{"id":1043086,"date":"2011-07-24T15:58:13","date_gmt":"2011-07-24T15:58:13","guid":{"rendered":"http:\/\/www.longevitymedicine.tv\/loco-and-fly-longevity\/"},"modified":"2024-08-17T17:02:01","modified_gmt":"2024-08-17T21:02:01","slug":"loco-and-fly-longevity","status":"publish","type":"post","link":"https:\/\/www.euvolution.com\/futurist-transhuman-news-blog\/longevity-medicine\/loco-and-fly-longevity.php","title":{"rendered":"Loco and Fly Longevity"},"content":{"rendered":"<p>Here is another piece in the exceedingly complicated puzzle of metabolism and longevity, touching on some other pieces that have shown up here before, <a href=\"http:\/\/www.fightaging.org\/archives\/2007\/11\/ac5-longevity-mutants.php\">such as adenylate cyclase<\/a>: \"Despite the various roles of <a href=\"http:\/\/en.wikipedia.org\/wiki\/Regulator_of_G_protein_signalling\">regulator of G protein signaling (RGS) protein<\/a> in the G protein signaling pathway that have been defined, the function of RGS has not been characterized in longevity signaling pathways. We found that reduced <a href=\"http:\/\/en.wikipedia.org\/wiki\/Gene_expression\">expression<\/a> of Loco, a Drosophila RGS protein, resulted in a longer lifespan of flies with stronger resistance to stress, higher <a href=\"http:\/\/en.wikipedia.org\/wiki\/Superoxide_dismutase\">MnSOD<\/a> activity and increased fat content. In contrast, overexpression of the loco gene shortened the fly lifespan significantly, lowered stress resistance and reduced fat content, also indicating that the RGS domain containing <a href=\"http:\/\/en.wikipedia.org\/wiki\/GTPase-activating_protein\">GTPase-activating protein (GAP)<\/a> activity is related to the regulation of longevity. Interestingly, expressional changes of yeast RGS2 and rat RGS14, <a href=\"http:\/\/en.wikipedia.org\/wiki\/Homology_(biology)\">homologs<\/a> to the fly Loco, also affected <a href=\"http:\/\/en.wikipedia.org\/wiki\/Oxidative_stress\">oxidative stress<\/a> resistance and longevity in the respective species. It is known that Loco [reduces] activity of adenylate cyclase (AC) and RGS14 interacts with activated <a href=\"http:\/\/en.wikipedia.org\/wiki\/HRAS\">H-Ras<\/a> and <a href=\"http:\/\/en.wikipedia.org\/wiki\/C-Raf\">Raf-1 kinases<\/a>, which subsequently inhibits <a href=\"http:\/\/en.wikipedia.org\/wiki\/MAPK\/ERK_pathway\">ERK phosphorylation<\/a>. We propose that Loco\/RGS14 protein may regulate stress resistance and longevity as an activator in AC-cAMP-PKA pathway and\/or as a molecular scaffold that sequesters active Ras and Raf from Ras*GTP-Raf-MEK-ERK signaling pathway.
