WEDNESDAY, Dec. 24, 2014 (HealthDay News) -- Researchers could be closing in on a "fountain of youth" drug that can delay the effects of aging and improve the health of older adults, a new study suggests.

Seniors received a significant boost to their immune systems when given a drug that targets a genetic signaling pathway linked to aging and immune function, researchers with the drug maker Novartis report.

The experimental medication, a version of the drug rapamycin, improved the seniors' immune response to a flu vaccine by 20 percent, researchers said in the current issue of Science Translational Medicine.

The study is a "watershed" moment for research into the health effects of aging, said Dr. Nir Barzilai, director of the Institute for Aging Research at Albert Einstein College of Medicine in New York City.

Rapamycin belongs to a class of drugs known as mTOR inhibitors, which have been shown to counteract aging and aging-related diseases in mice and other animals.

Barzilai, who wasn't involved in the study, said this is one of the first studies to show that these drugs also can delay the effects of aging in humans.

"It sets the stage for using this drug to target aging, to improve everything about aging," Barzilai said. "That's really going to be for us a turning point in research, and we are very excited."

The mTOR genetic pathway promotes healthy growth in the young. But it appears to have a negative effect on mammals as they grow older, said study lead author Dr. Joan Mannick, executive director of the New Indications Discovery Unit at the Novartis Institutes for Biomedical Research.

When drugs like rapamycin are used to inhibit the effects of the mTOR pathway in mice, they "seem to extend lifespan and delay the onset of aging-related illnesses," Mannick said.

Dec. 20, 2014 - Ibuprofen, a common over-the-counter drug used to relieve pain and sold under the brand names of Advil, Motrin and others, could hold the keys to a longer healthier life, according to a study showing that regular doses of ibuprofen extended the lifespan of yeast, worms and fruit flies.

The study was led by researchers at the Buck Institute for Research on Aging in a collaboration with Texas A & M's Agrilife program and published in PLoS Genetics on December 18.

"There is a lot to be excited about," said Brian Kennedy, PhD, CEO of the Buck Institute, who said treatments, given at doses comparable to those used in humans, extended lifespan an average of 15 percent in the model organisms. "Not only did all the species live longer, but the treated flies and worms appeared more healthy," he said.

"The research shows that ibuprofen impacts a process not yet implicated in aging, giving us a new way to study and understand the aging process."

But most importantly, Kennedy said the study opens the door for a new exploration of so-called "anti-aging medicines." "Ibuprofen is a relatively safe drug, found in most people's medicine cabinets," he said. "There is every reason to believe there are other existing treatments that can impact healthspan and we need to be studying them."

Michael Polymenis, PhD, an AgriLife Research biochemist in Texas A & M's Agrilife program started the work in baker's yeast and then moved it into worms and flies.

Ibuprofen, a common over-the-counter drug used to relieve pain and fever, could hold the keys to a longer healthier life, according to a study by researchers at the Buck Institute for Research on Aging. Publishing in PLoS Genetics on December 18th, scientists showed that regular doses of ibuprofen extended the lifespan of yeast, worms and fruit flies.

"There is a lot to be excited about," said Brian Kennedy, PhD, CEO of the Buck Institute, who said treatments, given at doses comparable to those used in humans, extended lifespan an average of 15 percent in the model organisms. "Not only did all the species live longer, but the treated flies and worms appeared more healthy," he said. The work was the result of a collaboration between the Buck Institute and Texas A & M's Agrilife program. Michael Polymenis, PhD, an AgriLife Research biochemist started the work in baker's yeast and then moved it into worms and flies. Polymenis, who also is a professor in the biochemistry and biophysics department at Texas A&M University, said the three-year project showed that ibuprofen interferes with the ability of yeast cells to pick up tryptophan, an amino acid found in every cell of every organism. Tryptophan is essential for humans, who get it from protein sources in the diet. "We are not sure why this works, but it's worth exploring further. This study was a proof of principle, to show that common, relatively safe drugs in humans can extend the lifespan of very diverse organisms," he said. "Therefore, it should be possible to find others like ibuprofen with even better ability to extend lifespan, with the aim of adding healthy years of life in people."

"Dr. Polymenis approached me with this idea of seeing how his cell cycle analysis corresponded with our aging studies," said Kennedy. "He had identified some drugs that had some really unique properties, and we wanted to know if they might affect aging, so we did those studies in our lab," he said. "The Buck Institute is interested in finding out why people get sick when they get old. We think that by understanding those processes, we can intervene and find ways to extend human healthspan to keep people healthier longer to slow down aging. That's our ultimate goal."

Ibuprofen is in the class of compounds known as NSAID's - nonsteroidal anti-inflammatory drugs used for relieving pain, helping with fever and reducing inflammation. It was created in the early 1960's in England and was first made available by prescription and then, after widespread use, became available over-the-counter throughout the world in the 1980s. The World Health Organization includes ibuprofen on their "List of Essential Medications" needed in a basic health system. Although deemed relatively safe and commonly used, ibuprofen can have adverse side effects, particularly in the gastrointestinal tract and the liver at high doses.

Chong He, PhD, a postdoctoral fellow at the Buck Institute and lead author on the paper, said the extended lifespan in the model organisms would be the equivalent to another dozen or so years of healthy living in humans. "Our preliminary data in the worms showed that ibuprofen also extended their healthspan," she said. "Healthy worms tend to thrash a lot and the treated worms thrashed much longer than would be normally expected. As they aged, they also swallowed food much faster than expected."

Honeybee hive sealant promotes hair growth in mice

Hair loss can be devastating for the millions of men and women who experience it. Now scientists are reporting that a substance from honeybee hives might contain clues for developing a potential new therapy. They found that the material, called propolis, encouraged hair growth in mice. The study appears in ACS' Journal of Agricultural and Food Chemistry.

Ken Kobayashi and colleagues note that propolis is a resin-like material that honeybees use to seal small gaps in their hives. Not only does it work as a physical barrier, but it also contains active compounds that fight fungal and bacterial invasions. People from ancient times had noticed propolis' special properties and used it to treat tumors, inflammation and wounds. More recently, research has shown that the substance promotes the growth of certain cells involved in hair growth though no one had yet tested whether that in turn would result in new locks. Kobayashi's team wanted to find out.

When the researchers tested propolis on mice that had been shaved or waxed, the mice that received the treatment regrew their fur faster than those that didn't. The scientists also noticed that after the topical application, the number of special cells involved in the process of growing hair increased. Although they tried the material on mice that could grow fur rather than balding mice, the researchers note that hair loss conditions often result from abnormal inflammation. Propolis contains anti-inflammatory compounds, so they expect it could help treat balding conditions.

They add that further testing is needed to see if the beehive material affects human hair follicles.

Abstract Propolis is a natural honeybee hive product with the potential for use in the treatment of dermatological conditions, such as cutaneous abrasions, burns, and acne. In this study, we investigated whether propolis stimulates hair growth in mice. Ethanol-extracted propolis, which contains various physiologically active substances such as caffeic acid and kaempferol, stimulated anagen induction in shaved back skin. Anagen induction occurred without any detectable abnormalities in the shape of the hair follicles (HFs), hair stem cells in the bulge, proliferating hair matrix keratinocytes in the hair bulb, or localization of versican in the dermal papilla. Propolis treatment also stimulated migration of hair matrix keratinocytes into the hair shaft in HFs during late anagen in the depilated back skin. Organotypic culture of skin containing anagen stage HFs revealed significant stimulation of hair matrix keratinocyte proliferation by propolis. Furthermore, propolis facilitated the proliferation of epidermal keratinocytes. These results indicate that propolis stimulates hair growth by inducing hair keratinocyte proliferation.

Can face-slapping give you younger looking skin?

Slapping your face to look younger? The concept garnered a lot of attention for one California massage therapist. The idea is to increase circulation and unblock energy. But is this trendy technique truly based in ancient practices?

Gam Pukkalanun is a certified Thai massage therapist in West Los Angeles. At her Thai Sabai salon, slapping is part of the routine.

"We use a chopping motion along the energy lines and to relax the muscles and open the energy lines," Pukkalanun said.

In Thai medicine, meridians or energy lines are called "sen." Like the rest of your body, the face also has specific ones. But can stimulating them make you look younger?

"The face slapping thing is, in my opinion, kind of a fad. I don't think it's a good treatment if anti-aging is what you're going to look for," said Dr. Shirley Chi, a dermatologist.

Chi says it's certainly not appropriate if you have irritable skin like rosacea patient Kristen Munoz.

"My skin is very, very sensitive. If I touch my skin, you'll see the mark on my face," Munoz said.

But gentle facial massage is something rooted in ancient tradition and common sense. Chi said when washing or massaging your face, use an upward motion.

"The concept behind the upward motion is that you're going against gravity and trying to get your skin to feel upward movement instead of downward movement," Chi said.

We are reactive organisms at a base level. Resistance training of sufficient intensity stimulates an increase in testosterone production, and the anabolic, muscle-building process ensues.

Always err on the side of training less. So, how do you know just how much is enough? Are you training too often or just the opposite? In both cases, there will be failed gains. Thats right overtraining can stall progress! The answer? Pay meticulous attention to your progress, or lack thereof.

Chart your progress. Set training goals, both short-term and long-term, and accomplish them. Buy a log book or download an app (there are many available for free) and make a habit of recording every workout. You dont know where youre going unless you know where youve been.

Nutrition is as important as training. You must provide your body with adequate nutrition to rebuild itself. If your fitness goal is muscular hypertrophy and strength, youll make few if any gains in the context of poor nutrition.
