An international team of researchers from Bristol and China has prepared biocompatible protocells that generate nitric oxide gas a known reagent for blood vessel dilation - that when placed inside blood vessels expand the biological tissue.

In a new study published today in Nature Chemistry, Professor Stephen Mann and Dr Mei Li from Bristols School of Chemistry, together with Associate Professor Jianbo Liu and colleagues at Hunan University and Central South University in China, prepared synthetic protocells coated in red blood cell fragments for use as nitric oxide generating bio-bots within blood vessels.

Coating the protocells led to increased levels of biocompatibility and longer blood circulation times. Critically, the team trapped an enzyme inside the protocells which, in the presence of glucose, produced hydrogen peroxide. This was then used by haemoglobin in the protocell membrane to degrade the drug molecule hydroxyurea into nitric oxide gas.

When placed inside small pieces of blood vessels, or injected into a carotid artery, the protocells produced sufficient amounts of nitric oxide to initiate the biochemical pathways responsible for blood vessel vasodilation.

Although at a very early stage of development, the new approach could have significant benefits in biomedicine, cellular diagnostics and bioengineering.

Professor Stephen Mann, Co-Director of the Max Planck Bristol Centre for Minimal Biology at Bristol, said: This work could open up a new horizon in protocell research because it highlights the opportunities for creating therapeutic, cell-like objects that can directly interface with living biological tissues.

Associate Professor Jianbo Liu at Hunan University added: We are all really excited about our proof-of-concept studies but there is a lot of work still to be done before protocells can be used effectively as bio-bots in therapeutic applications. But the potential looks enormous.

A note about the following letter:The Letter from Concerned Gonzaga Faculty emerged from a grassroots effort among faculty who attended several of the listening sessions held over the last two weeks. On one hand, the letter alone is just another statement; on the other hand, it points toward immediate actions, and it signals an intention from a large group of faculty to hold ourselves accountable, to hold each other accountable and importantly, to hold administration accountable for long overdue institutional changes.

The signatures of support for this letter are certainly incomplete; power dynamics certainly prevented some supportive colleagues from signing this letter. Faculty without the protection of tenure, adjunct faculty and instructional staff occupy vulnerable and precarious employment positions. Some faculty work in hostile climates to this type of engagement and may fear repercussions from colleagues.

The letter circulated only among faculty in recognition of the many academic challenges related to diversity, equity and inclusion that must be addressed. However, the need for change spans all corners of the University, and all employeesfaculty, staff and administrationmust work collaboratively as equal partners to create lasting change on this campus.

A letter from concerned Gonzaga Faculty...

On Nov. 8 Gonzagas Black Student Union was attackedGU was attacked. As GU faculty we are witnesses to the pain students experience semester after semester as a result of racist and homophobic attacks and exclusions. Students have voiced a loss of trust in our institution; their lack of trust is understandable. They fear this latest incident will pass unaddressed without the individual and communal care this moment requires, and that GU will move on and resume normal operations without the institutional changes that the history of these cyclical incidents demand.

Many faculty share students anger, hurt and frustration. We need swift action in the short term and serious change in the longer term. We acknowledge the messages of solidarity from GUs president, provost, head of student affairs and provost for diversity and inclusion. We also acknowledge that the institution regularly responds with important opportunities to listen to those who are hurt.

Messages of solidarity are not enough. Listening is not enough. Those strategies still have not interrupted the recurring, long-standing experiences of exclusion. In place of reactive responses, we need proactive measures and commitments.

We need stronger, clearer actions and commitments from this point forward. First, we expect the GU administration to swiftly and thoroughly address the tangible actions called for by the BSU. In addition to these student demands, we expect the administration to provide the campus with regular updates about this investigation and your findings. We want you to follow through on the support offered from the local NAACP, the alliance of independent colleges, the States political officials and Spokanes local neighborhoods.

Additionally, GU must affirm that the experiences of oppression, exclusion and pain transcend this particular hate attack; the systemic roots of these problems must be acknowledged and confronted lest these experiences continue to plague our campus community.

Without stronger action, accountability and education for every single member of our community, these problems fester. As we have already seen, what should be a holistic and collaborative space for reflective and critical thought, lifelong learning, spiritual growth, ethical discernment, creativity and innovation, has become divisive, disconnected and imperiled.

For years we have watched GU respond to racism on campus with passive, broad affirmations of diversity, equity, inclusion and our Jesuit values that do not manifest in sustained anti-racist action or community accountability. Moreover, our reluctance to expect accountability from our colleagues maintains the status quo year after year and ensures these experiences remain embedded in our culture. Therefore, we expect immediate commitments and changes grounded in proven strategies for equity and inclusion in higher education and the following unequivocal actions:

Upholding institutional expectations and values are not matters of academic freedom or personal choice; they are requirements for everyone who enjoys the privilege of working for this institution.

We expect GU administration and leadership to deliver on the things youve been tasked to do (and more). Its fundamental to who we are. Fr. Ignacio Ellacura, SJ, asked in a 1982 commencement address at Santa Clara University: What then does a university do, immersed in this reality [of oppression and injustice]? Transform it? Yes. Do everything possible so that liberty is victorious over oppression, justice over injustice, love over hate? Yes. Without this overall commitment, we would not be a university and even less so would we be a Catholic university.

Our values and mission mean little without actions that affirm them. But we as an intellectual community have the imagination and creativity together to discover the remedies to our problems (Fr. Ellacura). It is time to hold ourselves, our colleagues and the administration accountable. This institution must reaffirm our commitment to the common good and prove that it is the institution our mission promises.

ABSTRACT

Introduction: COVID-19 is a novel and devastating disease. Its manifestations vary from asymptomatic to lethal. Moreover, mortality rates differ based on underlying health conditions and ethnicity. We investigated the biochemical rationale behind these observations using machine reasoning by the sci.AI system (https://sci.ai/). Facts were extracted and linked from publications available in nlm.nih.gov and Europe PMC to form the dataset which was validated by medical experts. Results: Based on the analysis of experimental and clinical data, we synthesized detailed biochemical pathways of COVID-19 pathogenesis which were used to explain epidemiological and clinical observations. Clinical manifestations and biomarkers are highlighted to monitor the course of COVID-19 and navigate treatment. As depicted in the Graphical Abstract, SARS-CoV-2 triggers a pro-oxidant (PO) response leading to the production of reactive oxygen species (ROS) as a normal innate defense. However, SARS-CoV-2s unique interference with the antioxidant (AO) system, through suppression of nitric oxide (NO) production in the renin- angiotensin-aldosterone system (RAAS), leads to an excessive inflammatory PO response. The excessive PO response becomes critical in cohorts with a compromised AO system such as patients with glucose-6-phosphate dehydrogenase deficiency (G6PDd) where NO and glutathione (GSH) mechanisms are impaired. G6PDd develops in patients with metabolic syndrome. It is mediated by aldosterone (Ald) which also increases specifically in COVID-19. Conclusion: G6PD is essential for an adequate immune response. Both G6PDd and SARS-CoV-2 compromise the AO system through the same pathways rendering G6PDd the Achilles heel for COVID-19. Thus, the evolutionary antimalarial advantage of the G6PDd cohort can be a disadvantage against SARS-CoV-2.

Clearing the Course for Glycans in Development of Flu Drugs – UC San Diego Health

Researchers demonstrate molecular binding mechanism that could change approach to designing influenza treatments

There is no hole-in-one drug treatment when it comes to the flu, but that doesnt stop scientists from trying to sink one. Especially since as many as one in five Americans gets the flu. The reported estimated cost of this illness is $10 billion each year in medical expenses and another $16 billion in lost earnings in America alone, according to researchers at UC San Diego.

Teeing up on the science behind the flu virus is Rommie Amaro and J. Andrew McCammon, both professors of chemistry and biochemistry, and graduate student Christian Seitz. Together with co-workers Lorenzo Casalino, Robert Konecny and Gary Huber, they studied the effect of glycansgroups of sugar moleculeson the binding of antiviral drugs to viral neuraminidase. An enzyme found on the surface of flu viruses, neuraminidase enables the viruses to exit their diseased host cells and infect and replicate in new, previously healthy host cells. The glycans help to prevent large antibody molecules from binding to the enzyme.

The researchers findings, published in Biophysical Journal, likely apply more generallyincluding to the SARS-CoV-2 virus that causes COVID-19. Amaro will soon be releasing new findings about her latest research on the virus spike protein.

According to the scientists, influenza neuraminidase is the target for three FDA-approved influenza drugs in the U.S., but drug resistance and low drug effectiveness merit more drug development. Generally, however, drug developers do not include glycans in their development pipelines. They know glycans exist, but they have ignored glycans when designing new drugs without a basis for doing so and without evidence that glycans do not affect drug binding.

With their focus on glycans, the team thought it would be prudent to test the assumption about glycans in drug design relative to neuraminidase. Their results show that their proposed binding mechanism can help shed light on the complexity of the interplay between glycans and ligand binding.

Traditionally, glycans have been difficult to study experimentally or theoretically due to a number of technological constraints, which are beginning to be lifted, explained Seitz, first author of the study. Because of this recent emergence of glycans, we still have a lot to learn about them.

The superposition of four glycan conformations onto the static neuraminidase structure shows the conformational variability of the glycans, partially blocking binding site access. Metaphorically, the glycans are the rocks on the mini-golf course and the binding sites are the holes. The glycans are in blue, red, green and gray; the binding sites are in purple and orange, and neuraminidase is in teal. This representation is simplified to emphasize the relationship between the glycans and the binding sites, showing half of the neuraminidase structure and less than one-fifth of the glycans present in the full study. To better understand glycans in the context of this particular study, the team created all-atom in silico systems of influenza neuraminidase, consisting of four different glycan configurations and one glycan-free system. They observed a two- to eight-fold decrease in the rate of ligand binding to the primary binding site of neuraminidase After examining neuraminidases binding sites, the scientists noted that drugs prefer the primary binding site over the secondary binding site.

Personally, I found two things to be quite surprising. Glycans are flexible and can reside very close to the drug binding sites, so I thought that the glycans would completely block drug binding. However, we found the glycans acted more like a screen or a curtainthings can get through, it will just take a bit longer, said Seitz. Secondly, there are two binding sites on influenza neuraminidase; one is the primary (catalytic) site needed for the viral replication cycle to continue, and the other, secondary site, is not well understood. Some previous studies had initially concluded that ligands would reach the secondary site significantly faster than the primary site.

Seitz noted that McCammon was the first person to run a molecular dynamics study with a protein, and the Brownian dynamics software used in this study was developed in his lab. Additionally, Amaro is known as a world-leading expert in molecular dynamics virus simulations, and her virus work has recently been covered in The New York Times.

Combining these rich basins of knowledge we are able to gain new insights into a global disease right here in San Diego, said Seitz.

The graduate student likened the study to a mini-golf course. We have the obvious goal of wanting to get the ball in the hole except, in our study, the golf ball is an influenza drug and the hole is the protein receptor the drug must find to kill the virus. One can often find large rocks on the greens acting as gatekeepers to make it more difficult to get the ball in the hole. In our flu analogy, these rocks are the tiny sugars called glycans.

Just as a groundskeeper can change the position of the rocks near the hole, glycans can change position on the protein. So, the researchers wanted to know if changing the position of the glycans would change how easy or difficult it is for the influenza drug to find its target.

To start, we found common positions of these glycans. However, just as you would not change the position of the rocks on a mini-golf course and do one putt before declaring it easier or harder, we knew we would have to repeat this process many times (600 million, actually) to reach a statistically significant conclusion. Each Brownian dynamics trajectory in our study represented one putt on our mini-golf course, and we simply measured if the ball reached the hole, Seitz explained.

The scientists found that changing the positions of the glycans did make it somewhat harder for the drug to reach the target, but not as much as expected. This means that drug developers do not need to account for glycans when designing new small-molecule drugs for influenzasomething that was unclear before.

For a long time, I thought this couldn't be true and ran numerous tests to disprove it, but all these tests consistently said the same thing, that most small ligands are able to evade the glycans and bind to the enzyme, Seitz said. Thus, this work is one small step in helping to ameliorate the yearly human and economic cost in our nation and our world. This is paid for by our own taxes so each of us has made a tiny contribution to this progress.

Clearview AIs First Amendment Theory Threatens Privacyand Free Speech, Too – Slate

What could be one of the most consequential First Amendment cases of the digital age is pending before a court in Illinois and will likely be argued before the end of the year. The case concerns Clearview AI, the technology company that surreptitiously scraped 3 billion images from the internet to feed a facial recognition app it sold to law enforcement agencies. Now confronting multiple lawsuits based on an Illinois privacy law, the company has retained Floyd Abrams, the prominent First Amendment litigator, to argue that its business activities are constitutionally protected. Landing Abrams was a coup for Clearview, but whether anyone else should be celebrating is less clear. A First Amendment that shielded Clearview and other technology companies from reasonable privacy regulation would be bad for privacy, obviously, but it would be bad for free speech, too.

The lawsuits against Clearview are in their early stages, but there does not seem to be any dispute about the important facts. The company assembled a vast database of images scraped from the internetincluding from social media networks, news sites, and employment siteswithout the consent or knowledge of the people pictured. When a user of Clearviews app uploads a photo of a face, the app converts the image into a series of coordinates and shows images from the database that share similar coordinates. The app also supplies links to the websites from which the company obtained those similar images. The technology is powerful and fast, which is why federal immigration authorities and hundreds of police departments are already using it.

The people whove sued Clearview contend that the company is violating an Illinois privacy law that regulates the collection, use, and dissemination of biometric information. The company argues in defense that its business practices involve the kinds of activities that the First Amendment has been held to protect in the pastcollecting publicly available information, analyzing it, and sharing the conclusions of that analysis. In a brief filed in October, it likened its app to a search engine and contended that its judgment about what information will be most useful to users is an editorial judgment akin to those made by newspapers.

That Clearview is reaching for the First Amendment is not a surprise. In recent years, the Supreme Court has been willing to extend the First Amendments protection to an ever-expanding range of activities. Technology companies have learned that an effective way to protect lucrative business practices from regulation is to characterize those practices as free speech. Google has been arguing, with some success in the lower courts, that judges should deal with any effort to regulate its search engine in the same way theyd deal with efforts to censor the Wall Street Journal. In Maine, internet service providers are arguing that the First Amendment protects their right to use and sell their customers sensitive data without their consent. Earlier this fall, President Donald Trump issued an executive order meant to shut down TikTok, the video-sharing platform. The company sued, arguing that the order violated the First Amendment because TikTok runs on code, and code is speech.

These arguments may sound audacious, but its more difficult than you might imagine to draw lines between these companies business practices and the kinds of activities that most of us believe the First Amendment must protect. Why, exactly, should the First Amendment treat Googles decisions about which search results to highlight differently from the New York Times decisions about which articles to feature on its front page? Why is it, precisely, that the First Amendment should protect journalists who scrape the web in the service of their journalism but not Clearview when it scrapes the web in the service of its app? Principled line-drawing in this context is hard. (Go on, try it yourself.) The slopes are slippery. Even skeptics of the technology companies arguments may legitimately worry about the broader implications of allowing judges to place those companies activities outside the First Amendments protection.

But if line-drawing is difficult here, its also absolutely necessary. When courts conclude that a given activity is speech within the meaning of the First Amendment, they protect it from government regulation, which makes it more difficult for the government to address social harms that may be associated with it. Its important, then, that First Amendment protection be reserved for the kinds of activities that actually further the ends the First Amendment was meant to serve.

Is Clearview engaged in activity that the First Amendment should care about? This is a hard question, but the companys arguments dont immediately persuade us. The company says its engaged in the creation and dissemination of information, but American courts havent extended First Amendment protection mechanically to every activity that involves information, data, or even speech in the colloquial sense, as scholars have observed. Instead, courts have looked to the social meaning of the activity in question, asking, for instance, whether the activity belongs to a recognized medium of expression; whether it is intended to convey a message and whether that message is likely to be understood; and, perhaps most important, whether the activity has the effect of informing public discourse.

It was considerations like these that led the courts to extend the First Amendments protection to flag burning, video games, and picketingthough none of these things is speech in the ordinary sense of the word. And it was considerations like these that led courts to withhold First Amendment protection from the solicitation of hitmen, threats of immediate violence, and agreements to fix pricesthough most people would describe all of these things as speech. It has always mattered to courts, in other words, what an activity signifies, and what it is, and what it does.

With all of this in mind, Clearviews claim to First Amendment protection is less than compelling. The companys extraction of biometric data from photos scraped from the internetwhich is the specific activity that the Illinois law regulates heredoes not belong to any recognized mode of expression. And it is not intended to inform public discourse, even if on occasion it might do so incidentally. For understandable reasons, Clearview wants courts to think of the company as an editor and the companys app as a search engine. But the companys arguments seem to rely on obscuring differences that matter.

The bigger problem for the company, though, may be that even if the companys business activities are speech within the meaning of the First Amendment, the Illinois law is a reasonable regulation of that speech. In its legal papers, Clearview argues that any law regulating its business activities should be subject to the most stringent form of constitutional scrutinythe same kind of scrutiny courts would apply to a law censoring the press. But while laws censoring the press are speech-suppressive almost by definition, this isnt true of laws protecting individual privacy. To the contrary, privacy is a precondition for all First Amendment freedoms, including the freedom of speech, and so laws protecting privacy can sometimes be speech-enhancing. As the Supreme Court has observed, in many privacy cases there are free speech interests on both sides of the balance. In those cases, the question the courts should ask is not whether the law can survive the most stringent scrutiny but whether it balances those interests in a reasonable way.

The Illinois law does this. It focuses narrowly on certain forms of nonconsensual surveillance that pose an especially serious threat to individual privacy. Facial recognition in particular is an immensely powerful form of surveillance whose abuse could fundamentally undermine civil liberties, including the liberties the First Amendment is meant to protect. Clearviews technology highlights these dangers. The companys app would allow anyone to identify the protesters who attended a particular political rally, or to identify the people who entered a particular house of worship or medical clinic. According to the New York Times, one of the apps founders tried to sell access to a white supremacist who was running for Congress, telling him, quite accurately, that the app could be used to conduct extreme opposition research. Against this background, Illinois decision to restrict the nonconsensual collection and use of biometric information is perfectly understandable. The law is a straightforward response to technology whose unregulated deployment would, as the New York Times observed, end privacy as we know it.

This is not to say that the Illinois law is perfect. It doesnt restrict the collection and use of biometric data by government agencies, for example. It doesnt include exceptions for journalismand, consequently, its possible that a journalist might be able to challenge the application of the law to a specific investigative project. On the whole, though, the law is a reasonable effort to balance competing interests, including those related to the First Amendment. It would be disappointing, to say the least, if the courts let Clearview use the First Amendment to kneecap the freedoms it was meant to protect. Carefully drawn privacy laws are a precondition for free speech in the digital age, not a threat to it.

More broadly, it would be terrible for the freedoms of inquiry, association, and speech if the courts didnt think very carefully before allowing companies like Clearview to wrap their business models in the First Amendment. It hardly needs to be said that our speech environment looks radically different today than it did 50 years ago, when the Supreme Court issued many of the rulings that have come to define the First Amendment. Most political speech now takes place online. A small number of tech companies serve as the gatekeepers of online public discourse. Their business model entails pervasive surveillance of what we read, what we say, and whom we associate and correspond with. These developments demand that legislatures think creatively about how the digital public sphere can be kept moored to democratic values and the public interest. It would be a mistake to allow the First Amendment to become an obstacle to laws essential to protect the integrity and vitality of free speech in the digital age.
