Mitochondrial function requires the coordinated <a href=\"http:\/\/en.wikipedia.org\/wiki\/Gene_expression\">expression<\/a> of hundreds of <a href=\"http:\/\/en.wikipedia.org\/wiki\/Nuclear_DNA\">nuclear genes<\/a> and a few dozen <a href=\"http:\/\/en.wikipedia.org\/wiki\/Mitochondrial_DNA\">mitochondrial genes<\/a>, many of which have been associated with either extended or shortened life span. Impaired mitochondrial function resulting from <a href=\"http:\/\/en.wikipedia.org\/wiki\/Mitochondrial_DNA\">mtDNA<\/a> and <a href=\"http:\/\/en.wikipedia.org\/wiki\/Nuclear_DNA\">nuclear DNA<\/a> variation is likely to contribute to an imbalance in cellular energy <a href=\"http:\/\/en.wikipedia.org\/wiki\/Homeostasis\">homeostasis<\/a>, increased vulnerability to <a href=\"http:\/\/en.wikipedia.org\/wiki\/Oxidative_stress\">oxidative stress<\/a>, and an increased rate of <a href=\"http:\/\/en.wikipedia.org\/wiki\/Senescence\">cellular senescence<\/a> and aging. The complex genetic architecture of mitochondria suggests that there may be an equally complex set of <a href=\"http:\/\/en.wikipedia.org\/wiki\/Epistasis\">gene interactions (epistases)<\/a> involving genetic variation in the nuclear and mitochondrial genomes.
