser visionto cap off her arc. With all due respect to Aya Cash, who delivered an incredible performance, Id be fine if that was the last we ever see of her.

Season 2 was a transitional period for The Boys. After all, What I Know saves its biggest mic drop for last: Victoria Neuman, the representative reminiscent of Alexandria Ocasio-Cortez whos been trying to hold Vought accountable for its actions, has powers herself. And not only that, shes the character responsible for the series of head-exploding assassinations this season. (Her final kill of the season was the leader of the shows equivalent of the Church of Scientology.)

While Neumans motivations arent entirely clear, it appears that Vought is covering all bases: Since the company cant stop the government from interfering in its nefarious plans, they might as well have a mole in charge of the newly established Office of Supe Affairs. If a Nazi becoming the most popular and social-media-savvy member of the Seven was The Boys appetizer, the shows main course looks like it will be Voughta conglomerate with shades of Disney and Lockheed Martin that turns a blind eye to fascists and murderers in its ranksvying for complete global domination.

Having a young representative in the mold of AOC secretly being a ruthless assassin who can explode heads with her mind is, uh, definitely in line with The Boys provocative sensibilities. But the finales Neuman twist also reaffirms what the show has hammered home from the very beginning: Whether its superheroes, celebrities, or politicians, you should always have a healthy dose of skepticism for authority figures and the institutions that put them on a pedestal. And if you ever find yourself confronted by a Nazi, punch them square in the face.
