On Wednesday October 24, 2012, the Ornithine Laboratory has made a new video available to the public on its website for the 'Japan Otsukare University, Faculty of Ornithine' (literally University of Fatigue, Faculty of Ornithine - http://www.otsukare-univ.jp/). The site was established for the fictional university, with a goal of 'eliminating fatigue in the world'. This video depicts a showdown between Kageki Shimoda and Pe Hayashiya in a powerful, colorful 100-meter dash like never before seen. Shimoda is a holder of one of the most prestigious literary awards in Japan the Naoki Prize. Hayashiya is a comic-chat artist, one of Japan's historic traditional arts.

At the site (available in Japanese only) Shimoda plays the 'tireless' character; a school principal who combines radical fashion sense belying his age of 72 years, and intelligence. His challenger is 70 year-old Pe Hayashiya, a comedian known for his trademark pink outfits.

Low vitamin D levels linked to longevity, surprising study shows

ScienceDaily (Nov. 5, 2012) Low levels of vitamin D may be associated with longevity, according to a study involving middle-aged children of people in their 90s published in CMAJ (Canadian Medical Association Journal).

"We found that familial longevity was associated with lower levels of vitamin D and a lower frequency of allelic variation in the CYP2R1 gene, which was associated with higher levels of vitamin D," writes Dr. Diana van Heemst, Department of Gerontology and Geriatrics, Leiden University Medical Center, Leiden, the Netherlands, with coauthors.

Previous studies have shown that low levels of vitamin D are associated with increased rates of death, heart disease, diabetes, cancer, allergies, mental illness and other afflictions. However, it is not known whether low levels are the cause of these diseases or if they are a consequence.

To determine whether there was an association between vitamin D levels and longevity, Dutch researchers looked at data from 380 white families with at least 2 siblings over age 90 (89 years or older for men and 91 year or older for women) in the Leiden Longevity Study. The study involved the siblings, their offspring and their offsprings' partners for a total of 1038 offspring and 461 controls. The children of the nonagenarians were included because it is difficult to include controls for the older age group. The partners were included because they were of a similar age and shared similar environmental factors that might influence vitamin D levels.

The researchers measured levels of 25(OH) vitamin D and categorized levels by month as they varied according to season. Tanning bed use, which can affect vitamin D levels, was categorized as never, 1 times per year and 6 times per year. The researchers controlled for age, sex, BMI (body mass index), time of year, vitamin supplementation and kidney function, all factors that can influence vitamin D levels. They also looked at the influence of genetic variation in 3 genes associated with vitamin D levels.

"We found that the offspring of nonagenarians who had at least 1 nonagenarian sibling had lower levels of vitamin D than controls, independent of possible confounding factors and SNPs [single nucleotide polymorphisms] associated with vitamin D levels," write the authors. "We also found that the offspring had a lower frequency of common genetic variants in the CYP2R1 gene; a common genetic variant of this gene predisposes people to high vitamin D levels.

These findings support an association between low vitamin D levels and familial longevity." They postulate that offspring of nonagenarians might have more of a protein that is hypothesized to be an "aging suppressor" protein. More research is needed to understand the link between lower vitamin D levels, genetic variants and familial longevity.

New Rochelle, NY, November 5, 2012At least 2 million people worldwide will be infected with HIV this year, driving the need for better HIV prevention strategies to slow the global pandemic. A better understanding of how to prevent HIV transmission using antiviral drugs led to approval of the first oral pill for HIV prevention, and microbicides delivered as topical gels or via intravaginal rings are in clinical testing and have yielded both positive and negative results. The complex factors involved in the sexual transmission of HIV, the urgent need for new preventive approaches, and the most promising methods currently in development are examined in a special issue of AIDS Research and Human Retroviruses, a peer-reviewed journal published by Mary Ann Liebert, Inc, publishers. Guest Editor Patrick Kiser, University of Utah, Salt Lake City, and coauthors Pedro Mesquita and Betsy Herold, Albert Einstein College of Medicine, New York, NY, provide an overview of the scientific and developmental gaps in the field of drug discovery, formulation, and delivery to prevent sexual transmission of HIV. In the article "A Perspective on Progress and Gaps in HIV Prevention Science," they review the prophylactic agents in development and their advantages and limitations, and they present recommendations for future research directions. "The advances we are seeing today are exciting and justify a continued focus on the science and technologies that can make a difference in this public health crisis," says Dr. Kiser.

Peter Anton and coauthors, David Geffen School of Medicine at UCLA and the School of Public Health (Los Angeles, CA), University of Pittsburgh and Medical School (PA), University of North Carolina, Chapel Hill, Johns Hopkins University School of Medicine (Baltimore, MD), Alpha StatConsult (Damascus, MD), Columbia University (New York, NY), and CONRAD (Arlington, VA), compared the oral and topical administration of the microbicide tenofovir, given as a pill or rectal gel. A mucosal tissue sample was removed from each participant and exposed to HIV to assess if treatment prevented infection. While the patients preferred the oral drug, one-time rectal exposure to the microbicide led to 6-10 times greater drug concentrations in the sampled tissue, and this correlates with reduced infectibility.

The methods and materials used to administer microbicidal drugs can affect whether or not they will be effective. Meropi Aravantinou et al. (Population Council and Rockefeller University, New York, NY; Tulane University, Covington, LA; National Cancer Institute, Frederick, MD) demonstrate this in the article "The Nonnucleoside Reverse Transcription Inhibitor MIV-160 Delivered from an Intravaginal Ring, But Not from a Carrageenan Gel, Protects Against Simian/Human Immunodeficiency Virus-RT Infection."

Line Vibholm and colleagues from Aarhus University and Aarhus University Hospital, Denmark, present the results of a study designed to evaluate in a female mouse model the effects of a topical gel containing 1% tenofovir, a microbicide previously shown to reduce substantially the transmission of both HIV and herpes simplex virus (HSV). In "Antiviral and Immunological Effects of Tenofovir Microbicide in Vaginal Herpes Simplex Virus 2 Infection," the authors provide data to support the suitability of this model for testing future microbicidal drug candidates.
