Researchers develop the first DNA computer capable of pinpointing several antibodies in the blood and carrying through resultant calculations.

A team of researchers has developed an intelligent DNA computer that can enable controlled drug delivery into the bloodstream. The team, from Eindhoven University of Technology in the Netherlands, believes this new method will lead to the development of smart drugs that more effectively treat autoimmune diseases like rheumatoid arthritis and Crohn's disease. The team, led by Professor of Biochemical Chemistry Maarten Merkx, published their findings in the Nature Communications journal.

DNA Computing

DNA computing is a largely experimental field of scientific study. Using a combination of DNA, biochemistry, and molecular biology hardware, researchers seek to use DNA sequencing for data storage and to perform complex calculations.

The goal of this new field is to create nano-computers using DNA instead of mechanical parts. The resulting machines would be able to store immense amounts of data and make complex calculations faster than inorganic equipment. Some scientists believe DNA computers may one day replace silicon-based hardware.

Possible Applications in Medical Treatment

DNA computing methods, due to their organic nature, allow medicines to work more effectively with the bodys natural defenses. Researchers compare the body to a security system with facial recognition. Previous research has focused on creating a machine that is recognized by the body, similar to the way a security system would recognize an authorized person.

Professor Merkxs method adds the ability to recognize the presence of antibodies. The identification and measurement of antibodies is a crucial step in diagnosing and treating disease. Antibody levels are also a factor when calculating the dosage of therapeutic drugs.

With DNA nano-computers able to detect and measure antibodies while performing complex analyses, researchers are hopeful machines can be programmed to carry out actions in response to certain measurements. Merkxs team successfully used DNA computing to control the activity of enzymes. The professor believes it is possible to apply this technique to therapeutic antibodies. This should result in better absorption of therapeutic substances. When used as a delivery system, these computers will be able to decide how much medicine to release into the bloodstream, based on the antibodies in the bloodstream.

Future Applications in the Treatment of Chronic Disease

Researchers are working on ways to use the technology in the fight against chronic diseases like rheumatoid arthritis and Crohns Disease. These conditions are often treated with the same drugs. The dosage is dependent on the condition it is meant to treat. Smart drugs would be able to deliver the most accurately therapeutic dose of medication. This would allow for more effective treatment. It would also reduce the overall cost of treatment and reduce side effects.

Testosterone Anti-Aging Studies: Some Benefits, But None to Mental … – KQED

Testosterone treatment did not improve older mens memory or mental function in the latest results from landmark government research that challenges the anti-aging claims of popular supplements.

While testosterone use for one year appeared to strengthen bones and reduce anemia, it also showed signs of worsening artery disease. Questions also remain about other potential risks. The researchers said more studies are needed to determine long-term effects the kind of research the U.S. Food and Drug Administration has already asked supplement makers to conduct.

I dont think anybody would interpret these results as saying, Wow, this is a fountain of youth, this is a magical anti-aging potion,' said study co-author Susan Ellenberg, a University of Pennsylvania researcher.

The results are from the final four studies in a seven-part project mostly funded by the National Institute on Aging, involving nearly 800 U.S. men aged 65 and older with low testosterone levels. The goal was to see if rubbing testosterone gel on the skin daily for a year could treat problems linked with low levels of the male hormone, which declines with age. Half the men in each group used the real thing and half used fake gel.

Results published a year ago from the same research linked testosterone with mostly modest improvement in sexual performance, walking strength and mood.

The key new findings:

The overall health benefits, however, remain to be determined, the researchers said.

The studies were published Tuesday in the Journal of the American Medical Association and JAMA Internal Medicine. AbbVie Pharmaceuticals provided its gel for the study and helped pay for the research but had no other role in the study.

The research was not designed to look at risks and does not apply to younger men or those with normal levels of testosterone, said study leader Dr. Peter Snyder, a University of Pennsylvania hormone specialist. Its also not known if other forms of testosterone supplements would have similar effects in older men with low levels.

Prescription testosterone products, including gel, are approved only for men with low levels of the hormones caused by various medical conditions. Benefits and risks are unknown in men whose levels are low due only to aging, the FDA says. The agency requires testosterone labels to include possible risks for heart attacks and strokes, based on some previous studies.

A separate study published Tuesday in JAMA Internal Medicine found that men using prescription testosterone gel, patches or injections had fewer heart attacks and strokes than non-users during about three years of follow-up. But this was only observational data in men aged 40 and up, not rigorous research testing the hormone against a placebo.

Clarifying testosterones effects on heart problems, fractures and age-related disability will require larger, longer studies, said Dr. Evan Hadley of the National Institute on Aging. He said decisions about whether to use testosterone should take into account mens individual risks for conditions the hormone could affect.

Details of the Global Collaborative Study

A team of researchers from around the world determined vitamin D supplements combat the onset of numerous respiratory diseases. Carlos Camargo from Massachusetts General Hospital's Department of Emergency Medicine served as the study's senior author. The research was funded through a grant provided by the United Kingdom's National Institute of Health Research. The worldwide collaborative study confirms that vitamin D supplements help ward off acute respiratory infections.

The study consisted of a meta-analysis of 25 randomized trials with over 11,000 participants. The study's details and results were recently published on the web in The BMJ. It is widely known that vitamin D is vital for muscle and bone health. However, it was unclear as to whether vitamin D played a critically important role in combating acute respiratory infections. Such infections result in millions of hospital visits and deaths across the world each year. Clearing up Conflicting Data

A handful of observational studies that monitored patients across time without implementing a specific treatment had linked low levels of vitamin D to a higher susceptibility of acute respiratory infections. Several clinical trials have also been conducted to study vitamin D supplements' protective ability. Some of these studies determined the vitamin really does provide a protective quality. However, some studies showed that it does not have any protective effect. Meta-analyses performed of these clinical trials aggregated information from numerous studies that have differing designs and/or participant qualifications produced conflicting results.

The global research team referenced above cleared up these conflicting results. Queen Mary University of London's Adrian Martineau led the team. He performed a data meta-analysis of individual participants from over a dozen nations. Participants hailed from the United Kingdom, the United States, Canada and beyond. Prior meta-analyses pit average data from subjects in each study. Alternatively, individualized data meta-analysis distinguishes data from each participant, generating what is considered as a higher-resolution analysis of information across all studies.

The Results

It was determined that weekly and daily vitamin D supplements provided the optimal benefit for those who suffered from a severe vitamin D deficiency. This deficiency is defined as a blood level with 10 mg/dl or less. Vitamin D supplementation reduced the odds of a respiratory infection by one-half for such individuals suffering from a vitamin D deficiency. The study also found that every single participant enjoyed at least a minor benefit from consuming vitamin D supplements on a regular basis. It is worth noting that consuming occasionally high doses of the vitamin did not generate extensive benefits.

The hope is that people will ramp up their consumption of vitamin D supplements to reduce the risk of colds, the flu, and other respiratory maladies. This course of preventative action would prove especially helpful for those who are vitamin D deficient. Autologous hematopoietic stem cell transplantation halted progression of Multiple Sclerosis for 5 years in 46% of patients.

Recent research indicates that the transplant of autologous hematopoietic stem cells (AHSCT) is an excellent treatment for multiple sclerosis. It has been determined that the procedure stops disease progression for half a decade in nearly 50 percent of multiple sclerosis patients.

About the Study

The study was spearheaded by Dr. Paolo Muraro from the Imperial College London's Department of Medicine. Dr. Muraro and his colleagues revealed their findings through JAMA Neurology. These results were released on the heels of a separate study that found the success of a similar treatment in patients suffering from relapsing-remitting multiple sclerosis (RRMS). Dr. Muraro and his research team are quick to point out that additional trials are necessary to gauge the efficacy and safety of AHSCT. It is important to note that some patients perished within the first 100 days of treatment.

About AHSCT

AHSCT involves the harvesting of a patient's own stem cells. The patient is subjected to a powerful dose of chemotherapy to destroy any diseased cells. The next step is the return of harvested stem cells to the patient's blood. The goal is to restart the production of normal blood cells.

In layman's terms, AHSCT is best understood as a resetting of the body's immune system. Though it was already known that this style of treatment resets the immune system and poses certain risks, the length of its benefits was not fully understood. We now have a better picture of these benefits. AHSCT Results

The research team studied data from over two dozen treatment centers in 13 countries. They pinpointed 281 patients who suffered from multiple sclerosis and underwent AHSCT from 1995 to 2006. Exactly 78 percent of these patients had a progressive form of multiple sclerosis. The team made use of the Expanded Disability Status Scale (EDSS) to analyze patients' survival after five years of treatment as well as improvements in their multiple sclerosis symptoms. A whopping 46 percent of these patients enjoyed zero disease progression in the five years following treatment. Those with RRMS, characterized by flare-ups (inflammatory attacks) and periods of remission enjoyed the optimal outcomes.

An amazing 73 percent of these patients did not endure worsening of symptoms in the 5 years following AHSCT. The findings described above clearly show promise for the AHSCT use in individuals who suffer from multiple sclerosis. The research team would like to make it perfectly clear that some patients died in the 100 days following AHSCT. Exactly eight patients perished in this time period. It is assumed that the deaths were related to treatment. AHSCT makes use of aggressive chemotherapy that significantly weakens the immune system and spikes one's risk for infection. Since multiple sclerosis is not a disease that is immediately life-threatening, the risk of death posed by AHSCT must be weighed by all multiple sclerosis patients.

What's next Dr. Muraro is adamant that a follow-up study must be performed that includes a group of multiple sclerosis patients who have not received AHSCT. It is clear that additional studies are required to accurately gauge the efficacy and safety of AHSCT. Ideally, a massive randomized controlled trial of AHSCT will be performed in the coming months.

CHICAGO (AP) Testosterone treatment did not improve older men's memory or mental function in the latest results from landmark government research that challenges the anti-aging claims of popular supplements.

While testosterone use for one year appeared to strengthen bones and reduce anemia, it also showed signs of worsening artery disease and questions remain about other potential risks. The researchers said more studies are needed to determine long-term effects the kind of research the U.S. Food and Drug Administration has already asked supplement makers to conduct.

"I don't think anybody would interpret these results as saying, 'Wow, this is a fountain of youth, this is a magical anti-aging potion,'" said study co-author Susan Ellenberg, a University of Pennsylvania researcher.

The results are from the final four studies in a seven-part project mostly funded by the National Institute on Aging, involving nearly 800 U.S. men aged 65 and older with low testosterone levels. The goal was to see if rubbing testosterone gel on the skin daily for a year could treat problems linked with low levels of the male hormone, which declines with age. Half the men in each group used the real thing and half used fake gel.

Results published a year ago from the same research linked testosterone with mostly modest improvement in sexual performance, walking strength and mood.

The key new findings:

Testosterone had no effect on memory or mental function, based on tests given before, halfway and at the end of treatment to nearly 500 men with age-related memory decline.

Among almost 140 men who underwent heart artery imaging tests to see if the hormone slowed progression of plaque, those who used testosterone had more plaque buildup and narrower arteries after a year than the fake gel group. Those changes could signal increased chances for heart attacks although none occurred in the study. Men in this sub-study were already more vulnerable for heart problems because of conditions including artery disease, obesity and high blood pressure.

Among about 200 men given bone imaging tests before and at the end of treatment, those on testosterone showed increases in bone density and strength, especially in the spine, while minimal changes were found in the group that used fake gel. The improvement was similar to bone changes seen with treatment for osteoporosis, although most men studied did not have that bone-thinning condition, which can lead to fractures.

Among 126 men with anemia, a fatigue-linked condition involving inadequate red blood cells, those on testosterone showed substantial improvement. By the study's end, anemia had vanished in almost 60 percent of men on testosterone compared with 22 percent of the fake gel group. The hormone group also reported having more energy. "The overall health benefits, however, remain to be determined," the researchers said.

The studies were published Tuesday in the Journal of the American Medical Association and JAMA Internal Medicine. AbbVie Pharmaceuticals provided its gel for the study and helped pay for the research but had no other role in the study.

The research was not designed to look at risks and does not apply to younger men or those with normal levels of testosterone, said study leader Dr. Peter Snyder, a University of Pennsylvania hormone specialist. It's also not known if other forms of testosterone supplements would have similar effects in older men with low levels.

Potential of Adipose-derived Stem Cells in Anti-Aging Treatments – Technology Networks (press release) (registration) (blog)

Adult stem cells collected directly from human fat are more stable than other cells such as fibroblasts from the skin and have the potential for use in anti-aging treatments, according to researchers from the Perelman School of Medicine at the University of Pennsylvania. They made the discovery after developing a new model to study chronological aging of these cells.

Chronological aging shows the natural life cycle of the cells as opposed to cells that have been unnaturally replicated multiple times or otherwise manipulated in a lab. In order to preserve the cells in their natural state, Penn researchers developed a system to collect and store them without manipulating them, making them available for this study. They found stem cells collected directly from human fat called adipose-derived stem cells (ASCs) can make more proteins than originally thought. This gives them the ability to replicate and maintain their stability, a finding that held true in cells collected from patients of all ages.

Our study shows these cells are very robust, even when they are collected from older patients, said Ivona Percec, MD, director of Basic Science Research in the Center for Human Appearance and the studys lead author. It also shows these cells can be potentially used safely in the future, because they require minimal manipulation and maintenance.

Stem cells are currently used in a variety of anti-aging treatments and are commonly collected from a variety of tissues. But Percecs team specifically found ASCs to be more stable than other cells, a finding that can potentially open the door to new therapies for the prevention and treatment of aging-related diseases.

Unlike other adult human stem cells, the rate at which these ASCs multiply stays consistent with age, Percec said. That means these cells could be far more stable and helpful as we continue to study natural aging.

ASCs are not currently approved for direct use by the Food and Drug Administration, so more research is needed. Percec said the next step for her team is to study how chromatin is regulated in ASCs. Essentially, they want to know how tightly the DNA is wound around proteins inside these cells and how this affects aging. The more open the chromatin is, the more the traits affected by the genes inside will be expressed. Percec said she hopes to find out how ASCs can maintain an open profile with aging.

Saitama clinic ordered to halt unauthorized stem cell 'anti-aging' therapy using umbilical cord blood – The Japan Times

SAITAMA The health ministry on Monday ordered a Saitama clinic to stop injecting stem cells of human umbilical cord blood into patients as an anti-aging treatment.

Saitama Medical Clinic in Tokorozawa, Saitama Prefecture, was found to have been engaged in such therapy without getting permission from the ministry. The regenerative medicine safety law mandates that all hospitals and clinics submit plans for stem cell therapy and get permission from the ministry beforehand.

The ministry conducted an on-site inspection of the clinic on Friday, based on tips from an outsider, and confirmed the clinic offered the unauthorized therapy, boasting that umbilical cord cell shots would boost health and help with anti-aging.

Several men and women are believed to have undergone treatment there. The ministry has asked the clinic to report the number of cases, any health damage and the kind of stem cells used and how they were obtained.

For clinics to receive approval for medical procedures using cells of others, they need to submit a detailed plan and have it screened by a panel of experts under the health ministry. The clinic had not submitted such a plan.
