The machine is an automated, high-throughput system for counting blood cells and measuring biochemical and immunological components in the blood. This resource is part of the UK Open Educational Resource project and was made in collaboration between the Leicester Royal Infirmary and De Montfort University.

Scientists Unlock How Insulin Interacts With Cells

Newswise The discovery of insulin nearly a century ago changed diabetes from a death sentence to a chronic disease.

Today a team that includes researchers from Case Western Reserve University School of Medicine announced a discovery that could lead to dramatic improvements in the lives of people managing diabetes.

After decades of speculation about exactly how insulin interacts with cells, the international group of scientists finally found a definitive answer: in an article published today in the journal Nature, the group describes how insulin binds to the cell to allow the cell to transform sugar into energyand also how the insulin itself changes shape as a result of this connection.

These findings carry profound implications for diabetes patients, said Case Western Reserve biochemistry professor and department chair Michael A. Weiss, MD, PhD, MBA, one of the leaders of the team. This new information increases exponentially the chances that we can develop better treatmentsin particular, oral medications instead of syringes, pens or pumps.

Weiss, also the Cowan-Blum Professor of Cancer Research at the School of Medicine, is renowned worldwide for his work on insulin. In 1991 he used nuclear magnetic resonance techniques to describe the structure of insulin; more recently he has developed a preliminary version of the hormone that does not need to be refrigerated, a critical breakthrough for those with diabetes in the developing world.

The results published today, however, represent among the most promising for Weiss and an entire generation of scientists devoted to enhancing care for those suffering from diabetes. They have sought to solve mystery of how the hormone bound to cells since 1969, when the late Dorothy Hodgkin and colleagues at the University of Oxford, first described insulins structure.

Theres been a logjam in our understanding since then, Weiss said. We hope that weve broken the logjam.

The magnitude of the challenge is, in part, evidenced by the diversity of the team required to overcome it. Weiss partnered with Associate Professor Mike Lawrence, of the Walter and Eliza Hall Institute of Medical Research in Melbourne, Australia, to lead the project. They, in turn, engaged scientists from the University of Chicago, the University of York in the United Kingdom, and the Institute of Organic Chemistry and Biochemistry in Prague in the Czech Republic.

The scientists recognized that cells absorb sugar from food as energy for the body, yet glucose cant penetrate a cells membrane without help from insulin, a hormone secreted from endocrine cells in the pancreas. To absorb the sugar, most cells have insulin receptors that bind the hormone as it flows through the bloodstream.

The researchers tested structural models using molecular-genetic methods to insert probes that, in turn, are activated by ultra-violet light into the receptor. The procedure creates highly detailed, three-dimensional imageswhich provided critical answers for Weiss, Lawrence and their colleagues.

Health&Fitness Eileen White, Ph.D., associate director for basic science at The Cancer Institute of New Jersey and professor of molecular biology and biochemistry at Rutgers,The State University of New Jersey, and colleagues from Princeton University have been awarded a new five-year, $2.4 million dollar research grant (1R01CA 163591-01) from the National Cancer Institute.The funding will support research to identify metabolic vulnerabilities of particularly aggressive cancers those with mutations in the Ras cancer gene.The Cancer Institute of New Jersey is a Center of Excellence of the University of Medicine and Dentistry of New Jersey-Robert Wood Johnson Medical School (RWJMS).

Led by principal investigators Dr. White and Joshua D. Rabinowitz M.D., Ph.D., professor in the Department of Chemistry and the Lewis-Sigler Institute for Integrative Genomics at Princeton University, the project is an extension of a collaborative effort between their two laboratories.That work, which has examined cancer metabolism, was initially funded through a National Institutes of Health Challenge Grant.

For nearly half a century, it has been known that altered metabolism distinguishes cancer cells from normal cells. Only recently have scientists learned that activation of cancer genes and loss of tumor suppressor genes reprograms cell metabolism.With this altered metabolism, cancer cells acquire the ability to make building blocks for production of new cancer cells and produce the energy needed for cancer growth.

The current project aims to identify the Achilles heel in the altered metabolism of cancer cells that are specifically fueled by mutations in the Ras gene. Cancer-causing mutations in Ras proteins activate Ras signaling that promotes cancer cell growth. Ras mutations cause the most deadly cancers such as pancreatic, lung and colon cancers.Targeting Ras proteins directly with intended therapeutics has been unsuccessful, thus prompting investigators to instead look at blocking the downstream consequences of Ras function. But in order to do that, they must first identify what Ras needs to convert normal cells into tumor cells, including how Ras alters metabolism in ways that are essential for tumor growth.

The White and Rabinowitz laboratories recently examined Ras activation and its impact on tumor cells.They found that Ras-driven cancers are dependent on a self-preservation process known as autophagy, in which cells cannibalize themselves in order to survive stressful conditions during tumor growth and in response to therapy. In this project, White and Rabinowitz aim to further explore metabolic dependence and autophagy addiction as metabolic vulnerabilities that can be exploited in the development of new cancer therapies.

White, who also is the program leader of the Cell Death and Survival Signaling Program at The Cancer Institute of New Jersey and an adjunct professor of surgery at RWJMS, notes,We have already shown that Ras-driven cancers are vulnerable to interference in specific metabolic pathways. By further examining these metabolic liabilities, we will be able to identify additional weaknesses that can be targeted therapeutically to provide new treatments for Ras-driven cancers.

The grant runs through June 2017.

Beer brewing classes grow in popularity

For some South Florida college students, beeris more than an extracurricular activity -- it's a science class.

"It's essentially an applied biochemistry class in that we will look at the chemistry, biochemistry, physics, mechanics of all the brewing operation," said Dr. Barry Gump, who teaches at Florida International University's Chaplin School of Hospitality & Tourism Management.

With South Florida's thirst for craft beer growing, FIU began offering its brewing science course year round instead of summer-only. Turning out brewmasters helps make South Florida an up and coming hub in the craft beer industry.

FIU students Matt Weintraub and Zhilong Yang plan to open their own brew pub after graduation.They already are gathering experience brewing beers for private parties.

"A lot of people just want a local vibe in their beer, using local ingredients, local ideas, local companies," said Weintraub.

During the semester, students study the different nuances of beers by keeping tasting notes, and learning about sanitation, quality control and packaging.

The class brews about six different beers. As a final project, they then brew their own recipe.

Lemongrass, strawberries, and pumpkin are all ingredients the students have used.

"I just love the endless possibilities you can make in your beer," said Weintraub.
