Reversing hearing loss with regenerative therapy | MIT News | Massachusetts Institute of Technology – MIT News

Most of us know someone affected by hearing loss, but we may not fully appreciate the hardships that lack of hearing can bring. Hearing loss can lead to isolation, frustration, and a debilitating ringing in the ears known as tinnitus. It is also closely correlated with dementia.

The biotechnology company Frequency Therapeutics is seeking to reverse hearing loss not with hearing aids or implants, but with a new kind of regenerative therapy. The company uses small molecules to program progenitor cells, a descendant of stem cells in the inner ear, to create the tiny hair cells that allow us to hear.

Hair cells die off when exposed to loud noises or drugs including certain chemotherapies and antibiotics. Frequencys drug candidate is designed to be injected into the ear to regenerate these cells within the cochlea. In clinical trials, the company has already improved peoples hearing as measured by tests of speech perception the ability to understand speech and recognize words.

Speech perception is the No. 1 goal for improving hearing and the No. 1 need we hear from patients, says Frequency co-founder and Chief Scientific Officer Chris Loose PhD 07.

In Frequencys first clinical study, the company saw statistically significant improvements in speech perception in some participants after a single injection, with some responses lasting nearly two years.

The company has dosed more than 200 patients to date and has seen clinically meaningful improvements in speech perception in three separate clinical studies. Another study failed to show improvements in hearing compared to the placebo group, but the company attributes that result to flaws in the design of the trial.

Now Frequency is recruiting for a 124-person trial from which preliminary results should be available early next year.

The companys founders, including Loose, MIT Institute Professor Robert Langer, CEO David Lucchino MBA 06, Senior Vice President Will McLean PhD 14, and Harvard-MIT Health Sciences and Technology affiliate faculty member Jeff Karp, are already gratified to have been able to help people improve their hearing through the trials. They also believe theyre making important contributions toward solving a problem that impacts more than 40 million people in the U.S. and hundreds of millions more around the world.

Hearing is such an important sense; it connects people to their community and cultivates a sense of identity, says Karp, who is also a professor of anesthesia at Brigham and Womens Hospital. I think the potential to restore hearing will have enormous impact on society.

From the lab to patients

In 2005, Lucchino was an MBA student in the MIT Sloan School of Management and Loose was a PhD candidate in chemical engineering at MIT. Langer introduced the two aspiring entrepreneurs, and they started working on what would become Semprus BioSciences, a medical device company that won the MIT $100K Entrepreneurship Competition and later sold at a deal valued at up to $80 million.

MIT has such a wonderful environment of people interested in new ventures that come from different backgrounds, so were able to assemble teams of people with diverse skills quickly, Loose says.

Eight years after playing matchmaker for Lucchino and Loose, Langer began working with Karp to study the lining of the human gut, which regenerates itself almost every day.

With MIT postdoc Xiaolei Yin, who is now a scientific advisor to Frequency, the researchers discovered that the same molecules that control the guts stem cells are also used by a close descendant of stem cells called progenitor cells. Like stem cells, progenitor cells can turn into more specialized cells in the body.

Every time we make an advance, we take a step back and ask how this could be even bigger, Karp says. Its easy to be incremental, but how do we take what we learned and make a massive difference?

Progenitor cells reside in the inner ear and generate hair cells when humans are in utero, but they become dormant before birth and never again turn into more specialized cells such as the hair cells of the cochlea. Humans are born with about 15,000 hair cells in each cochlea. Such cells die over time and never regenerate.

In 2012, the research team was able to use small molecules to turn progenitor cells into thousands of hair cells in the lab. Karp says no one had ever produced such a large number of hair cells before. He still remembers looking at the results while visiting his family, including his father, who wears a hearing aid.

I looked at them and said, I think we have a breakthrough, Karp says. Thats the first and only time Ive used that phrase.

The advance was enough for Langer to play matchmaker again and bring Loose and Lucchino into the fold to start Frequency Therapeutics.

The founders believe their approach injecting small molecules into the inner ear to turn progenitor cells into more specialized cells offers advantages over gene therapies, which may rely on extracting a patients cells, programming them in a lab, and then delivering them to the right area.

Tissues throughout your body contain progenitor cells, so we see a huge range of applications, Loose says. We believe this is the future of regenerative medicine.

Advancing regenerative medicine

Frequencys founders have been thrilled to watch their lab work mature into an impactful drug candidate in clinical trials.

Some of these people [in the trials] couldnt hear for 30 years, and for the first time they said they could go into a crowded restaurant and hear what their children were saying, Langer says. Its so meaningful to them. Obviously more needs to be done, but just the fact that you can help a small group of people is really impressive to me.

Karp believes Frequencys work will advance researchers ability to manipulate progenitor cells and lead to new treatments down the line.

I wouldn't be surprised if in 10 or 15 years, because of the resources being put into this space and the incredible science being done, we can get to the point where [reversing hearing loss] would be similar to Lasik surgery, where you're in and out in an hour or two and you can completely restore your vision, Karp says. I think we'll see the same thing for hearing loss.

The company is also developing a drug for multiple sclerosis (MS), a disease in which the immune system attacks the myelin in the brain and central nervous system. Progenitor cells already turn into the myelin-producing cells in the brain, but not fast enough to keep up with losses sustained by MS patients. Most MS therapies focus on suppressing the immune system rather than generating myelin.

Early versions of that drug candidate have shown dramatic increases in myelin in mouse studies. The company expects to file an investigational new drug application for MS with the FDA next year.

When we were conceiving of this project, we meant for it to be a platform that could be broadly applicable to multiple tissues. Now were moving into the remyelination work, and to me its the tip of the iceberg in terms of what can be done by taking small molecules and controlling local biology, Karp says.

For now, Karp is already thrilled with Frequencys progress, which hit home the last time he was in Frequencys office and met a speaker who shared her experience with hearing loss.

You always hope your work will have an impact, but it can take a long time for that to happen, Karp says. Its been an incredible experience working with the team to bring this forward. There are already people in the trials whose hearing has been dramatically improved and their lives have been changed. That impacts interactions with family and friends. Its wonderful to be a part of.

Scavenging for resources, keeping yourself fed and hydrated, crafting tools and building a base, and staying alive in a harsh environment is what survival games are all about. But what about altering that environment until it's not as harsh as you found it? What if you could turn a hostile desert planet into a green and leafy paradise through terraforming?

The Planet Crafter, now in Steam Early Access, drops you on a barren, arid world that looks a lot like Mars, and your job is not to merely survive but ultimately to make the planet habitable for other humans. As you explore and collect minerals and other resources, and keep yourself fed and oxygenated, you'll craft tools and gear, build a base, and fabricate machines and modules that can begin warming up the planet. Eventually you'll be able to create a breathable atmosphere and bring life to the world in the form of trees, plants, lakes, and even wildlife. You may start off growing crops in hydroponic tubes inside your base, but the end goal is to be able to farm them outside under a bright blue sky.

The Planet Crafter has an appealing Subnautica vibe to it, and there's a nice chunky sci-fi look to the base modules and other machines you build. And unlike many survival games where you spend a lot of time hunting animals for food or defending against attacks, The Planet Crafter is a non-violent game. There's plenty of challenges when it comes to surviving the elements and progressing through the higher stages of terraforming technology, but you won't have to kill anything to do it. Nice.

The sci-fi survival game will spend between one and two years in Early Access, according to the two-person dev team at Miju Games, though that may change based on player feedback, which so far is "Overwhelmingly Positive" on Steam. There are plans to add small life forms, more environmental threats, a vehicle, and more story elements during its development.

All The Ways Living On Mars Could Kill You – 2oceansvibe News

The possibility of life on Mars has really stretched peoples imaginations and skillsets as they try and figure out how to build a home on the inhospitable planet.

One does not simply pitch a tent and get on with life as we know it.

Nope, not with Mars gruelling temperatures, carbon dioxide-plagued atmosphere, radiation, and intensely low atmospheric pressure.

But still, engineers, scientists, astronomers, and generally aspiring Martians have been racking their brains for the past couple of years, thinking outside the box in order to take the human species beyond Earth.

Before we get the big ideas,CNEToutlined all the possible ways that we can die if we try to make life on Mars a thing right off the bat:

If you were teleported to Mars with just basic camping gear, youd eventually die of radiation poisoning or cancer. But youd freeze to death long before then, most likely on the first night when temperatures dip to Antarctic levels.

Before that, youd suffocate trying to breathe the atmosphere made up of mostly carbon dioxide. Butbefore even that, the very low atmospheric pressure on Mars would cause your blood to literally boil, regardless of the outside temperature.

At this rate, it would be safer to just stay on the spacecraft once it lands on Mars.

This is actually an idea that a substantial amount of people are leaning towards, initially at least.

Elon Musk and space envision astronauts have thought of living out of SpaceX Starships at the start (see the image above) while planning and constructing a more permanent human settlement on Mars:

[Starships] are very valuable on the surface of Mars, said Paul Wooster, the companys principal Mars development engineer, in 2018 at a Mars Society convention.

Youd actually be having most of the ships stay and youd be operating using the various systems on them to support the activities there.

Next on the agenda would be terraforming, which is basically changing the planets environment to be more Earth-like and expanding the habitable bubble across the entire planet.

But first, one just needs to make it through the first night on Mars.

Any Martian home needs to consider shielding us from the intense radiation:

Former NASA physician Jim Logan estimatesputting our fragile, fleshy bodies behind or beneath about 9 feet (2.7 meters) of Martian soil should suffice.

Zubrin has also suggested using thick bricks made from Martian regolith to construct shelter, adding a uniquely medieval castle vibe to the more traditionally sleek and futuristic vision of a Mars outpost.

In the case of dust and solar storm sweeping across the planet, the old lava tubes and underground caves can make for good enough homes.

Otherwise, 3D printing a shelter is also something to consider:

NASA held a 3D printed habitat challenge in 2019, with New Yorks AI SpaceFactory (which bills itself as a multi-planetary architectural and technology design agency) winning the top prize for a system that built a lightweight but strong structure using autonomous robots requiring almost no human guidance.

Going underground or within hardcore structures wont allow agriculture to flourish, a practice that will be essential to survival on the Red Planet.

Thats because plants need light:

Mechanical engineerAndrew Geiszler suggested at the 2015 Mars Society conventionthat geodesic glass domes could be the answer. Mars provides all the raw materials needed to create glass, plastic and metals that can then be turned into dome homes.

Ultimately were going to need to use native materials. Its very feasible. Theyre there for the taking.

Building a permanent base will require that we can harvest resources and materials from the rugged landscape, which is a whole other story:

Very little that pertains to living on Mars in the early years will involve off-the-shelf equipment and supplies from Earth, writes Stephen Petranek in his bookHow Well Live on Mars. Almost every tool or device in use on Mars will need to have been carefully thought out.

Finding water will be crucial, which will provide the ability to make oxygen, grow food, and produce fuel and other raw materials.

Apparently, some water molecules have been found in Martian soil, in trace amounts in the air, and present in significant amounts near and below ice deposits, but it is a matter of extracting enough of that to last a human lifetime or more.

Weighing up the factors, possible impacts, effects, situations, problems, challenges, et cetera, of bringing human life to Mars is staggering and overwhelming.

I mean, astronauts have even been found to lose about 1% of the mass of some bones per month spent in space.

They found a solution for that in a human parathyroid hormone (PTH) peptide-rich space lettuce, but still, it just goes to show how much there is to think about before packing up for interplanetary travel and living.

Not to mention how the chief aspiring Martian (Musk) seems more preoccupied with posting memes.

Our curriculum is the basis for a city-wide STEM competition in San Antonio, Texas, but you can easily use this guide for your own classroom or program showcase. It was just over 30 years ago mankind discovered its first one.

When we were in kindergarten two things happened: We became best friends and we fell in love with Mars. From that point on we knew that we wanted to become scientists and make contributions to change the world. We began by pretending that we were scientists and started inventing things. Our love for science, STEMand anything related to outer space stayed with us all the way into middle school.

Thats when our guidance counselor, Dr. John MacDonald, introduced us to the American Student Assistance (ASA) Solve Together Career Exploration Competition. Its a national challenge that has kids work together with an adult to solve a real world problem that might lead to a future career. We signed up right away and decided that our project would be to act as a team of botanists to create a livable colony on Mars. We called ourselvesthe Galactic Girls, and our team of five went on to win first place and $10,000 for our school.

In the beginning, our project to make hydroponic farms went well, but we hit a wall trying to figure out what type of power source to use. Dr. MacDonald knew that we were onto something big and reached out to someone with a connection to NASA.

The next thing we knew, we were meeting (virtually) with a former NASA scientist and current KBR senior vice president of science and space, Mr. Todd May. He taught us a lot about the power source that would help grow crops on Mars, and he also taught us about pushing hard to reach our goals.

Here are some lessons we learned from Dr. MacDonald and Mr. May that we hope will inspire other future scientists, especially girls, in middle school and beyond:

Do what you love. Our love for all things outer space began when we were little, and its something that we have been passionate about ever since. For years, we haddreamed about becoming real life scientists so when the opportunity came, we grabbed it. We got to actually do the scientific work that we plan to do in high school, in collegeand afterward.

Be different. One of the ASA Solve Togethers requirements is that you form a team with up to five students. We knew that two of us were all in and that we needed to carefully pick other team members to ensure our success. We ended up inviting three girls who we knew shared a passion for outer space and had the curiosity, energyand focus to make it happen. We all had different cultural backgrounds and perspectives, which made our overall group strong in at least five ways.

Make every mentor count. We were lucky because we had seven mentors for this project: Dr. MacDonald, Mr. Mayand the five of us.We learned from ASA that a mentor isnt just someone you go to for help, or whom you wait to find when you are older. Mentors can be anywhere, though sometimes you have to go out of your way to find one even to outer space.Since the competition, lots of kids have turned to us for advice, so weve already become mentors ourselves.

Learn from people and with people. We started this project with lots of information and then worked hard to get the parts that were missing. When we talked to Mr. May, for instance, we did a lot of preparation so we could learn what we really needed to know. We made sure that we listened hard, recording our session with him and playing it over and over to make sure we got the science right. We also learned from each other every single day.

Mr. May said a lot of things to us, but the one we will always remember is that he couldnt wait to see us walk on Mars. He didnt say it like it was a maybe. He said it because he knew that if we wanted to, and if we worked hard enough, we would eventually get there.
