DNA confirms Greece girl's parents

Posted: October 28, 2013 at 10:45 am

DNA tests have confirmed a Bulgarian Roma couple as the biological parents of Maria, a mystery blond girl whose discovery in a Greek Roma camp last week made global headlines.

'DNA samples showed that Sasha Ruseva is the biological mother and Atanas Rusev is the biological father of the child called Maria,' Bulgaria's interior ministry chief of staff Svetlozar Lazarov told reporters.

The girl, aged somewhere between four and six, was found living with a Greek Roma couple in a camp near the town of Farsala.

DNA tests showed that the 39-year-old man and 40-year-old woman taking care of her were not her real parents.

The couple were accused of abducting the girl, amid an international effort to find here real parents that gave hopes to hundreds of missing children parents across the world.

The two denied the accusations, saying that the little girl was voluntarily handed over to them by her Bulgarian Roma mother who could not care for her.

Following reports in the Greek media pointing to the identity of the mother, Bulgarian police tracked down on Thursday a Bulgarian Roma couple from the central town of Nikolaevo, and questioned them on the Maria case.

The woman, Sasha Ruseva, 35, told police she had given birth to a baby girl while working in Greece several years ago and left it there when it was seven months old, the interior ministry said.

Ruseva has said she was not completely certain that Maria was her child but that she resembled some of her other nine children - also blond, fair-skinned and light-eyed.

The woman said she was ready to take Maria back if DNA tests confirmed that she was her real mother.

Traces of DNA exposed by twisted light

ANN ARBORStructures that put a spin on light reveal tiny amounts of DNA with 50 times better sensitivity than the best current methods, a collaboration between the University of Michigan and Jiangnan University in China has shown.

Highly sensitive detection of DNA can help with diagnosing patients, solving crimes and identifying the origins of biological contaminants such as a pathogen in a water supply.

"It really does not matter where the target DNA is from," said Nicholas Kotov, the Joseph B. and Florence V. Cejka Professor of Chemical Engineering at U-M. "In order to detect a specific DNA, we just need to know a small portion of its sequence."

Current DNA analysis methods rely on copying segments of a strand of DNA. The process unzips the double helix and then short, lab-made 'primer' DNA strands attach to each half of the original DNA. These primers kick-start the copying process, using the unzipped DNA as a template. Targeted segments of DNA can be replicated in this way, doubling every cycle. If enough DNA is produced before copying errors become a major problem, then further analysis can show whether the sample matches a suspect, for example.

But if the primers were very selective for the suspected DNA sequence, then a match could be determined by simply detecting whether the DNA had copied or not. Studies revealed that small amounts of DNA could be observed when spherical gold nanoparticles were attached to the primers. If the DNA matched suspicions, strings of particles bound together with DNA would form in the replication process. The nanoparticle solution would change color from red to blue, due to the way the strings of particles interact with light.

"Impressive detection limits were attained for short DNAs with nanoparticles; however, not for long DNA," Kotov said.

The problem, he explained, is that if the particles are further apart than a few nanometers, or millionths of a millimeter, "they do not interact strongly and the blue color does not happen." Longer strands are needed to differentiate between species and individuals with greater accuracy.

Kiwifruit Became More Nutritious Following Genome Duplication Events

redOrbit Staff & Wire Reports Your Universe Online

Scientists working to sequence the genome of the kiwifruit have revealed that the berry has recently undergone a pair of whole-genome duplication events, according to new research published earlier this month in the journal Nature Communications.

Zhangjun Fei, an associate professor at the Boyce Thompson Institute at Cornell University, and his colleagues also discovered that there were many similarities between the kiwis more than 39,000 genes and those of other plant species, including tomatoes and potatoes.

However, they were surprised to learn of the duplication events. When genes are duplicated, they explain, the extra genes can wind up mutating in order to perform new functions previously lacking in the organism. This process, which is known as neofunctionalization, turned out to be beneficial to the kiwi, according to Fei.

The duplication contributed to adding additional members of gene families that are involved in regulating important kiwifruit characteristics, such as fruit vitamin C, flavonoid and carotenoid metabolism, he said in a statement Friday. Previous research had extensively analyzed the metabolic accumulation of those substances, but this marks the first time that genome sequence data had been available.

Fei was one of several US and Chinese researchers who participated in the project, which used a Chinese variety of kiwifruit known as Hongyang in order to produce the draft sequence. After comparing its genome to those of grape, rice, tomato, and a type of mustard weed called Arabidopsis, they found 8,000 genes that were common among all five species, including some related to fruit growth and ripening, nutrient metabolism, and disease resistance.

One of the most remarkable findings of the study was uncovered when scientists observed a high percentage of similarities within the kiwifruit DNA, the university noted. The data revealed two unusual mishaps that occurred in the process of cell division about 27 and 80 million years ago, when an extensive expansion of genes arose from an entire extra copy of the genome, followed by extensive gene loss.

Fei, who called the kiwi an economically and nutritionally important fruit crop, said that the genome sequence he and his colleagues have created will serve as a valuable resource for kiwifruit research and may facilitate the breeding program for improved fruit quality and disease resistance.

The fruit, which has fuzzy brown skin and green flesh and a taste similar to that of a strawberry, originated from the mountains of southwestern China and did not become known throughout the world until the 20th century. According to the researchers, it is a type of berry that, like grapes, grows on woody vines. The kiwifruit is a member of the order Ericales, a classification it shares with blueberries, tea bushes and Brazil nuts.

Campaign launched on World Psoriasis Day

Posted: at 10:43 am

Dubai: Psoriasis sufferers will be able to better understand the chronic skin condition during the unveiling of a campaign titled Break away from Psoriasis on Tuesday.

The campaign by the dermatology department at the Dubai Health Authority (DHA), in association with AbbVie Pharmaceuticals, is timed with World Psoriasis Day, a global observance every October 29.

The observance gives an international voice to the 125 million psoriasis patients around the world by increasing understanding, improving access to treatments and building unity among the psoriasis community.

According to figures from the DHAs Dermatology Centre, in 2012, out of 12,000 patients, 966 patients had psoriasis. Globally, psoriasis affects approximately two to three per cent of the population.

The National Psoriasis Foundation, an international advocacy organisation, states psoriasis is a non-contagious, chronic, inflammatory, painful, disfiguring and disabling disease for which there is no cure.

The organisations research links patients with psoriasis with depression, anxiety, fatigue, suicidal thoughts and sleep impairment, as well as self-conscious, embarrassed and helpless feelings. It also links the condition to social discrimination and humiliation, and poor sexual activity.

UAE-based dermatology specialists said people who have psoriasis, a chronic skin condition marked by raised, red bumps covered with white, flaking scales, may feel stigmatised socially and emotionally.

Speaking to Gulf News, Dr Anwar Al Hammadi, Consultant and Head of Dermatology at the DHA and Mediclinic City Hospital and speaker for the campaign, said that because the condition has no cure, some patients are unable to cope, leading to depression, anxiety and rejection.

Referring to psoriasis as a cycle of despair, he explained that the condition can affect ones social, personal and occupational life adversely.

The campaign aims to educate the community that the disease in not contagious yet it needs community understanding. Schools write to us asking whether a child with psoriasis can swim with other children. [Yes, as it is not contagious.] Further psoriasis is associated with co-morbidities like hypertension, obesity and high cholesterol, he said.
