Banned fountain of youth drug may be making a comeback

July 3, 2013 Despite it being more than 30 years since the "fountain of youth drug" Gerovital H3 was banned in the United States, it may be making a comeback. In an editorial published in this month's Journal of the American Geriatrics Society BUSM researcher Thomas Perls, MD, points out that a few U.S.-based anti-aging and longevity clinics have begun to advertise Gerovital H3 in pill form and as intravenous infusions despite the fact that the U.S. Food and Drug Administration (FDA) banned it in 1982.

Gerovital H3 is the dental anesthetic procaine hydrochloride (novocaine), yet in the 1950s, this drug was abused among Hollywood stars. According to Perls governmental bodies such as the FDA are relied upon to protect their citizens from drugs that do not do what they are claimed to do or are unsafe, yet in communist Romania, the opposite occurred with the state-sponsored marketing of Gerovital H3. In 1956, a paper titled "A new method for prophylaxis and treatment of aging with Novocain-eutrophic and rejuvenating effects" was published in the now discontinued journal Therapiewoche by Ana Aslan, director of the Geriatrics Institute of Bucharest. As a result the communist regime established an anti-aging resort and clinic for foreigners.

Perls explains that by the 1970s, the National Institute on Aging commissioned a thorough evaluation of the studies and claims surrounding Gerovital H3. "The conclusion of that work was that, except for a possible mild monoamine oxidase (MAO) inhibitor effect that would potentially ameliorate depression, there was no scientifically credible evidence supporting the claims that procaine hydrochloride is beneficial in treating age-related diseases or syndromes," said Perls, professor medicine at BUSM and director of the New England Centenarian and Supercentenarian Studies at Boston Medical Center. Perls points out that a plausible explanation for why some subjects might have experienced some improvements in health was that, in addition to receiving Gerovital H3, they were receiving other interventions such as exercise, stress reduction and healthy nutrition. "Thus, a glaring problem for the demonstration of any benefit associated with Gerovital H3 or similar compounds is the absence of any double-blind, placebo controlled trials demonstrating a significant improvement in the outcomes that anti-aging doctors and entrepreneurs claimed," he added.

Additional studies were later performed to further investigate a possible MAO inhibitory effect from procaine hydrochloride and any subsequent neurocognitive benefit but there was no evidence that procaine and its preparations could treat or preventing cognitive impairment or dementia.

"Gerovital H3 appears to have experienced a recent rebound in marketing and sales in the United States, primarily because of Internet-based marketing. A search for "Gerovital H3" on Google or Yahoo! results in more than 300,000 hits. Inquiring patients and the public need to be informed about the approved and unapproved uses of procaine hydrochloride and aware that there is no scientific evidence supporting any systemic health benefits or "anti-aging" effects of the drug," said Perls.

The Fellowship in Anti-Aging and Regenerative Medicine (FAARM) Module 1C: Advanced Metabolic Endocrinology was held last weekend. The workshop featured speakers Gary Huber, DO; Director of the Fellowship Pam W. Smith, MD, MPH; Elizabeth Stuller, MD; and David Zava, PhD.

Genomes of cholera bacteria from Haiti confirm epidemic originated from single source

The strain of cholera that has sickened thousands in Haiti came from a single source and was not repeatedly introduced to the island over the past three years as some have thought, according to a new study published in mBio, the online open-access journal of the American Society for Microbiology.

The results of this latest study are consistent with earlier findings that indicate Vibrio cholerae bacteria were introduced to Haiti by United nations soldiers between July and October 2010, when Nepalese soldiers arrived to assist recovery efforts after the January 2010 earthquake in that country. The genome sequences of V. cholerae strains from Haiti reveal they have not gained any new genetic material since their introduction and that they have a limited ability to acquire genes from other organisms through a process called transformation.

This new information may help public health authorities understand future cholera outbreaks in Haiti and elsewhere, according to the authors. "The use of high resolution sequence data that is amenable to evolutionary analysis will greatly enhance our ability to discern transmission pathways of virulent clones such as the one implicated in this epidemic," write the authors.

The earthquake in January 2010 killed tens of thousands of Haitians, and it was followed several months later by an outbreak of cholera, a disease that had never before been documented in Haiti. Studies of the outbreak indicate that poor sanitation at a United Nations camp resulted in sewage contamination of local water supplies, and phylogenetic analysis of the Haiti V. cholerae strains and strains from around the globe indicate the strain was most likely accidentally brought to the camp by U.N. troops from Nepal.

Earlier "fingerprinting" of Haiti's V. cholerae isolates using pulse-field gel electrophoresis (PFGE) has shown the bacterium has changed somewhat since the epidemic began in October 2010, but because of the nature of PFGE, the significance of those changes was not known. Were the changes meaningful? Were the bacteria gaining or losing genes that could impact the course of disease? Did they gain genes from other bacteria in the environment? Are their genomes rearranged? The answers could make a difference in the severity of future outbreaks.

The authors of the study in mBio set out to study in greater detail how V. cholerae may have evolved since its introduction to the island nation, and whether it has acquired genes that bestow new abilities. They sequenced the genomes of 23 different V. cholerae isolates from Haiti that represent multiple PFGE "fingerprint" patterns and were taken from a variety of locations and at various time points during the epidemic.

When compared with the genome sequences of V. cholerae strains from around the world, the Haiti isolates and three Nepal isolates are tightly related, forming a monophyletic group to which no other genome sequences belong.

This result indicates that "Nepalese isolates are the closest relatives to the Haiti strain identified to date, even when placed into a phylogeny with a larger collection of isolates representing recent cholera epidemics," write the authors. This means that the outbreak originated from a single introduction of bacteria, and PFGE variants arose from gradual evolution of the organisms, not from any secondary introduction.

Now she can literally walk a mile.

Lawyer Charito Planas, 83, used to have a hard time getting up. But she can now walk without pain, thanks to stem cell therapy that cost her only five figures.

While hospitals here and abroad charge millions for the treatment, a wellness center in Tagaytay offers the service 10 times cheaper, Planas said.

The cane I used to walk with is just an accessory now, for I walk freely, without pain, without limping, said Planas, a political and human rights activist who sought refuge in the United States during the Marcos martial law administration.

My heavily grey hair has now regained its ebony shade; my skinboth body and facial skinhas regained a youthful, shining glow; and my wrinkles are slowly fading away, she added.

Planas underwent six sessions of stem cell therapy from March to April at Green and Young Health and Wellness Center, owned by Dr. Antonia Carandang Park.

VIP patients

Former President Gloria Macapagal-Arroyo reportedly visited the Filipino physician a day after she was released from detention at Veterans Memorial Medical Center (VMMC) in Quezon City after posting a P1-million bail in July last year. She was taken back to VMMC in October.

Other well-known personalities, such as justices, physicians and a senator, also reportedly frequented the wellness center.
