Kanazawa University research: Engineering biological pattern formation with synthetic morphogens – PRNewswire

KANAZAWA, Japan, Nov. 5, 2020 /PRNewswire/ --Researchers at Kanazawa University report, in Science how concentration gradients of particular molecules ('synthetic morphogens') can be produced in biological systems. Such gradients can be exploited for programming pattern formation, offering the promise of controlled tissue engineering.

Multicellular organisms can grow and develop thanks to so-called morphogens: molecules that provide positional information. They are produced in a source, from which they diffuse and build up a concentration gradient in their environment. Nearby cells can 'read' this gradient, therefore 'know' where they are in the organism and 'act' accordingly. Now, Satoshi Toda from Kanazawa University and colleagues report the successful engineering of a synthetic morphogen system that can be programmed.This achievement not only helps to understand how exactly morphogens encode positional information, but is also promising in the context of engineered tissue formation.

Toda who is a PI at the NanoLSI WPI at Kanazawa University, has in a few years become an expert on programming cell patterning and tissue self-assembly, and his team tackled the question of what a minimal synthetic morphogen signaling system looks like, with the synthetic system operating 'orthogonally' to biological (endogenous) morphogens, meaning that the endogenous and the synthetic system don't interfere.

The researchers adapted an existing synthetic receptor system called synNotch so that it worked for soluble green fluorescent proteins (GFPs) as the morphogen molecules.(The original synNotch system requires morphogen molecules to be connected to a cell membrane, i.e. be non-soluble.) They did so by designing so-called anchor cells that can capture a GFP molecule in solution. An anchor cell carrying a trapped GFP molecule is then detected by a receiver cell (through chemical docking).Toda and colleagues demonstrated that this approach worked in vitro: a concentration gradient of GFP morphogen and activated cells formed within about 24 hours.

The scientists then investigated how the shape of the morphogen gradient can be regulated. They found that for different anchor protein densities, different gradient shapes were obtained. In addition, the spatial distribution of the gradient could be influenced through the use of inhibitors (molecules blocking the anchorreceiver mechanism).Toda and colleagues concluded that synthetic morphogen gradients can be tuned so that shapes similar to those occurring in vivo are obtained.

Finally, the research team around Toda showed that it is possible to alter the 'interpretation' of the gradient. They designed different kinds of feedback circuits, for example one in which GFP detection leads to more GFP production, or one in which GFP detection leads to GFP inhibitor production.Via such circuit engineering combined with the morphogen and inhibitor sources, the scientists were able to produce binary and ternary domain structures.

The results of Toda and colleagues have promising application potential. Citing the researchers: "These synthetic morphogen platforms can program positional information without cross-talk to endogenous signaling pathways.Thus, it may be possible to deploy them in vivo as inert tools to probe or redirect [tissue] development."

Morphogens

Morphogens are molecules that are essential for the biological process of pattern formation. They are produced in a source, after which they diffuse through surrounding tissue in an embryo, setting up concentration gradients in the process. These gradients then drive the biochemical reactions that ultimately lead to the formation of all the various tissues and organs in an organism. Kanazawa University's Satoshi Toda, a pioneer of programming multicellular self-organizing structures, and colleagues have now shown that it is possible to design synthetic morphogen systems that have programming functionality but at the same time do not interfere with biological (non-synthetic) morphogen systems.

Green fluorescent protein

Green fluorescent protein (GFP) is a molecule displaying green fluorescence upon exposure to blue-to-ultraviolet light. GFP is often used in biomedical protein expression experiments. For instance, it has been shown that the GFP gene can be expressed in specific cells, particular organs, or whole organisms. Now, Toda and colleagues have shown that (soluble) GFP can be used as a synthetic morphogen.

ABSTRACT

BACKGROUND: Drug-loaded nanoparticles have established their benefits in the fight against multiple myeloma; however, ligand-targeted nanomedicine has yet to successfully translate to the clinic due to insufficient efficacies reported in preclinical studies.

METHODS: In this study, liposomal nanoparticles targeting multiple myeloma via CD38 or CD138 receptors are prepared from pre-synthesized, purified constituents to ensure increased consistency over standard synthetic methods. These nanoparticles are then tested both in vitro for uptake to cancer cells and in vivo for accumulation at the tumor site and uptake to tumor cells. Finally, drug-loaded nanoparticles are tested for long-term efficacy in a month-long in vivo study by tracking tumor size and mouse health.

RESULTS: The targeted nanoparticles are first optimized in vitro and show increased uptake and cytotoxicity over nontargeted nanoparticles, with CD138-targeting showing superior enhancement over CD38-targeted nanoparticles. However, biodistribution and tumor suppression studies established CD38-targeted nanoparticles to have significantly increased in vivo tumor accumulation, tumor cell uptake, and tumor suppression over both nontargeted and CD138-targeted nanoparticles due to the latters poor selectivity.

CONCLUSION: These results both highlight a promising cancer treatment option in CD38-targeted nanoparticles and emphasize that targeting success in vitro does not necessarily translate to success in vivo.

Those excuses Fauria was referencing began during an interview Belichick had with Charlie Weis on Sirius XM radio last week, where Belichick referenced youth, the NFL salary cap, Covid-19 opt-outs, and injuries while discussing his teams rough 2-5 start to the 2020 season.

The Weis interview opened the portal for OMF Monday to press Belichick on his newly found in-season introspection.

It was obvious when he talked to Charlie [Weis] that he was willing to go there, said Fauria. It was on his mind. Because he doesnt do anything unless he wants to. How long have we been interviewing him and how many times have we asked him tough questions and nothing, no information, no explanation? My rabbit ears went up with the Weis interview, and then talking with other people who were listening to it or covering the team for years, they all said the response was odd. It was off script. Different from what were used to. I just felt like it was weird.

But would Belichick give them that information after an agonizing loss to the Bills? That was no extra point.

I didnt think it should go without it at least being said, Fauria explained about his pre-interview thoughts. And I respect Bill like crazy but that was something that was too much in the open. In my head I was like, This really sounds like a bunch of excuses. Is that the case?

Fauria continued: I felt like as a show, we couldnt go into that situation without asking or wed lose credibility. We talked about it beforehand, there were a couple of things we wanted to get in there. We always kind of assume [Belichicks] not going to answer anything. We have a question and a follow-up, always, youve always got to keep one in your back pocket. And then when Glenn [Ordway] got into it.

Ordway indeed started the line of questioning about why Belichick had been so revealing to Weis, but he received a terse response.

It seems pretty obvious to me, Belichick grumbled. Whats the question?

Why are you doing it now? Ordway asked.

I mean its pretty obvious," Belichick said. "It was an honest answer to an honest question. No more, no less than that. Im sure you can read a lot into it if you want to. That wasnt the intent.

To Faurias credit, his follow-up made all the difference.

What it sounds like is a lot of excuses, Fauria declared. Covid excuses, things youve never said before.

And then, after a few awkward seconds, the reveal.

"I didn't say it as an excuse. I never said that," said Belichick. "Look, we paid Cam Newton a million dollars. It's obvious we didn't have any money. It's nobody's fault. Anatomy of a Goal: Artur secures an early lead – Massive Report

Welcome back to the Anatomy of a Goal, where each week we dissect one goal (or near goal) from the Columbus Crews previous match.

For match 21 of the 2020 MLS Season, we take a look at Arturs 37th minute goal that gave the Crew a 1-0 lead as part of their 2-1 win against the Philadelphia Union on Sunday.

Heres a look at the goal from Columbuss midfield stalwart.

For the first time since the last time the team played Philadelphia, Columbus started a full-strength lineup. Darlington Nagbe, Lucas Zelarayan, Eloy Room and Gyasi Zardes all returned to the lineup as the team tries to round itself into playoff form.

The first few minutes of Sundays match showed a lot of promise for the Black & Gold before the Union settled into the match and began to create some chances. Philadelphias attack is at its most effective when the trio of attacking midfielders and two strikers can play quick passing combinations up the middle of the field. Fortunately, the Crews biggest strength is its central core which helped limit the effectiveness the Union attack.

Arturs goal begins with a throw-in on the sideline.

Milton Valenzuela runs up the sideline as Pedro Santos quickly throws the ball out ahead of the Argentine left back.

Olivier Mbaizo outpaces and out-muscles Valenzuela, winning the ball deep in his own defensive territory.

Mbaizo hits a quick pass to Jakob Glesnes who has already turned his attention to his next pass up the field.

Glesnes attempts to clear the ball toward Alejandro Bedoya, but Santos steps in front of the American winger.

Santos intercepts the pass to Bedoya and quickly finds Zelarayan.

Zelarayan takes turns, takes a touch toward the middle of the field, and finds himself with five options. He can play a drop to Artur, continue carrying the ball toward the middle of the field, hit a square pass to Derrick Etienne, play a through pass to Gyasi Zardes or make a pass toward Valenzuela.

Zelarayan sees Etienne call for the ball and plays a quick pass toward the winger.

Before we get to what Etienne does with his first touch, lets take a look at Valenzuelas positioning. As you can see above, Valenzuela appears to be in an offside position at this moment. This picture is taken at the moment the ball leaves Etiennes foot, so if the pass is to Valenzuela rather than Zardes, it should be whistled for offside.

Before Valenzuela can make a touch on the ball, it is intercepted by Bedoya and touched toward Glesnes.

Bedoya takes another touch on the ball and then turns to attempt a clearance up the field.

Valenzuela just gets in front of Bedoyas clearance, deflecting the ball up and toward the top of the 18-yard box.

So, should this play have been blown dead for offside? Maybe. In the case of Video Assitant Review, plays can be reviewed if there is a clear and obvious error. In this instance, Bedoya intercepts the pass before it has a chance to touch Valenzuela. As always, the offside law is tricky when the offending player does not touch the ball. The whistle can be blown if the offside player interferes with play or gains an advantage from being in the offside position, but that is a judgment call by the referee in this case and most likely not a clear and obvious error.

Valenzuelas deflection pops up toward Jamiro Monteiro at the top of the penalty box. Artur heads toward Monteiro to try and intercept the ball.

Artur and Monteiro turn but Artur is in the better position to win the ball.

The Columbus midfielder shields off Monteiro and plays the ball back to Harrison Afful.

Monteiro shifts toward Afful who has four options. He can continue to carry the ball, play a through pass to Etienne, hit a looping pass over the top to Zardes or a simple pass back to Artur.

Afful fakes a pass to Etienne causing Monteiro to throw up a leg in defense. This gives Afful a chance to further survey the field and for Artur to open himself up for a simple touch.

The Ghanian right back his a pass right back to his teammate.

Artur picks up the ball and surveys the field.

Artur takes a touch to set himself up for his next move and can do a variety of diffrent thins from this position. The midfielder can play a diagonal pass to Zelarayan, play a through pass to Zardes, have a shot on goal, play a diagonal pass to Etienne or a pass the ball right back to Afful.

Artur takes aim at the bottom left corner and hits a low, rolling shot toward the goal.

Kai Wagner isnt able to get in front of the shot, leaving old friend Joe Bendik as Philadlphias last line of defense.

Bendik starts to lean as the ball approaches the penalty spot, apparently not expecting Artur to take a shot 26 yards from goal.

Bendik isnt able to get to the ball as it rolls under his hand . . .

. . . into the back of the net!

The Anatomy of Account Takeover Attacks in Banking and Fintech – Banking Exchange

COVID-19 has already proved its substantial impact on online-based fraud. According to recent analyses of real user sessions and attack patterns on my companys fraud and abuse prevention network, digital attack rates rose by 20% in the first quarter of this year, with payment attacks up by 49% compared to the end of 2019. Over the past six months, our network recorded double the volume of attacks since the second half of 2019, with a 25% attack rate across all online transactions.

Fraudsters are proving to be exceptionally nimble and determined, quickly adapting to ever-changing realities and turning new situations such as the pandemic into an opportunity to widen their reach and maximize profits. To overcome the prevalence of anti-fraud and security solutions in the highly regulated industry of banking and fintech, fraudsters are planning and orchestrating attacks more diligently than ever before, and taking advantage of the new socio-economic context to carry out successful fraud as a result. In fact, financial institutions on the Arkose Labs network saw attack levels rise notably in Q2 of 2020, after an initial dip in April.

To navigate this new era of online fraud and realistically stand a chance at combating persistent fraudsters, organizations need to familiarize themselves with common attack vectors. Knowledge and proactivity are especially critical for organizations within the banking and fintech sector, as the potential gain for fraudsters is higher.

Common Attack Vectors and ATO Use Cases

Two common attack vectors for harvesting credentials in fintech and banking include phishing and malware. With phishing attacks, customers receive emails that appear to come from a trusted organization. Theyre asked to click a link and login because theres an important message or issue that requires immediate attention. Few will notice that the link isnt safe, and once they log in to the fake website, their credentials are recorded into the attackers database and customers are redirected to the real login page. With malware attacks, customers are enticed to install malware on their computer, which listens for keystrokes as the customer logs in to various web sites. Any credentials collected are reported to a database owned by the attacker.

Once credentials are acquired via phishing or malware, attackers may use a botnet or automated attack tool such as Sentry MBA to carry out their account takeover (ATO). They may also leverage cheap human labor as part of the ATO attack process to remain under the radar. Attackers will then use the compromised accounts to commit fraud or simply resell them to the highest bidder. Accounts that have been taken over are exploited as follows:

User-Friendly, Naturally Evolving Defense Strategies are Key

Ultimately, all online accounts are at risk and fraudsters will leverage any number of techniques to achieve their goals. Dont wait for an ATO attack to occur before deciding to protect a login endpoint. Protect all login endpoints equally whether they handle web or mobile traffic. Continuously re-evaluate resilience levels and assess where new attack points could emerge across various customer touchpoints. Given the financial hardships already brought on by COVID-19 in addition to the sensitive collateral involved, fintech and banking organizations cant rely on their customers to be sufficiently security savvy. Investing in user-friendly fraud protection and defense strategies that are capable of naturally evolving with emerging attack patterns is paramount, now more than ever.

Heres when Greys Anatomy, NCIS, Law & Order: SVU return, plus more TV highlights (The Crown, Shame – OregonLive

When everyone seems to be fighting, yelling at each other, and the future is uncertain, the idea of settling in to watch new episodes of familiar TV shows sounds like the entertainment equivalent of putting on your most comfy pajamas and snuggling up in a blanket.

Like everything else, TV schedules have been impacted by the coronavirus, as filming was forced to stop in the spring. That meant that, instead of a wave of returning series coming back to our screens in September and early October, season premieres were delayed because of the slow pace involved in getting shows back into production, and steps required for safety protocols.

Returns and premieres of new shows have generally been more staggered than usual, as some series were able to film before others. But, after several weeks of space-filling fare, including repurposed series from cable (L.A.'s Finest), streaming (Star Trek: Discovery) and imports (Transplant), the networks are finally bringing back some viewer favorites, along with intriguing new offerings.

For those of us lucky enough to be safe and able to work from home, November and December are bringing back TV meat and potatoes fare, along with some more gourmet items. Here are some of the most anticipated, and most promising, shows coming to our screens in the final months of this difficult year.

Anatomy of a Play: How the Dolphins beat Jared Goff with Cover-0 pressure – Touchdown Wire

Coming into Sundays game against the Dolphins, per Sports Info Solutions, Rams quarterback Jared Goff had faced Cover-0 defenses a high-blitz scheme with no deep safeties on just three of his dropbacks. On those three dropbacks, Goff attempted two passes and completed one to 49ers cornerback Jason Verrett in Week 6, on a red zone situation that obviously calls for no deep safety.

So, Cover-0 is not something Goff had seen a lot this season. But in 2019, he completed 7 of 16 passes for 71 yards, five touchdowns, and no interceptions against Cover-0. That said, Dolphins head coach Brian Flores and defensive coordinator Josh Boyer decided to throw more of it right at Goff, and it turned out beautifully in the Dolphins 28-17 win, Goff completed just 35 of 61 passes for 355 yards, one touchdown in garbage time, two back-breaking interceptions, and two lost fumbles one of which was returned for a touchdown. In the first half, when Miami brought most of its pressure, Goff completed 15 of 32 passes for 136 yards, no touchdowns, both of those picks, and a passer rating of 32.8.

As edge-rusher Emmanuel Ogbah, who caused the fumble that led to linebacker Andrew Van Ginkels 78-yard return, said, The key to this game was attack Jared Goff.

After the game, Rams head coach Sean McVay talked about how difficult it was to handle what the Dolphins were bringing the pressures that forced McVay out of his preferred empty formations in certain instances, and took Goff out of his ability to cut the field in half with boot-action.

They were bringing zero pressures, McVay said. We had some answer. We didnt execute them, and ultimately, the answers were not good enough on my part. That falls on me we were not in alignment, and the communication wasnt on par with what the expectations were with how we execute those plays. those situations never allowed us to get into rhythm.

The first interception came out of zero pressure Miami brought an extra pressure look, and then dropped defensive tackle Christian Wilkins into coverage. Goff didnt pick that up on a quick pass attempt to Cooper Kupp, and Wilkins had his first career pick.

It wasnt just the zero blitzes that took Goff out of his game Flores and Boyer showed him all kinds of blitzes with all kinds of coverages behind but as McVay said, it was the zero blitzes, and the ability to combine pressure with the illusion of pressure, that made the difference in this game.

The Women of Greys Anatomy on Their Favorite Episodes, Filming With COVID-19 Protocols and the Shows Legacy – Variety

For VarietysPower of Women cover story about Greys Anatomy, Ellen Pompeo, Chandra Wilson, Debbie Allen and showrunner Krista Vernoff sat down to discuss the show its past and present.

They also talked about some of their favorite episodes over the years.

Vernoff, who was head writer for the first seven seasons of Greys Anatomy and returned for Season 14 as showrunner, said a two-part Season 3 episode about the death of George OMalleys dad was her favorite thing shes done. It was very much my story, Vernoff said. And its the thing Im most proud of.

Vernoff also prompted Allen to talk about a Season 15 episode shed directed called Silent All These Years that focused on sexual assault. The hallway of women, Allen said, summoning imagery from the episode. Silent All These Years, this episode we did about rape and women who are survivors, not just victims, but survivors and we changed the face of the planet with that episode. And around the world we encouraged people to stand up for themselves, and not to be ashamed.

Wilson pickedSliding Doors,an episode she had directed from Season 10, Sandra Ohs final year on the show. In the episode, Ohs Cristinanarrates the episode, and sees two parallel paths her life might have taken.Sandra wanted me to take care of her for that, Wilson said. She looked at it as her last episode, but we gave her more to finish the season. So that was an honor for me to be in that position.

For Pompeo, she cited being directed by Denzel Washington in Season 12s The Sound of Silence.

Being directed by Denzel was definitely a highlight of all 17 seasons for me, Pompeo said. Hes one of my acting idols, and such an incredible talent and force, and to have him have the humility to come in here, because of Miss Allen, and want to direct an episode of our little show, I thought, was so exciting.

And it was really a boost, I think, she continued. They knew I needed something that year! I was really losing my steam, and they knew that I needed something, and Debbie came through, like she always does, and gave me the gift of Denzel, and that episode. And it was really fantastic. So that would definitely be one of my highlights, for sure. And made me cry. I always cry when I watch.

Since Pompeo toldVarietythat it may be the shows final season, the group also discussed its legacy.

Sam Ford/THEREVIEWIn a country that focuses heavily on politics, where do sports fans lie on the political spectrum?

Where do sports fans lie on the political spectrum? Will sports fans of certain leagues come out in droves to vote for either Donald Trump or Joe Biden in the 2020 general election?

These questions come during a year in which sports and politics seem to be linked more than usual. Between the current election, athletes standing up for social justice, and leagues creating initiatives for increased voter participation, that relationship has been augmented.

But could sports or athletes alienate a portion of their fans by involving political and social messages? That question leads to understanding where fans of certain sports lean on the political spectrum.

In 2017, FiveThirtyEight looked at the search interest of seven sports and the Trump vote share in markets (cities and towns) across the United States. This comparison was then used to indicate which of the sports lean Democratic and which lean Republican.

For instance, the NBA is considered more popular in Democratic markets because markets that were more likely to search NBA news had a relatively low share of votes for Trump in the 2016 election. Meanwhile, markets with higher trends for college football searches saw higher vote shares for Trump . Thus, college football is considered more popular in Republican markets.

Six of the seven sports looked at by FiveThirtyEight showed a significant lean to either side of the political spectrum. However, the NFL showed no significant skew; search interest for the league was essentially the same in all markets, no matter if the market had a 20% or an 80% Trump vote share.

This middle ground in its fan base could be an explanation for the NFLs hesitancy to be heavily involved in political or social issues. Other sports, like the NBA and MLB, are perhaps offered more flexibility in political and social statements.

This past summer, commissioner Adam Silver did not hesitate at all in making the NBA an avenue for social justice awareness. Players were permitted to put messages of social justice on their jerseys, and Black Lives Matter was written on all courts in support of the movement for racial justice.

The NBA, along with the MLB, underwent a boycott of games following the shooting of Jacob Blake on Aug. 23 in Kenosha, Wisconsin . Silver said that he wholeheartedly support[ed] the leagues players shining a light on important issues of social justice.

Over the course of the summer, though, the NBA has experienced a decline in ratings a trend that has led President Trump to publicly call out and criticize the league. But with a rearranged schedule, the ongoing effects of the pandemic and declines in sports ratings across America, not all the blame for lost viewership can be put on the increased involvement of social and political messages.

In fact, a large portion of fans remain likely to still watch sports that include political messaging from athletes. Schuyler Reed, an avid NBA, NFL and soccer fan, and a registered Democrat in New Jersey, does not mind athletes voicing their opinions.

I believe that athletes have the platform and the following where they should be able to stand for issues that they believe in, Reed said. The way leagues, such as the NBA, have helped [athletes get their message across] has been very supportive.

Other fans look at sports as an escape from political and social hostility in the country. Dorothy McKain, a lifelong MLB fan, leans on the side of keeping politics out of sports, despite being a registered Democrat in Delaware and planning to vote for Joe Biden.

She said she watches sports to get away and that the two should be separate. On Trumps public criticism of the NBA, she said he came across as having poor judgement and looking unprofessional.

Andrew Surowiec, a Michigan voter who leans center right politically, looks at sports the same way in terms of an escape from national news. He has been supportive of the NHL striking a balance.

I feel like the NHL has done a good job at condemning hate and racism and everything in between but keeping the politics out of the sport, Surowiec said. Politics dont belong in sports, so anyone that criticizes it is in the right.

While the NHL has seen less political and social involvement as a whole, and from its players, ratings have still dropped. Increased competition from other leagues and a lack of scheduling normalcy are two possible factors independent of social messaging.

During such an unprecedented time, the Marist Poll conducted a polling of sports fans to see how certain factors would increase their viewing habits. Athletes speaking out on political issues saw the second highest percentage of making sports fans less likely to watch live sports.

Still, nearly half of respondents noted that athletes speaking out would cause no difference to their viewing, and a substantial 21% would be more likely to watch given athletes voicing their political opinions.

The above table supports the idea that politicizing of sport is not the leading factor in ratings decline seen across the board during 2020. Even so, are the four major professional sports alienating a portion of fans by refusing to eliminate political and social issues from live broadcasts?

The answer to that question remains somewhat unclear, as declining ratings have many possible explanations. What is known is that the four major sports leagues will run the risk of angering and driving away fans on both sides of the spectrum, no matter how they handle the evolving social and political climate.

Marathon Preliminary Hearing Ends as Judge Rules Accused Mobile Phone Robbers Should Stand Trial

SACRAMENTO In a preliminary hearing that stretched over weeks, and a case that is more than two years old, a Sacramento County Superior Court judge found two men guilty enough to stand trial for allegedly participating in a one-day mobile phone store robbery spree.

They will be arraigned Nov. 13 on charges, and a trial date set.

Defendants David Fritz and Glenn Burgler faced Judge Stacy Boulware Eurie in Dept. 18, with Fritz appearing in person along with his public defender, while Burgler called in to the hearing via Zoom out of custody. Deputy District Attorney Matthew Moore joined the call via Zoom.

Both Fritzs defense attorney, John Gonzales, and Burglers defense attorney, Carmen Butler made unsuccessful attempts at reducing the charges and striking the enhancements against their clients.

The co-defendants are charged in a four-count felony complaint with robbing a T-Mobile store and a Verizon store on June 10, 2018. Burgler is alleged to have been personally armed with a handgun. It is alleged the duo stole $39,000 worth of cell phones.

Gonzales argued against Fritzs enhancement, claiming that out of the four other members who took part in the robbery, they already pled out two other people for non-personal use, so they have to put personal use on somebody, and the only person left is my client.

John Dossey and Kyle Mangiola were both found guilty of their involvement in the robbery and are both serving jail sentences in Folsom State Prison and San Quentin State Prison, respectively, since March of 2020.

According to Gonzales, all of the physical evidence, including the gun, and the clothes and motorcycle helmet identified from security footage, was found at Mr. Mangiolas house, the other African American person involved in this four person conspiracy to robbery, so I dont know how we come to the conclusion its Mr. Fritzs whos the one personally using this firearm other than we have Mr. Mangiola trying to save himself, make a deal with the DA, and he lied four times prior.

DDA Moore admitted that its clear that [Mangiola] is lying at times to protect himself but that neither defense attorney has pointed to any reason that he would falsely (implicate) the two defendants in this case.

Gonzales also argued that the gun was never even used at the Verizon store, the second stop in the robbery, even though the prosecution includes it in their case against Fritz.

He pushed the argument further saying, simply having a gun in your pocket, in your waistband, is not personal use.

Gonzales also described the scene from inside the Verizon store, which was caught on the stores surveillance system, and said, There were very large sweatshirts that were covering their waistbands, you cant even see a gun.

Moore fired back.

In terms of the use of the firearm, I disagree with Mr. Gonzales, he said, clearly (defendant) uses it in the first case, and by the jury instruction which simply requires that he display it in a menacing manner, he used it in the second case as well. And that actually is captured on the surveillance, you can see him motioning towards his waistband, in the exact way that the victim described, and that was done for one purpose, to intimidate the victim.

Judge Eurie turned to Butler to make any remaining points.

All of the items connected with the robbery were found at the homes of the two gentlemen who already pled out, said Butler, affirming what Gonzales suggested earlier. Kyle Mangiola, she continued, a loaded firearm was found at his home and its actually registered to him.

Overall, Butler asked the judge to focus her attention on the robbery that occurred at 11:45 a.m., where there has been insufficient evidence that Mr. Glenn had any connection to that incident.

DDA Matthew Moore admitted that its clear that [Mangiola] is lying at times to protect himself but that neither defense attorney has pointed to any reason that he would falsely implement the two defendants in this case.

Earlier in the week, Det. Anthony Brantley said, based on the T-Mobile surveillance footage, the first suspect that brandished the firearm was wearing all black with a blue and yellow multi-colored motorcycle helmet.

He said the second suspect was wearing dark clothing and a backpack with distinctive vertical zippers. A white t-shirt was also seen on the suspect. The suspects shoes were black with a white sole. The motorcycle helmet was black with a gold design.

Based on the Verizon surveillance footage, the first suspect was wearing a helmet that resembled the black and gold helmet that the second suspect in the T-Mobile robbery was wearing. The first suspect also had the same build as the first suspect in the T-Mobile robbery.

The second suspect in the Verizon robbery was seen wearing the same clothing seen on the second suspect in the T-Mobile robbery, except now he was wearing a clown motorcycle helmet. A Nike symbol was also observed on the suspects black shoes with white soles, which was not as visible in the T-Mobile robbery.

Moore asked Det. Brantley about two witnesses that he spoke with on June 18, 2018, and July 13, 2020. They knew two individuals that will be called suspect A and suspect B, both separate suspects than those seen in the robberies.

On June 10, 2018, the witnesses were at suspect As house for his childs birthday party. One of the witnesses saw a man coming out of the house upon arrival, to which he was introduced as suspect As boy named Dave. An individual named Glenn was also at the house.

The witness was shown a six-pack photo lineup, which contained defendant Fritz. The witness picked Fritz out of the lineup as Dave, the person he was introduced to at the party. The same witness was also shown a six-pack photo lineup that contained defendant Burgler, to which he knew as Glenn that was seen at the house.

Det. Brantley claimed that the witness saw suspect A and Fritz leave the house in a van, while suspect B and Burgler left on their motorcycles. The four men returned to the house later that evening.

According to the witness, suspect A seemed upset and went inside briefly with suspect B. Burgler and Fritz stayed outside. The witness followed suspect A and suspect B once they went back outside. The witness saw Fritz leave in a separate vehicle other than the van and the motorcycles.

When the witness went back into the house, suspect B came into the house shortly after with a black and blue duffel bag. The witness overheard suspect B say, you can call me El Chapo.

Burgler, suspect A, and suspect B all went into a separate bedroom. The witness went into the room and saw that the black and blue duffel bag contained numerous cell phones and tablet devices that filled the whole bag, where suspect A stated that they just had a come up.

Defense attorney Carmen Butler, who represented Burgler, objected for multiple levels of hearsay. I believe this is the Det. testifying about what the witness told him that he heard the other individuals say, so there are multiple levels there of hearsay.

It is a co-conspirator statement. Suspect A conspired with suspect B and defendants Burgler and Fritz to do the robbery in this case, and these are ongoing statements following that, so that is the one level of hearsay and of course the conveyance of that from the witness to Det. Brantley is under (Proposition) 115. Moore concluded.

Suspect A is not here to be cross-examined about whether or not that statement is accurate, Butler replied. I dont see what other purpose it is offered for, other than to say the come up is the equivalent of, we committed this robbery and obtained this property.

Judge Eurie overruled. While Moores questioning continued, Det. Brantley was asked about another Det. who executed a search warrant at suspect Bs residence with Det. Brantley.

Moore shar
ed his screen on Zoom to show evidence of the clown helmet, the scorpion helmet, the backpack, and the duffel bag. Det. Brantley confirmed that these were the items that matched the culprits in the Verizon robbery. The black shoes with the white soles and Nike symbol were also found.

Det. Brantley also spoke with another Det. that conducted a search on the same day at suspect As residence. This Det. found a black Springfield 40 caliber handgun in suspect As closet, which was consistent with the gun in the T-Mobile robbery. The Det. also found the blue and yellow helmet that was consistent with the helmet in the T-Mobile robbery.

Moore asked if Det. Brantley spoke with suspect A about the robberies, which he did after his arrest and during a proper interview. Suspect A admitted to being a lookout, and indicated that suspect B, Fritz, and Burgler were involved in the robberies.

Brantley said that suspect A said Burgler was also a lookout, while Fritz and suspect B went into the stores. Burgler and Fritz were on the Harley-Davidson, suspect A was in a van, and suspect B was on the Yamaha. Suspect A claimed that Fritz was armed.

Det. Brantley reviewed two cell phone downloads that were conducted by a different Det.. These downloads were seized during the arrest of suspect A and suspect B. Based on the review of the contents, Det. Brantley was able to determine that the cell phones belonged to suspect A and suspect B.

In suspect Bs phone, there was a text conversation with an individual named David Walmart. In suspect As phone, the same phone number was saved as David OG. Suspect A confirmed that the contact was defendant Fritz.

When Moore shares his screen on Zoom to show the contents of the conversation between suspect Bs phone and David Walmart, Butler objects as hearsay.

There is no proof that these are conversations between them, these are text messages. Theres no indication as to who actually texted these text messages. Butler argued.

The cell phone download comes from a cell phone seized from suspect B. The Det.s review of that phone and the contents of it shows that it belongs to suspect B. The phone number that he is communicating with, first of all is labeled as David, which is the defendant Fritzs first name. Moore explained after Judge Eurie asked about his foundational argument.

In addition, suspect A has the same phone number saved in his phone, which he indicates belongs to defendant FritzTheres no reason to believe that same phone number goes to two different people. Moore continued.

I dont believe suspect B ever made any statements admitting to sending any texts to Mr. Fritz, Butler responded.

These are messages that someone sent from a phone labeled, on one phone, David Walmart. Theres no last name, its not David Fritz, and even if it is Mr. Fritzs phone number, theres no proof that Mr. Fritz is the person who is actually sending the text messages. Butler concluded.

Judge Eurie asked Moore to ask the questions again in order to establish Det. Brantleys testimony to show that David OG and David Walmart are the same person as Fritz. Brantley said that suspect A confirmed that the phone number belonged to Fritz, and upon a photograph of Fritz shown to suspect A, the photo was confirmed to be David OG in his phone. Judge Eurie ruled it to be sufficient for submission.

Further into the testimony, Moore also asked if Det. Brantley saw a text conversation between suspect B and a contact labeled as Glenn. Det. Brantley said yes, and he also saw the same phone number in suspect Bs phone that was labeled as Glenn (Dan Mechanic) in suspect As phone.

Butler objected once again to lack of foundation and hearsay. There is no indication that Mr. Burgler was the person who sent these messages to suspect B. We dont have a statement from suspect B saying, these are messages that I received from Glenn Burgler. concluded Butler.

The reason Im overruling it is in part, suspect A has confirmed by phone number and identification that this phone number belongs to Mr. Burgler. The court does find that there is sufficient foundation laid in terms of the hearsay. reasoned Judge Eurie.

The COVID-19 pandemic hits the first responders of Seattle in a premiere crossover event on Thursday, November 12, on ABC, and the first photos out show just how safe everyone is being.

It all begins inStation 19's Season 4 premiere, "Nothing Seems the Same," at 8/7c, when the crew lean on each other for support while facing the health emergency. Meanwhile, Andy (Jaina Lee Ortiz) "tries to make sense" of the fact that her mother is actually alive, and Sullivan (Boris Kodjoe) settles into "a new routine" following his surgery.

Then, in the first of the two-hour Grey's Anatomy Season 17 premiere, "All Tomorrow's Parties," we see how the pandemic turns the lives of Meredith (Ellen Pompeo), Bailey (Chandra Wilson), and the rest of the Grey Sloan doctors upside down a month in.

What brings it all together is a group of teenagers who unintentionally start a wild fire (cuing the need for Station 19's firefighters and pandemics) and are brought into the hospital for treatment (enter theGrey's doctors).

In literature, film and TV, the vampire has varied in appearance and abilities, although a few key traits remain the same. The vampire must consume human blood to survive, their power is at its peak during the night and a human must consume the blood of a vampire to become one themselves.The Vampire Diaries features some of the time-honored traditions of vampire lore but the show also introduces its own elements to the bloodthirsty fiends.

The rule that vampire blood is necessary to kick-start the transformation from human to bloodsucker has existed for generations of vampire stories, and The Vampire Diaries is no different. To become a vampire, a human must die with a vampire's blood in their system. The matter of the human's death itself is irrelevant -- although the body does have to be somewhat intact. However, humans hoping to become a creature of the night must either consume vampire blood orally or through injection, or have a vampire's blood poured on an open wound.

The Vampire Diariesestablishes early on that in order to complete the transformation from human to vampire, a newly created vampire must consume human blood. Animal blood is not sufficient to sustain the transformation, so new vampires who want to adhere to a strict diet of animal blood are out of luck -- at least initially. A human need not die in the process, and vampires can utilizeunconventional methods to obtain the needed blood, likestealing blood donor bags or feeding from a recently deceased human. Regardless, a newly created vampire only has 24 hours to have their first taste of human blood or they will die permanently.

In theTheVampire Diariesuniverse,all vampires descend from the six Original vampires known as the Mikaelson Family -- Mikael, Finn, Elijah, Kol, Rebekah and Klaus. After the Originals were created in 1001 AD, they fed on human blood and thus created the first bloodlines, through which all vampires are connected. The magical properties of the Originals' blood is passed down through each vampire they sire and the ones their progeny sire, meaning that vampires are all one big family. Unfortunately, due to every vampire's link to the Mikaelsons, if one of the Originals die, their entire bloodline follows suit in less than a hour.

These 2 ‘Grey’s Anatomy’ Actors Are Married in Real Life, But Their Characters Never Met – Showbiz Cheat Sheet

Greys Anatomyis known for epic romances. Fans get very invested in the relationships at Grey Sloan Memorial Hospital. But some cast members have relationships off-screen, too. Fans on Reddit recently discussed how two recurring characters are married in real life.

Although Denny Duquette and Dr. Lauren Boswell never appeared on screen together, the actors who play them are married in real life, and fans think they make an adorable couple.

Jeffrey Dean Morgan, the actor who played Izzies love interest Denny Duquette, is married to Hilarie Burton, the actress who played Dr. Lauren Boswell. The two were together for a decade before making it official in October of 2019.

Even though both of these actors have appeared onGreys Anatomy,they didnt meet on set. After starring in the first season ofGreys,Morgan worked on another hit show,Supernatural.

Burton had just ended her work on One Tree Hillin 2009, when she went to visit friend Danneel Ackles. The actress starred alongside Morgan inSupernatural,and thought he and Burton would enjoy getting to know each other.

They met briefly before Burton left for a trip to Paris, and couldnt stop thinking about one another. Morgan and Burton were living together just a few months later. Shortly after that, they welcomed their first child, a son named Gus.

Burton and Morgan still hadnt met when he first appeared onGreysas a patient named Denny Duquette. But Denny did go back toGreys,and it seems like that second appearance happened after he and Burton had gotten together.

In 2009, Denny returned to the set ofGreysto re-hash his romance with Izzie. Of course, Denny died in the first season ofGreys,so he ended up coming back as a ghost, or maybe a hallucination of Izzies.

Fans loved Dennys character, so they were excited to see him back on the show, even if it was in spectral form. However, when Izzie decided to have carnal relations with the ghost Denny, fans were not on board. That scene is still thought of as one of the cringiest Greys Anatomymoments, even though it has been more than a decade since the episode aired.

Still, fans didnt change their opinion on Denny. Hes still one of the most loved recurring characters onGreys,according to fans on reddit.

Morgan may have played one of the most loved recurring characters on Greys Anatomy, but his wife Burton may have played one of the most hated. She played the role of Dr. Lauren Boswell, an all-star surgeon who came to work at Grey Sloan Memorial.

Fans are unclear on why, but Lauren broke up Arizona and Callie. She flirted relentlessly with Arizona, despite learning that the other doctor was married.

During a blackout, Lauren and Arizona slept together, ending Arizona and Callies marriage. Fans are still bitter. Although fans are all agreed that Lauren is not a most loved character, some fans do defend her.

Commenters on reddit pointed out that Arizona is the one to blame for cheating on Callie, since she was the one in the relationship. Others think that Laurens confidence was enthralling, and would like to see her back on the show.

Burton may come back to reprise her role as Dr. Lauren Boswell. The doctor was a consultant, so her stay and Grey Sloan was intended to be short. She left under normal circumstances, when the case she was called in on was resolved. Morgans character Denny died.

Without Izzie to see him as a ghost, its unlikely he will return, although anything is possible on Greys Anatomy.

Greys Anatomy first premiered on television in 2005 and has been at the forefront of pop culture ever since.
