By examining midlife <a href=\"http:\/\/en.wikipedia.org\/wiki\/Hepatic\">hepatic<\/a> <a href=\"http:\/\/en.wikipedia.org\/wiki\/Gene_expression\">gene expressions<\/a> in mice under different dietary conditions, which resulted in different lifespans and aging-related <a href=\"http:\/\/en.wikipedia.org\/wiki\/Phenotype\">phenotypes<\/a>, we were able to identify genes and pathways that modulate the aging process. We found that pathways <a href=\"http:\/\/en.wikipedia.org\/wiki\/Transcription_(genetics)\">transcriptionally<\/a> correlated with diet-modulated lifespan and physiological changes were enriched for lifespan-modifying genes. Intriguingly, <a href=\"http:\/\/en.wikipedia.org\/wiki\/Mitochondrion\">mitochondrial<\/a> gene expression correlated with lifespan and anticorrelated with aging-related pathological changes, whereas <a href=\"http:\/\/en.wikipedia.org\/wiki\/Peroxisome\">peroxisomal<\/a> gene expression showed an opposite trend. Both <a href=\"http:\/\/en.wikipedia.org\/wiki\/Organelle\">organelles<\/a> produce <a href=\"http:\/\/en.wikipedia.org\/wiki\/Reactive_oxygen_species\">reactive oxygen species<\/a>, a <a href=\"http:\/\/www.fightaging.org\/archives\/2006\/10\/how-age-damaged-mitochondria-cause-your-cells-to-damage-you.php\">proposed causative factor of aging<\/a>. This finding implicates a contribution of peroxisome to aging. Consistent with this hypothesis, lowering the expression levels of peroxisome proliferation genes decreased the cellular peroxide levels and extended the lifespan of <a href=\"http:\/\/en.wikipedia.org\/wiki\/Drosophila_melanogaster\">Drosophila melanogaster<\/a> and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Caenorhabditis_elegans\">Caenorhabditis elegans<\/a>.
