ABSTRACT

Bitter taste perception is critical to prevent animals from ingesting potentially harmful substances. The aim of this study was to characterize the evolution of T2R4 and test the hypothesis that different regions of the T2R gene are subject to disparate selective pressures, with extracellular regions (ECs) being erratic while transmembrane (TMs) and intracellular regions (ICs) being constrained. Thus, we examined the selective pressures acting on T2R4 and its different regions in 37 primates, and discovered that T2R4 and ECs were subject to neutral evolution and purifying selection, respectively, whereas both TMs and ICs showed purifying selection, as suggested by the hypothesis. We attribute this result to the relatively conservative property of T2R4 gene and the limited number of bitter tastants that T2R4 can respond to. Furthermore, we found that positive selection had acted on the first loop of extracellular regions (EL1). In contrast, the second loop (EL2) and transmembrane region-3, -6, -7 (TM367) were subject to purifying selection, and the third loop (EL3) was subject to neutral evolution. This discovery is probably because EL2, EL3, and TMs play a crucial role in the ligand-binding process, and EL1 is involved in the tastant recognition process. We further tested whether the of T2R4 differs among species with different diets and found that a specialized diet affected the evolution of T2R4. Feeding habits, fewer T2Rs, and a dietary shift may account for the results. This study can help to uncover the evolution of T2Rs during the primate evolutionary course.

Stanford depression treatment nearly 80% effective – The Stanford Daily

Stanford neuromodulation therapy (SNT), an experimental, accelerated version of magnetic pulse brain stimulation developed at Stanford, may provide a revolutionary treatment for severe depression, according to researchers at Stanfords School of Medicine.

The researchers optimism comes on the heels of an Oct. 29 study in which the treatment was found to have induced remission in nearly 80% of participants. The study tested the efficacy of SNT on patients with long-term, moderate to severe depression who were also treatment resistant, meaning that they had unsuccessfully tried several other conventional forms of treatment. For many of these patients, SNT was the first treatment that successfully alleviated their depression in a significant, lasting way, researchers said.

My brain has been rebooted, said one patient treated with SNT who asked to remain anonymous due to concerns regarding the stigmatization of mental health treatment. Its like this cloud of depression has been lifted from me.

SNT is an adaptation of an already existing, noninvasive form of brain stimulation called transcranial magnetic stimulation (TMS). TMS delivers magnetic pulses to specific locations within the brain, activating neural circuits that show decreased activity during depressive episodes. According to researchers, while TMS has been approved for use by the Food and Drug Administration (FDA) since 2008, its efficacy is limited, and it can take weeks for patients to show improvement.

SNT differs from TMS in several key ways. First, SNT relies on functional neuroimaging to map out the brain, allowing researchers to pinpoint where to deliver magnetic pulses. Second, SNT deploys an especially efficient stimulation pattern that relies on fewer pulses to change neural circuits. Third and most crucially SNT dramatically increases the rate at which patients receive brain stimulation. While TMS involves one brain stimulation session per day for 36 weekdays, SNT requires only 10 stimulation sessions per day over a five-day period.

There have been a lot of innovations in the TMS technology space, said Nolan Williams, the lead investigator in the Oct. 29 study. The question we tried to answer was, how do we take what we now understand and reengineer TMS? And thats what we did.

Williams, who is the Director of the Stanford Brain Stimulation Lab and an assistant professor of psychiatry and behavioral sciences, said that the research team plans to submit the data to the FDA, which has already deemed SNT a breakthrough therapy. Williams said that the timeline for the treatments deployment will depend on whether or not it receives FDA approval, although he is optimistic about its potential to treat a broad range of neurological disorders.

One application [of SNT] is for people who are in psychiatric emergencies, Williams said. Weve been doing this in obsessive compulsive disorder and borderline personality disorder. We think that this is a brain tool.

SNTs versatility makes it a much more compelling alternative to other treatments, said Kristin Raj, the co-chief of Stanford Mood Disorders and chief of the Stanford Bipolar Clinic. As one of the researchers in the study, Raj said that the studys results demonstrate that SNT is far more effective than medication, which often produces diminishing returns after the first few doses. Moreover, SNT has none of the side effects that come with other popular treatments for depression like electroconvulsive shock therapy or ketamine therapy, which are still viewed as controversial.

Ive had many patients tell me how much hope it gives them to hear about SNT, Raj said.

Still, despite overwhelmingly positive results, SNT is not a miracle cure.

Researchers have said that not all patients respond in the same way to the treatment while some patients are still in remission years after being treated, others have reported relapse after only a few weeks.

Most individuals fall somewhere in the middle, said psychiatry and behavioral sciences professor Brandon Bentzley, who offered one hypothesis as to why responses to SNT might vary.

What were doing in SNT is trying to move the brain from the default mode network, which is dominant during depression, to the central executive network, Bentzley said, another researcher in the study. My speculation is that different people have a different propensity to shift back to the default mode network.

The question of how to extend the positive effects of SNT to all patients, Bentzley said, remains an active research focus.

David Carreon, another researcher for the study and a clinical assistant professor of psychiatry and behavioral sciences, said that he has already adapted SNT to his private practice, albeit without the functional neuroimaging techniques used in the Oct. 29 study. While the treatment is not as effective as the full version of SNT, it is still showing success in patients he said.

While SNT might offer hope for depression treatment, the stigma that hangs over mental health issues remains dangerous, according to the same patient who requested anonymity. The patient, who is a physician, said that this stigma is especially prevalent within the healthcare industry, which takes an almost militaristic approach to denying the existence of mental health issues in its workforce.

You would think that the healthcare system, since its so good at treating others, would be better at providing treatment for physicians, the patient said. But its more like, Everyone else has problems, but we dont.

Carreon said that many of his patients, including those who have gone into remission, have refused interviews with media outlets looking to report on SNT due to this stigma.

People dont want to be on national TV as the guy who was depressed, Carreon said. Even if it was in the past.

The stigma surrounding mental health has real-world dangers; according to Enas Dakwar, a clinical psychologist at the Vaden Health Center, social stigma prevents many individuals from seeking support for mental health. This is exacerbated by the fact that many individuals with depression are high-functioning, Dakwar said, which can lead them to try to fix their depressive state on their own. Without proper support, these individuals often fail, taking them deeper into the rabbit hole of depression, Dakwar said.

CBN Stops Banks from Processing Form ‘A’ Hard Copies, NCX on TMS – THISDAY Newspapers

The Central Bank of Nigeria (CBN) has directed that banks are no longer allowed to process hard copies of Form A and Non commercial Export (NCX) Form on the Trade Monitoring System (TMS) with a N5,000 processing charge also taking effect.

The apex bank in two separate circulars sent to the authorised dealers, the Nigeria Custom Service (NCS), shipping lines, airlines National Museum and Monuments as well as the general public, said henceforth, the forms will only be processed in electronic form.

In the circulars signed by the Director, Trade and Exchange department, CBN, Mr. O.S Nnaji, the central bank stated that the electronic form A will replace the hard copy of Form A for invincible transactions such as Personal and business travel allowance (PTA/BTA) medicals, education, other remittances amongst other while the e-Form NCX shall replace the hard copy of Form NCX for non-commercial exports, with effect from November 30, 2021.

For those filling the e-Form A, the CBN said they are required to obtain o valid Bank Verification Number (BVN) from their banks, as the BVN is a prerequisite for customers to access the Trade System for e-Form A application;

According to the central bank, The e-Form A is web based and allows the General Public to initiate the Form from their offices/homes and submit same to the Authorized Dealer Bank. A charge of N5,000 as fee per declaration of e-Form A is applicable with effect from November 30, 2021 and henceforth.

There will be a direct debit of the processing banks current account for each declaration which should be recovered from the customer by the bank. However, the charge on the Customer for the e-Form A should be separated from other bank charges;

All hard copies of Forms A established on or before November 30, 2021, prior to the commencement of the e-Form A shall be utilized within 15 working days of the establishment of the Form. For avoidance of doubt, all established hard copies of Forms A for which disbursement had not been made within the transition period of 15 working days shall be deemed cancelled.

For those filling out the e-Form NCX, the apex bank said, Authorized Dealer Banks are to ensure that their customers obtain a valid Tax Identification Number (TIN) from Federal Inland Revenue Service (FIRS)/Joint Tax Board (JTB). The TIN is a prerequisite for customers to access the Trade System for e-Form NCX application.

The N5,000 charge is also applicable for the e-Form NCX and there will be a direct debit of the processing banks current account for each declaration which should be recovered from the customer by the bank. However, the charge on the customer for the e-Form NCX should be separated from other bank charges.

All hard copies of Forms NCX established on or before November 30, 2021 (prior to the commencement of the e-Form NCX) shall be utilized within 90 days of the establishment of the Form. For avoidance of doubt, all established hard copies of Forms NCX for which shipment has not taken place within the transition period of 90 days shall be deemed cancelled.

Faith Evans Exits The Masked Singer: Honored To Be Asked – The Blast

R&B singer Faith Evans was finally unmasked as the Skunk on The Masked Singer.

Fans already put the clues together weeks ago. With her strong vocals, the judges proclaimed that Skunk might have been one of the strongest performers in TMS history.

The Group A finals that aired on December 1, saw Skunk coming face to face against the Bull, whohas yet to be unmasked. He will go on to face the winner of next weeks Group B finals to compete for a chance at being the season six winner in the finale airing on Wednesday, December 15.

Although there were only two competitors in last nights episode, the show switched it up a little bit by adding a duet with a celebrity guest. Skunk sang with legendary talent Michael Bolton, while Bull performed with the season 3 runner-up, Turtle, better known as Jesse McCartney.

Celebrity judges Robin Thicke and Nicole Scherzinger recognized Evans signature vocals in her last performance before the Thanksgiving break; however, judges Jenny McCarthy Wahlberg and Ken Jeong went with a solid guess of Mary J. Blige.

However, they were proven wrong when host Nick Cannon helped remove the Skunk mask to reveal a breathless Evans!

In a post-unmasking interview with Variety, Evans described herself as elated when her manager finally got the call to be on TMS because she always thought that would be a fun show to be a part of.

I havent been performing at all in the last three or more years, she added. So it was exciting to be able to do a show that I already enjoy watching and its local in L.A. And I had enough lead time to map out plans. I was totally excited and honored to be asked.

Although some performers might turn up their nose at appearing in a skunk costume, Faith said that she loved it.

I loved the idea of it, she continued. I loved the idea of it. And once I got into it, it was cumbersome. It was hot. But it was still sexy. And the reason I chose the skunk was because not only was it just a cute little sexy skunk, but because Im biracial. I thought that the black and white thing, you know, but that was a little play on things.

Although various guesses included Janet Jackson, Toni Braxton, Yolanda Adams, Queen Latifa, and Jennifer Hudson, Thicke thought of Evans from the very beginning.

Evans said that she was not surprised to hear that Robin recognized her voice.

Weve worked together a couple of times before and aside from the record that we put out; we had a duet together ten years ago, she explained. But there was a time when I used to just go by his house. Hes kind of like family, so I assumed he would know.

In one of Evans last videos, she said that just as many people have been inspired by my person as by my music.

She described a time when she was at church with the late Whitney Houston and they encountered a young lady who was crying. At first, Evans thought that she was crying because she was encountering the great Whitney Houston.

And so the lady got up and she was saying, my song got her through so much. And she was literally crying, I know God is real because I never thought Id ever see you in my life! And then I start crying because Im thinking shes talking about Whitney. I was like, oh my God! It made me realize that my actions and the hardships Ive been through didnt just make me stronger, they helped other people too. And Ill take that with me forever.

The Masked Singer Group B semi-finals will air on Wednesday, December 8 on Fox at 8 PM EST.

This will be the first one youll get out of the other three methods. To more easily access this, head on over to the entrance of this route from Celestic Town. Follow the route to the narrow Bikeways. Youll face a Ninja Boy named Davey. Keep following the path and break the rocks with Rock Smash. TM 30 Shadow Ball will be right there.

Battle Park

After beating the Elite Four, you will have access to the islands detached from the rest of Sinnoh. Specifically, at the Battle Park, there are vendors from who you can buy all sorts of different endgame items. One of them sells TMs as part of her wares, costing 64 BP per item. You can earn BP from battling through the Battle Tower, earning more points per successful streak.

Grand Underground NPCs

A much less reliable way to get Shadow Ball is to dig up tons of spheres and trade them over to random vendors who venture the Grand Underground. Oftentimes, these sellers might want to trade spheres for things like pedestals and some will trade TMs with you. If you happen to run into one with Shadow Ball, it wont come cheap!
