This seminal work was followed by numerous studies reporting alternative (non-insertional) reprogramming methods, and various conditions to improve the efficiency of reprogramming. These studies have contributed little to an understanding of global mechanisms underlying reprogramming efficiency. Here we report that <a href=\"http:\/\/www.fightaging.org\/archives\/2009\/07\/latelife-administration-of-rapamycin-extends-life-in-mice.php\">inhibition of the mTOR (mammalian target of rapamycin) pathway by rapamycin or PP242<\/a> enhances the efficiency of reprogramming to <a href=\"http:\/\/en.wikipedia.org\/wiki\/Induced_pluripotent_stem_cell\">induced pluripotent stem cells (iPSCs)<\/a>. Inhibition of the <a href=\"http:\/\/www.fightaging.org\/archives\/2008\/10\/some-papers-on-igf1-and-insulin-in-aging.php\">insulin\/IGF-1 signaling pathway<\/a>, which like <a href=\"http:\/\/en.wikipedia.org\/wiki\/Mammalian_target_of_rapamycin\">mTOR<\/a> is involved in control of longevity, also enhances reprogramming efficiency. In addition the small molecules used to inhibit these pathways also significantly improved longevity in <a href=\"http:\/\/en.wikipedia.org\/wiki\/Drosophila_melanogaster\">Drosophila melanogaster<\/a>. We further tested the potential effects of six other longevity-promoting compounds on iPSC induction, including two <a href=\"http:\/\/en.wikipedia.org\/wiki\/Sirtuin\">sirtuin<\/a> activators (<a href=\"http:\/\/www.fightaging.org\/archives\/2010\/10\/resveratrol-doesnt-extend-life-limited-benefit-to-rapamycin.php\">resveratrol<\/a> and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Fisetin\">fisetin<\/a>), an <a href=\"http:\/\/www.fightaging.org\/archives\/2010\/02\/spermidine-and-another-vote-for-autophagy.php\">autophagy inducer (spermidine)<\/a>, a <a href=\"http:\/\/www.fightaging.org\/archives\/2010\/03\/pi3-kinase-and-the-prospect-of-reversing-memory-loss-in-alzheimers.php\">PI3K (phosphoinositide 3-kinase) inhibitor (LY294002)<\/a>, an <a href=\"http:\/\/www.fightaging.org\/archives\/2008\/09\/antioxidants.php\">antioxidant<\/a> (<a href=\"http:\/\/en.wikipedia.org\/wiki\/Curcumin\">curcumin<\/a>) and an <a href=\"http:\/\/www.fightaging.org\/archives\/2008\/09\/metformin-as-calorie-restriction-mimetic.php\">AMPK (activating adenosine monophosphate-activated protein kinase) activator (metformin)<\/a>. With the exception of metformin, all of these chemicals promoted somatic cell reprogramming, though to different extents.
