{"id":1042411,"date":"2011-04-10T15:53:04","date_gmt":"2011-04-10T15:53:04","guid":{"rendered":"http:\/\/www.longevitymedicine.tv\/mitochondrial-mechanisms-and-aging\/"},"modified":"2024-08-17T16:57:19","modified_gmt":"2024-08-17T20:57:19","slug":"mitochondrial-mechanisms-and-aging-2","status":"publish","type":"post","link":"https:\/\/www.euvolution.com\/futurist-transhuman-news-blog\/longevity-medicine\/mitochondrial-mechanisms-and-aging-2.php","title":{"rendered":"Mitochondrial Mechanisms and Aging"},"content":{"rendered":"<p>The evidence points toward <a href=\"http:\/\/en.wikipedia.org\/wiki\/Mitochondrion\">mitochondrial structure and function<\/a> being <a href=\"http:\/\/www.fightaging.org\/archives\/2006\/10\/how-age-damaged-mitochondria-cause-your-cells-to-damage-you.php\">very important in the progression of aging<\/a> within a species and <a href=\"http:\/\/www.fightaging.org\/archives\/2008\/04\/metabolic-rate-mitochondria-and-mammalian-lifespan.php\">differences in life span between species<\/a>. Here researchers review some of the mechanisms involved: \"Mitochondria are considered major regulators of longevity, although their exact role in aging is not fully understood. Data from different laboratories show a negative correlation between <a href=\"http:\/\/en.wikipedia.org\/wiki\/Free-radical_theory\">reactive oxygen species (ROS)<\/a> generated by <a href=\"http:\/\/en.wikipedia.org\/wiki\/Electron_transport_chain#Complex_I\">complex I<\/a> and lifespan. This suggests that complex I has a central role in the regulation of longevity. Here, we review data that both support and refute the role of complex I as a pacemaker of aging. We include data from our laboratory, where we have manipulated ROS production by the <a href=\"http:\/\/en.wikipedia.org\/wiki\/Electron_transport_chain\">electron transport chain (ETC)<\/a> in <a href=\"http:\/\/en.wikipedia.org\/wiki\/Drosophila_melanogaster\">Drosophila melanogaster<\/a>. The by-pass of complex I increases the lifespan of the fruit fly, but it is not clear if this is caused by a reduction in ROS or by a change in the <a href=\"http:\/\/en.wikipedia.org\/wiki\/Nicotinamide_adenine_dinucleotide\">NAD+ to NADH ratio<\/a>. We propose that complex I regulates aging through at least two mechanisms: (1) an ROS-dependent mechanism that <a href=\"http:\/\/www.fightaging.org\/archives\/2006\/10\/how-age-damaged-mitochondria-cause-your-cells-to-damage-you.php\">leads to mitochondrial DNA damage<\/a> and (2) an ROS-independent mechanism through the control of the NAD+ to NADH ratio. Control of the relative levels of NAD+ and NADH would allow the regulation of (1) glyco- and (2) lipoxidative-damage and (3) the activation of <a href=\"http:\/\/en.wikipedia.org\/wiki\/Sirtuin\">sirtuins<\/a>.\" Amongst other things, the NAD+ \/ NADH ratio determines <a href=\"http:\/\/www.fightaging.org\/archives\/2006\/10\/how-age-damaged-mitochondria-cause-your-cells-to-damage-you.php\">how much in the way of damaging free radicals a cell exports<\/a> into the surrounding environment.<\/p>\n<p><span>Link: <a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/21471732\">http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/21471732<\/a><\/span><\/p>\n","protected":false},"excerpt":{"rendered":"<p>The evidence points toward mitochondrial structure and function being very important in the progression of aging within a species and differences in life span between species.
