Like a range of other mitochondrial manipulations, altering levels of uncoupling proteins can extend life in laboratory animals, and here researchers suggest this works via <a href=\"http:\/\/en.wikipedia.org\/wiki\/Hormesis\">hormesis<\/a>, causing just enough damage to spur repair mechanisms to greater ongoing effects for a net overall gain:<\/p>\n<blockquote><p><i>Ectopic <a href=\"http:\/\/en.wikipedia.org\/wiki\/Gene_expression\">expression<\/a> of <a href=\"http:\/\/en.wikipedia.org\/wiki\/UCP1\">uncoupling protein 1 (UCP1)<\/a> in skeletal muscle (SM) mitochondria considerably increases lifespan in high fat diet fed UCP1 <a href=\"http:\/\/en.wikipedia.org\/wiki\/Transgenesis\">transgenic (TG)<\/a> mice in comparison to wildtype (WT).<\/p>\n<p>In order to clarify the underlying mechanisms we investigated substrate metabolism as well as oxidative stress damage and antioxidant defense in SM of low fat and high fat fed mice. TG mice [showed] elevated <a href=\"http:\/\/en.wikipedia.org\/wiki\/Lipid_peroxidation\">lipid peroxidative protein modifications<\/a> with no changes in <a href=\"http:\/\/en.wikipedia.org\/wiki\/Advanced_glycation_end-product\">glycoxidation<\/a> or direct protein oxidation. This was paralleled by an induction of <a href=\"http:\/\/en.wikipedia.org\/wiki\/Catalase\">catalase<\/a> and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Superoxide_dismutase\">superoxide dismutase<\/a> activity, an increased <a href=\"http:\/\/en.wikipedia.org\/wiki\/Redox_signaling\">redox signaling<\/a> (<a href=\"http:\/\/en.wikipedia.org\/wiki\/Mitogen-activated_protein_kinase\">MAPK<\/a> signaling pathway), and increased expression of stress protective <a href=\"http:\/\/en.wikipedia.org\/wiki\/Heat_shock_protein\">heat shock protein 25<\/a>. <\/p>\n<p>We conclude that increased skeletal muscle mitochondrial uncoupling <a href=\"http:\/\/en.wikipedia.org\/wiki\/In_vivo\">in vivo<\/a> does not reduce the <a href=\"http:\/\/en.wikipedia.org\/wiki\/Oxidative_stress\">oxidative stress<\/a> status in the muscle cell. Moreover it increases <a href=\"http:\/\/en.wikipedia.org\/wiki\/Lipid_metabolism\">lipid metabolism<\/a> and reactive lipid-derived <a href=\"http:\/\/en.wikipedia.org\/wiki\/Carbonyl\">carbonyls<\/a>. This stress induction in turn increases the endogenous antioxidant defense system and redox signaling.
