Employing <a href=\"http:\/\/en.wikipedia.org\/wiki\/Proton_NMR\">(1)H NMR spectroscopy<\/a>, we characterized the plasma metabolic phenotype (metabotype) of three long-lived murine models: 30% <a href=\"http:\/\/www.fightaging.org\/archives\/2001\/11\/calorie-restriction-explained.php\">dietary restricted (DR)<\/a>, <a href=\"http:\/\/www.fightaging.org\/archives\/2008\/10\/some-papers-on-igf1-and-insulin-in-aging.php\">insulin receptor substrate 1 null (Irs1(-\/-))<\/a>, and <a href=\"http:\/\/www.fightaging.org\/archives\/2011\/08\/on-mitochondrial-function-in-ames-dwarf-mice.php\">Ames dwarf (Prop1(df\/df))<\/a>. A panel of metabolic differences were generated for each model relative to their <a href=\"http:\/\/en.wikipedia.org\/wiki\/Scientific_control\">controls<\/a>, and subsequently, the three long-lived models were compared to one another. Concentrations of mobile <a href=\"http:\/\/en.wikipedia.org\/wiki\/Very_low-density_lipoprotein\">very low density lipoproteins<\/a>, <a href=\"http:\/\/en.wikipedia.org\/wiki\/Trimethylamine\">trimethylamine<\/a>, and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Choline\">choline<\/a> were significantly decreased in the plasma of all three models. Metabolites including glucose, choline, <a href=\"http:\/\/en.wikipedia.org\/wiki\/Glycerophosphocholine\">glycerophosphocholine<\/a>, and various <a href=\"http:\/\/en.wikipedia.org\/wiki\/Lip\">lipids<\/a> were significantly reduced, while <a href=\"http:\/\/en.wikipedia.org\/wiki\/Acetoacetate\">acetoacetate<\/a>, <a href=\"http:\/\/en.wikipedia.org\/wiki\/Hydroxybutyrate\">d-3-hydroxybutyrate<\/a> and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Trimethylamine_N-oxide\">trimethylamine-N-oxide<\/a> levels were increased in DR compared to <a href=\"http:\/\/en.wikipedia.org\/wiki\/Ad_libitum\">ad libitum<\/a> fed controls. Plasma lipids and glycerophosphocholine were also decreased in Irs1(-\/-) mice compared to controls, as were <a href=\"http:\/\/www.fightaging.org\/archives\/2009\/07\/too-much-methionine-appears-to-be-bad-for-mammals.php\">methionine<\/a> and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Citrate\">citrate<\/a>. In contrast, <a href=\"http:\/\/en.wikipedia.org\/wiki\/High_density_lipoprotein\">high density lipoproteins<\/a> and glycerophosphocholine were increased in Ames dwarf mice, as were methionine and citrate. Pairwise comparisons indicated that differences existed between the metabotypes of the different long-lived mice models. Irs1(-\/-) mice, for example, had elevated glucose, acetate, <a href=\"http:\/\/en.wikipedia.org\/wiki\/Acetone\">acetone<\/a>, and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Creatine\">creatine<\/a> but lower methionine relative to DR mice and Ames dwarfs. Our study identified several potential candidate biomarkers directionally altered across all three models that may be predictive of longevity but also identified differences in the metabolic signatures.
